Sclerotinia rot occurred severely on some vegetable crops grown in Namyangju, Yangpyung, and Yangiu areas in Korea in 2001-2002. The crops infected with Scterotinia sp. were Adenophora remotiflora, Armoracia lapathfolia, Angelica acutiloba, Angelica archangelica, Anthriscus sylvestris, Aster tataricus, Beta vulgaris var. cicla, Brassica campestris var. marinosa, Brassica juncea var. laciniata, Chicholium intybus, Lactuca indica var. dracoglossa, Lactuca sativa var. oak-leaf, Petroselinum crispum, and Phyteuma japonicum. The fungus associated with the disease was identified as Sclerotinia sclerotiorum, based on the morphological characteristics of the pathogen. The symptoms were water-soaked spots that enlarged later and became a watery soft rot. Infected parts became yellow and then turned brown, followed by death of the whole plant. White mycelia developed on the upper petioles and leaves and on the soil where these plant parts lay. Then black sclerotia in variable size and shape formed from the mycelial mass. Pathogenicity of the fungus was proven by artificially inoculating each crop. This is the first report of Sclerotinia rot on the listed vegetable crops in Korea.

Cultural conditions during production of compost, using wheat straw and chicken litter as raw materials, will affect the microbial and biochemical characteristics, leading to a wide variation in mushroom productivity. Over the past 10 years, chemical and instrumental methods, suitable for assessing compost quality have been studied in Northern Ireland. In addition, the use of near subject of investigation over the past 4 years. Previous studies have shown that NIRS can be used fer assessing quality of dried and milled composts. The aim of the current investigation is to develop NIR calibrations for key quality parameters such as dry matter, pH, nitrogen, carbon, ash, microbial population and fibre factions during the two stages of production using spectra of fresh composts. Near infrared reflectance measurements of fresh composts prepared by 6 producers were made during a two-year period. Although the spectra of fresh composts were dominated by two moisture peaks at 1450 nm and 1940 nm, good calibrations for determining moisture content, conductivity, pH, nitrogen, carbon and fibre fractions were developed. The results of quality assessment during commercial production using the calibrations will be presented and discussed.
The ratio of occurrence of azalea (Rhododendron mucromultum T.) was determined on the north and south facing slopes on a mountain in Koyang county, Kyungi Province. Data was actually recorded at the eight aspects facing N, NE, E, SE, S, SW, W and NW on the mountain. In each of the eight aspects, six 10m transect tapes were placed in parallel directly down the slope, each 1m apart, and fifty 1$m^2$ quadrats were constructed between them. Frequency of the azalea in each of these quadrats was determined by counting the central stumps of the bushes Soil samples were also taken from pits dug in each plot to use for determination of the following: Soil pH, soil moisture loss on ignition, total nitrogen, and available phosphorus. The occurrence rate of azalea on the northern slop e was found to be for higher on the northern slope than on the southern slope. The abundance of azalea on the northern slope was found to be partially correlated with soil moisture(r=0.93; 0.01>p) and the amount of humus in the soil (r=0.04;0.01>p). The content of total nitrogen, soil PH, and available phosphorus in the soil, however, were found to have no evident relationship with the abundance of azalea. The conditions for growth in the species of azalea Rhododendron mucronulatum T'. were generally found to be more favorable on the northern slope.

This study analyzed the status of climate-change indicator plants native to the main islands of the Korean peninsula, while elucidating their distribution characteristics. Information on flora from over 129 island locations, comprising more than 100 species of native plants, was collected, compiled into a database, and utilized as raw data. The distribution of 193 climate-change indicator plants was confirmed. The distribution area of broadleaf evergreen trees and ferns, including Mallotus japonicus and Cyrtomium falcatum, was relatively wide. In contrast, the distribution of common northern plants such as Corydalis turtschaninovii and Malus baccata was limited. If global warming persists, northern plant distribution is expected to decrease rapidly in the Korean Peninsula island region, while the northern limit line of the southern plants is expected to migrate further northward. During this process, it is likely that the plant congregation structure and species diversity within the island region will change dynamically. In this study, comparative analyses between species and regions were conducted by assessing the relative frequency of their occurrence, and six types of botanical geographic distribution patterns were noted.
In vitro grown microtubers of potato (cv Jaju) were used for introduction of herbicide resistance gene using bombardment with DNA-coated particles. The apical shoot-tip area of newly sprouted microtubers were intensively bombarded. After bombardment, microtubers were germinated and transplanted in a greenhouse. Northern blot analysis indicated that bar gene was expressed in two plantlets. After 5 weeks of growing, commercial herbicide Basta was sprayed to screen the resistant plants. All untransformed potato plants died after 7 days while two transgenic plants survived.

