Construction of T$_1$ Map Image (T1 이완시간의 영상화)

  • The T1 mapping of an human anatomy may give a characteristic contrast among the various tissues and the normal/abnormal tissues. Here, the methodology of constructing T1 map out of several images with different TRs, will be described using non-linear curve fitting. The general curve fitting algorithm requires the initial trial values T1t and Mot for the variables to be fitted. Three different methods of suppling the trial T1t and Mot are suggested and compared for the efficiency and the accuracy. The curve-fitting routine was written in ANSI C and excuted on a SUN workstation. Several distilled-water phantoms with various concentrations of Gd-DTPA were prepared to examine the accuracy of the curve-fitting program. An MR image was used as the true proton density image without any random noise, and several images with different TRs were generated with the theoretical T1 relaxation times 250, 500, and 1000msec. The random noise of 1, 5, and 10% were embedded into the simulated images. These images were used to generate the T1 map, and the resultant T1 maps for each T1 were analyzed to study the effect of the random noise on the T1 map.

A Comparison of Postharvest Physiology and Storability of Paprika Fresh-Cut Made from Disordered and Normal Fruits (착색단고추 생리장해과와 정상과의 수확 후 생리 및 신선편이의 저장성 비교)

  • The study was conducted to compare the postharvest physiology and storability of fresh cut paprika fruits classified by normal, blossom end rot(BER), and misshapen (or knots) fruit. Some disordered paprika fruits that were produced frequently during high temperature season in highland, were sorted out to non-marketable products. These fruits are mostly wasted, but some of them may be used for fresh cut. The respiration rate of fresh cut paprika fruits was lower and ethylene production rate was higher in normal fruits than in disordered fruits, but there was no significant difference. The fresh-cut paprika fruits were stored in MAP conditions at $4^{\circ}C$, $9^{\circ}C$ and room temperature in 25 ${\mu}m$ and 50 ${\mu}m$ thickness ceramic film packaging. The fresh weight of fresh cut paprika fruits decreased below to 1.1% regardless of fruit types, but the fresh weight loss increased in thinner packaging materials and lower storage temperatures. There were not significant different carbon dioxide and oxygen contents in MAP of all fruit types, while $4^{\circ}C$ storage temperature treatment and 25 ${\mu}m$ thickness ceramic film treatment had lower carbon dioxide and higher oxygen contents. Moreover, the carbon dioxide and oxygen contents were changed rapidly at 9 days in $4^{\circ}C$ storage and at 6 days in $9^{\circ}C$ storage when the visual quality of fresh cut decreased dramatically. The ethylene concentration of packages was below 7 ${\mu}l{\cdot}l^{-1}$ in all treatments during storage, while the treatments of thinner packaging material and lower storage temperature showed lower ethylene concentration. The fresh cut of disordered fruits showed less visual quality than normal fruit treatment in both $4^{\circ}C$ and $9^{\circ}C$ storage temperatures, but there was no significant difference. The value of $4^{\circ}C$ treatment that measured 12 days in storage was higher than $9^{\circ}C$ treatment that measured 9 days in storage. The results suggest that the disordered fruits may be used to fresh cut product without any concerns that they will decreased the value of commodities more quickly than the fresh cut made of marketable paprika fruits. As the fresh cut paprika fruits stored in MAP condition, the more effective storage temperature is $4^{\circ}C$ that may have induced chilling injury a whole fruit of the paprika.

Comparison of Metabolic Profiles of Normal and Cancer Cells in Response to Cytotoxic Agents

  • Together with radiotherapy, chemotherapy using cytotoxic agents is one of the most common therapies in cancer. Metabolic changes in cancer cells are drawing much attention recently, but the metabolic alterations by anticancer agents have not been much studied. Here, we investigated the effects of commonly used cytotoxic agents on lung normal cell MRC5 and lung cancer cell A549. We employed cis-plastin, doxorubicin, and 5-Fluorouracil and compared their effects on the viability and metabolism of the normal and cancer cell lines. We first established the concentration of the cytotoxic reagents that give differences in the viabilities of normal and cancer cell lines. In those conditions, the viability of A549 decreased significantly, whereas that of MRC5 remained unchanged. To study the metabolic alterations implicated in the viability differences, we obtained the metabolic profiles using $^1H$-NMR spectrometry. The $^1H$-NMR data showed that the metabolic changes of A549 cells are more remarkable than that of MRC5 cells and the effect of 5-FU on the A549 cells is the most distinct compared to other treatments. Heat map analysis showed that metabolic alterations under treatment of cytotoxic agents are totally different between normal and cancer cells. Multivariate analysis and weighted correlation network analysis (WGCNA) revealed a distinctive metabolite signature and hub metabolites. Two different analysis tools revealed that the changes of cell metabolism in response to cytotoxic agents were highly correlated with the Warburg effect and Reductive lipogenesis, two pathways having important effects on the cell survival. Taken together, our study addressed the correlation between the viability and metabolic profiles of MRC5 and A549 cells upon the treatment of cytotoxic anticancer agents.

