  • Diabetic nephropathy remains a major cause of morbidity and mortality in the diabetic population and is the leading cause of end-stage renal failure. Despite current therapeutics including intensified glycemic control and blood pressure lowering agents, renal disease continues to progress relentlessly in diabetic patients, albeit at a lower rate. Since synthetic drugs for diabetes are known to have side effects, fungal mushrooms as a natural product come into preventing the development of diabetes. Our previous report showed the hypoglycemic effect of extracellular fungal polysaccharides (EPS) in streptozotocin (STZ)-induced diabetic rats. In this study, we analyzed the differential expression patterns of rat kidney proteins from normal, STZ-induced diabetic, and EPS-treated diabetic rats, to discover diabetes-associated proteins in rat kidney. The results of proteomic analysis revealed that up to 500 protein spots were visualized, of which 291 spots were differentially expressed in the three experimental groups. Eventually, 51 spots were statistically significant and were identified by peptide mass fingerprinting. Among the differentially expressed renal proteins, 10 were increased and 16 were decreased significantly in diabetic rat kidney. The levels of different proteins, altered after diabetes induction, were returned to approximately those of the healthy rats by EPS treatment. A histopathological examination showed that EPS administration restored the impaired kidney to almost normal architecture. The study of protein expression in the normal and diabetic kidney tissues enabled us to find several diabetic nephropathy-specific proteins, such as phospholipids scramblase 3 and tropomyosin 3, which have not been mentioned yet in connection with diabetes.

  • Purpose: Familial hypokalemic periodic paralysis (HOKPP) is an autosomal dominant channelopathy characterized by episodic attacks of muscle weakness and hypokalemia. Mutations in the calcium channel gene, CACNA1S, or the sodium channel gene, SCN4A, have been found to be responsible for HOKPP; however, the mechanism that causes hypokalemia remains to be determined. The aim of this study was to improve the understanding of this mechanism by investigating the expression of calcium-activated potassium ($K_{Ca}$) channel genes in HOKPP patients. Methods: We measured the intracellular calcium concentration with fura-2-acetoxymethyl ester in skeletal muscle cells of HOKPP patients and healthy individuals. We examined the mRNA and protein expression of KCa channel genes (KCNMA1, KCNN1, KCNN2, KCNN3, and KCNN4) in both cell types. Results: Patient cells exhibited higher cytosolic calcium levels than normal cells. Quantitative reverse transcription polymerase chain reaction analysis showed that the mRNA levels of the $K_{Ca}$ channel genes did not significantly differ between patient and normal cells. However, western blot analysis showed that protein levels of the KCNMA1 gene, which encodes $K_{Ca}$1.1 channels (also called big potassium channels), were significantly lower in the membrane fraction and higher in the cytosolic fraction of patient cells than normal cells. When patient cells were exposed to 50 mM potassium buffer, which was used to induce depolarization, the altered subcellular distribution of BK channels remained unchanged. Conclusion: These findings suggest a novel mechanism for the development of hypokalemia and paralysis in HOKPP and demonstrate a connection between disease-associated mutations in calcium/sodium channels and pathogenic changes in nonmutant potassium channels.

  • New occupational qualification system built on NCS(National Competency Standard) aimed to induce industries, the final appliers of qualification, to play a leading role in designing qualification and to spontaneously apply such qualification standard to employment and promotion so as to reinforce universality of qualification. In this study, we verified the validity of NCS level of qualification categories for 130 occupational fields by redesigning NCS-based new occupational qualification system with a focus on 7 major areas, including mechanical field, which were found to have wide-ranging social ripple effect throughout society and high acceptability in 2014. The results of study on suitability at qualification level suggested the followings: First, there is a difference in number of competency units for each qualification category, depending on area. New qualification covering 7 major areas included 15.3 competency units for each qualification category, which exceeds the number of competency units(about 10 units) in subdivided NCS. By field, some difference was found with 7.9 competency units for cultural field and 22.6 competency units for information and communication area. Second, preference above normal level was observed in the criteria, procedure and method for determining qualification redesigning. However, preference below normal level was exhibited in connection with possibility of linkage with overseas qualifications, although preference was above normal level in relation to the suitability of design/qualification level and qualification categories based on lifelong career development path within scope of NCS as the criteria for determining the redesign of qualification categories. Second, we compared NCS level and SC(Sector Council level, and the results showed that NCS level was found to be somewhat well-defined. For the qualifications with different definitions, it was deemed difficult to determine superiority between NCS level and SC level. However, majority opinion indicated that it would be desirable to follow NCS level because NCS was created through collection of opinions for a prolonged period.

