The Viability Change of Yeast Cell Responding to Trehalose Accumulation and Maintaining Neutral Trehalase Activity under Extracellular pH Acidified by $H_2SO_4$

  • Saccharomyces cerevisiae KNU5377 (KNU5377) and S. cerevisiae ATCC24858 (ATCC24858) were exposed to $H_2SO_4$ as a stress, which was added at various concentrations to a YPD media. The growth of KNU5377 was reduced to approximately 60% in the YPD media containing 40 nm sulfuric acid when compared to the non-stressed condition. When their growth was monitored during an overnight culture, two strains, KNU5377 and ATCC24858, could not grow when exposed to over 50 mM of sulfuric acid. After a short exposure to this acid for 1 h, KNU5377 exhibited stronger resistance against $H_2SO_4$ than ATCC24858. The neutral trehalase activity of KNU5377 unchanged despite under various concentrations of $H_2SO_4$. In contrast, It at of ATCC24858 was much low at higher $H_2SO_4$concentrations. Trehalose, a non-reducing disaccharide, was maximally accumulated after a short exposure to 60 nm $H_2SO_4$ for KNU5377, but it was reduced under more severe stressful conditions. These results suggest that KNU5377 should modulate the trehalose concentrations under the severe stress condition of high sulfuric acid concentrations. The most highly induced protein in the KNU5377 exposed to sulfuric acid was found to be an approximately 23 kDa protein, which was revealed to be the 605 large subunit ribosomal protein, Ll3 by FASTA search results.

Transcription Profiles of Human Cells in Response to Sodium Arsenite Exposure

  • Arsenic exposure is associated with several human diseases, including cancers, atherosclerosis, hypertension, and cerebrovascular diseases. In cultured cells, arsenite, an inorganic arsenic com-pound, was demonstrated to interfere with many physiological functions, such as enhancement of oxidative stress, delay of cell cycle progression, and induction of structural and numerical changes of chromosomes. The objective of this study is to investigate the effects of arsenic exposure on gene expression profiles by colorimetric cDNA microarray technique. HFW (normal human diploid skin fibroblasts), CL3 (human lung adenocarcinoma cell line), and HaCaT (immortalized human keratinocyte cell line) were treated with 5 $\mu\textrm{M}$ or 10 $\mu\textrm{M}$ sodium arsenite for 6 or 16 h, respectively. By a dual-color detection system, the expression profile of arsenite-treated cultures was compared to that of control cultures. Several genes expressed differentially were identified on the microarray membranes. For example, MDM2, SWI/SNF, ubiquitin specific protease 4, MAP3K11, RecQ protein-like 5, and Ribosomal protein Ll0a were consistently induced in all three cell types by arsenite, whereas prohibitin, cyclin D1, nucleolar protein 1, PCNA, Nm23, and immediate early protein (ETR101) were apparently inhibited. The present results suggest that arsenite insults altered the expression of several genes participating in cellular responses to DNA damage, stress, transcription, and cell cycle arrest.

  • Colorants in green tea were extracted freeze-dried and analyzed to investigate the possibility of using as a natural dye. Fractionation of the colorants was carried out by column chromatograpy. Colorants in green tea were eluted into five fractions. All the fractions except fraction F2 showed absorption peakat 280nm. Fraction f2 showed absorption peak at 270nm and broad peak at 350nm, From the IR analysis it is speculated that fractions F2-F5 having similar stucture but different molecular weight are catechis. Silk fabrics dyes with fractions F1-F4 showed yellowish red color while sample dyed with fraction F5 showed red color. The colorants from green tea infusion was applied to silk wool nylon cotton and rayon fabrices. It showed relatively good affinity to protein and polyamide fibers bur low affinity to cellulose and regenerated cellulose fibers.

