Screening of the Physiological Activity of Solvent Extracts of Paulownia coreana Bark and Antioxidative Effect of the Extracts on an Edible Oil

  • Paulownia coreana is a medicinal, edible and industrial plant with the largest leaf, and is native to Korea. We evaluated the reducing power activities, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities (RSAs), nitrite scavenging activities (NSAs) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation RSAs by solvent extraction of P. coreana bark by using $50^{\circ}C$ hot water and $25^{\circ}C$ methanol. The antioxidative effect of P. coreana bark extract on corn seed oil was evaluated using the Rancimat test. The extraction yields on dry weight basis with 15 folds of hot water and methanol were 23.88% and 5.30%, respectively; further the flavonoid content in the hot water extract was over 2.5 times more than that in the methanol extract. The DPPH RSA of the methanol extract was substantially higher than that of the water extract, whereas the NSA of the water extract was higher than that of the methanol extract at pH 1.2. The ABTS RSAs of the two extracts were almost the same as that of ascorbic acid and 2,6-ditertiarybutyl hydroxytoluene. The two extracts of P. coreana bark in this study were found to slightly improve the oxidation stability of corn seed oil.

한국산 삽주의 기능성 (Functional Properties of Korean Atractylodes japonica Koidz)

  • 생삽주의 기능성을 분석한 결과, 추출 수율은 ethanol 추출물에 14.8%와 water 분획물에 17.7%를 나타내었다. Acetone 추출물과 butanol 분획물의 수소공여 능은 각각 72.9%와 74.2%로 높게 나타났으며, methanol 추출물과 butanol 분획물의 아질산염 소거 효과는 각각 95.0%와 79.2%로 가장 높게 나타났다. 생삽주의 용매별 추출물 중 methanol 추출물이 높은 항균 활성을 나타내었는데, Bacillus subtilis에 대해 20 mm clear zone을 나타내었으며, Pseu. aeruginosa에 대해 19 mm clear zone을 나타내었다. 항균 활성이 가장 높은 methanol 추출물을 계통분획하여 항균 활성을 확인한 결과, butanol 분획물이 높은 항균 활성을 나타내었는데, Bac. subtilis, V. parahaemolyticus에 대해 각각 18 mm clear zone을 나타냈다. Methanol 추출물과 butanol 분획물을 이용하여 최소저해농도를 확인한 결과 대체로 gram(+)균에 대해 2 mg/disc와 4 mg/disc에서 나타났구 gram(-)균에 대해서는 6 mg/disc에서 저해활성을 나타내었다.

녹차와 보이차 추출물의 항산화 효과 (Antioxidative Effect on the Green Tea and Puer Tea Extracts)

  • 녹차에 대한 보이차의 항산화력을 알아보기 위해 물 추출물과 메탄올 추출물의 항산화 효과를 비교$\cdot$조사하였다. 총 폴리페놀의 함량은 물 추출물의 경우 녹차와 보이차가 비슷하였으며 메탄올 추출물의 경우는 보이차의 함량이 녹차의 약 $58\%$정도 밖에 되지 않았다. 전자공여능은 물 추출물의 경우 녹차와 보이차가 거의 유사하였으며 메탄올추출물의 경우 보이차가 녹차의 $54\%$정도였다. SOD 유사 활성은 용매간에 많은 차이가 있었는데 물 추출물의 활성은 $13.46\~14.07\%$로 녹차와 보이차간에는 거의 차이가 없었으나 메탄올 추출물의 활성은 녹차가 $59.63\%$, 보이차가 $48.93\%$로 물 추출물보다 $3\~4$배 높았으며 또한 녹차가 활성이 높았다. 아질산염 소거능은 물 추출물의 경우 녹차와 보이차간에 거의 차이가 없었으며 메탄올 추출물은 보이차가 녹차보다 다소 효과가 높았다. ACE 저해 활성은 물 추출물의 경우 보이차가 녹차보다 약 $38\%$ 정도 활성이 높았으나 메탄올 추출물은 반대로 녹차가 보이차보다 활성이 높았다. 이상의 연구 결과 보이차의 물추출물 항산화력은 녹차와 비슷하였으나 오히려 ACE 저해활성만은 높은 값을 나타내었고 메탄올 추출물에서는 보이차는 녹차보다 항산화력이 전반적으로 낮았으나 아질산염 소거능만은 높았다.

