Effect of Variable Feed Allowance with Constant Protein Input on Water Quality in Channel Catfish Production Ponds

  • This study was carried out to evaluate the effect of feeding higher protein feeds with lesser amount, but feeding the constant total protein input for all treatments, on water quality and nitrite toxicity in channel catfish ponds. There was no significant difference in survival rate among treatments $(P>0.05)$. Specific growth rate (SGR) for Treatment 1$(28\%\;protein\;and\;100\%\;of\;satiation)$ was significantly higher $(P>0.05)$ than for Treatment 3$(36\%\;protein\;and\;87.5\%\;of\;satiation)$, but not significantly higher than for Treatment 2 $(32\%\;protein\;and\;77.8\%\;of\;satiation)$ at constant digestible energy (DE), 3.08kcal/g (treatments 1, 2 and 3). At constant DE/P (treatments 4, 2 and 5), no significant difference in SGR was observed among treatments. Feed conversion ratio (FCR) slightly improved or improved as dietary protein level increased from $28\%$ to $32\%$ and feed allowance decreased by $12.5\%$, but did not improve as dietary protein level increased from $32\%$ to $36\%$ and feed allowance decreased by $22.2\%$, at constant DE and constant DE/P. There was no significant difference in water quality variables, such as total ammonia nitrogen (TAN), nitrite, chlorophyll a, soluble phosphorous concentrations among treatments, but significant difference in water quality variables over time as amount of feed fed increased $(P<0.0001)$. There was a trend toward increase in TAN and nitrite over time. A strong linear regression was observed between mean total ammonia nitrogen and nitrite for all treatments Y (Nitrite) =$0.04\times (TAN)+0.01$, $R_2=0.89$. Methemoglobin percent in the blood of catifish was not significantly different among treatments. And its mean value was $7.5\%$, which was relatively low, so that it was not serious problem in catfish production pond under these experiment conditions. There was the stronger linear regression between the percentage of Methemoglobin and the molar ratio of nitrite to chloride rather than nitrite alone: $Y\;(Methemoglobin\;\%)\;=\;58.45\;\times\;(NO^{2-}/Cl^-)\;+\;0.41,\;R^2=0.60$. These results indicate that deterioration of water quality has no strong impact on poor weight gain for $36\%$ dietary protein in this study.

Anti-inflammatory Activities of Chopi (Zanthoxylum piperitum A.P. DC) Essential Oil: Suppression of the Inducible Nitric Oxide Synthase and Cellular Adhesion

  • The aim of this study is to elucidate the anti-inflammatory activities of chopi (Zanthoxylum piperitum A.P. DC.) essential oil. Essential oil (EO) of chopi was extracted by steam distillation method, and its major constituents were limonene and geranyl acetate. Chopi-EO decreased approximately 38% of nitrite production, as compared to the lipopolysaccharde (LPS)-induced nitrite production. However, chopi-EO and its components did not quench nitric oxide (NO) chemically in cellfree system, and markedly inhibited approximately 40.4% of inducible nitric oxide synthase (iNOS) mRNA transcription. In addition, the inhibition of E-selectin gene transcription by chopi-EO caused the suppression of cellular adhesion. These results suggest that chopi-EO may exert potential anti-immunological inflammatory activity.

Color Stability of Chinese-Style Sausage Inoculated with Staphylococcus Carnosus and Staphylococcus Xylosus

  • This study investigate the effects of starter cultures on the color stability of Chinese-style sausage. The samples were inoculated with $10^7cfu\;g^{-1}$ meat of either Staphylococcus carnosus or Staphylococcus xylosus. After mixing, curing at either $4^{\circ}C$ or $20^{\circ}C$ for 20 h and then drying at $50^{\circ}C$ for 5 h, the samples were then either vacuum packed or hung at $4^{\circ}C$ and $25^{\circ}C$ (85% R. H.). The pH, nitrite content, nitrosyl pigment content, metmyoglobin and L-, a-, b- values were measured. The pH value still remained above 6.0 during storage. Nitrite residue of all samples decreased after storage at $25^{\circ}C$ for 7 days. The samples inoculated with S. carnosus and S. xylosus had higher nitrite content (20.9-34.7 ppm) than the control (p<0.05). Samples inoculated with S. carnosus and S. xylosus had higher nitrosyl pigment content and lower metmyoglobin content than those of the control. The L- and b- values of all samples decreased but the a- values increased with storage time. The result suggested that S. carnosus and S. xylosus starter cultures be used to improve color stability of Chinese-style sausage.

