  • The global RNA transcription profiles of Bacillus lentimorbus WJ5 under an in vitro co-culture with Colletotrichum gloeosporioides were analyzed in order to study the antagonistic bacteria-fungi interactions. Using a filter membrane system, B. lentimorhus WJ5 was exposed to the spores of C. gloeosporioides at the late exponential stage. The transcription profiles of the B. lentimorhus WJ5, both with and without a challenge from C. gloeosporioides, were analyzed using custom DNA chips containing 2,000 genome fragments. A total of 337 genes were expressed, with 87 and 47 up- and down-regulated, respectively. Of these, 12 genes, which were involved in central carbon metabolisms, and 7 from minor catabolism were relatively highly up-regulated (> 10 fold) and down-regulated (< 0.2 fold), respectively. Nine genes, which were thought to be related to the antifungal activity, were also up-regulated, but their levels were not so high (2.0 - 9.7 folds). From the results, during the early stage of the co-culture of B. lentimorbus WJ5 and C. gloeosporioides, nutrient competition seemed to occur; therefore, the genes from central carbon metabolisms could be up-regulated, while those from minor catabolism could be down-regulated.

  • This study investigated the effects of carnitine and/or ${\gamma}$ -aminobutyric acid (GABA) supplementation on lipid profiles and some immune related nutrient in mice. Balb/c male mice were orally treated with either an AIN-76 diet (Con), a control diet plus carnitine (CS, 0.5 g/kg bw), a control diet plus GABA (GS, 0.5 g/kg bw) or a control diet plus carnitine plus GABA (CGS, 0.25 g/kg bw, respectively) for 6 weeks. There were no significant differences in feed consumption, energy intake, body weight gain or feed efficiency ratio among the groups during the experimental period. However, abdominal fat deposits were smaller in CS, GS and CGS groups compared with the Con group. Serum and liver triglycerides also were lower in CS, GS and CGS and serum total cholesterol was significantly lower in the CGS group compared with the Con group. Serum LDL cholesterol was lower in the CGS group and liver HDL cholesterol was significantly higher in the CS group compared with Con group. In serum, stearic acid and selecholeic acid were lower, but arachidic acid was higher in the CS group. Liver stearic acid was higher but oleic acid lower in CGS group compared with Con group. In carnitine supplemented groups, serum and liver nonesterified carnitine (NEC), acidsoluble acylcarnitine (ASAC), total carnitine (TCNE) concentrations were higher in only the CS group, not CGS group. Serum vitamin A and E concentrations were not different among the groups. These results may suggest that carnitine and/or GABA supplementation improves lipid profiles in mice, but did not affect the immune-related nutrients that we measured under the experimental conditions of this study.

  • Objectives: This study was conducted to survey the related factors of metabolic syndrome of Chinese workers aged 20 years and above. Methods: The study was conducted at three locations in Shandong, China, currently working and took the physical examination (PE) within one year in the area as target participants. Personal characteristics, physical and biochemical results based on the PE, lifestyle habits, and food intake of the participants were used to analyze the relationship with metabolic syndrome. Results: Results showed that overall, thirty-one subjects (22.5%) had metabolic syndrome, twenty males (32.7%) and eleven females (14.2%). Metabolic syndrome was related to age, gender, educational level and occupational type with more risk in male (P < 0.05), people of older age (P < 0.001), low educational level (P < 0.05) and non-office workers (P < 0.01). According to the life style scores, lifestyle evaluation showed specifically alcohol consumption and smoking (P < 0.001) and stress management (P < 0.05) as important factors that were associated with the metabolic syndrome. High calorie (P < 0.01) and carbohydrate (P < 0.01) intakes were observed on male participants with metabolic syndrome in comparison to the non-metabolic syndrome but no significant difference on female participants. Conclusions: This results of this study can be used as significant supporting data to prevent and control metabolic syndrome in Chinese workers.

  • To evaluate the seasonal differences of nutrient intake and serum biochemical indices (total cholesterol, HDL-, LDL- cholesterol, TG, MDA : malondialdehyde, TAS : total antioxidant status, folate) in Korean college women with a mean age of 20.15y, this study was conducted twice: once in winter and a second time in summer. Anthropometric assessment was also measured in the two seasons. As a result body fat mass (p<0.05), body fat% (p<0.05), and abdominal fat% (p < 0.01) in the summer were significantly lower than those in winter. Intake of fat (p < 0.05), polyunsaturated fatty acid (p < 0.01), zinc (p < 0.05), and vitamin E (p < 0.05) were at significantly higher levels in the summer period compared with the winter period. In addition, nutrients with an intake level less than 75% RI of KDRIs were folate in winter and calcium, and folate in the summer. Total cholesterol (p < 0.05) and HDL-cholesterol in summer, according to lipid profiles, were significantly lower than those in winter. Although MDA, one of the products of lipid peroxidation, remained unchanged, TAS was significantly higher in summer than in winter. The level of folate in summer showed significantly lower than that in winter. The present study suggests that nutrient intake of college women women differs according to the seasons and anthropometric indices and serum biochemical indexes were associated with seasons. Thus nutritional education programs on the basis of season are needed for college women.