Antioxidant and Anti-inflammatory Activities of Ethanol Extract from Leaves of Cirsium japonicum

  • Antioxidant and anti-rheumatoid activities of Cirsium japonicum leaf extract (CJLE) were investigated in this study. CJLE had similar DPPH radical scavenging activity and reducing power to ascorbic acid and several flavonoids. Rheumatoid arthritis (RA) is a chronic inflammatory tissue-destructive disease, partly related with functions of hyaluronidases (HAases) and collgenases. CJLE ($1,000\;{\mu}g/mL$) had approximately 60.7 and 31.9% inhibition of HAase and collagenase activity, respectively. Also, CJLE inhibited lipopolysaccharide (LPS)-induced nitrite production in a dose-dependent manner, and CJLE ($1,000\;{\mu}g/mL$) suppressed approximately 70% of LPS-induced nitrite production effectively in RAW 264.7 macrophage cells. CJLE had inhibitory effects on the adherence of monocytic THP-1 to human umbilical vein endothelial cell (HUVEC) monolayers to the basal level. Inhibitory effect of CJLE on the adhesion was caused by suppression of tumor necrosis factor-a-upregulated expression of vascular cellular adhesion molecule-1 (VCAM-1) and E-selectin. We expect that CJLE may alleviate the inflammatory process in rheumatoid synovium, and these findings will raise the possibility of the usage of C. japonicum as a traditional pharmaceutical of anti-rheumatoid arthritis.

제주 재래식된장의 생리적 특성 (Physiological Properties of Jeju Traditional Doenjang)

  • 곰취(Ligularia fischeri, GC), 고추잎(Capsicum annuum L., GCY), 취나물(Aster scaber, CNM), 머위대(Petasites japonicus S. et Z. Max, MYD) 및 고구마순(Ipomoea batatas L. (Lam), GGM)과 같은 산채들의 물 추출물의 항산화능력을 평가하고 이들 동결건조 블록 물 추출물들의 항산화력과 비교하였다. 산채 물 추출물들과 그들의 동결건조 블록물 추출물들의 항산화력 측정은 2,2-diphenyl-1-picrylhydrazyl(DPPH) radical 소거작용, hydroxyl radical 소거작용 및 아질산염 소거작용과 같은 방법에 의해 알아보았다. 산채 물 추출물은 그들의 동결건조 물 추출물보다 총 페놀함량이 더 높았다. GC, GCY, CNM, MYD 그리고 GGM 물추출물들의 총 페놀 함량은 각각 $471.66{\pm}3.52\;{\mu}g/mg,\;141.33{\pm}2.51\;{\mu}g/mg,\;177.33{\pm}2.88\;{\mu}g/mg,\;238.66{\pm}9.50\;\mu}g/mg\;그리고\;122.67{\pm}3.51\;{\mu}g/mg$이었다. 1000 ppm GC, GCY, CNM 그리고 GGM 물 추출물의 DPPH radical 소거작용은 그들의 동결건조 블록 물 추출물보다 더 높았고, 1000 ppm CNM, GC, GCY, MYD 그리고 GGM의 물 추출물의 DPPH radical 소거작용은 각각 90.9%, 89.9%, 76.6%, 71.1% 그리고 57.4%였다. 10000 ppm GC, GCY, CNM, MYD 그리고 GGM 물 추출물들은 hydroxyl radical 소거작용을 각각 38.8%, 33.4%, 35.9%, 34.3% 그리고 33.8%까지 증가시켰고, GCY, CNM 그리고 GGM의 물 추출물은 동결건조 블록 물 추출물과 유사한 활성을 나타내었으나 GC와 MYD의 물 추출물이이들 동결건조 블록들의 물 추출물의 hydroxyl radical 소거작용보다 약간 더 영향력이 있었다. 산채 물 추출물들과 이들 동결건조 블록 물 추출물들은 실험된 모든 농도에서 DPPH radical 소거작용 및 hydroxyl radical 소거작용을 나타내었다. GC 물 추출물의 아질산염 소거작용은 현저하게 농도 의존적으로 증가하였고, GC 물 추출물의 아질산염 소거작용이 그것의 동결건조 블록 물 추출물의 아질산염 소거작용보다 높았다. 이상의 결과들로부터 동결건조블록이 산채와 비교하여 산채가 가지고 있는 항산화력을 유지하고 있다는 것을 알수 있었다.