Uncertainty of Agrometeorological Advisories Caused by the Spatiotemporally Averaged Climate References (시공간평균 기준기후에 기인한 농업기상특보의 불확실성)

  • Agrometeorological advisories for farms and orchards are issued when daily weather exceeds a predefined range of the local reference climate, which is a long-term average of daily weather for the location. The reference climate at local scales is prepared by various simplification methods, resulting in uncertainty in the agrometeorological advisories. We restored daily weather data for the 1981-2010 period and analyzed the differences in prediction results of weather risk by comparing with the temporal and spatial simplified normal climate values. For this purpose, we selected the agricultural drought index (ADI) among various disaster related indices because ADI requires many kinds of weather data to calculate it. Ten rural counties within the Seomjin River Basin were selected for this study. The normal value of 'temporal simplification' was calculated by using the daily average value for 30 years (1981-2010). The normal value of 'spatial simplification' is the zonal average of the temporally simplified normal values falling within a standard watershed. For residual moisture index, temporal simplification normal values were overestimated, whereas spatial simplification normal values were underestimated in comparison with non-simplified normal values. The ADI's calculated from January to July 2017 showed a significant deviation in terms of the extent of drought depending on the normal values used. Through this study, we confirmed that the result of weather risk calculation using normal climatic values from 'simplified' methods can affect reliability of the agrometeorological advisories.

Locating QTLs controlling overwintering seedling rate in perennial glutinous rice 89-1 (Oryza sativa L.)

  • A new cold tolerant germplasm resource named glutinous rice 89-1 (Gr89-1, Oryza sativa L.) can overwinter using axillary buds, with these buds being ratooned the following year. The overwintering seedling rate (OSR) is an important factor for evaluating cold tolerance. Many quantitative trait loci (QTLs) controlling cold tolerance at different growth stages in rice have been identified, with some of these QTLs being successfully cloned. However, no QTLs conferring to the OSR trait have been located in the perennial O. sativa L. To identify QTLs associated with OSR and to evaluate cold tolerance. 286 $F_{12}$ recombinant inbred lines (RILs) derived from a cross between the cold tolerant variety Gr89-1 and cold sensitive variety Shuhui527 (SH527) were used. A total of 198 polymorphic simple sequence repeat (SSR) markers that were distributed uniformly on 12 chromosomes were used to construct the linkage map. The gene ontology (GO) annotation of the major QTL was performed through the rice genome annotation project system. Three main-effect QTLs (qOSR2, qOSR3, and qOSR8) were detected and mapped on chromosomes 2, 3, and 8, respectively. These QTLs were located in the interval of RM14208 (35,160,202 base pairs (bp))-RM208 (35,520,147 bp), RM218 (8,375,236 bp)-RM232 (9,755,778 bp), and RM5891 (24,626,930 bp)-RM23608 (25,355,519 bp), and explained 19.6%, 9.3%, and 11.8% of the phenotypic variations, respectively. The qOSR2 QTL displayed the largest effect, with a logarithm of odds score (LOD) of 5.5. A total of 47 candidate genes on the qOSR2 locus were associated with 219 GO terms. Among these candidate genes, 11 were related to cell membrane, 7 were associated with cold stress, and 3 were involved in response to stress and biotic stimulus. OsPIP1;3 was the only one candidate gene related to stress, biotic stimulus, cold stress, and encoding a cell membrane protein. After QTL mapping, a total of three main-effect QTLs-qOSR2, qOSR3, and qOSR8-were detected on chromosomes 2, 3, and 8, respectively. Among these, qOSR2 explained the highest phenotypic variance. All the QTLs elite traits come from the cold resistance parent Gr89-1. OsPIP1;3 might be a candidate gene of qOSR2.

Step Trajectory/Indoor Map Feature-based Smartphone Indoor Positioning System without Using Wi-Fi Signals (Wi-Fi 신호를 사용하지 않고 보행자 궤적과 건물내 지도 특성만을 이용한 스마트폰 실내 위치 측정 시스템)

  • In this paper, we proposed indoor positioning system with improved accuracy. The proposed indoor location measurement system is based pedestrian location measurement method that use the embedded sensor of smartphone. So, we do not need wireless external resources, such as GPS or WiFi signals. The conventional methods measure indoor location by generating a movement route of pedestrian by step and direction recognition. In this paper, to correct the direction sensor error, we use the common feature of the normal indoor floor map that the indoor path is lattice-structured. And we quantize moving directions depending on the direction of indoor path. In addition, we propose moving direction measuring method using geomagnetic sensor and gyro sensor to improve the accuracy. Also, the proposed step detection method uses angle and accelerometer sensors. The proposed step detection method is not affected by the posture of the smartphone. Direction errors caused by direction sensor error is corrected due to proposed moving direction measuring method. The proposed location error correction method corrects location error caused by step detection error without the need for external wireless signal resources.

