Determination of 25 EDs in Frog and Fish Tissue by GC-MS (SIM)

  • A gas chromatography/mass spectrometric assay method was developed for the simultaneous determination of neutral and bacis twenty-five disruptors $(ED_S)$ in frog and fish. Afther homogenization and sonication of 5 g of sample, purification was achieves in one step with a solid phase extraction procedure using silica gelflorisl. Eluton was performed with 50mL of acetone : n-hexane (1 : 9) solution. The eluate was concentrated to approximately 10uL and dissolves with 100 uL of hexane and analyzed by GC-MS (SIM). The peaks had good chromatographic properties and the extraction of these compounds from sample also gave relatively high recoveries with small variatoins. Detection limits were 0.1 ng/g for 4-nitrotoluene, benzophenone, hexachlorobenzene, atrazine, malathion, o,p-DDT, o,p-DDT and permethrin, and 0.2 ng/g for heptachlor epoxide, γ-chlordane, α-chlordane, p,p'-DDE, p,p'-DDD, cypermethrin and fenvalerate, and 0.3 ng/g for trifluralin, metribuzin, alachlor, dieldrin and p,p'-DDT, and 0.5 ng/g for heptachlor, aldrin and parathion, and 0.7 ng/g for endrin, and 0.8 ng/g for nitrofen. The recoveries were between 33 and 109%. The method was used to analyze twenty-five frogs and forty-six fishes fishes samples caught from various regions in Korea. Benzophenone was detected at concentration of up to 17.2 ng/g in frog or fish. Heptachlor, aldrin, γ-chlordane, p,p'-DDE, p,p'-DDD, endrin and o,p-DDD were detected at concentrations of 0.7-12.5 ng/g in frog or fish. Also significant leveles of dieldrin (up to 22.5 ng/g) were observed. The developed method may be valuable to be used to the national monitoring project of EDS in biota samples.

Induction of Imposex in Rock Shell, Thais clavigera, Exposed to Organotins and Other Endocrine Disrupting Chemicals

  • Interactions of both feminizing and masculinizing chemicals inducing imposex in gastropod were investigated with a long-term exposure experiment. Imposex-free rock shell(Thais-clavigera)was exposed to TBT and other antagonistic chemicals (nonylphenol and DDT), P450 inducer (phenobarbital) and natural sex hormone (estrogen) mixtures for 60 days. None of imposexed-females were found in control, sham (ethanol) and testosterone exposure group, on the other hand imposex was induced in the TBT and all the TBT+antagonist mixture groups. After 60-day exposure, frequency of imposexed-female ranged from 31-68%. Female mean penis length (FMPL) of imposexed-female ranged from 0.87-2.58 mm, and relative penis length indices were from 7.2 to 21.2%. However, the degrees of imposex were different among the exposure groups. The FMPLS of three exposure groups (TBT+estrogen, + DDT and +nonylphe- not) were less than that of the TBT exposure group. Ethoxyresorufin-O-deethylase (EROD) activities in gastropod decreased in the TBT and all the TBT+antagonist ekposure groups compared to the control group. A negative correlation was abtained between TBT body residue and EROD activity, while a positive relationship was obtained between 737 body residue and the degree of imposex except for the TBT +DDT exposure group. Although 737 concentration was relatively high in the TBT+DDT exposure group, the TBT +DDT exposure group demonstrated low EROD activity and low degree of imposex. These results indicate that a certain antagonistic effect of DDT occurred in induction of imposex by TBT.

Estrogenic Potentials of Organochlorine Pesticides which Detected in Korean Adipose Tissue (한국인의 지방조직에서 검출되는 유기염소제 농약의 에스트로겐 활성)

