Mannan-binding lectin of the sea cucumbers Stichopus japonicus has common antigenic determinants with human serum mannan-binding lectin

  • The host defense system or immune system of all modern animals has their roots in very ancient organisms. After analyzing literature data concerning properties of invertebrates and vertebrates lectins we suggest that mechanism of mannans recognition may exist in marine invertebrates, as a universal mechanism for homeostasis maintenance and host defense, and mannan-binding lectins family of vertebrates has ancient precursor, as was shown for another S-type lectins family. We carried out the screening of mannan-binding type lectin among different species of echinoderms inhabiting in Piter the Grate Bay, the sea of Japan. As a result, the C-type lectins (SJL-32) specific for high mannose glycans was isolated from the coelomic plasma of the sea cucumbers Stichopus japonicus by ion-exchange chromatography on a DEAE-Toyopearl 650M, affinity chromatography on a mannan-Sepharose 6B and gel filtration on a Sephacryl S-200. SJL-32 is homodimer with molecular mass about 32 kDa on SDS-PAGE under non-reducing conditions. Protein part of the lectin has high conteins Asn, Glu, Ser. Hemagglutination of trypsin-treated O blood group human erythrocytes by SJL-32 was competitively inhibited by high-branched -D-mannan composed of -1,2 and -1,6 linked D-mannopyranose residues. In contrast, a variety of mono-, oligo-, and polysaccharides composed of residues of galactose and fucose showed absence or little inhibitory activities. The lectin activity strong depends on Ca2+ concentration, temperature and pH. Monospecific polyclonal antibodies were obtained to the lectin. As was shown by ELISA assay, antibodies to SJL-32 cross-reacted with human serum mannan-binding lectin. This data allows making conclusion about common antigenic determinants and structural homology of both lectins. In our opinion, SJL-32 belongs to evolutionary high conservative mannan-binding lectins (MBLs) family and takes part in the host defense against pathogenic microorganisms.

Porphyromonas gingivalis biofilm에 대한 면역혈청의 침투력에 대한 Fusobacterium nucleatum의 조절효과 (Fusobacterium nucleatum modulates serum binding to Porphyromonas gingivalis biofilm)

  • P. gingivalis를 단독면역하거나 또는 Fusobacterium nucleatum 선면역 후 P. gingivalis 항혈청을 각각 얻어냈다. 두 종류의 항혈청이 P. gingivalis biofilm을 침투해 들어가는 능력을 confocal laser scanning microscope를 이용하여 비교 감증하였다. 항혈청의 P. gingivalis에 대한 avidity index도 측정하였다. 결과적으로 F. nucleatum의 선면역은 P. gingivalis 특이 항혈청에 대해 세균성 biofilm의 침투능력을 저하시키고, 동일한 세균에 대한 avidity도 감소시켰다.

Functional properties of an alternative, tissue-specific promoter for rice NADPH-dependent dihydroflavonol reductase

  • A deletion analysis of the Oryza sativa dihydroflavonol reductase (DFR) promoter defined a 25 bp region (-386 to -362) sufficient to confer pericarp-specific expression of ${\beta}$ -glucuronidase(GUS) reporter gene in transgenic rice. Site-specific mutagenesis of these conserved sequences and subsequent expression analysis in calli which transiently expressed the mutated promoter::GUS gene showed that both bHLH (-386 to -381) and Myb (-368 to -362) binding sites in the DEL3 (-440 to 70) promoter were necessary for complete expression of the GUS gene including the tissue-specific expression of DFR::GUS gene. The GUS gene was expressed well in the mutated Myb (-368 to -362) binding site, but not as strong as in normal condition, implying that the Myb is also necessary to express GUS gene fully. Also, we found the non-epistatic relation between Rc and DFR. There were no changes of expression patterns GUS under the Rc and rc genotypes. Thus, DFR expression might be independent of the presence of functional Rc gene and suggested that Rc and Rd (DFR) share the same pathway controlling the regulation of flavonoid synthesis but not a direct positive transcriptional regulator of DFR gene.

