Effects of Electron Donors on Nitrate Removal by Nitrate and Nitrite Reductases

  • Effects of artificial electron donors to deliver reducing power on enzymic denitrification were investigated using nitrate reductase and nitrite reductase obtained from Ochrobactrum antroyi. The activity of nitrite reductase in the soluble portion was almost the same as that in the precipitated portion of the cell extract. Nitrate removal efficiency was higher with benzyl viologen than with methyl viologen or NADH as an artificial electron donor. The turn-over numbers of nitrate and nitrite reductase were 14.1 and 1.9 umol of nitrogen reduced/min$.$mg cell extracts, respectively when benzyl viologen was used as an electron donor.

가토(家兎)에서 nitrite에 의한 methemoglobinemia에 미치는 selenium의 영향(影響) (Protective influence of selenium on nitrite-induced methemoglobinemia in rabbits)

  • The protective of influences of sodium selenitc ($Na_2SeO_3$) against the methemoglobinemia with sodium nitrite were investigated on hemoglobin, methemoglobin, glutathione peroxidase and NADH-methemoglobin reductase activity in rabbits which were given 0,1,3 and 9ppm sodium selenite of drinking water for a week. Dietary selenium did not alter total hemoglobin in the blood of rabbits. Selenium was found to decrease nitrite-induced methemoglobin in a dose-dependent manner. The glutathione peroxidase activity was also increased by selenium in all the experimental groups. However, the NADH-methemoglobin reductase activity by selenite did not show significant differences as concerns the methemoglobinemia. These results showed that selenium could inhibit nitrite-induced methemoglobinemia. Its influence of inhibition is suggested that the effect of the reduction of methemoglobin was greatly stimulated by glutathione peroxidase activity.

Modified BAF 공정을 이용한 독립적인 무산소조에서 탈질미생물 군집의 특성 (Characterization of Denitrifier Community in Independent Anoxic Reactor Using Modified BAF Process)

  • 최근 수계의 총질소(T-N) 규제가 강화되면서 기존 BAF 공정의 개선을 위해 독립적인 무산소조가 추가로 도입되었다. 본 연구에 사용된 공정은 유기물과 질산화 중심으로 개발 된 기술인 $Biobead^{(R)}$공법으로 상용화된 상향류의 BAF공정의 하나이다. 독립적인 무산소조의 도입의 타당성을 검토하기 위해 분자생물학적 방법의 하나인 PCR-DGGE기법이 수행되었다. 두 가지 type의 nitrite reductase genes를 통해 진행되었는데, nirS로 암호화된 cytocrome $cd_1$ nitrite reductase gene과 nirK로 암호화된 Cu를 함유한 nitrite reductase gene이다. 이러한 탈질 기능유전자를 이용하여 PCR-DGGE를 통해 탈질 목적으로 순화된 독립적인 무산소조의 탈질미생물의 군집을 해석하였다. PCR 증폭결과, 탈질을 수행하는 무산소조 내에서는 nirS와 nirK유전자 가운데 nirS유전자만 검출되었고, DGGE 분석결과, 최초 식종원으로 이용된 활성슬러지에서는 상대적으로 많은 band들이 검출되는 반면, 무산소조 내에서는 운전일수와 nitrate 부하량이 증가할수록 단일 band로 우점화 하는 경향을 나타내었다. DGGE band에 대한 염기서열 분석결과, 식종 슬러지의 경우 다양한 uncultured bacteria가 나타났으나, nitrate 제거율이 높은 안정화된 무산소조에서는 alcaligenes faecalis 등 특정 탈질미생물이 우점화 되는 것으로 확인되었다. 결론적으로 이러한 탈질미생물 군집특성을 가지는 무산소조의 도입은 96%이상의 안정적인 탈질을 가능하게 하였으며, BAF 공정 개선을 위한 독립적인 무산소조의 도입은 적절한 것으로 판단되었다.