  • 유기염소계 농약은 화학적으로 안정하고, 지용성이 크며 체내 대사에 대해 저항성을 가지고 있어서 인체 및 생태계의 생물체 중 축적성이 매우 크다. 또한 대부분 내분비계 장애물질로 분류되어 있어 관심의 대상이 되고 있다. 유기염소계 농약은 잔류성으로 인하여 사용이 금지되었음에도 불구하고 인체에서 검출되고 있으며 한국인의 지방조직을 분석한 결과 9종의 유기염소계 농약이 주로 축적되어 있음이 보고된 바 있다. 본 연구에서는 이들 유기염소계 농약의 에스트로겐 활성을 MCF-7 BUS cell을 이용한 E-screen assay competitive binding assay 및 pS$_2$ gene experession assay에 의해 조사하였다. o,p'-DDT,p, p'-DDT,p,p'-DDD,p,p'-DDE등 4종의 유기염소계 농약은 에스트로겐 수용체에 대한 ligand의존적인 작용기전에 의해 에스트로겐 활성을 가지며, $\alpha$-, $\beta$-, ${\gamma}$-, $\delta$-BHC, dieldrin등 5종의 유기염소계 농약은 lingand-비의존적 작용기전에 의해 에스트로겐 활성을 보였다. 또한 이들 유기염소계 농약을 혼합 투여하여 에스트로겐 활성을 관찰한 결과 DDT류의 경우에는 단독투여시 보다 그 대사체와 혼합 투여할 때 에스트로겐 활성에 상승적 효과가 나타났으며 o,p'-와 p,p'-DDT의 두 이성질체를 혼합 투여할 경우가 단독 투여시 보다 상승적 효과가 나타났다. 따라서 지방조직에서 검출되는 유기염소계농약은 상호작용에 의해 개별 물질이 나타내는 내분비계장애작용 보다 실제로는 강한 효과를 나타낼 것으로 추정되었다.

Resistance of Culex pipiens Mosquito Larvae to Insecticide in Seoul city (서울시내 빨간집모기(Culex pipiens) 유충의 살충제에 대한 저항성에 대하여)

  • The present status of insecticide resistance of Korean mosquitoes is still unknown. The experiment cited below is a part of our series of resistance tests on Korean mosquitoes. The test was carried out during the summer of 1959 to ascertain the degree of resistance of Culex pipiens to insecticides currently used. Culex pipiens eggs were collected from ditches in Seoul City and reared in laboratory with media consisted of the following : Distrilled water 1.0 liter, Magnesium sulfate (MgSO$_4$.7$H_2O$) 1.0gram , Calcium sulfate (CasO$_4$.2$H_2O$) 0.5gram ; Sodium cholride 0.5 gram. A Sufficient amount of beer yeast was also used as a larvar food. Late third and early fourth instar larvae were selected for testing. The results are summarized in the following. THe LC-50 of Culex pipiens to insecticides were 0.061 ppm in p, p-DDT , 0.045 ppm in ${\gamma}$-BHC and0.029 ppm in malathion. Comparing these with LC-50 doses of Orlando laboratory strain, it can be pointed out that the present resistance of Seoul strain shows about a ten-fold increase in DDt, and a fourfold increase in ${\gamma}$-BHC . The resistance development of the Seoul strain has, however, not arisen so markedly as compared with data observed in other parts of Far Eastern areas received heavy insecticidal operations.

A Survey for Pesticide Residues in Major Rivers of Korea (주요하천수(主要河川水)에 대(對)한 농약잔류(農藥殘留) 실태조사(實態調査))

  • To obtain basic information for the safe use of pesticides, the aquatic environment of 5 major rivers was surveyed in April and August 1982 for organochlorine and organophosphorus pesticide residues. Regardless of sampling times and sites, pesticide residue levels were very low. Of the organophosphorus pesticides surveyed, only IBP and diazinon were detected in water samples while fenthion, fenitrothion, chlorpyriphos, chlorpyriphos-methyl, chlorfenvinphos, phenthoate, and edifenphos were not detected in any water samples. Residue levels and detection frequencies of IBP or diazinon were much higher in water samples collected in August than those collected in April. Detection frequencies of organochlorine pesticides, on the other hand, were high, but their actual residue levels were very low. BHC was detected nearly in all water samples while aldrin was not found in any samples. Detection frequencies of heptachlor, endosulfan, and o.p`-DDT were considerably high but dieldrin and p.p`-DDT were detected in a few water samples and their residue levels were also very low.