Desiree and Atlantic, were infected with a Agrobacterium MP90 strain containing chimeric gene construct, consisting of antibiotic and low temperature regulated gene (H28) for transformation. regenerated multiple shoots were selected on a medium containing kanamycin and carbenieillin after exposure to Agrobacterium. Both PCR analysis of NPT Ⅱ, H28 genes and northern blot analysis indicated that the genes coding for the enzyme were successfully integrated into the potato genome and could be expressed in potato plants.
To understand plant-pathogen interactions, a complete set of hot pepper genes differentially expressed against pathogen attack was isolated. As an initial step, hundreds of differentially expressed cDNAS were isolated from hot pepper leaves showing non-host resistance against bacterial plant pathogens (Xanthomonas campestris pv. glycines and Pseudomonas syringae pv. syringae) using differential display reverse transcription polymerase chain reaction (DDDRT-PCR) technique. Reverse Northern and Northern blot analyses revealed that 50% of those genes were differentially expressed in pepper loaves during non-host resistance response. Among them, independent genes without redundancy were micro-arrayed for further analysis. Random EST sequence database were also generated from various CDNA libraries including pepper tissue specific libraries and leaves showing non-host hypersensitive response against X. campestris pv. glycines. As a primary stage, thousands of cDNA clones were sequenced and EST data were analyzed. These clones are being spotted on glass slide to study the expression profiling. Results of this study may further broaden knowledge on plant-pathogen interactions.
In March 2000, gray mold was found on day lily (Hemerocallis fulva L.) in Korea. Among the symptoms observed was blight or early rot with chlorotic halo of the leaves. All the isolates obtained from the lesions of the diseased plant parts were identified as Botrytis elliptica, based on the morphological characteristics of conidia. Conidia that formed on conidiogenous cells were not in chains, hyaline to pale brown, unicellular, ellipsoidal to obovate with a single hilum at the base, entirely verruculose, and 21-31 x 12-$23\mu\textrm{m}$ in size. Pathogenicity of the fungus was established by artificial inoculation on day lily plants. This is the first record of gray mold on day lily caused by B. elliptica in Korea.
A leaf spot disease occurred on Allium tuberosum Roth. (Chinese chive) in Korea. All the isolates of Stemphylium sp. from the lesions of the diseased plant parts were identified as S. vesicarium (Waller.) Simmons, based on the morphological characteristics of conidia and conidiophores. Pathogenicity of the fungus was proved by artificial inoculation on Chinese chive plants. This is the first record of leaf blight on Chinese chive caused by S. vesicarium in Korea.
To investigate flowering related genes in winter-type oilseed rape (Brassica napus L. cv. Tammi), differentially expressed genes were isolated from leaves of the plant after low temperature treatment which is requirements for floral induction. As a result of suppression subtractive hybridization (SSH), 288 clones were randomly selected from SSH library. Using reverse Northern blot analysis, 150 of 288 clones were identified to be differentially expressed. Out of these 150 clones, 45 clones showed very high identities with the known genes. Four clones showed very high identities over 90% with metallothionein-like gene that is related to flowering-induced genes. Of these 4 clones, the cDNA clone, rfs-13, revealed high identity with meotallothionein-like protein in Arabidopsis thaliana (98%) and Brassica compestris (89%). Furthermore, gene expressed in immature flower stages was confirmed by Northern blot analysis.