  • An anomalous systemic arterial supply to the normal basal segments of the left lower lobe without sequestration is a rare congenital anomaly. It differs from classical bronchopulmonary sequestration in that the involved lung retains a normal connection to the bronchial tree, although some place this entity exists within the broad framework of pulmonary sequestration. We experienced a case of a woman who presented with a nodular lesion on a chest X-ray. Contrast-enhanced CT diagnosed her as having an anomalous systemic arterial supply to the normal basal segments of the left lower lobe. This case is reported with a brief literature review.

  • In this study, a prediction model for the effective compressive strength of corner columns with intervening normal strength concrete slabs was developed. A structural analogy between high-strength concrete column-normal strength concrete slab joint and brick masonry was used to develop the prediction model. In addition, the aspect ratio of slab thickness to column dimension was considered in the models. The reliability of the new prediction model was evaluated by comparison with experimental results and its superiority was demonstrated by comparison with previous models proposed by design codes and other researchers. As a result, with average test-to-predicted ratios of 1.09, a standard deviation of 0.15, the newly developed equation provided superior predictions in terms of accuracy and consistency over all of the existing effective strength prediction approaches including KCI structural concrete design code (2012).

  • In proportion as the growth of the wireless sensor network applications, we need for more accuracy wireless channel information. In the case of indoor or outdoor wireless sensor networks, multipath propagation causes severe problems in terms of fading. Therefore, a path-loss model for multipath environment is required to optimize communication systems. This paper deals with log-normal path loss modeling of the indoor 2.4 GHz channel. We measured variation of the received signal strength between the sender and receiver of which separation was increased from 1 to 30m. The path-loss exponent and the standard deviation of wireless channel were determined by fitting of the measured data. By using the PRR(Packet Reception Rate) of this model. Wireless sensor channel is defined CR(Connect Region), DR(Disconnected Region). In order to verify the characteristics of wireless channel, we performed simulations and experiments. We demonstrated that connection ranges are 24m in indoor, and 14m in outdoor.

  • The computer simulation for the fault current limiting characteristics of the superconducting fault current limiter (SFCL) using the magnetic coupling of two coils was performed. The magnetic fluxes generated from two coils were canceled out during a normal time. However, the resistance generation of high-$T_c$ superconducting (HTSC) element after a fault occurrence keeps up the magnetic fluxes of two coils and contributes to the fault current limiting operation. Through the computer simulation for the fault current limiting characteristics based on its electrical equivalent circuit, its operational current and the limiting impedance could be improved by adjusting the inductance ratio between two coils.

  • A study on the construction of a CAD/CAM system operated by 16 Bit PC basic language, for machining sculptured surface of die, was carried out. The system consists of 2 steps i.e., process for geometric modelling by wire frame and process for machining data generation. Geometric modelling for sculptured surface is made by the point data fitting, parallel sweeping, normal sweeping and linear connection of cross section curve. Machining data are gained by cutter off-set of geometric model data and machining carried out by DNC. This system is to be proved enough for rough cutting by actual machining experiment. But, for becoming a high level system, another method of cutter off-set has to be regarded and system must be reconstructed by another program language.

  • Proper orthogonal decomposition(POD) is a method for extracting bases for modal decomposition from the ensemble of signals. We verified the connection of the proper orthogonal modes(POMs) and the linear normal modes(LNMs) through MATLAB simulation for the simple cantilever and AFM microcantilever models. Using the POMs, we can analyze and model effectively the dynamic mode of AFM microcantievers.