The Risk Assessment of Residual Pesticides and Heavy Metals in Rehmanniae Radix Preparata (숙지황 중 잔류농약과 중금속의 안전성 평가)

  • To obtain the risk assessment of hazardous materials in Rehmanniae Radix Preparata, the residual pesticides and heavy metals in samples on the Korea and China are surveyed. Group I ($BHC-{\delta}$, $BHC-{\beta}$, Fenitrothion, Penthoate, Endosulfan-${\alpha}$, Dieldrin, Endosulfan-${\beta}$ and Endosulfan-sulfate), Group II (BHC-${\gamma}$, Aldrin, DDD, DDT-p,p Permethrin and Fenvalerate), Group III(BHC-${\alpha}$, Chlorpyrifos, Tolyfluanid, Captan and DDT-o,p) and Group IV(Quintozene, Vinclozolin, DDE and Chlorfenapyr) could analysed on gas chromatography-ECD for evaluation of residual pesticides. Qualified detection concentration on the GC-ECD are $0.45 ng/g{\sim}2.50 ng/g$. Group I, Group II, Group III and Group IV are not detected in Rehmanniae Radix Preparata on the Korea and China. Concentration of As, Cd and Pb in Rehmanniae Radix Preparata. on the Korea are 3.06%, 7.00% and 5.78% for Korea Food & Drug Administration(KFDA). Concentration of As, Cd and Pb in Rehmanniae Radix Preparata. on the China are 5.16%, 5.33% and 6.50% for Korea Food & Drug Administration(KFDA). The hazardous materials in Rehmanniae Radix Preparata on the Korea and China were verified the safety of the residual heavy metals and pesticides compare with Korea Food & Drug Administration (KFDA) advisory level.

Heavy Metals and Pesticides with Maturing Process of Rehmanniae Radix Preparata (숙지황 제조과정에서 중금속과 잔류농약)

  • To obtain the risk assessment of hazardous materials with maturing process of Rehmanniae Radix Preparata, the residual heavy metals and pesticides in samples are surveyed. Concentration of As, Cd and Pb during the maturing process of Rehmanniae Radix Preparata of Korea are $0.147{\pm}0.007\;mg/L,\;0.025{\pm}0.001\;mg/L\;and\;0.193{\pm}0.021\;mg/L$, respectively. Concentration of As, Cd and Pb during the maturing process of Rehmanniae Radix Preparat of China are $0.0145{\pm}0.014\;mg/L,\;0.023{\pm}0.002\;mg/L\;and\;0.299{\pm}0.013\;mg/L$, respectively. Concentration of As, Cd and Hg remain unchanged during the maturing process of Rehmanniae Radix Preparata. Concentration of As, Cd and Pb in Rehmanniae Radix Preparat on the Korea are 3.53%, 6.66% and 5.16% for Korea Food & Drug Administration (KFDA) advisory level, respectively. The residual pesticides, such as aldrin, $BHC-{\alpha},\;BHC-{\beta},\;BHC-{\gamma},\;BHC-{\delta}$, captan, chlorfenapyr, chlorpyrifos, DDD, DDE, DDT-o,p, DDT-p.p, dieldrin, endosulfan-sulfate, $endosulfan-{\alpha}$, $endosulfan-{\beta}$, fenitrothion, penthoate, permethrin, quintozene, tolyfluanid and vinclozolin, are not detected in Rehmanniae Radix Preparat on the Korea and China. The hazardous materials in Rehmanniae Radix Preparata on the Korea and China are verified the safety of the residual heavy metals and pesticides compare with Korea Food & Drug Administration (KFDA) advistory level.

Gas-particle Partitioning of Organochlorine Pesticides in Atmosphere (대기 중 유기염소계 살충제의 가스-입자 분배)

  • This study was performed to estimate the gas-particle partitioning of organochlorine pesticides (OCPs) in atmosphere, the samples were collected by PUF high volume air sampler for two years from June, 2000 to June, 2002. The gas phase fraction of ${\alpha/\gamma}-HCH$, heptachlor epoxide, ${\alpha/\gamma}-chlordane$ and trans-nonachlor was over 90%. But the gas phase fraction of ${\beta}-HCH$, p,p'-DDE, endosulfan sulfate, p,p'-DDD and p,p'-DDT was range of 20% through 80%, which means the gas phase fraction of OCPs components described above is sensitive to temperature. The correlation between the gas phase fraction and molecular weight of each OCPs component was not found in this research. The slope of regression line between gas-particle partitioning coefficient(${\log}K_p$) and subcooled liquid vapor(${\log}{P^o}_L$), gal-particle partitioning coefficient(${\log}K_p$) and octanol-air partitioning coefficient(${\log}K_{oa}$) which show -0.54 and 0.43 was not steep. So the equilibrium state between gas and particle was not reached and in this state the particulate fraction was low.