붉나무 껍질 에탄올 추출물의 추출특성과 항산화 활성에 관한 연구 (Extraction Characteristics and Antioxidant Activity of Ethanol Extract of Rhus javanica Bark)

  • 붉나무의 유용성에 대한 기초자료를 마련하고자 붉나무 수피를 에탄올로 추출하여 추출물의 몇 가지 이화학적 특성과 항산화 활성을 측정하였다. 추출물의 가용성 고형분의 함량은 73.5 mg/100g, 당도는 $17.8^{\circ}Brix$로 측정되었으며, 총방향족 화합물, 총플라보노이드 함량 및 총페놀성 화합물의 함량은 각각 흡광도 0.508, 49.88 mg/100g 및 296.6 mg/100g으로 측정되었다. 추출물의 환원력은 대조구로서 추출물의 가용성 고형분과 동량의 ascorbic acid의 27.5 %로 측정되었으나, 철환원력은 ascorbic acid와 유사하게 측정되었다. 추출물의 DPPH라디칼 소거능은 95.16 %로서 ascorbic acid와 거의 유사하였고, 금속이온 봉쇄능은 ascorbic acid의 74.73 %보다 높은 81.58 %로 측정되었다. 그리고 pH 2에서 추출물의 아질산염 소거능은 51.76 %로 측정되었으며, Rancimat test에 의한 대두유 산화 억제효과가 있음을 확인하였다.

토사자(Cuscuta japonica Choisy, 菟絲子)의 화장품 약리활성에 관한 연구 (Study of the Cosmeceutical Activities of Cuscuta japonica Choisy)

  • Objectives : The cosmeceutical activity of Cuacuta japonica Choisy were investigated for cosmetics industrial application. Methods : Cosmeceutical activities include antioxidant, tyrosinase inhibition effects and nitrite-scavenging ability were examined through the Cuacuta japonica Choisy Results : The result were obtained as follows : The electron donating ability of water and ethanol extract was 69.5%, 78.3% at 1,000ppm. In SOD-like activity, water extract showed low effect, but ethanol extract scored 63.4% at 1,000ppm. In inhibition activity of xanthine oxidase, water extract showed low effect, which is 27.2% at 500ppm, in contrast to ethanol extract high effect: 99.5% at 500ppm, in nitrite-scavenging ability test, 500ppm of C. japonica Choisy water extract showed the effect of 51.3%, while 500ppm of C. japonica Choisy ethanol extract showed the effect of 72.7%. From the test of inhibitory effect of tyrosinase, both water and ethanol extract showed insufficient inhibitory rate. Conclusion : According to all the test results that have been given, it is possible that the extract of C. japonica Choisy can be used as a new natural material of the cosmeceutical industry.

천연 색소 고정화 기술을 활용한 오미자와 자소엽 추출물 함유 음료의 항산화 활성 연구 (Antioxidative Activities of Mixture of Schisandrae Fructus (SF) and Perilae Folium (PF) using Natural Color fixation technic)