A Comparison of Cardiovascular Effects between Orotracheal Intubation and Nasotracheal Intubation (경구기관삽관법과 경비기관삽관법의 심혈관계 영향에 대한 비교)

  • Background: This prospective study was designed to compare the cardiovascular response to endotracheal insertion of either an orotracheal tube or a nasotracheal tube Methods: 120 ASA physical status I and II surgical patients requiring general anesthesia and tracheal intubation were studied and assigned to two groups: orotracheal intubation group (n = 60) and nasotracheal intubation group (n = 60). Patients were premedicated with midazolam 0.05 mg/kg and glycopyrrolate 0.005 mg/kg intramuscularly and anesthesia was induced with thiopental sodium 5 mg/kg and succinylcholine 0.1 mg/kg intravenously. Systolic blood pressure (SBP), diastolic blood pressure (DBP). mean arterial pressure (MAP) and heart rate (HR) were assessed noninvasively before induction of anesthesia and immediately after intubation, 1 min, 2 min, 3 min, and 5 min after intubation. Results: Cardiovascular responses such as SBP, DBP, MAP and HR were similar for both techniques and no significant differences between two groups were observed until 5 min after intubation. Conclusions: In healthy ASA I and II patients with normal blood pressure, induction doses of thiopental sodium 5 mg/kg and succinylcholine 0.1 mg/kg didn't attenuated the cardiovascular response to laryngoscopy and tracheal intubation. Insertion of an endotracheal tube may be the most invasive stimulus during intubation procedures. (JKDSA 2001; 1: 10-15)

A Study on Serum Zinc Concentration in some Methamphetamine Abusers (Methamphetamine 남용자에 있어서 혈청 아연농도에 관한 연구)

  • The author investigated the zinc concentrations in sera of 110 methamphetaimine abusers (male 87, female 23) and of 41 healthy official workers(male 29, female 12) using inductively coupled plasma emission spectrometer. The serum zinc concentrations were analysed according to hepatitis C virus antibody(Anti HCV), hepatitis B surface antigen(HBsAg) and alanine aminotransferase(ALT) value in both groups. The analytical results were obtained as follows 1. The mean zinc concentration of MAP abuser group($298.5{\pm]246.9{\mu}g/100ml$) was 2.5 times higher than that of control group($118.8{\pm}25.4{\mu}g/100ml$). 2. The distributions of zinc concentration in both groups were skewed pattern. 3. The zinc concentrations were higher in Anti HCV-positive group and abnormal ALT value group than Anti HCV-negative group and normal ALT value group. But HBs-Ag positive group had lower zinc concentration than HBs-Ag negative group. 4. The number of anti HCV-positive subjects was increased and the zinc concentrations were decreasd in proportion to the abusing duration of MAP.

  • YBCO do superconducting quantum interference device (SQUID) magnetometers based on bicrystal junctions have been fabricated for magnetocardiograph (MCG) measurements. We could fabricate YBCO SQUID magnetometers having magnetic field noise of about $20fT/Hz^{1/2}$ at white noise region. We have developed an MCG system employing the high performance SQUID magnetometers. The lightweight MCG system, requiring liquid nitrogen as a coolant, consists of 6-channel SQUID sensors, an adjustable patient bed with sliding motion, and data analyses software. The MCG system could record quite clear MCG signals in a room with moderate magnetic shielding. In normal operation with multi-position MCG measurements, we could obtain clear 48-point mappings of magnetic field map and current source map with high enough signal qualities far clinical trials.

R and T Wave Amplitude as a Parameter to Detect Coronary Artery Disease (관상동맥질환을 진단하기 위한 R파와 T파의 크기에 대한 연구)

  • Multi-channel magnetocardiography (MCG) has been proposed to detect ischemic heart disease because its sensitivity is quite high comparing with other conventional diagnostic tools. Especially, current map and magnetic field map of MCG provide crucial information on whether myocardiac muscles maintain the normal conduction pathway. In addition, MCG parameters derived from repolarization are useful to detect coronary artery disease. Recently, there was a study reporting that R- and T- wave amplitude are highly correlated with ischemic heart disease. In this study, we studied R- and T-wave amplitude and their ratio as well as MCG parameters. MCG data from 20 young, 20 age-matched controls, and 20 myocardial infarction (MI) patients were analyzed. As a result, MCG parameters showed significant change in MI patients comparing to those of controls. R- and T-wave amplitude of MI patients showed a feature of severe ischemic heart disease even though it was difficult to find consistent values. Further study is needed to reveal the relations between small T-wave amplitude and coronary artery disease.