NMR Studies on N-terminal Domain of DNA2

  • Saccharomyces cerevisiae Dna2 protein has biochemical activities: DNA-dependent ATPase, DNA helicase and DNA nuclease and is essential for cell viability. Especially, Pro$\^$504/ is determined as an important residue in ATPase, helicase, and nuclease activity. We synthesized and determined the three-dimensional solution structure of N-terminal domain comprising residues of Val$\^$501/ -_Phe$\^$508/ (Dna2$\^$pep/) using two-dimensional $^1$H-NMR and dynamical simulated annealing calculations. On the basis of a total of 44 experimental restraints including NOEs, $^3$J$\_$$\alpha$$\beta$/ and $^3$J$\_$$\alpha$$\beta$/ coupling constants, the solution structures of Dna2$\^$epe/ were calculated with the program CNS. The 23 lowest energy structures were selected out of 50 final simulated-annealing structures. The atomic RMSDs of the final 23 structures fur the individual residues were calculated with respect to the average structure. The mean RMSDs for the 23 structures were 0.042 nm for backbone atoms and 0.316 nm for all heavy atoms, respectively. The Ramachandran plot indicates that the $\Phi$, Ψ angles of the 23 final structures are properly distributed in energetically acceptable regions. Solution structure of Dna2$\^$pep/ showed a single unique turn spanning residues of Asn$\^$503/ Val$\^$506/.

Effect of Ethanol/water Solvent Ratios on the Morphology of Zein Nanofiber Mats and their Wettability

  • Zein is a hydrophobic protein produced from maize and has great potential in a number of industrial applications, such as food, food coating and food packaging. To obtain suitable electrospinning conditions for thinner and uniform zein nanofiber mats, a series of experiments was conducted on various volume ratios (v/v) of ethanol/water solutions with different zein concentrations. The prepared zein nanofiber mats were characterized by field emission scanning electron microscopy and contact angle measurements. Uniform zein fibers with a average diameter in the nanometer scale (300~500 nm) could be prepared from 30 wt.% zein in 7/3 (v/v) ethanol/water solutions.

볶음처리에 의한 인삼박의 이화학적 성분변화 (Changes in Physicochemical Components of Ginseng Marc by Roasting Process)

  • This study was carried out in order to utilize ginseng marc (GM), a byproduct produced during the preparation of ginseng extract. For the utilization of GM as coffee-replacing beverage, GM was roasted at 140-23$0^{\circ}C$, for 10-30 min to generate coffee-like flavor and taste. The physicochemical changes in GM induced by roasting process was measured. Weight and moisture contents of GM abruptly decreased while roasting at various temperature and period of time, and the amount of water-extracted solid substances decorated beyond 23$0^{\circ}C$. Among the proximate components, total and free sugar decreased remarkably, while crude fat content increased. Crude protein content did not change by the roasting process. pH derides to pH 4.9 after 30 min at 20$0^{\circ}C$, but increased at higher temperatures. Hunter color L value of the GM powder decreased, whereas a and b values, increased up to 10 times as compared to that of the control. The brown color(O, D 490 nm) in 50% ethanol extract of the roasted GM was 46 times higher than that of the control.

들깨 유전자원의 단백질함량과 아미노산조성 (Variation of Protein Content and Amino Acid Composition in Perilla Germplasm)

  • 들깨 장려품종과 지방수집재래종 도입종 등을 대상으로 단백질 함량과 아미노산 조성을 분석 조사하여 고단백 양질 다수성 품종육성에 필요한 기초자료를 얻고자 시험을 실시하였던 바 그 결과를 요약하면 다음과 같다. 1. 들깨 유전자원의 단백질 함량은 최저 17.9%에서 최고 28.1%까지 10.2%의 변이폭을 보여 주었고 평균 함량은 23.6%였으며 대부분 21-25% 범위에 분포하였다. 본 연구 결과 단백질 함량이 높은 남지종, 산동종, 은진종 등 7품종을 선발하였다. 2. 단백질 함량은 유래별로는 전남지역에서 수집된 품종이 가장 높았고 숙기에 따라서는 차이가 없었으며, 종파색은 은회색이, 1,000립중은 무거운 대립종에서 높은 경향이었다. 3. 단백질 함량과 임실율은 유의적인 부의 상관을 보였고 단백질 함량과 기름함량은 유의적인 정의 상관을 보였으며 단백질 함량과 1,000 립중간에는 정의 상관을 보였으나 유의성은 인정되지 않았다. 4. Micro-kjeldahl법과 비교한 NIR법의 단백질 calibration 정밀도는 0.27%였다. 5. 들깨 95품종을 대상으로 micro-kjeldahl법과 NIR 분석법에 의한 단백질 함량 차이는 유의성이 보이지 않았으며 표준편차 차이는 0.34%를 보여 주었고, 분석방법간에 고도의 정상관이 인정되었다. 6. 엽실들깨등 4품종의 아미노산 조성은 FAO권장량보다 높아 그 우수성이 인정되었으며 필수아미노산에서 총량은 품종간에 차이가 크지 않았으나 lysine과 methionine은 다소 차이가 있었다. 7. 들깨의 주요 아미노산은 glutamic acid와 arginine아었다.

