Variations in carbon and nitrogen stable isotopes and in heavy metal contents of mariculture kelp Undaria pinnatifida in Gijang, southeastern Korea

  • Korean mariculture Undaria pinnatifida was collected during the months of January, February, March, and December of 2010, as well as from January of 2011 to investigate the changes in the carbon and nitrogen stable isotope ratios (${\delta}^{13}C$ and ${\delta}^{15}N$) and heavy metal with respect to it growth and to identify the factors that influence such changes. The blades of U. pinnatifida showed ${\delta}^{13}C$ and ${\delta}^{15}N$ in the range (mean) of -13.11 to -19.42‰ (-16.93‰) and 2.99 to 7.57‰ (4.71‰), respectively. Among samples with the same grow-out period, those that weighed more tended to have higher ${\delta}^{13}C$ suggesting a close association between the carbon isotope ratio and growth rate of U. pinnatifida. Indeed, we found a very high positive linear correlation between the monthly average ${\delta}^{13}C$ and the absolute growth rate in weight ($r^2=0.89$). Nitrogen isotope ratio tended to be relatively lower when nitrogen content in the blade was higher, probably due to the strengthening of isotope fractionation stemming from plenty of nitrogen in the surrounding environment. In fact, a negative linear correlation was observed with the nitrate concentration in the nearby seawaters ($r^2=0.83$). Concentrations of Cu, Cd, Pb, Cr, Hg, and Fe in the blades showed a rapid decrease in their concentration per unit weight in the more mature U. pinnatifida. Specifically, compared to adult samples, Cu, Hg, and Pb were concentrated by 30, 55, and 73 folds, respectively, in the young blades. Therefore, U. pinnatifida tissue ${\delta}^{13}C$ is as an indirect indicator of its growth rate, while ${\delta}^{15}N$ values and heavy metal concentrations serve as tracers that reflect the environmental characteristics.

Operation Mode in Sequencing Batch Reactor for Nitrogen Removal (질소제거를 위한 연속회분식 반응조의 운전방식 연구)

  • This research investigated the effect of COD/N ratio on nitrogen removal, and the use of organics in raw wastewater as a carbon source for denitrification in SBR(Sequencing Batch Reactor) systems. Four laboratory scale reactors were operated in three modes. Only the difference between modes were; Mode I operated in aerated condition during fill while Mode II in anoxic condition and Mode III operated on two fills per cycle in anoxic condition. When COD/N ratio increased, total nitrogen removal efficiencies increased from 8.7 to 57.7 percent in Mode I, from 28.9 to 83.2 percent in Mode II and from 42.7 to 97.8 percent in Mode III, respectively. COD removal efficiencies ranged from 93 to 98 percent throughout the study. SBR operation in Mode III of feeding twice per cycle in anoxic condition was an effective operating method for nitrogen removal and nitrogen concentration in effluent can be estimated using influent COD and nitrogen concentrations.

  • In this study, inter-laboratory comparison was done using elemental analyzer-isotope ratio mass spectrometers (EA-IRMSs) to determine carbon and nitrogen contents as well as stable carbon and nitrogen isotopic compositions (${\delta}^{13}C$ and ${\delta}^{15}N$) of five environmental samples containing lake and marine sediments, higher plant leaves, and fish muscle, and one organic analytical standard (Protein (Casein) Standard OAS). Five national laboratories participated in this comparison study, and each laboratory analyzed all five samples and the analytical standard. Results showed that variations in total organic carbon (TOC) and total nitrogen (TN) contents as well as ${\delta}^{13}C_{TOC}$ and ${\delta}^{15}N_{TN}$ values among the laboratories were large compared to the analytical uncertainties. The results highlighted the inhomogeneity of the test samples and thus, the need to select suitable standard reference materials for future inter-laboratory studies. Further inter-laboratory comparison exercises could promote good measurement practices in the acquisition of stable carbon and nitrogen isotopic composition data.

Feasibility Study of Applying EMMC Process to Recirculation Water Treatment System in High Density Seawater Aquaculture Farm through Laboratory Scale Reactor Operation (실험실규모 반응조 운전을 통한 고밀도 해산어 양식장 순환수 처리공정으로서 EMMC공정의 적용 가능성 연구)