  • This study was performed to comprehend the plasma proteins expressed specifically during early pregnancy in pregnant or non-pregnant Hanwoo using proteomic analysis technique. Plasma samples (0, 2, 3, 4, 7, and 11 weeks after AI) were obtained from pregnant (P, n=3) or non-pregnant (NP, n=4) Hanwoo, respectively. To evaluate proteins differentially expressed, 2-dimensional electrophoresis (2DE) was conducted. Normalized protein spots were selected for the significant expression variation deviated over two fold in its expression level between two groups; Molecular functions of the proteins were DNA binding, protein binding, hemoglobin binding, ferrochelatase and transporter activity and arylestera, respectively. According to western blotting, haptoglobin was specifically expressed only in NP group during early pregnancy; however, paraoxonase 1 was highly expressed in pregnant group. Based on these results, pregnancy was maintained successfully by the activation of specific plasma proteins associated with immune system and antioxidant regulation during early pregnancy in Hanwoo.

MicroRNA Target Recognition: Insights from Transcriptome-Wide Non-Canonical Interactions

  • MicroRNAs (miRNAs) are small non-coding RNAs (~22 nucleotides) regulating gene expression at the post-transcriptional level. By directing the RNA-induced silencing complex (RISC) to bind specific target mRNAs, miRNA can repress target genes and affect various biological phenotypes. Functional miRNA target recognition is known to majorly attribute specificity to consecutive pairing with seed region (position 2-8) of miRNA. Recent advances in a transcriptome-wide method of mapping miRNA binding sites (Ago HITS-CLIP) elucidated that a large portion of miRNA-target interactions in vivo are mediated not only through the canonical "seed sites" but also via non-canonical sites (~15-80%), setting the stage to expand and determine their properties. Here we focus on recent findings from transcriptome-wide non-canonical miRNA-target interactions, specifically regarding "nucleation bulges" and "seed-like motifs". We also discuss insights from Ago HITS-CLIP data alongside structural and biochemical studies, which highlight putative mechanisms of miRNA target recognition, and the biological significance of these non-canonical sites mediating marginal repression.

  • E.coli에서 발견된 ATP-dependent 효소인 Clp효소 중에서 Clp A의 ATPase 활성에 대한 영향을 검토하였다. Clp효소의 limiting amount으로 나타난 specific 활성은 일정하게 증가하는 효소의존성을 보였다. ATPase 활성을 나타내고 있는 ClP A는 casein에 의하여 활성화되어지며 2분자의 ATP가 결합하고 ATPase 활성을 나타내기 위한 ATP의 분해는 Clp효소의 단백질 분해 활성에 필요하다.

Heterogeneous Sequences of Brain Cytoplasmic 200 RNA Formed by Multiple Adenine Nucleotide Insertions

  • Brain cytoplasmic 200 RNA (BC200 RNA), originally identified as a neuron-specific non-coding RNA, is also observed in various cancer cells that originate from non-neural cells. Studies have revealed diverse functions of BC200 RNA in cancer cells. Accordingly, we hypothesized that BC200 RNA might be modified in cancer cells to generate cancerous BC200 RNA responsible for its cancer-specific functions. Here, we report that BC200 RNA sequences are highly heterogeneous in cancer cells by virtue of multiple adenine nucleotide insertions in the internal A-rich region. The insertion of adenine nucleotides enhances BC200 RNA-mediated translation inhibition, possibly by increasing the binding affinity of BC200 RNA for eIF4A (eukaryotic translation initiation factor 4A).

파워 유형과 파트너십 연계를 통한 공급사슬관리 개선방안 모색 (Searching for the SCM Improvement Directions through the Power Attribute and Partnership)

