In order to study the effects of green juice of Angelica keiskei Koidz on the toxicity of several selected elements, experiments were conducted with mice for a period of six weeks. The results obtained from the experiment are summarized as follows ; 1) Mice fed with 50 ppm of cadmium showed no adverse effects on the growth rate and mortality , but cadmium accumulation into the internal organs. 2) mice fed with 500ppm of lead showed no reduced effect on the growth rate and mortality, but lead accumulation was increased in the internal organs. The administration of green juice slightly reduced lead accumulation in the internal organs. 3) Mice fed with 50 ppm of selenium also showed no serious adverse effect of the growth rate and mortality , but selenium accumulation occurred in the internal organs. The administration of green juice slightly reduced selenium accumulation in the internal organs, but the modes and degrees of reduction in selenium accumulation were not consistent with the internal organs, liver, kidney and pancreas. 4) Mice fed with 500 ppm of chromium showed, numerically , an adverse effect on the growth rate, but showed no statistically significant difference. The administration of green juice rather increased the accumulation of chromium instead of reduction of chromium accumulation in the internal organs.

BACKGROUND/OBJECTIVES: Cadmium is a toxic metal that is an occupational and environmental concern especially because of its human carcinogenicity; it induces serious adverse effects in various organs and tissues. Even low levels of exposure to cadmium could be harmful owing to its extremely long half-life in the body. Cadmium intoxication may be prevented by the consumption of dietary components that potentially reduce its accumulation in the body. Dietary chitosan is a polysaccharide derived from animal sources; it has been known for its ability to bind to divalent cations including cadmium, in addition to other beneficial effects including hypocholesterolemic and anticancer effects. Therefore, we aimed to investigate the role of dietary chitosan in reducing cadmium accumulation using an in vivo system. MATERIALS/METHODS: Cadmium was administered orally at 2 mg (three times per week) to three groups of Sprague-Dawley rats: control, low-dose, and high-dose (0, 3, and 5%, respectively) chitosan diet groups for eight weeks. Cadmium accumulation, as well as tissue functional and histological changes, was determined. RESULTS: Compared to the control group, rats fed the chitosan diet showed significantly lower levels of cadmium in blood and tissues including the kidneys, liver, and femur. Biochemical analysis of liver function including the determination of aspartate aminotransferase and total bilirubin levels showed that dietary chitosan reduced hepatic tissue damage caused by cadmium intoxication and prevented the associated bone disorder. CONCLUSIONS: These results suggest that dietary chitosan has the potential to reduce cadmium accumulation in the body as well as protect liver function and bone health against cadmium intoxication.
The effect on body fat accumulation on male Wistar rats undergoing continuous feeding with a histidine-enriched diet was investigated. Five-week-age rats were assigned to two groups and were fed either the control diet (purified diet AIN-$76^{TM}$) or the histidine-enriched diet containing 3% histidine for 28 days. It was observed that both adipose tissue masses in retroperitoneal and epididymal areas of rats fed histidine-enriched diet significantly decreased (p<0.05) compared to those of control rats, while there was no significant difference in the food efficiency ratio between them. The blood levels of histidine derivatives of 3-methylhistidine and carnosine were significantly (p<0.05) increased in the rats fed a histidine-enriched diet, whereas there were no significant different between the histidine-enriched diet and control groups in the general amino acid distribution. Our results demonstrate that a histidine-enriched diet suppresses body fat accumulation in rats.
The objective of this study was to evaluate novel usability as natural anti-obesity supplement of Selaginella tamariscina extract. The total phenol contents and total flavonoid contents were $60.29{\pm}3.11GAE\;mg/g$ and $14.90{\pm}0.34QE\;mg/g$, respectively. To evaluate anti-obesity activity of Selaginella tamariscina extract, pancreatic lipase inhibition activity as well as its inhibition effect of lipid accumulation in adipocytes were conducted by Oil Red O staining and lipolysis assay. The result of pancreatic lipase inhibition activity of S. tamariscina extract showed a wide range between 40 and 73% dose dependently. While the incubation of 3T3-L1 cells with S. tamariscina extract inhibited differentiation of preadipocytes and reduced lipid accumulation, the level of released free glycerol into culturing medium was increased in multiple concentrations. These results showed that S. tamariscina extract inhibit adipogenesis and pancreatic lipase activity. Thus, S. tamariscina extract can be a candidate for regulating lipid accumulation in obesity.
This study was carried out to evaluate the effect of Acanthopanacis Cortex boiling extract solutions on the fat accumulation in the obese rats induced by the oral high fat administration for six weeks. Total cholesterol, neutral fat and adipose acid of ACR groups were lower than the control group. During the feeding experiment, LDL and VLDL were increased while HDL was decreased in all groups. Insulin and cortisol were higher than the control group, due to the fat accumulation. Based on the above results, it was shown that it is possible to improve fat accumulation induced by high fat dietary through using the oral administration of Acanthopanacis Cortex boiling extract solutions.
In an effort to elucidate the differential actions of blueberry(BB) in both normal and cancer cells, we utilized human breast cell lines to assess the accumulation of radical oxygen species(ROS) and ROS-associated apoptosis in both human normal breast epithelial(MCF10A) and breast cancer(MCF7) cells. BB extract was added to the cultures at a final concentration of $20{\mu}g/m{\ell}$ for 0(control), 6, 12, and 24 hr intervals. The MCF10A cells evidenced no marked ROS accumulation in the presence of BB, whereas the MCF7 cells evidenced clear ROS accumulation upon BB treatment from 12 hours forward. The number of dying or dead cells did not increase in the BB-treated MCF10A cell groups, whereas that number increased profoundly from 12 hr forward. Furthermore, the expression levels of certain stress-related, and pro- and antiapoptotic gene products evidenced differential responses to BB treatment between the MCF10A and MCF7 cell groups. These results indicate that the components of BB extract differentiate cancer cells by not preventing ROS accumulation within cells and by inducing ROS-associated cell death in cancer cells. However, no marked ROS accumulation or induction of cell death was noted in the normal breast epithelial cells. The fact that BB extract exerted a differential effect on cancer cells opens further directions of research regarding the specific components that exert the differential BB-mediated effects in the selective prevention of normal cells and therapy for cancer tissues in the physiological body.
