The Effect of Far Infrared Ray-Vacuum Drying on the Quality Changes of Pimpinella bracycarpa (원적외선건조와 원적외선-진공건조를 이용한 참나물의 품질변화 특성)

  • This study was conducted to determine the effect of far infrared-vacuum drying on the quality changes of Pimpinella bracycarpa, such as drying efficiency (weight loss), color differences, browning degrees, rehydration and sensory evaluation. Wehn Pimpinella bracycarpa was dried for designated time at 5$0^{\circ}C$, 6$0^{\circ}C$ and 7$0^{\circ}C$, drying time of far infrared-vacuum drying was decreased more than 17% compared to that of infrared drying. The color changes increased as drying temperature increased and far infrared-vacuum drying made less color changes than infrared drying. Also, the total microbial counts and the number of yeast and mold were gradually reduced as drying temperature increased and drying time was longer, but there was no significant differences in microbial changes among drying methods. The rehydration rates of Pimpinella bracycarpa increased as drying temperature increased and was better at far infrared-vacuum drying than infrared drying. Also, according to the sensory evaluation after rehydration of Pimpinella bracycarpa, the hihger scores were obtained at lower temperature and far infrared-vacuum drying, especially the color was obtained much hihger score in the far infrared-vacuum drying than infrared drying. Thus, the results showed that drying efficiency and stability of rehydration and color changes was increased and the quality deterioration of Pimpinella bracycarpa could be minimized by using far infrared-vacuum drying.

Determination of Microbial Contamination in the Process of Rice Rolled in Dried Laver and Improvement of Shelf-life by Gamma Irradiation (김밥 제조공정에서의 미생물 오염도 평가 및 감마선 조사를 이용한 김밥의 보존안정성 향상)

  • Determination of regional microbial contamination in the process of rice rolled in dried laver (Kimbab) and effects of gamma irradiation on the improvement of hygienic quality and shelf stability were investigated. Total aerobic bacterial distribution of raw materials of Kimbab were; 10$^{6}$ ∼10$^{7}$ CFU/g in dried laver, 10$^3$ CFU/g in cucumber and below 10 CFU/g in steamed rice, ham, fried egg, and salted radish. Total coliform bacteria were 10$^3$ CFU/g in dried laver and detected below detection limit (10 CFU/g) in other raw materials. And it was arithmetically calculated that the levels of total aerobic bacteria and coliform bacteria in Kimbab does not exceed 10$^{5}$ CFU/g and 10$^1$ CFU/g under the aseptic process, respectively. However, microbial contamination levels in just prepared Kimbab in a market were about 10$^{6}$ CFU/g of total aerobic and coliform bacteria. Therefore, it was considered that microbial contamination of Kimbab is mainly originated from environmental uptake during the preparation. The representative media for putrefying bacterial growth were steamed rice. Coliform bacteria were mainly increased in ham and fried egg during storage. The bacteria in dried laver were radio-resistant and survived at 3 kGy of gamma irradiation. Coliform bacteria on EMB agar plate were eliminated at the dose of 2 kGy. The sensory acceptability of 2 kGy irradiated Kimbab was stable and the Kimbab can be preserved for 24 hour at 15$^{\circ}C$. Therefore, it was considered that optimal irradiation dose for radicidation of Kimbab was 2 kGy.

Studies on immobilization and application of beta-galactosidase I. Conditions for production and properties of the enzyme from Aspergillus niger CAD 1 (beta-Galactosidase의 고정화 및 응용에 관한 연구 제1보: Aspergillus niger CAD 1의 효소생산 조건 및 효소학적 성질)

  • A strain of Aspergillus niger CAD 1 which produces considerable amount of beta-galactosidase was selected from extracellular beta-galctaosidase producing fungi isolated from soil. Optimal conditions for the enzyme from Aspergillus niger CAD 1 were the growth in wheat bran supplemented with 0.5% skim milk powder at $30^{\circ}C$ for 72 hrs. The crude enzyme was purified 1,387 fold through DEAE-cellulosc and Sephadex G-100 chromatographr and its recovery was 6.2%, The optimal pH and temperature for the purified enzyme were pH 4.5 ana $45^{\circ}C$, respectively. The Km and Vmax on ONPG were $3.57{\times}10^3M$ and 33.0 unit/mg protein, whereas those on lacose were $83.3{\times}10^3M$and 15.33 unit/mg protein, respectively, The activation energy for the enzyme was 9,900 cal/mol and the enzyme had no metal ion requirement for its activity and stability. The hydrolysis of lactose in skim milk, 4.8% lactose solution and acidic whey were 65%, 70% and 78% after 10 hrs incubation at $45^{\circ}C$, when 182 units of the enzyme were used 50ml of the substrate solutions.