  • 신개간간척답토양(新開墾干拓畓土壤)에서 볏짚 및 석고시용시(石膏施用時) 토양중(土壤中) Urease활성(活性), 초산(硝酸) 및 아초산환원효소활성(亞硝酸還元酵素活性)에 미치는 영향(影響)을 알고저 실내(室內) 시험(試驗)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 간척답토양(干拓畓土壤)의 Urease활성(活性)은 일반답토양(一般畓土壤)에 비(比)하여 1/10정도(程度)로 낮고 담수초기(湛水初期)보다 담수후기(湛水後期)에 높았으며 볏짚시용(施用)은 석고시용구(石膏施用區)보다 Urease 활성(活性) 증가효과(增加效果)가 높았다. 2. 초산환원효소활성(硝酸還元酵素活性)은 삼요소구(三要素區)에 비(比)하여 석고(石膏) 및 볏짚시용(施用)으로 증대(增大)되었으며 그 효과(效果)는 볏짚보다 석고시용(石膏施用)으로 더욱 증대(增大)되었다. 그러나 볏짚과 석고병용시(石膏倂用時)는 효소(酵素)의 활성(活性)이 현저(顯著)히 감소(減少)되었다. 3. 아초산환원효소(亞硝酸還元酵素)의 활성(活性)은 담수초기(湛水初期) 볏짚과 석고시용(石膏施用)으로 현저(顯著)히 증대(增大)되었으며 볏짚과 석고병용효과(石膏倂用效果)는 볏짚단용구(單用區)와 비슷한 결과(結果)를 보였다. 4. 토양중(土壤中) 효소활성간(酵素活性間)의 관계(關係)를 보면 Urease활성(活性)과 Ammonia농도간(濃度間)에는 정상관관계(正相關關係)($r=0.550^*$) Urease활성(活性)과 아초산환원효소활성간(亞硝酸還元酵素活性間)에는 부상관관계(負相關關係)($r=0.555^*$) 그리고 초산환원효소(硝酸還元酵素)와 아초산환원효소간(亞硝酸還元酵素間)에는 정상관관계(正相關關係)($r=0.296^*$)를 보였다. 5. 볏짚과 석고시용시(石膏施用時) 토양중(土壤中) 초산(硝酸)과 아초산(亞硝酸)의 생성(生成)은 토양미생물(土壤微生物)의 산화(酸化) 및 환원작용(還元作用)에서 First Order Kinetic에 의한 효소반응식(酵素反應式)으로 밝혀졌다.

The pH as a Control Parameter for Oxidation-Reduction Potential on the Denitrification by Ochrobactrum anthropi SY 509

  • The pH as a control parameter for oxidation-reduction potential (ORP) was investigated through the denitrification by Ochrobactrum anthropi SY509 under non-growing condition. The optimal pH of nitrate reductase was 7.0, and the minimal ORP level was -250 mV for the denitrification under aerobic condition. In the case of anaerobic condition, the optimal pHs of nitrate and nitrite reductase were shifted to 10.0 and 9.0, respectively, and the minimal ORP levels of nitrate and nitrite reductase were decreased to -370 mV and -340mV, respectively. In the case of alkaline pH and anaerobic condition, the denitrification efficiency of nitrate was increased up to about 2-fold over that of neutral pH and anaerobic condition. Therefore, the combined control of pH and ORP in the anaerobic condition is shown to be an important parameter in the biological denitrification process.

