The Effects of Sodium Chloride and the Cultivation Method on Antioxidant Compounds and Activities in Wheat (Triticum aestivum) Sprouts (염화나트륨 처리 및 재배방법이 새싹밀의 항산화 성분 및 활성에 미치는 영향)

  • Sprouts have various health benefits. Specifically, wheat sprouts are rich in bioactive compounds, such as vitamins and polyphenols. Elicitation induces and enhances secondary metabolite biosynthesis in plants. Therefore, in this study, we investigated the effects of sodium chloride (NaCl) treatments on the growth profile, free amino acid content, and antioxidant activity of germinated wheat (Triticum aestivum). Wheat seeds were germinated at 20℃ for 10 days and treated with 0, 2, 4, 7.5, and 10 mM of NaCl 10 days before harvesting. Treating the soil bed with NaCl increased the nutritional component amounts, such as free amino acids and γ-aminobutyric acid. The chlorophyll a and b concentrations were the highest in the hydroponic system treated with 7.5 mM NaCl. In addition, the polyphenol and flavonoid contents of sprouts treated with 2 and 7.5 mM NaCl were 1.94 and 1.34 times higher than that of the control sprouts (0 mM NaCl, water only), respectively. These results suggest that 2 to 4 mM NaCl treatments improve the nutritional and food quality of wheat sprouts more than water only.

Effect of UV-B Radiation on the Leaf Growth of Rice Seedling (벼 유묘의 엽생장에 대한 자외선의 효과)

  • Rice seedlings were used to examine the effect of UV-B radiation on leaf elongation and development. Leaf elongation in both rice seedlings showed differently depending on each leaf age. UV-B radiation strongly reduced leaf elongation, 58-66% compared to without UV-B radiation, of two rice seedlings, therefore, those seedlings could not grow further. Both control and plants grown under UV-B regime showed a diurnial fluctuation in growth rate, showing maximum growth during the light period and minimum during the dark period. Leaf growth at the third leaf stage by UV-B treatment was considerably reduced by 1.7-fold than the control whereas at the fifth leaf stage was not changed. Hydrogen peroxide was considerably increased in the second phase of UV-B-induced response as catalase and peroxidase are deactivated with an increase of UV-B radiation.

Induction of Nitric Oxide and TNF-$\alpha$ by Herbal Plant Extracts in Mouse Macrophages (일부 한약재의 생쥐 대식세포 일산화질소와 TNF-$\alpha$ 생산 유도)

  • In this experiment, we show the effects of herbal plant extracts on the production of nitric oxide (NO) and TNF-$\alpha$. The extracts of Angelica gigas, Astragalus membranaceus, Acanthopanax sessiliflorus and Houttuynia cordata had no effect on NO synthesis by itself in mouse macrophage cell line (RAW264.7). However, the stimulation with these extracts in the presence of murine interferon-${\gamma}$(mIFN-${\gamma}$) resulted in increased NO synthesis. When these extracts were used in combination with mIFN-${\gamma}$, there were a marked cooperative induction of NO and TNF-$\alpha$ synthesis in a dose-dependent manner. The same results were obtained in the mouse peritoneal macrophages used. The optimal concentration of these extracts on NO synthesis was shown at 100$\mu\textrm{g}$/mL with 100U/mL of mIFN-${\gamma}$. NO synthesis was inhibited by NG-monomethyl-L-arginine. When cell lines were treated with extracts, the expression of inducible NO synthetase (iNOS) was markedly increased in RT-PCR analysis. In addition, synergy between mIFN-${\gamma}$ and extracts was dependent on extracts-induced tumor necrosis factor-$\alpha$(TNF-$\alpha$). These results suggest that water extracts of herbal plants can induce iNOS, NO and TNF-$\alpha$ synthesis of mouse macrophage cell line (RAW264.7) and peritoneal macrophages in combination with mIFN-${\gamma}$.