  • The beta-galactosidase of Aspergillus niger CAD 1 described in previous report was immobilized to cellulose triacetate. The optimal conditions for immobilization of the enzyme were obtained when 13%(v/v) of the soluble enzyme (349 unit/ml) was entrapped in fiber prepared from 10%(w/v) cellulose triacetate in methylene chloride. The optimal temperature and pH for the activity of the immobilized enzyme were $45^{\circ}C$ and 4.5, respectively, which stowed the same values as those of the soluble enzyme. It was found that Km of the immobilized enzyme for lacotse exhibited high value compared with that of the soluble enzyme. The immobilized enzyme showed good storage stability, reusability, and also hydrolysis of lactose when introduced into skim milk and acidic whey.

  • To evaluate the quality of commercial dehydrated tea-products, the relationships between particle sizes, densities, moisture absorption & desorption and color appearance were studied by using Hunter-lab tristimulus colorimeter and spectrophotometer. Among the tea-products was held no significant relation between particle sizes and color appearance but red ginseng extract powder (RGEP) was included L, a and b values when was reduced particle size. appearance color of tea-products indicated red-orange color, L, a and b values were ranged 32.7 to 48.0, 4.0 to 10.0 and 5.6 to 18.0, respectively, densities of tea-products ranged 0.232 to 0.898 g/ml and increased L values, Hunter's a/b ratio values was included in 0.61 to 0.90. Color stability in this products was well agreed with decrease of total color difference value ($\Delta$E) and chromaticity difference value ($\Delta$C) of the Hunter-lab color data.

In vitro Culture of Adventitious Roots from Dioscorea nipponica Makino for the Production of Steroidal Saponins (부채마 (Dioscorea nipponica Makino)의 부정근 배양조건과 Steroidal Saponin의 기내생산)

  • Effects of growth regulators on growth of adventitious roots and accumulation of steroidal saponins, such as dioscin, prosapogenin A and prosapogenin C, in cultures of Dioscorea nipponica were determined. The maximum growth of adventitious roots was observed in MS medium supplemented with 30 g/L sucrose and 1.0 mg/L NAA. Addition of BA in combination with NAA appeared to be no effective in the growth of adventitious roots. Among the twenty different adventitious roots formed from different seeds, strain No. 10 was selected based on production ability of dioscin, and its stability through the successive liquid culture. During the first 4 weeks of incubation, contents of steroidal saponins in adventitious roots were negligible but the contents were markedly increased at 5 weeks of incubation. Dioscin and prosapogenin C content in IBA-treated adventitious roots were significantly higher than those in NAA-treated roots. However, content of prosapogenin A was not significantly different among NAA or IBA level. Results provide that liquid culture of adventitious roots of D. nipponica have a potential for mass production of dioscin including prosapogenin A and prosapogenin C.

Comparison on Physicochemical Properties of Amaranth Starch with Other Waxy Cereal Starches (아마란스 전분과 곡류 찰전분의 특성 비교)

  • Physicochemical properties of amaranth starch were compared with those of waxy rice and waxy corn starches. Amaranth starch granules vary from $1.1{\sim}1.9\;{\mu}m$ in diameter and are polygonal in shape. Total amylose contents from waxy rice, waxy corn and amaranth starches were 0.01, 0.03 and 0.07%, respectively. Swelling power of amaranth starch granule was slightly different from waxy rice. The swelling power of amaranth increased at $70^{\circ}C$. X-ray diffraction patterns of amaranth and other waxy cereal starches showed an A-type crystalline structure. Relative crystallinities of their starches were similar. According to pasting properties by Rapid Visco-Analyzer, amaranth starch showed a very high gelatinization temperature $(75.1^{\circ}C)$ and lower viscosity and higher stability than other waxy cereal starches during heating and cooling cycle. Peak onset temperatures (To) of starches from waxy rice, waxy corn and amaranth in DSC thermograms were $58.7{\sim}64.0$, 67.2 and $71.5^{\circ}C$, respectively, and their peak enthalpies were similar. Enthalpy of reheated amaranth starch after 3 day storage at $4^{\circ}C$ was higher than those of waxy corn and rices starchs.