  • Objectives : This study focused on Schisandrae Fructus (SF) and Perilae Folium (PF), traditional medicine herbs and health functional food in Korea, Japan and China. We investigated various pharmacological activities that include a potential source of free radical scavenging, anti-viral, anti-microbial, anti-allergy and anti-inflammatory activities. Methods : We conducted an investigation of total contents of phenolic and flavonoid compounds in these single herbal extraction with/without combined to mixture. We also measured antioxidant activities such as DPPH free radical scavenging, SOD-like scavenging, nitrite scavenging and hydroxyl radical scavenging activities, xanthine oxidase inhibition, linoleic acid peroxidation inhibition, and reducing power. Results : As the results, contents of total phenoilc compounds and flavonoids were higher in those of PF than those of SF. Those of SF+PF mixture showed the synergy effects compared with those of SF and PF single extractions. Activities of DPPH free radical and SOD-like scavenging in 1 mg/mL concentration increased in dose dependent manners. That of SF increased compared with that of PF. That of SF and PF mixture also increased compared with that of BHA as a positive control. The other antioxident activities also showed similar to patten of activity of DPPH free radical scavenging. When combined to SF and PF extractions, there was showed synergic effect compared with those of BHA, excepted activities of xanthine oxidase inhibition and reducing power. Taken together, SF and PF have high phenolic and flavonoid compounds content furthermore, antioxidant activities in SF and PF mixture showed more synergy effect compared with those of BHA. Conclusions : Therefore, these findings suggest that SF and PF mixture may offer functional materials potential for development of functional beverage. But further studies are needed for the identification of the active compounds.

싸리꽃 에탄올 추출물의 피부(皮膚) 생리활성(生理活性) 효과 (Dermal Bioactive Properties of the Ethanol Extract from Flowers of Lespedeza bicolor)

  • Objective : In this study, the ethanol extract from flowers of Lespedeza bicolor was investigated for their dermal bioactive properties related to cosmeceuticals such as depigmentation and radical scavenging effect. Results : The ethanol extract from flowers of Lespedeza bicolor showed considerable radical scavenging activity ($SC_{50}:\;10\;{\mu}g/ml$) and inhibited the production of nitric oxide (NO) in Raw 264.7 macrophages activated with the endotoxin lipopolysaccharide (LPS). Although the proliferation of B16/F10 cells was slightly decreased by the ethanol extract from flowers of Lespedeza bicolor at the concentration of $100\;{\mu}g/ml$, it did not appear necrosis. The ethanol extract from flowers of Lespedeza bicolor down-regulated melanin formation effectively. Methods : The free radical scavenging activity of Lespedeza bicolor was assayed in cell free systems using a stable free radical, 1,l-diphenyl-2-picrylhydrazyl (DPPH). Nitrite accumulated in culture medium was measured as an indicator of NO production using a Griess reaction. Cell viability was measured by MTT assay and melanin content was assessed using the method of Hosei with some modifications. Conclusions : These results suggest that the ethanol extract from flowers of Lespedeza bicolor is a potent depigmetation agent and it may be a candidate for antioxidant and anti-inflammatory agent.

야콘 분말 추출물의 항산화 효과 및 항균 활성 분석 (Analysis of Antioxidant Effects and Antimicrobial Activity of Extracts from Yacon (Polymnia sonchifolia) Powder)

  • The purpose of this study was to analyze the antioxidant effects and antimicrobial activity of extracts from yacon powder. The total polyphenol contents of yacon powder extracted with methanol was 38.5 mg/g, while extracted with water was 24.0 mg/g. Extractions with methanol were therefore more effective than with water. The total flavonoid contents of yacon powder extracted with methanol was 9.2 mg/g, while that with water was 7.8 mg/g. The electron donating ability of yacon powder extracts, natural antioxidant, and artificial antioxidant at 1.0 mg/mL arranged in order of decreasing concentration were ascorbic acid (98.7%) > BHT (44.7%) > methanol extract (12.1%) > water extract (7.8%). Nitrite-scavenging ability of yacon powder extracted with methanol at pH 1.2 was 13.1%, while that of powder extracted with water at pH 1.2 was 6.8%. And, the hydroxyl radical scavenging activities of yacon powder with methanol at 5 mg/mL was 40.2%. The antimicrobial activity of yacon powder extracted with methanol could be detected on Bacillus cereus ($10,000{\mu}g/disc$) and Staphylococcus aureus ($10,000{\mu}g/disc$).