Screening of Some Plant Extracts for Inhibitory Effects on HIV-1 and Its Essential Enzymes (Human Immunodeficiency Virus Type I에 대한 수종 식물 추출물의 억제활성 검색)

  • In order to elucidate the relationship between anti-HIV-1 enzyme activity and inhibition of HIV-1 replication by natural sources, extracts from some plants using the foods and oriental medicines were tested for inhibitory effects on the viral replication, reverse transcriptase (RT), protease and ${\alpha}-glucosidase$. In the anti-RT test, water extracts of Ficus carica (leaf), Houttuynia cordata (aerial part) and Ixeris tamagawaensis (aerial part) showed more than 79% inhitibion at a concentration of $100\;{\mu}g/ml$. The protease and ${\alpha}-glucosidase-inhibiting$ samples in the screening were water extract of Syringa dilatata (leaf) and methanol extract of Hibiscus syriacus (leaf and stem), which showed more than 40% inhibition at a concentration of $100\;{\mu}g/ml$. In the primary anti-HIV-1 test, water extracts of Equisetum arvense (aerial part), Hibiscus syriacus (leaf), Ixeris tamagawaensis (aerial part) and Pueraira thunbergiana (leaf) showed the potent inhibition against HIV-1 induced cytopathic effects.

Antioxidant activities of brown teff hydrolysates produced by protease treatment

  • Purpose: Various plants, herbal medicines, and marine foodstuffs have been used in kimchi preparation to improve its overall quality. Teff, which is rich in minerals and starches, facilitates stable blood glucose levels and is well-suited for use in gluten-free products; hence, it can be used to reinforce the mineral composition of kimchi. In this study, we probed the antioxidant activities of hydrolysates prepared by treatment of brown teff with three proteases under different conditions. Methods: The mineral composition of brown teff was determined by inductively coupled plasma spectrophotometry-mass spectrometry, and we established optimal hydrolysis conditions by determining the total phenol and flavonoid contents of teff hydrolysates obtained using three different proteases (protamax, flavourzyme, and alcalase), two different protease concentrations (1 and 3 wt%), and three different incubation times (1, 2, and 4 h). The antioxidant activity of the hydrolysates was further investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, total antioxidant capacity (TAC), and ferrous reducing antioxidant power (FRAP) assays. Results: Brown teff was rich in I, K, Mg, and Ca, and the highest total phenol content ($24.16{\mu}g/mL$), total flavonoid content ($69.08{\mu}g/mL$), and TAC were obtained for 1 wt% protamax treatment. However, the highest DPPH scavenging activity and FRAP values were observed for hydrolysates produced by alcalase and flavourzyme treatments, respectively. Conclusion: Treatment of brown teff with proteases affords hydrolysates with significantly increased antioxidant activities and high total phenol and flavonoid contents, and these antioxidant activities of teff hydrolysates have the potential to enhance the quality and functionality of kimchi in future applications.

Expression and Purification of Soybean Protein from Escherichia coli (콩 단백질의 대장균 발현과 정제)

  • One of the major objectives of the food industry is the enrichment of the functional properties and nutritional value of soybean protein. To attain this goal, an expression system of cDNA encoding native and protein-engineered soybean proteins in a microorganism must be developed and the function then ability of self-assembly and the functionalities of the expressed proteins should be evaluated before the modified genes are transfered to soybean plants. The pro-$\beta$-conglycinin synthesized in E. coli BL21(DE3) comprised approximately 20% of the total bacterial proteins and the expressed protein are formed soluble and trimer such as native protein in E. coli cells. The highly expressed protein was purified to homogeneity by salt precipitation with 20~40$ Ammonium sulfate ion-exchange chromatography with Q-Sepharose and hydrophobic column chromatography with Butyltoyopearl. Therefore, we concluded that the high-level expression system of $\beta$-conglycinin cDNA was established and a relatively simple and rapid method for purifying pro-$\beta$-conglicinin was also developed.

The Effect of Plant Ethanolic Extracts on Oxidation of Soybean Oil (다류원류 식물류의 에탄올 추출물이 대두유의 산화에 미치는 영향)

  • This study was planned to investigate the effect of 40 plant ethanolic extracts on antioxidant activities in vitro. The total phenolics, $\beta$-carotene, $\alpha$-tocopherol and selenium contents were also determined . Antioxidant activities fo the ethanolic extracts(0.02%, w/w) in the soybean oil were measured both by determining the peroxide value (POV) during 35 days of storage at 4$0^{\circ}C$ in a forced draft air-incubator and by determining changes in conductivity at 11$0^{\circ}C$(Rancimat method.). Soybean oil without any additives was used as a control and that treated with 0.02% BHT was used as a positive control. Based on the POV determination, green tea extract was found to be the most effective in stabilizing soybean oil, then followed by long tea, which both of them showed higher antioxidant activities compared to the BHT treatment. The antioxidant activities of them showed higher antioxidant activities compared to the BHT treatment. the antioxidant activities of coffee, cinnamomi cortex, acanthopanacis cortex, black tea, orange peel , instant coffee, peony and crni fructus extracts were stronger compared to the control .By the Rancimat method, green tea leaf and oolong tea leaf, foxglove, acanthopanacis cortex and peony extracts. Compared to other extracts, green tea leaf, black tea leaf, foxglove, acanthopanacis cortex and peony extracts had stronger antioxidative effects in both the POV and Rancimat methods used in this study. ethanolic extracts which showed the stronger antioxidative effect also has the higher contents of total phenolics, $\beta$-carotene, and/or $\alpha$-tocopherol. The antioxidative effect of ethanolic extracts was found to be due to the combined effect of various antioxidants.