  • Cigarette smoking has a negative effect for nutrient intakes and nutritional status of antioxidant nutrients including vitamin C. This study has been performed to compare the dietary nutrient intakes, serum vitamin C concentration and serum cotinine concentration of smoking teenage girls(n=104) with those of non-smoking teenage girls(n=791). The subjects were 895 high school girls(15-19 years of age) living in a rural community in Korea. Of the subjects, 11.6% stated that they were currently smoking. The frequency of taking alcohol, soda and coffee was higher in smokers than in non-smokers, respectively. The usage rate of vitamin/mineral supplements was higher in smokers than in non-smokers. The smokers tended to skip breakfast and lunch and to take snack more often than did the non-smokers. The dietary intakes of calorie, protein, vitamin A, vitamin B, vitamin C, calcium and iron were lower in smokers than in non-smokers. Daily mean intakes of these nutrients were less than the 2/3 of the Korean RDAs in smokers while these intakes were similar to or more than the RDAs in non-smokers. The ratio of under weight or lean tended to be higher in smokers. Serum vitamin C concentration was lower in smokers than in non-smokers. These findings show that smokers have unsound eating habits and poor nutirtional status. In particular, nutritional status of vitamin C is significantly poor in smokers, and this tendency can be found more notably in heavy smokers. Therefore, oxidative stress may be present in young teenage girls with short smoking history as evidence by above results. (Korean J Nutrition 34(3) : 338∼347, 2001)

  • Ginseng (Panax ginseng CA. Meyer) hairy root cultures, which are established via the infection of ginseng root discs with Rhizobium rhizogenes, have been used to construct profiles of both biomass growth and nutrient consumption in flask cultures. In a 250 mL shake flask culture, the maximum biomass was observed on the 59th day of the culture period, at 216.8 g (fresh wt) per liter or 11.4 g (dry wt) per liter. The hairy roots were determined to have a growth rate of 0.355 g-DW/g cells/day during the exponential growth phase and a maximum specific growth rate on day 7. Total ginseng saponin and phenolic compound contents were noted to have increased within the latter portion of the culture period. Linear correlations between increases in biomass weight and nutrient uptake were used to imply the conductivity yield $2.60g-DW/(L{\cdot}mS)$ and carbon yield 0.45 g-DW/(g sugar) in the 250 mL flask cultures. The biomass yield when two different nitrogen sources were used (ammonia and nitrate) was shown to remain approximately constant. at $0.47g-DW/(L{\cdot}mM\;NH_4$) and $0.33g-DW/(L{\cdot}mM\;NO_3$); it remained at these levels for 16 days with the ammonia. and for 24 days with the nitrate. The biomass yield when a phosphate source was used was also shown to remain approximately constant for 9 days, at $3.17g-DW/(L{\cdot}mM\;PO_4$), with an $R^2$ of 0.99.

  • This study was conducted to investigate the anthropometric data, bone density, serum profiles, nutrient intakes and diet quality of premenopausal and postmenopausal women. For the study, we obtained data for analysis from the combined 2008~2011 Korean National Health and Nutrition Examination Survey (KNHANES). The height and weight were 157.8 cm and 58.7 kg in premenopausal women and 155.5 cm and 58.3 kg in postmenopausal women, respectively. The obesity rate was 27.49% in premenopausal women and 34.98% in postmenopausal women (p<0.001). Total cholesterol, LDL-cholesterol, triglyceride, GOT, GPT and alkaline phosphatase in postmenopausal women were significantly higher than those in premenopausal women (p<0.001). The prevalence of osteoporosis was 0.0~0.89% in premenopausal women and 0.48~13.22% in postmenopausal women (p<0.001). In postmenopausal women, rates of hypertension, stroke, myocardial infarction, depression, and diabetes were significantly higher than those in premenopausal women. Water, fat. ash, sodium, retinol, thiamin, riboflavin and niacin intakes in premenopausal women were significantly higher than those in postmenopausal women. Water, fiber, Ca, and, K intakes were below KDRIs (Dietary Reference Intakes for Koreans) in both groups. The mean adequacy ratio (MAR) of premenopausal women was higher than that of postmenopausal women (p<0.001). The index of nutritional quality (INQ) in premenopausal women was also higher than that of postmenopausal women except iron and vitamin C. Therefore dietary guidelines and an education program should be developed for desirable improvement of health, bone density, nutrient status and dietary quality of postmenopausal women.