Effects of Nitrite Exposure on Plasma Nitrite Levels and Hepatic Drug-metabolizing Enzymes in the Carp, Cyprinus carpio (아질산 노출이 이스라엘잉어 혈장내 아질산 농도 및 간장 약물대사효소에 미치는 영향)

  • Effects of ambient nitrite, NO$_2$$\^$-/, at 1, 3, 10 and 30 mg/1, on the changes of plasma nitrite/nitrate and on hepatic drug - metabolizing enzyme activity were examined in the juvenile Israeli carp, Cyprinus carpio. When the fish were exposed to 1 and 3 mg/1 NO$_2$$\^$-/, there was an exposure duration-dependent increase in plasma NO$_2$$\^$-/ over the 96-hr period reaching 6∼7 fold excess the ambient concentration. In the fish exposed to 10 mg/1, a plateau concentration of less than 2-fold of the environment was attained in 12 hr. With 30 mg/1, however, the maximal plasma NO$_2$$\^$-/ was 41.25 mg/1 at 12 hr followed by a gradual decline. There was a concentration-dependent increase in methemoglobin (metHb) level in all NO$_2$$\^$-/ -exposed groups and a significant decrease in hematocrit value in 30 mg/l group after 96-hr exposure. Apart from the blunted increase in plasma NO$_2$$\^$-/ with higher NO$_2$$\^$-/ (10 and 30 mg/1) exposure, the ratio of plasma NO$_3$$\^$-/ to NO$_2$$\^$-/ was signifirantly higher in these groups compared to 1 and 3 mg/1. The imbalance in the plasma NO$_3$$\^$-//NO$_2$$\^$-/ at higher NO$_2$$\^$-/ exposure suggests a possible accelerated conversion of NO$_2$$\^$-/ to NO$_3$$\^$-/. Nitrite exposure did not affect the hepatic drug-metabolic activities in juvenile Israeli carp. All these data indicate that disposition of NO$_2$- differ depending upon exposed concentration and that metHb production may not be the exclusive toxic mechanism in carp.

  • The changes of amine, nitrate and nitrite nitrogen in yellow corvenia were studied during its processing and storage in order to clarify the precursors of N-nitrosamine(NA) formation in the salted and dried yellow corvenia(Gulbi), prepared y using the different salting method like dry and brine salting by pure and curde salt. As a result, during the processing and storage of Gulbi, DMA and TMA contents were significantly increased in the yellow corvenia. And after 40 days storage the increase rate showed 25.7∼45.7, 3.3∼5.6 times higher than those of 0.3, 2.4mg/kg, respectively, while nitrite contents, during its processing and storage, were scarcely changed in the salted and dried yellow corvenia. During the processing and storage, of Gulbi, DMA and TMA contents were less produced in brine salted and dried yellow corvenia using crude salt than in sample prepared using were scarcely changed in the salted and dried yellow corvenia. During the processing and storage of Gulbi, DMA and TMA contents were less produced in brine salted and dried yellow corvenia using crude salt than in sample prepared using pure salt, while the former were more effective than the latter in inhibiting the production of nitrate and nitrite. Therefore, it was revealed that reduction of NA precursors such as DMA, TMA, nitrate and nitrite were more effective in preparing with the brine salting method than with the dry salting method.

Effect of Soused Shrimp and Cooked Glutinous Rice Flour on the Changes of Low Molecular Nitrogen Compounds Content during Kimchi Fermentation (새우젓 및 찹쌀풀 첨가가 김치발효중 저급 질소화합물 함량에 미치는 영향)

  • This study was intended to observe the effect of the addition of soused shrimp(5%, 10%) and/or cooked glutinous rice flour(2.5%) on the changes of low molecular nitrogen compounds content of Kimchi during fermentation at $10^{\circ}C$ for 14 days. The pH, total acidity, free sugar, nitrite, dimethylamine(DMA) and volatile basic nitrogen(VBN) contents were measured. The nitrite contents were approximately 0.16 to 1.31 mg/Kg at the inital stage of fermentation. It was found that the nitrite contents of Kimchi samples with 10% soused shrimp$(S_2,\;GS_2)$ were relatively high among the Kimchi samples. However, nitrite was not detected after the stage of optimum maturity in all Kimchi samples. The dimethylamine(DMA) content of Kimchi samples were increased during the fermentation with increasing the soused shrimp concentration. Cooked glutinous rice flour enhanced the DMA production of Kimchi with soused shrimp. DMA content were 0.93-1.2 mg/kg at initial stage end 1.6-2.26 mg/kg at final stage of fermentation of the Kimchi with soused shrimp. As the fermentation period increased, the VBN contents of Kimchi samples with soused shrimp$(S_1,\;S_2,\;GS_1,\;GS_2)$ were apparently higher than those of Kimchi samples without it(C, G). Though the soused shrimp enhanced the volatile basic nitrogen production but cooked glutinous rice did not affected VBN production during Kimchi fermentation.