Amelioration of $Cd^{++}$ Toxicity by $Ca^{++}$ on Germination, Growth and Changes in Anti-Oxidant and Nitrogen Assimilation Enzymes in Mungbean(Vigna mungo) Seedlings

  • The present study describes the ameliorating effect of $Ca^{++}\;on\;Cd^{++}$ toxicity on the germination, early growth of mungbean seedlings, nitrogen assimilation enzyme. s-nitrate reductase (NR), nitrite reductase (NIR), anti-oxidant enzymes (POD, CAT and SOD) and on the accumulation of hydrogen peroxide and sulphydryls. $Cd^{++}$ inhibited seed germination and root and shoot length of seedlings. While NR activity was down- regulated, the activities of NIR, POD and SOD were up- regulated with $Cd^{++}$ treatment. $Cd^{++}$ treatment also increased the accumulation of sulphydryls and peroxides, which is reflective of increased thiol rich proteins and oxidative stress. $Ca^{++}$ reversed the toxic effects of $Cd^{++}$ on germination and on early growth of seedlings as well as on the enzyme activities, which were in turn differentially inhibited with a combined treatment with calcium specific chelator EGTA. The results indicate that the external application of $Ca^{++}$ may increase the tolerance capacity of plants to environmental pollutants by both up and down regulating metabolic activities. 아질산에 노출된 뱀장어(Anguilla japonica)의 혈액학적 반응과 항산화효소의 활성 변화 (Changes in Hematological Responses and Antioxidative Enzyme Activities of Japanese Eel Anguilla japonica Exposed to Elevated Ambient Nitrite)

  • The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

  • 질산염은 많은 유전자의 발현을 조절하고 생장과 발육과정에서 매우 중요한 영양소이자 시그널 분자이다. 본 연구는 고등식물에서 질산 신호에 의한 유전자발현제어 관련 전사인자의 연구현황을 소개하고자 한다. 질산 환원 효소는 질소 동화 경로상의 효소이며, 질산이온을 아질산이온으로 환원하는 과정을 촉매한다. 질산이온, 빛, 대사산물, 식물호르몬, 저온, 가뭄 등의 여러 요인이 질산환원효소 유전자의 발현 수준과 생리적 역할과 같은 질산환원효소 활성을 조절한다. 최근 질산 환원 효소 유전자의 발현제어에 관여하고 있는 몇몇 전사인자들이 식물에서 분리되었다. NODULE-INCEPTION-like proteins (NLPs)는 질산 환원 효소 유전자의 질산 유도성 발현에 관여하는 전사인자이다. NLPs는 질산 수송체, 아질산 수송체, 아질산 환원 효소에 관련된 유전자의 질산 유도성 발현을 제어한다. 질산 환원 경로와 관련된 유전자의 발현 수준은 질산에 반응하여 NLPs에 의해 협조적으로 조절된다. 따라서 식물에서 질산염의 기능을 이해하면 질소 사용량이 적은 작물을 육성할 수 있다.

Detection of Nitrate/Nitrite Bioavailability in Wastewater Using a luxCDABE-Based Klebsiella oxytoca Bioluminescent Bioreporter

  • In the present study, we have constructed a bioluminescent bioreporter for the assessment of nitrate/nitrite bioavailability in wastewater. Specifically, an approximately 500-bp DNA fragment containing a nitrate/nitrite-activated nasR-like promoter (regulating expression of genes encoding nitrite reductase in the genus Klebsiella) was fused upstream of the Vibrio fischeri luxCDABE gene cassette in a modified mini-Tn5 vector. Characterization of this strain, designated W6-1, yielded dose-dependent increased bioluminescence coincident with increased nitrate, nitrite, and ammonium added to the growth medium from 1 to 11 ppm. Bioluminescence in response to nitrogen species addition was light dependent up to 10, 7, and 8 ppm with nitrate, nitrite, and ammonium, respectively. This response was linear in the range from 1 to 8 ppm for nitrate ($R^2=0.98$), 1 to 6 ppm for nitrite ($R^2=0.99$), and 1 to 7 ppm for ammonium ($R^2=0.99$). A significant bioluminescent response was also recorded when strain W6-1 was incubated with slurries from aged, nitrate/nitrite contaminated wastewater. Thus, bioreporter strain W6-1 can be used to elucidate factors that constrain the use of nitrate/nitrite in wastewaters.