  • Treatability tests were conducted to study the feasibility of EMMC process as a recycling-water treatment system in high density seawater aquaculture farm. To study the effect of organic and ammonia nitrogen loading rate on system performance, hydraulic retention time was reduced gradually from 12hr to 10min. The conclusions are can be summarized as follows. When the system HRT was reduced from 12hr to 2hr gradually, there was little noticeable change(reduction) in ammonia nitrogen removal efficiencies. However, removal efficiencies were decreased dramatically when the system was operated under the HRT of less than 2hr. In case of organics(COD), there was no dramatic change in removal efficiencies depending on HRT reduction. COD removal efficiencies were maintained successfully higher than 9% when the system was operated at tile HRT of 10 min. System performances depending on media packing ratio in the reactors were also evaluated. There were little differences in each reactor performances depending on media packing ratio in reactor when the reactors were operated under the HRT of longer than 1hr. However, differences in reactor performances were considerably evident when the reactors were operated under the HRT of shorter than 1hr. When comparing reactor performance among 25%, 50%,7 5% packed reactor, it can be judged that media packing ratio more than 50% plays no significant role in increasing reactor performance. For this reason, packing the media less than 50% is more reasonable way in view of economic. Such a tendency shown in COD removal efficiencies well agreed with the variation of ammonia-nitrogen removal efficiencies according to the media packing ratio in reactors at each HRT. Difference in effluent ammonia-nitrogen concentration between 50% media packing reactor and 75% media packing reactor was negligible. When comparing with the results of 25% packing reactor, difference was not so great.

  • A study was carried out to select a nitrogen source and the optimize the C/N ratio for the maximum cell growth of Phaffia rhodozyma in fed-batch culture. The yeast extract was the best organic nitrogen source. In the batch culture experiments, the highest cell yield was obtained 0.575 g-cell/g-glucose from 10 g/L and 10 g/L yeast extract. In the fed-batch experiments, the maximum cell concentration was obtained 33.1 g/L from the C/N ratio of 2:1 while the astaxanthin concentration of cell was Increased by increasing the C/N ratio, of feed medium.

  • We try to distribute air pollution sources at target area. The complex method is used to distribute air pollution sources. Most of people, However, want to simple indicator as standard for express air pollution source. In many area, air pollution is caused by different types of sources. The general type is point source, such as tall stack of power plants and oil refineries stacks. A second type is areal source, such as local industry and transportation. In this aspect, the ratio of sulfur dioxide to nitrogen oxides (RSN =SO$_2$/NOx)is an indicator of air pollution source. the role of this ratio is to show the characteristics at target area of the relationship the point and the areal source.   • These studies were conducted to investigate the changes of starch, sugar and nicotine content in relation to maturity and cultural conditions and correlation between growth characteristics and chemical components in order to improve the leaf quality. The results were summarized as follows; 1. Nicotine content, total nitrogen content and the ratio of total nitrogen to nicotine of the green leaf were increased with heavier fertilizer, wider planting space and lower topping, whereas starch, total sugar and the ratio of sugar to nicotine were decreased. 2. Starch content, sugar content and the ratio of total sugar to nicotine(T-S/Nic) in the cured leaf were decreased with milder planting space, less application of MH and delaying harvesting date. 3. It was estimated that 58.1% of starch of the harvested leaf was converted into sugar of the cured leaf, and also they were significantly correlated each other.

  • The Anaerbic Anoxic/oxic process is one of the biological treatment methods to remove nitrogen and phosphorus effectively which are nutritional elements for eutrophication. Supernatant of primary sediment of Anaerobic digester is used as a carbon source instead of methanol methanol supply in usual A$_{2}$/O process. The efficiency of the following treatment processes are as follow : 1) Changing recycle ratio in the usual A$_{2}$/O process without the stage of Anaerobic digester. 2) Changing recycle ratio in the usual A$_{2}$/O process with the supernatant supply of the Anaerobic digester. In the result of comparison, changing recycle ratio is almost no effect in the removal of phosphorus, however the effect of removal in nitrogenous substance are remarkable, and the effect of Anaerobic digester is not as effective as expected because the BOD removed in the digester partly, the rate of phosphorus to the BOD exceed pertinent range.

  • The pullulan production and morphological change of Aureobasidium pullulans ATCC 9348 were investigated both in batch fermentation and in continuous fermentation. The best carbon source for pullulan production was sucrose among seven different carbon sources. The pullulan production of A. pullulans was increased with increasing the carbon to nitrogen ratio of the medium using sucrose as a carbon source. In batch fermentation, production of pullullan occurred following exhaustion of the nitrogen source from the medium. The continuous fermentation showed that the pullulan production was closely parallelled with cell growth and was most effective at a dilution rate of 0.06~0.07 hr$^{-1}$-. The ratio of yeast-like cells(blastospores) of A. pullulans increased with the increase of growth rate, and reached 100% over the growth rate of 0.07 hr$^{-1}$. The growth rate, within a certain range, affected not only on the cell morphology, but on the specific pullulan productivity of A. pullulans.

  • It has been acknowledged that fertilizer and animal waste have different ratio of \ulcornerN/\ulcornerN which is presented as a symbol of $\delta$\ulcornerN, and that the values of $\delta$\ulcornerN for fertilizer and animal waste are placed less than +8 $\textperthousand$ and higher than +10$\textperthousand$, respectively. By this reason, the demarcation range for the source nitrogen mass ratio is between +8$\textperthousand$ and +10$\textperthousand$. In this study, we would suggest a possible method to trace the major pollution basis using the nitrogen isotope analysis method for management of water quality.