하천수 BOD 예측을 위한 용존 자연유기물질의 synchronous 형광 스펙트럼 분석 (Examining Synchronous Fluorescence Spectra of Dissolved Organic Matter for River BOD Prediction)

  • Fluorescence measurements of dissolved organic matter (DOM) have the superior advantages over other analysis tools for the applications to water quality management due to their rapid analysis. It is known that protein-like fluorescence characteristics are well corelated with microbial activities and biodegradable organic matter. In this study, potential biochemical oxygen demand (BOD) predictor were explored using the fluorescence peak intensities and/or the integrated fluorescence intensities derived from synchronous fluorescence spectra and the first derivative spectra of river samples. A preliminary study was conducted using a mixture of a river and a treated sewage to test the feasibility of the approach. It was demonstrated that the better BOD predictor can be derived from synchronous fluorescence spectra and the derivatives when the difference between the emission and the excitation wavelengths (${\Delta}{\gamma}$) was large. The efficacy of several selected fluorescence parameters was rivers in Seoul. The fluorescence parameters exhibited relatively good correlation coefficients with the BOD values, ranging from 0.59 to 0.90. Two parameters were suggested to be the optimum BOD predictors, which were a fluorescence peak at a wavelength of 283 nm from the synchronous spectrum at the ${\Delta}{\gamma}$ value of 75 nm, and the integrated fluorescence intensity of the first derivatives of the spectra at the wavelength range between 245 nm and 280 nm. Each BOD predictor showed the correlation coefficients of 0.89 and 0.90, respectively. It is expected that the results of this study will provide important information to develop a real-time efficient sensor for river BOD in the future.

서해안 양식장에서 분리한 Vibrio alginolyticus의 특이 bacteriophage에 대한 구조적 특성 (Morphological characterization of Vibrio alginolyticus specific bacteriophage isolated from fish farms on west coast of Korea)

  • Vibrio alginolyticus (V. alginolyticus) 는 우리나라 전 연안에서 높은 빈도로 발견되며 인간 및 어패류에 감염을 유발시키는 박테리아의 일종이다. 본 연구는 서해안 양식장의 해수와 어패류에서 V. alginolyticus와 이에 대한 특이적인 용균성 파아지를 분리하였으며 분리된 파아지의 형태학적 특성을 전자현미경으로 확인 하였다. 또한 파아지의 핵산의 종류 및 구조 단백질의 특성 대한 연구가 수행되었다. 분리된 파아지는 형태학적으로 60 nm의 육각형 두부와 20 nm의 짧은 미부를 가진 podoviridae과로 분류되었다. 핵산을 분리한 결과 23 Kb 크기의 DNA로 판명 되었으며 구조 단백질은 37.8 kDa과 198 kDa 사이에 7종류의 단백질 분획이 존재함을 확인 하였다.

청국장 점질물의 이화학적 특성 (Physico-Chemical Properties of Viscous Substance Extracted from Chungkook-jang)

  • 국내에 아직 연구보고된 바 없는 청국장 점질물의 이화학적 특성을 조사하기 위하여 B. natto와 B. subtilis로 발효시킨 청국장으로부터 각각 6.3%, 5.0%의 점질물을 얻었다. 이 점질물의 조단백질 함량은 모두 약 61% 정도이었으며, 점질물의 단백질 중 아미노태 질소함량은 약 32%이었고, 구성아미노산 중에는 글루탐산이, 무기질 중에는 K함량이 특히 높았다. 점질물의 점도는 5% 농도에서는 설탕 15% 용액의 점도보다도 높게 나타났다. B. natto와 B. subtilis로부터 분리한 점질물은 pH5.0과 7.0, pH5.0에서 각각 최저 투과도를 나타내었고, 280nm에서 측정한 흡광도는 각각 pH 5.5, pH 4.0과 10.5에서 최저흡광도를 나타내었다. 점질물의 fibrinolytic activity는 B. natto가 0.438 unit/mg protein, B. subtilis가 0.163 unit/mg protein이었고, $100^{\circ}C$에서 5분간 열처리하여도 효소활성이 90% 정도 유지되었으며, 30분간 열처리하였을 때도 45% 정도의 효소활성이 남아 있었다.