Effects of Cyclooxygenase Inhibitors on Vascular Reactivity and Alterations of Cyclooxygenase Expression (혈관 반응성에 대한 Cyclooxygenase 억제제 효과와 Cyclooxygenase 발현 변화)

  • Background: There is controversy regarding whether COX-2 specific inhibitors are associated with elevation of blood pressure. We compared the effects of aspirin, indomethacin, and celecoxib for vascular reactivity induced by phenylephrine. We also tested the effects of indomethacin and NO donor on COX-1 and COX-2 protein expression, as well as nitrite production in culture medium of vascular smooth muscle cells. Materials and Methods: In this experiment, we used the isometric tension study for vascular reactivity. After 45 minutes of pretreatment with aspirin, indomethacin, celecoxib, and phenylephrine induced contractions were tested. COX-1 and COX-2 protein expressions were analyzed by Western blot and nitrite production by the Griess reaction. Results: Although celecoxib pretreatment caused enhanced arterial contraction, aspirin pretreatment induced more potent arterial contraction than celecoxib in the isometric tension study of rabbit femoral artery. COX-1 protein expression was unchanged by indomethacin, SNP and NOR-3; COX-2 protein expression was increased by the addition of indomethacin, SNP, and NOR-3. Especially, NOR-3, a NO donor, significantly increased COX-2 protein expression with unstimulated conditions as well as LPS stimulation. Induction of nitrite production was higher with NOR-3 treatment than SNP treatment with LPS stimulation. Conclusion: These results suggest that aspirin caused more potent vascular contraction than celecoxib and indomethacin. COX-2 expression in VSMC depended on the types of NO donor and LPS stimulation.

Nitrite Removal by Autotrophic Denitrification Using Sulfur Particles (황입자를 이용한 독립영양탈질에서의 아질산성질소 탈질 조건 탐색)

  • Swine wastewater contains high amounts of organic matter and nutrients (nitrogen and phosphorus). The biological nitrogen removal can be achieved by nitrification and denitrification processes. Nitrification-denitrification can be performed via nitrite which is called as the short-cut process. This Short-cut process saves up to 25% of oxygen and 40% of external carbon during nitrification and denitrification. In this study, the batch tests were conducted to assess the different parameters for the nitrite sulfur utilizing denitrification, such as alkalinity, temperature, initial nitrite concentration, and dissolved oxygen. The experimental results showed that the nitrite removal efficiency of the reactor was found to be over 95% under the optimum condition ($30^{\circ}C$ and sufficient alkalinity). Autotrophic nitrate denitrification was inhibited at low alkalinity condition showing only 10% removal efficiency, while nitrite denitrification was achieved over 95%. The nitrite removal rates were found similar at both $20^{\circ}C$ and $30^{\circ}C$. In addition, nitrite removal efficiencies were inhibited by increasing oxygen concentration, but sulfate concentration increased due to sulfur oxidation under an aerobic condition. Sulfate production and alkalinity consumption were decreased with nitrite compared those with nitrate.

Antioxidant Effects and Anti-inflammation Effects of Lophatheri Herba Water Extracts Via Reducing iNOS Synthesis Induced by LPS in RAW 264.7 Cell (담죽엽의 항산화 효과와 RAW 264.7 세포에서 LPS로 유도된 iNOS 발현에 미치는 영향)

  • We studied to know the anti-inflammation effect on water extracts of Lophatheri Herba which was growing in every places in our country. We objected free radical scanvenger effect and nitrite eliminate effect of the Lophatheri Herba water extracts, and the cell viabillity, the effects of Lophatheri Herba water extracts on NO production, iNOS synthesis induced by LPS. Free radical scavenger effects were $27.91{\pm}0.12%$, $38.96{\pm}0.10%$, $46.22{\pm}0.15%$ depend on 0.5, 1.0, 2.0 mg/ml each dose of Lophatheri Herba water extracts. Nitrite eliminate effects were $9.86{\pm}0.3%$, $80.61{\pm}0.23%$, $97.62{\pm}0.56%$ in 0.1, 1.0, 2.0 mg/ml Lophatheri Herba water extracts on pH 1.2. NO production and iNOS synthesis induced by LPS were reduced in RAW 264.7 cell by Lophatheri Herba water extracts. As the above results, Lophatheri Herba water extracts have anti-inflammation effects via NO production decrease, iNOS synthesis decrease mechanism. So Lophatheri Herba water extracts will be used as the protection or treatment in chronic inflammation desease like a asthma, stomatitis etc.