  • This study was carried out to compare the performance of two types of sequencing batch biofilm reactors (SBBRs), anoxic-oxic-anoxic-oxic $(AO)_2$ SBBR and anoxic-oxic-anoxic $A_2O$ SBBR, on the biological nutrient removal. The TOC removal efficiency in $A_2O$ SBBR was higher than that in $(AO)_2$ SBBR. At the 1st non-aeration period, the release of ${PO_4}^{3-}-P$ in $A_2O$ SBBR was higher than that in $(AO)_2$ SBBR because of the high TOC removal. At the 1st aeration-period, the nitrification was not completed in $(AO)_2$ SBBR, however, it was completed in $A_2O$ SBBR and the nitrification rate in $A_2O$ SBBR was higher than that in $(AO)_2$ SBBR. The release and uptake of ${PO_4}^{3-}-P$ in $A_2O$ SBBR was much higher than in $(AO)_2$ SBBR. Also, the profiles of DO and pH in reactors were used to monitor the biological nutrient removal in two SBBRs. The break point in DO and pH curves at the aeration period coincided with the end of nitrification.

  • This experiment was conducted to investigate the effects of palm kernel expellers on growth performance, nutrient digestibility, and blood profiles of weaned pigs. A total of 88 weaned pigs ($6.94{\pm}0.76kg$ body weight [BW]; 28 d old) were randomly allotted to 2 dietary treatments (4 pigs/pen; 11 replicates/treatment) in a randomized complete block design (sex as a block). The dietary treatments were a typical nursery diet based on corn and soybean meal (CON) and CON added with 20% of palm kernel expellers (PKE). Pigs were fed for 6 wk using a 3-phase feeding program with declining diet complexity and with phases of 1, 2, and 3 wk, respectively. Blood was collected from randomly selected 2 pigs in each pen before weaning and on d 7 after weaning. Pigs were fed respective dietary treatments containing 0.2% chromic oxide from d 29 to 35 after weaning. Fecal samples were collected from randomly selected 2 pigs in each pen daily for the last 3 days after the 4-d adjustment period. Measurements were growth performances, digestibility of dry matter, nitrogen and energy, white and red blood cell counts, packed cell volume, and incidence of diarrhea. The PKE increased average daily gain (ADG) (246 vs 215 g/d; p = 0.06) and average daily feed intake (ADFI) (470 vs 343 g/d; p<0.05) and decreased gain-to-feed ratio (G:F) (0.522 vs 0.628 g/g; p<0.05) during phase 2 compared with CON, but did not affect growth performance during phase 1 and 3. During overall experimental period, PKE increased ADG (383 vs 362 g/d; p = 0.05) and ADFI (549 vs 496 g/d; p<0.05) compared with CON, but did not affect G:F. However, no differences were found on digestibility of dry matter, nitrogen, and energy between CON and PKE. The PKE reduced frequency of diarrhea (15% vs 25%; p = 0.08) for the first 2 wk after weaning compared with CON. Similarly, PKE decreased white blood cells (8.19 vs $9.56{\times}10^3/{\mu}L$; p = 0.07), red blood cells (2.92 vs $3.25{\times}10^6/{\mu}L$; p = 0.09), and packed cell volume (11.1% vs 12.6%; p = 0.06) on d 7 after weaning compared with CON. In conclusion, addition of 20% palm kernel expellers to nursery diet based on corn and soybean meal had no negative effects on growth performance, nutrient digestibility, and blood profiles of weaned pigs.

  • A biomarker is needed to monitor and manage the health of pigs from heat stress (HS). Therefore, we investigated the effects of HS on growth performance, nutrient digestibility, and blood profiles in finishing pigs. A total of 12 finishing pigs (n = 12) were raised in thermal neutral (TN; 25℃) conditions for a 3-d adaptation period. After the adaption, 6 pigs were exposed to HS at 33℃ (HS33) for 5 d. The pigs were fed the same diet based on corn and soybean meal. Chromic oxide was added to all the diets at a level of 2 g·kg-1 as an indigestible marker for the determination of the apparent total track digestibility (ATTD) of nutrients and amino acids. Blood samples were collected after the adaptation and heat treatment to verify the blood profiles. The HS33 pigs had a lower (p < 0.01) average daily feed intake (ADFI) and higher (p < 0.05) rectal temperature compared to the TN pigs. However, there was no difference in the ATTD of nutrients and amino acids. The HS33 pigs had reduced (p < 0.05) levels of serum glucose, non-esterified fatty acids (NEFA), total protein, albumin, and calcium compared to the TN pigs. However, the level of total bilirubin was increased (p < 0.05) in the HS pigs. In conclusion, HS reduced the feed intake and had an adverse effect on health. Altered blood profiles as a result of a negative energy balance are expected to be biomarkers of HS in finishing pigs.