Purification and Characterization of Alkaline Protease from saewoo-jeot, salted and fermented shrimp (Acetes japonicus) (새우젓에서 alkaline pretease의 정제 및 특성)

  • This study was performed to elucidate the purification and characterization of pretease from saewoo-jeot, a Korean traditional salt-fermented shrimp product. The protease in saewoo-jeot (Acetes japonicus) were extracted, desalted through electrodialysis and purified by ammonium sulfate fractionation, Sephadex G-100 gel filtration and DEAE-cellulose column chromatography. Purified enzyme had specific activity of 8.4 unit/mg, yield of 14% and purification fold of 9.8. Purified enzyme was confirmed as single band protein by polyacrylamide gel electrophresis and the molecular weight was estimated to be about 24 kDa. The optimal pH and temperature for the enzyme activity were 8.0 and $40^{\circ}C$, respectively. The range of its stability to the pH and temperature were 7.0 to 10.0 and $30^{\circ}C\;to\;60^{\circ}C$, respectively. The activity of enzyme to synthetic substrate showed BAPNA and TAME. The enzyme was activated significantly by manganese ions, while inhibited by STI, TLCK. metals $(K^+,\;Li^+,\;Na^+,\;Ca^{++},\;Co^{++},\;Cu^{++},\;Mg^{++},\;Ba^{++},\;Hg^{++},\;Zn^{++},\;Fe^{+++})$. The Km value of the enzyme was $5.1{\times}10^{-7}\;M$ to hammersten casein. It's suggested that purified protease from saewoo-jeot seemed to be trypsin-like enzyme.

Changes of Functional Properties of Garlic Extracts during Storage (마늘추출액의 저장 중 기능성성분 변화)

  • An invistigation was carried out to study the effects of heat treatment of garlic and storage temperature of garlic extracts on functional properties of garlic extracts. The garlic was heated at 40, 60, 80, 100, $120^{\circ}C$ for 10 minutes and extracted with 50% ethanol. The extracts were stored at 4, 25 and $50^{\circ}C$ for 30 days, and then electron donating ability(EDA), nitrite-scavenging effects(NSE) and total thiosulfinates contents were determined. Among the functional properties, total thiosulfinates were rapidly reduced as the heating and storage temperature increased. The total thiosulfinates in the extracts were not detected for those heat-treated at 100 and $120^{\circ}C$. The EDAs were also decreased to almost half of their initial level by heat treatment of garlic at higher temperature than $80^{\circ}C$ while NSE was decreased a little. As the storage temperature increased, EDA and thiosulfinate contents decreased. Particularly the thiosulfinates were ditected none after 11 days of storage at $25^{\circ}C$ and $50^{\circ}C$. Therefore, mild heat treatment of garlic at lower temperature than $60^{\circ}C$ and storage at refrigerated condition are recommendable for preparation and storage of garlic extracts.

Effects of Roasting Condition on the Quality Characteristics and Oxidative Stabilities of Rice Germ (볶음 조건이 쌀눈의 품질 특성 및 산화안정성에 미치는 영향)

  • In order to compare quality characteristics and oxidative stabilities of rice germs prepared under different roasting conditions, sensory evaluation, color value, tocols (tocopherol+tocotrienol) contents, and peroxide value were investigated. Optimum roasting temperatures for the best acceptability were 20, 10, and 6 min at 170, 180 and 190, respectively. Hunter color a values of rice germ increased as roasting temperature and time increased, whereas L value decreased. Peroxide values of unroasted, and roasted rice germs at $170^{\circ}C$ for 20 min, $180^{\circ}C$ for 10 min, and $190^{\circ}C$ for 6 min were 2.0, and 145.6, 169.5, and 182.9 meq/kg, respectively, after 9 days storage at $60^{\circ}C$. Four tocopherol and three tocotrienol isomers were identified, whereas no ${\beta}$-tocotrienol was detected. The major tocopherol and tocotrienol isomers in rice germ were ${\alpha}$-tocopherol and ${\alpha}$-tocotrienol, respectively. ${\alpha}$-Tocopherol content in roasted rice germ decreased significantly during storage, whereas those of ${\beta}$- and ${\gamma}$-tocopherols slowly decreased. ${\delta}$-Tocopherol had the highest stability among tocopherol isomers in roasted rice germ. Similar trends were observed in tocotrienol isomers.
