피부 여드름 치료제 개발을 위한 천연물의 항균활성 검색 (Screening of Anti-acne Activity of Natural Products against Propionibacterium acnes)

  • 피부 여드름 치료, 예방을 위한 천연물의 활성평가를 시도하였다. 335종의 약용 및 야생식물의 다양한 부위로부터 500종의 추출물을 조제하여, 여드름 원인세균인 Propionibacterium acnes에 대한 생육저지 활성을 평가한 결과, 21종 식물체에서 조제한 25종 추출물에서 활성을 확인하였다. 500종 추출물 중, 홀아비꽃대(지상부) 추출물에서 가장 강력한 활성이 나타났으며, 방기, 고삼, 오수유, 은행(씨), 상백피, 독활 및 호장근 추출물에서도 우수한 활성을 확인하였다. 특히 고삼, 상백피, 독활 등의 항여드름 활성은 기존 보고 및 민간요법과도 잘 일치하며, 홀아비꽃대 및 방기 추출물의 항여드름 균 활성은 본 연구에서 처음으로 확인하였다. 고삼을 대상으로 한 활성물질 분리결과, kuraridin과 kurarinone의 주요 활성물질임을 확인하였다. 본 연구결과는, 안정성이 확보된 식물추출물로부터 새로운 피부여드름 치료제 개발이 가능함을 제시하고 있으며, 현재 고삼 추출물 및 활성물질의 세포 독성 및 작용기전에 대한 연구가 진행중이다.

  • The inhibition of rat lens aldose reductase (AR) in vitro using methanol extracts from Korean folk plants was investigated. Among them, the extracts of Saussurea grandifolia and Rumex crispus showed highest inhibition of AR. $IC_{50}$ values of the extracts from S. grandifolia and R. crispus were demonstrated 0.07 and 0.05 mg/ml, respectively. Korean folk plants such as S. grandifolia and R. crispus has a possibility of new natural resources for the inhibition of AR.

  • The methanol extracts of 162 herbal medicines were screened for the inhibitory activity against VHR dualspecificity protein tyrosine phosphatase (DS-PTPase). Seventeen medicinal plants, Scutellaria baicalensis, Cuscuta chinensis, Caesalpinia sappan, Arecae pericarpium, Rubus coreanus, Machilus thunbergii, Amsonia elliptica Cinnamomum cassia, Arisaema erubescens, Pueraria thunbergiana, Dendrobium moniliforme, Mentha arvensis, Peucedanum japonicum, Salvia miltiorrhiza, Leonurus sibiricus, Siegesbeckia orientalis, Prunella vulgaris showed potent VHR DS-PTPase inhibitory activity.

Metabolic classification of herb plants by NMR-based metabolomics

  • Metabolomics is the systematic identification and quantification of all metabolites in an organism or biological sample. NMR has been used as a major application tool in plant metabolomics such as quality control, chemotaxonomy, and analysis of genetically modified plants. Herbal medicines are the important therapeutics and are used to manage common diseases such as cold, inflammation, pain, heart diseases, liver cirrhosis, diabetes and central nerve system diseases. Herb plants include various kinds of species such as geranium, mint, and thyme and so on and contain different kinds of metabolites. We performed NMR-based metabolomics study on the seven different species of herb plants using $^1H$ NMR experiments and OPLS-DA to understand the correlation between the classification of herb plants and their metabolite contents. This study showed clear metabolic discrimination among various herb plants. This metabolmics study found several diagnostic NMR signals which are able to be used as bio-markers for identification of the specific herb plants among various species. Clear metabolic discrimination of herb plants suggests three chemotaxonomic groups of herb species.

  • To search for the aldose reductase (AR) inhibitors from Korean folk plants, the inhibition of rat lens AR in vitro using the methanol (MeOH) extracts from Korean folk plants was investigated. Among fifty four Korean folk plants tested, the MeOH extract of Cedrela sinensis showed highest inhibition of AR ($IC_{50}$ value, 2.52 ${\mu}g/ml$). The plant C. sinensis has a possibility of new natural resources for the development of AR inhibitor for the prevention of diabetic complications.

  • There is a growing concern in the United Stares over the environmental and human health implications associated with heavy use of water, pesticides, and inorganic ferilizers in maintaining picture perfect golf courses. There is also a growing awareness that a beautiful course is not necessarily a healthy course. The following discussion reviews the interrelationship of turfgrass and the soil that supports it and provides basic information on currently available alternatives to turf management practices that feature intensive application of inorganic fertilizers. water and pesticides. Soil is a dynamic natural environment in which microorganisms play an important role. Soil contains a large mass of microorganisms which produce thousands of enzymes that can catalyze the transformation and degradation of many organic molecules. (In top soil under optimum conditions may contain 10 billion cells per gram of soil.). Turfgrass and the soil which supports it are interdependent. The natural organic cycle as applied to turf and soil begins with healthy vigorous grass plants storing up the sun's energy in green plant tissues as chemical energy. Animals obtain energy by eating plants and when plants and animals die, their wastes are returned to the soil and provide "food" for soil microorganisms. In the next step of the organic cycle soil microorganisms break down complex plant tissues into more basic forms and make the nutrients available to grass roots. Finally, growing plants extract the available nutrients from the soil. By free operation of this organic cycle, natural grasslands have some of the most fertile soils on earths.

데침조건에 따른 참나물의 생리활성 성분 변화 (Change of Biologically Functional Compounds of Pimpinella brachycarpa(Chamnamul) by Blanching Conditions)

  • Wild edible plants are consumed as raw and/or processed. Analytical data for raw biologically functional compounds were relatively well established. The changes on functional compounds during processing are, however, not well studied. This study was carried out to investigate the change of the quality of wild edible plants, Pimpinella brachycarpa(Chamnamul) at various conditions. Samples were blanched at the salt concentration of 0%, 1% or 2% for 1, 2, and 3 minutes each. The biologically active compounds, vitamin C, ${\beta}-carotene$, chlorophyll, flavonoids, polyphenols and minerals were analyzed. The concentration of vitamin C in Chamnamul decreased as blanching time increased. Beta-carotene in Chamnamul showed high concentration when Chamnamul was treated under longer blanching time. Higher salt concentration of blanching water, however, resulted in the increased concentration of ${\beta}-carotene$ in the blanched Chamnamul. Higher salt concentration of blanching water also reduced the loss of total flavonoids and total polyphenols from the blanched Chamnamul. The change of colors in the blanching water seemed to be corresponding to those of total flavonoids and total polyphenols concentrations in the blanching water.