Volatile flavor components of Dioscorea japonica (참마의 휘발성 풍미성분)

  • An attempt was made to derermine the volatile flavor components of Dioscorea japonica. Essential oils from roots of the samples were isolated by simultaneous steam distillation-extraction(SDE) method using diethyl ether as solvent. Concentrated samples were analyzed by gas chromatography(GC) and combined gas chromatography-mass spectrometry(GC-MS). Fifty nine volatile flavor components, including 35 hydrocarbons, 5 aldehydes, 1 ketone, 9 alcohols, 2 esters, 3 acids and 4 miscellaneous ones were confirmed in the young roots of Dioscorea japonica. Forty two components, including 23 hydrocarbons, 2 aldehydes, 7 alcohols, 1 ester and 8 acids and 1 miscellaneous one were confirmed in the roots of mature stage. ${\sigma}-3-Carene$ and dodecanoic acid were regarded as the most abundant components in young and mature roots repectively. The profile of volatile flavor components was markedly different in young and mature roots of Dioscorea japonica.

Screening of Natural Products that have Activities against Skin-Aging (피부노화 억제활성을 갖는 천연 추출물의 탐색)

  • To screen antioxidative and whitening effects of some medicinal plants, 70% ethanol extracts were tested for their electron donating abilities to DPPH ( ${\alpha}$, ${\alpha}$-diphenyl- ${\beta}$-picrylhydrazyl) and inhibitory activities against lipid peroxidation, and tyrosinase inhibition in vitro. The electron donating activities of 70% ethanol extracts at the concentration of 100 $\mu\textrm{g}$/ml were in the order of Forsythiae fructus > Polyporaceae > Glycyrrhizae radix > Schizandrae fructus > Ramulus mori > Carthami semen. The order was Forsythiae fructus > Carthami semen > Schizandrae fructus > Polyporaceae > Glycyrrhizae radix when tested at 10 $\mu\textrm{g}$/ml, indicating that the activities depend on the concentration. The electron donating activities of Forsythiae fructus and Carthami semen at the concentration of 1 $\mu\textrm{g}$/ml were 37% and 31% respectively. Lipid peroxidation inhibitory activities were in the order of Artemisia messerschimidtiana > Glycyrrhizae uadix > Rehmaniae radix > Ramulus mori. Tyrosinase inhibitory activities were in the order of Ramulus mori > Artemisia messerschimidtiana > Oyster mushroom. The results suggest that Forsythiae fructus, Artemisia messerschimidtiana and Ramulus mori could be used as materials for anti-skin aging functional cosmetics.

Antimicrobial Activity of Oleanolic Acid for Foodborne Bacteria (식중독 세균에 대한 Oleanolic Acid의 항균활성)

  • Oleanolic acid and its derivatives are pentacyclic triterpene acids, which are produced in many plants and herbs. These are considered safe and thus, oleanolic acid is now used for cosmetic and pharmaceutical industry. Oleanolic acid affects peptidoglycan in cell wall of bacteria. Hence, the antimicrobial activity of oleanolic acid is not very obvious to Gram-negative bacteria such as Escherichia coli, Yersinia enterocolitica, Shigella flexneri, and Shigella sonnei because the peptidoglycan is covered with outer membrane. However, oleanolic acid derivatives showed improved antimicrobial activity to Gram-negative bacteria. For Gram-positive bacteria such as Staphylococcus aureus and Listeria monocytogenes, oleanolic acid was very effective on reducing the cell counts of the pathogens. In addition, the cytotoxicity of oleanolic acid for human cell lines was minimal. Therefore, oleanolic acid should be considered as an antimicrobial food additive and a therapeutic agent to control foodborne pathogens.
