Whole-mount in situ Hybridization of Mitochondrial rRNA and RNase MRP RNA in Xenopus laevis Oocytes

  • In order to analyze the intracellu1ar localization of specific RNA components of ribonucleoproteins (RNP) in Xenopus oocytes, a modified protocol of whole-mount in situ Hybridization is presented in this paper, Mitochondria specific 12S rRNA probe was used to detect the amplification and distribution of mitochondria in various stages of the oocyte life cycle, and the results were found to be consistent with previously known distribution of mitochondria. The results with other specific probes (U1 and U3 small nuclear RNAs, and 5S RNA) also indicate that this procedure is generally effective in localizing RNAs in RNP complexes even inside organelles. In addition, the RNA component of RNase MRP, the RNP with endoribo-nuclease activity, localize to the nucleus in various stages of the oocyte life cycle. Some of MRP RNA, however, were found to be localized to the special population of mitochondria near the nucleus, especially in the active stage of mitochondrial amplification. It suggests dual localization of RNase MRP in the nucleus and mitochondria, which is consistent with the proposed roles of RNase MRP in mitochondrial DNA replication and in rRNA processing in the nucleolus.

Characterization of the Variability of Nucleoli in the Cells of Panax ginseng Meyer In Vivo and In Vitro

  • Results of karyological study of intact plants and some callus lines of Panax ginseng are presented. In the native plants of P. ginseng the nucleus with 1 nucleolus (90%) dominate, and nucleus with 2 nucleoli is rare. One nucleolar nucleus also dominate in interphase nuclei of cells of cultivated P. ginseng (from 2006), but we also found nucleus with 2 to 3 nucleoli in the same cell lines. Interphase nuclei of P. ginseng in long cultivated lines (from 1988) contain 1 to 9 nucleoli, with a predominance of nuclei containing from 3 to 4 nucleoli. It was shown that long-time cells (cultivated since 1988) had cytogenetic changes such as increase level of polyploid and aneuploid cells, increase of nucleoli number into interphase nucleus and decrease of nuclei/nucleoli ratio. These long-time cultivated cells had very low ginsenoside content.

  • Naturally occurring canine transmissible venereal tumors of genital organs in mature and regressive stages from 6 dogs were examined by transmission electron microscope. The tumor cells at the stage of maturation were comprised of large round and ovoid cells with prominent nuclei and nucleoli, a few spindle-shaped cells, and irregularly shaped cells. The mature round cells were characterized by the presence of a central ovoid to irregularly round nucleus with a large eccentric nucleolus, vesicular endoplasmic reticulum, round to oval swollen mitochondria with few cristae, Golgi's apparatus, and plasma membranes with numerous microvilli. As the tumor degenerated, the tumor cells were increased in the number of spindle-shaped, fibroblast-like and irregularly shaped cells, collagen bundles, and mainly lymphocytes, in contrast to those of the stage of maturation. Regressing tumor cells were characterized by the swelling and vacuolation of mitochondria and endoplasmic reticulum, membrane-bound granules, lamellar complex, tubular structures, and dense bundles of collagen. It was suggested that transformation might occur in the course of tumor growth causing morphological change from the round to the fibroblast-like cells, and that there was the evidence of cell-mediated tumor cell lysis by lymphocyte infiltration.

  • Ultrastructure of germ cell during gametogenesis of surf clam, Spisula sachalinensis were investigated by electron microscopic observations. Oogonia are oval in shape and 5-6 ${\mu}m$ in diameter. They contain a large nucleus and have several mitochondria in the cytoplasm. Vitellogenic oocytes grew attached by a stalk to the germinal epithelium in the ovarian sac and their cytoplasm contained many yolk granules, lipid granules and mitochondria. Mature oocytes measuring 50-60 ${\mu}m$ in diameter have a nucleus containing an electron dense nucleolus, and a lots of yolk granules, lipid granules, mitochondria, rough endoplasmic reticulum and cortical granules were present in the cytoplasm. The surface of the plasma membrane in mature oocytes was formed of microvilli approximately 1.5 ${\mu}m$ long and enveloped in the vitelline layer. Spematogonia are oval in shape and about 5 ${\mu}m$ in diameter. Sprmatogonia develop into spermatocyte, spermatid and spermatozoon. The spermatozoon consisted of the head, midpiece and tail. The sperm head with diameter of about 1.5 ${\mu}m$ was ovoid, and contained the nucleus which consisted of acrosome. The four mitochondria encircled the centrosome in midpiece. The flagellum measuring about 40 ${\mu}m$ long had the classical 9+2 axoneme structure.