The purpose of this study was to investigate the effects of green tea catechin on the cadmium accumulation in body, cadmium excretion and detoxification functions in chronic cadmium poisoned rats. Sprague-Dawley male rats weighing 100$\pm$10g were randomly assigned to one normal group and three cadmium poisoned groups. Cadmium groups were classified to catechin free diet (Cd-0C group), 0.25% catechin diet(Cd-0.25C group) and 0.5% catechin diet(Cd-0.5C group) according to the levels of catechin supplement. Animals were maintained on 0, 0.25 and 0.5% catechin diets for 20 weeks and simultaneously administered 50ppm Cd(sup)2+ dissolved in the drinking water. Body weight, food intakes and food efficiency ratio in Cd-0C group was lower than the normal group. The accumulation of cadmium in rat liver, kidney, and blood was reduced by catechin supplementation. The excretion of cadmium in urine and feces was increased by catechin supplementation. The metallothionein(MT) contents in liver and kidney were increased in all cadmium groups compared with that of normal group. The ratios of cadmium absorption and retention ratios were significantly decreased in catechin supplementation groups. Accordingly, catechin supplementation resulted to an excretion of cadmium in urine and feces and a lowered accumulation of cadmium in liver and kidney by increasing methallothionein synthesis that led to the significant decrease in cadmium absorption and retention ratios.(Korean J Nutrition 34(4) : 384~392, 2001)
The purpose of this study was to investigate the effect of vitamin E on the cadmium accumulation in body, cadmium excretion and detoxification functions in chronic cadmium poisoned rats. The accumulation of cadmium in rat liver, kidney and blood was reduced by sufficient vitamin E supplementation. The metallothionein (MT) content in liver and kidney were increased in all cadmium groups compared with that of normal group. The ratio of cadmium absorption and retention were significantly decreased in vitamin E supplementation groups. Accordingly, vitamin E supplementation resulted in an excretion of cadmium in urine and feces and a lowered accumulation of cadmium in liver and kidney. It can be suggested that increased MT synthesis lead to the significant decrease in cadmium absorption and retention ratios.s.
BACKGROUND/OBJECTIVES: The anti-diabetic activity of pear through inhibition of ${\alpha}-glucosidase$ has been demonstrated. However, little has been reported about the effect of pear on insulin signaling pathway in obesity. The aims of this study are to establish pear pomace 50% ethanol extract (PPE)-induced improvement of insulin sensitivity and characterize its action mechanism in 3T3-L1 cells and high-fat diet (HFD)-fed C57BL/6 mice. MATERIALS/METHODS: Lipid accumulation, monocyte chemoattractant protein-1 (MCP-1) secretion and glucose uptake were measure in 3T3-L1 cells. Mice were fed HFD (60% kcal from fat) and orally ingested PPE once daily for 8 weeks and body weight, homeostasis model assessment of insulin resistance (HOMA-IR), and serum lipids were measured. The expression of proteins involved in insulin signaling pathway was evaluated by western blot assay in 3T3-L1 cells and adipose tissue of mice. RESULTS: In 3T3-L1 cells, without affecting cell viability and lipid accumulation, PPE inhibited MCP-1 secretion, improved glucose uptake, and increased protein expression of phosphorylated insulin receptor substrate 1 [p-IRS-1, ($Tyr^{632})$)], p-Akt, and glucose transporter type 4 (GLUT4). Additionally, in HFD-fed mice, PPE reduced body weight, HOMA-IR, and serum lipids including triglyceride and LDL-cholesterol. Furthermore, in adipose tissue, PPE up-regulated GLUT4 expression and expression ratio of p-IRS-1 ($Tyr^{632})/IRS$, whereas, down-regulated p-IRS-1 ($Ser^{307})/IRS$. CONCLUSIONS: Our results collectively show that PPE improves glucose uptake in 3T3-L1 cells and insulin sensitivity in mice fed a HFD through stimulation of the insulin signaling pathway. Furthermore, PPE-induced improvement of insulin sensitivity was not accompanied with lipid accumulation.
Present study was investigate to evaluate the aluminum absorption effect on lactic acid bacteria(Lactobacillus acidophilus ATTC 4356, Lactogacillus bulgaricus ATTC 11842, Lactobacillus casei IFO 3533, and Streptococcus thermophilus KCTC 2185 ; LAB) and Clostridium perfringens ATCC 3627 (CP) in artificial intestinal tract. Their growth rate, aluminum accumulation and cellular distribution was studied under anaerobic broth system. All of above microbes were inhibited by adding 10 to 100ppm of aluminum. The degree of aluminum in LAB (Lactobacillus acidophilus ATCC 4356, Lactobacillus bulgaricus ATCC 11842, Lactobacillus casei IFO 3533, and Streptococcus thermophilus KCTC 2185) was higher than of CP. The largest amount of aluminum was accumulated in Lactobacillus bulgaricus ATCC 11842. Aluminum accumulation in LAB was distributed in 49.1% at cell wall, 27.3% at plasma membrane, and 23.6% at cytoplasm, respectively. This study suggests that LAB might help to eliminate the ingested aluminum in intestinal tract.
