Synthesis and Antibacterial Activities of 4-Hydroxy-o-phenylphenol and 3,6-Diallyl-4-hydroxy-o-phenylphenol against a Cariogenic Bacterium Streptococcus mutans OMZ 176

  • For the purpose of survey of the antibacterial activity against a cariogenic bacterium Streptococcus mutans OMZ 176 with the introduction of hydroxyl and allyl groups to o-phenylphenol (Fig. 2, 1), 4-hydroxy-o-phenylphenol (2), and 3,6-diallyl-4-hydroxy-o-phenylphenol (4) were synthesized, sucessively. The synthesized compounds, 2 and 4 showed more potent antibacterial activity than the starting material, 1. The hydroxyl group was supposed to the essential element for the antibacterial activity and the introduction of allyl group to phenolic ring to be another element to increase the activity.

  • Naturally occurring diallyldihydroxybiphenyl compounds, magnolol and honokiol were reported to have potent antibacterial activities against most of Gram positive bacteria (1-4). To developmore potent antibacterial agents, some allyhydroxybiphenyl derivatives were synthesized from the starting compounds, p-phenyl-phenol (I) p-phenylphenol (IV), o, o-biphenol (VII) and p. p'-biphenol (XII). Among the newly synthesized compounds (III, VI, IX, XI, XIV and XVI), the antibacterial activities of 2-allyl-p-phenylphenol (VI), 6-ally-o, 0'-biphenol (IX), 2, 2'-diallyl-o, o'-biphenol (XIV) and 2, 2', 6-triallyl-p, p'-biphenol (XIV) were more potent than those of magnolol and bonokiol against a cariogenic bacterium, Stroptococcus matans ATCC OMZ 176.

  • Dendritic macromolecules of the first to fourth generation were synthesized, using alkenylation and hydrosilation cycles with allylmagnesium bromide and dichloromethylsilane as building blocks and siloxane tetramer $(Me(CH_2=CH)SiO)_4$, as core molecule. By the reaction of the dichloromethylsilyl-capped generation (G4P) with p-bromophenol, p-phenylphenol and lithium phenyethynylide, dendrimers with specific functions (G4P-BP (Mw: 16,300), G4P-PP (16,121), and CAP-PA (11,764)) have been produced. Analysis of new dendrimers by NMR, UV and MALD1 mass spectrometry suggests that they are pure and unified.

  • Tetrahomodioxa p-phenylcalix(4)arene dihexylether(C/sub66/H/sub68/O/sub6/) has been synthesized and structurally characterized by X-ray diffraction. Reaction of tetrahomodioxa p-phenylcalix(4)arene with hexyl halide and NaH in DMF leads to the dihexyl derivatives, 7,13,21,27-tetraphenyl-29,31-dihexyloxy- 2,3,16,17-tetrahomo-3,17-dioxacalit(4)arenes. The crystal is orthorhombic, P2₁2₁2₁, a= 9.764(2), b=16.167(2), c=32.994(3) Å, V=5208(1) Å, Z= 4, Dc = 1.221 gcm/sup -3/. The structure was solved by direct methods and refined by full-matrix least squares. Refinement converged at R = 0.070 for 2009 observed reflections. This molecule has a C-1,2-alternate conformation with pseudo-centrosymmetry and has two pairs of opposite phenyl rings, which are approximately parallel to each other. The benzene rings A and B are up, and the rings C and D rings are down with respect to the plane of the macrocyclic ring.