  • To investigate ultrastructural characteristics of cancer cells of the nervous system, 25 cases; i.e. astrocytoma(9), oligodendroglioma(1), medulloblastoma(1), meningioma(5), pinealoma(2) and pituitary adenomas(7). The common findings were marked irregularity of nuclear membrane with pronounced infoldings, clumping of heterochromatin along inner nuclear membrane, enlargement of nucleolus, and frequent observations of nuclear bodies and nuclear inclusions. But these findings are also the signs that can be observed in hyperactive cells. Thus, ultrastructural characteristics of cancerous nucleus are the great variability of nuclear size, shape and composition. but none of them appear to be specific. Among cytoplasmic organelles, massive fibrils are characteristic of astrocytoma and meningiomas, cytoplasmic protofibrils such as glial process and microvesicles in oligodendroglioma, secretory granules are characteristic in pituitary adenomas, and fine filamentous fibrils and desmosomes are characteristic of fibroblastic type of meningioma. Intercellular relationships and cell membrane specialization are important features in the differential diagnosis of various undifferentiated tumors. The frequent resolution of difficult diagnosis problems by electron microscopy outweighs the disadvantages of this technique, such as the expense and time required.

Diethylnitrosamine Induced Tissue Damage and Change of Lipid Components in the Chick Embryo Liver (Diethylnitrosamine에 의한 계배 간 조직 손상 및 지질 성분의 변화)

  • Diethylnitrosamine (DEN) is known as a potential hepatic carcinogen by single administration. This study was designed to measure the effects of DEN-induced cell damage on the triglyceride and cholesterol concentration in the liver, excluding dietary effects. Fertilized chicken eggs, 10 days before hatching, were randomly divided into three groups (n=20) and each egg was injected 10 ${mu}ell$ of corn oil (vehicle control), 5 $\mu\textrm{g}$ of DEN/10 ${mu}ell$ of DEN/10 ${mu}ell$ into yolk via air sac. After 48 hr and 96 hr incubation, the damage of the chick-embryo liver cell was investigated by electron microscopy and by measuring the concentration of lipid components (total cholesterol, free cholesterol, phospholipid and triglyceride). For eggs administered 10 $\mu\textrm{g}$ of DEN and incuvated 96 hr, in hepatocyte, the nucleus membrane was roughed, the size of nucleolus was apparently increased and euchromatin was accumulated. Mitochondria were condensed and cristae, located mitochondiral inner membrane, were obscured. Additionally, the leaves of triglyceride and cholesterol classes were significantly increased depend on the amount treated with 10 $\mu\textrm{g}$ DEN at 96 hr, but phospholipids component of cell membrane, were decreased with significance. As a conclusion, carcinogen induced hepatic lesion was correlated with the changes in lipid component of liver.

A primo vessel-like structure in a dog with inflammatory pseudotumor

  • Inflammatory pseudotumor (IPT) is a term defining a mass characterized microscopically by a proliferation of bland mesenchymal spindle cells infiltrated by diffuse mixed inflammatory cells with a predominance of plasma cells and lymphocytes. Here, we show the primo vessel-like structure of the primo-vascular system (PVS) in a dog with IPT. A 6-years old male Mongrel dog was diagnosed with an abnormal mass (diameter 5.5 cm, weight 22 g) near left preputial area. The dog was submitted to the surgical detectomy of the mass. During the surgical operation, we observed primo vessel-like material. After fixations, the masses appeared macroscopically as lipoid-like, firm, white to grey masses, measuring $5{\times}8cm$. Histologically, cellular infiltration into the muscular layers was frequently seen. The mesenchymal proliferation remained the main component of the mass and was composed of myofibroblastic-like spindle cells characterized by globular, irregular nuclei containing open chromatin and a prominent nucleolus. On the basis of the histopathologic lesions, the subcutaneous mass was diagnosed as IPT. Also, we detected a primo vessel-like structures in some areas of the IPT tissues. These were observed as novel thread-like structures and bundle of tubular structures. To our knowledge, this report is the first case of primo vessel-like structure in a dog with IPT.