  • SCM 성과 극대화를 달성하기 위한 전제조건으로 파트너십 관계의 구축 및 유지가 중요하다. 기업 간 SCM 성과 제고를 위한 지속적인 노력에도 불구하고, 기업 간 힘의 불균형에 따라 발생할 수 있는 비효율적인 낭비 요소가 만연해 있다. 이러한 협업의 걸림돌이 되는 요소가 파워(Power)의 한 속성이다. 따라서 본 연구의 목적은 파워유형의 속성을 파악하고 파트너십 향상을 통해 공급사슬관리의 개선방안을 모색하고자 하였다. 선행연구를 통해 파워의 원천을 크게 구속적 파워와 비구속적 파워로 분류하여, 구속적 파워의 종류로는 행동 강제성, 구속적 보상, 관계 합법성으로 구분하였으며, 비구속적 파워의 종류로는 업무 전문성, 정보 우위성, 가치 준거성으로 구분하여 살펴보고자 한다. 따라서 파워의 유형을 살펴보고 파트너십 주요 변인인 신뢰 및 몰입에 미치는 영향과 협업 및 SCM 성과로 이어지는 인과관계를 밝혀 보았으며 구체적인 연구 결과는 다음과 같다. 첫째, 구속적 파워는 신뢰에 유의한 영향을 미치지 못하였다. 그러나 구속적 파워가 몰입에는 정(+)의 유의한 영향을 미치는 것으로 나타났다. 둘째, 비구속적 파워는 신뢰와 몰입 모두에 유의한 영향을 보였다. 총 효과 분석결과 역시 비구속적 파워는 협업 및 SCM 성과에 간접영향을 미치는 것으로 나타났다. 셋째, 신뢰와 몰입은 모두 협업에 유의한 영향을 미치는 것으로 나타났다. 조직 간 관계이론 중 사회교환이론 및 거래비용이론 측면에서 살펴본 신뢰, 몰입, 협업의 SCM 성과로 이어질 수 있으며 더불어 파워의 속성별 연계가 유의미한 결과로 이어지는 것을 확인할 수 있었다. 넷째, 신뢰와 협업은 SCM 성과에 유의한 영향을 미치는 것으로 나타났다. 그러나 몰입은 직접적으로 SCM 성과에는 영향을 미치지 못하였다. 다만 협업을 통한 SCM 성과에는 간접적으로 유의한 영향을 미치는 것으로 나타났다. 기업들은 파워의 원천, 불균형, 결과들에 대한 이해를 포함한 공급사슬 내 파트너기업에 대한 충분한 이해와 인식을 하여야 하는데, 이러한 파워의 적절한 사용은 공급사슬 구성원 간 협력관계를 공고히 구축하고 이에 따라 공급사슬 성과의 향상과 구성원 만족을 유도할 수 있을 것이다.

Assembly of Biomimetic Peptoid Polymers

  • The design and synthesis of protein-like polymers is a fundamental challenge in materials science. A biomimetic approach is to explore the impact of monomer sequence on non-natural polymer structure and function. We present the aqueous self-assembly of two peptoid polymers into extremely thin two-dimensional (2D) crystalline sheets directed by periodic amphiphilicity, electrostatic recognition and aromatic interactions. Peptoids are sequence-specific, oligo-N-substituted glycine polymers designed to mimic the structure and functionality of proteins. Mixing a 1:1 ratio of two oppositely charged peptoid 36 mers of a specific sequence in aqueous solution results in the formation of giant, free-floating sheets with only 2.7 nm thickness. Direct visualization of aligned individual peptoid chains in the sheet structure was achieved using aberration-corrected transmission electron microscopy. Specific binding of a protein to ligand-functionalized sheets was also demonstrated. The synthetic flexibility and biocompatibility of peptoids provide a flexible and robust platform for integrating functionality into defined 2D nanostructures. In the later part of my talk, we describe the use of metal ions to construct two-dimensional hybrid films that have the ability to self-heal. Incubation of biomimetic peptoid polymers with specific divalent metal ions results in the spontaneous formation of uniform multilayers at the air-water interface. We anticipate that ease of synthesis and transfer of these two-dimensional materials may have many potential applications in catalysis, gas storage and sensing, optics, nanomaterial synthesis, and environmentally responsive scaffolds.

Structure biology of selective autophagy receptors

  • Autophagy is a process tightly regulated by various autophagy-related proteins. It is generally classified into non-selective and selective autophagy. Whereas non-selective autophagy is triggered when the cell is under starvation, selective autophagy is involved in eliminating dysfunctional organelles, misfolded and/or ubiquitylated proteins, and intracellular pathogens. These components are recognized by autophagy receptors and delivered to phagophores. Several selective autophagy receptors have been identified and characterized. They usually have some common domains, such as motif, a specific cargo interacting (ubiquitin-dependent or ubiquitin-independent) domain. Recently, structural data of these autophagy receptors has been described, which provides an insight of their function in the selective autophagic process. In this review, we summarize the most up-to-date findings about the structure-function of autophagy receptors that regulates selective autophagy.