Reproductive ecology of the blackthroat seaperch, Doderleinia berycoides (Hilgendorf) in South Sea of Korean waters (한국 남해에 서식하는 눈볼대, blackthroat seaperch, Doderleinia berycoides (Hilgendorf)의 생식생태연구)

  • Maturation and spawning of the Blackthroat seaperch, Doderleinia berycoides were investigated based on the samples captured in Korean waters from January 2008 to December 2009. Gonadosomatic index began to increase in June, and reached maximum between July to September. After spawning it began to decrease from October. Reproductive season was estimated to July-September, with peak in August. Fecundity was proportional to the size of the female, with the clutch size varying from 115,500 eggs in the smallest female (TL〓28.2cm) to 652,000 eggs in the largest (TL〓33.5cm). Size at 50% sexual maturity ($TL_{50}$), determined from mature females, was 29.6cm. Annual reproductive cycles of this species could be divided into six successive stages; immature stage (October-May), nucleolus stage (June-July), yolk vesicle stage (July-August), vitellogenic stage (June-September), ripe and spent stage (August-October).

Metallothionein Induction in Liver Regeneration Stimulated by Partial Hepatectomy

  • Metallothionein (MT) is induced in the regenerating rat liver. We have investigated expression of MT gene by RT PCR as well as specific localization of MT by immunocytochemistry in regenerating rat liver after partial hepatectomy (PH). MT mRNA level started to increase from 1 h and reached the peak at 8 h after PH. The level decreased gradually by 24 h, and became similar to that of control group. In the immunocytochemical study, in all groups treated with primary antibody, immunogold particles indicating the presence of MT were evenly distributed throughout both cytoplasm and nucleus of the rat hepatocytes. Within the nucleus, the gold particles appeared to be intensely localized in the areas of euchromatin and nucleolus. Within the cytoplasm, gold particles did not seem to adhere to mitochondria or Iysosomes, but were freely distributed. However, rough endoplasmic reticulum was the obvious compartment on which the gold particles were localized. Time course of MT immunoreactivity revealed that distribution of gold particles in hepatocytes increased gradually by 24 h, and decreased at 48 h after PH. Briefly, PH resulted in the sharpest increase in the expression of MT mRNA at 8 h and in the immunoreactivity of MT at 24 h, respectively. It is suggested that the increase of MT mRNA expression, the intensity of immunoreactivity and the specific localization of MT may be associated with the compensatory cell proliferation followed by PH.

Mitochondrial Location of Severe Acute Respiratory Syndrome Coronavirus 3b Protein

  • Severe acute respiratory syndrome-associated coronavirus (SARS-CoV), a distant member of the Group 2 coronaviruses, has recently been identified as the etiological agent of severe acute respiratory syndrome (SARS). The genome of SARS-CoV contains four structural genes that are homologous to genes found in other coronaviruses, as well as six subgroup-specific open reading frames (ORFs). ORF3 encodes a predicted 154-amino-acid protein that lacks similarity to any known protein, and is designated 3b in this article. We reported previously that SARS-CoV 3b is predominantly localized in the nucleolus, and induces G0/G1 arrest and apoptosis in transfected cells. In this study, we show that SARS-CoV 3b fused with EGFP at its N- or C- terminus co-localized with a mitochondriaspecific marker in some transfected cells. Mutation analysis of SARS-CoV 3b revealed that the domain spanning amino acids 80 to 138 was essential for its mitochondria localization. These results provide new directions for studies of the role of SARS-CoV 3b protein in SARS pathogenesis.
