  • Phytotron and field experiments were conducted to determine the influence of morphological growth stage and environmental temperature on synthesis and accumulation pattern of cyanogenic glycosides in sorghum cv. Pioneer 931 and Sioux at Munich technical university from 1979 to 1980. Various growth stages of sorghum plants were grown in phytotron at 4 different temperature regimes of 30/25, 25/20, 28/18 and 18/8 degree C with 35,000 Lux over 13-h days. The results obtained are summarized as follows: 1. Cyanogenic glycosides in sorghum plants were shown to have a great synthetic rate at early growth stages. The highest concentrations of hydrocyanic acid (HCN) were found at 2-leaf stage with 2384 and 1800ppm (DM basis) for Pioneer 931 and Sioux respectively. The contents of HCN were, however, however decreased markedly as morphological development, which shows a value of 173ppm (Pioneer 931) and 70ppm (Sioux) at heading stages. 2. Changes of hydrocyanic acid in sorghum plants were positive correlated with leaf weight ratio and leaf area ratio ($P{\leqq}0.1%$), while plant height shows a negative correlation with HCN contents ($P{\leqq}0.1%$). 3. Cyanogenic glycosides were accumulated in young plants mainly in leaves. During the late maturities, the contents of HCN in leaves and stalks were shown, however, a similar distribution. 4. Synthesis rates of cyanogenic glycosides were increased under high temperature. Accumulated hydrocyanic acid in the plants was, however declined when temperature exceeded 30 degree C. 5. Synthesis rates of cyanogenic glycosides were affected by nitrogen reductase activity (NRA). The concentration of hydrocyanic acid in sorghum plants was associated with increasing of nitrate-N accumulation.

  • Nutrient balance during rice cultivation in the paddy of a local area under the environmental protection for drinking water supply was investigated. To compare nutrient balance in the paddy soil applied with different types of fertilization, 7 treatments were selected as followings : Recommended level of chemical fertilizers(R), Conventional fertilization(CF), Fresh cow manure(FCM), Cow manure compost(CMC), Straw compost+reduced chemical fertilizer(SCF), Fresh straw+recommended level of fertilizers(FSC), and no fertilization as control(C). Here, FCM, CMC and SCF were applied at the same level of total nitrogen as recommended in R. Rice yield was the highest in the recommendation(R) and fresh cow manure (FCM) treatments with $6,730kg\;ha^{-1}$(index 100), and followed by SCF (index 98), FSC (index 98), CMC(index 94), and CF(index 94). But statistically significant difference was not recognized among treatments except the control. Nitrogen infiltration loss was high in the simple chemical fertilizer treatments with $63kg\;ha^{-1}$ in CF and $58kg\;ha^{-1}$ in R during rice cultivation, respectively. Nitrogen infiltration loss was decreased below half level of chemical fertilizer treatments with cow manure treatments ($23kg\;ha^{-1}$ in FCM and $27kg\;ha^{-1}$ in CMC) and with reducing chemical fertilizer treatment by adding straw compost ($25kg\;ha^{-1}$). Phosphate was not leached during rice cultivation in paddy soil of a fluvial deposit type, in which oxidation horizon was developed broadly under around 15 cm depth of surface soil. Phosphate balance (A-B) was closed to 0 in all treatments except cow manure treatment (CMC), in which it was $+30kg\;ha^{-1}$ and show the possibility of over accumulation of phosphate by continuously replicated application of cow manure compost. Potassium balance was negative value in all but straw recycling treatment (FSC). It means that potassium was continuously supplied from soil minerals, uptaken by plants or eluted out of soil. In conclusion, by substituting inorganic fertilizer for organic fertilizer or reducing application rate of chemical fertilizer through mixing organic fertilizer, it would be possible to achieve the same rice yield as in the recommendation treatment and to decrease nutrient leaching below half level in rice paddy soil.

  • Field survey on the spatio-temporal distribution of water quality and chlorophyll a concentration, and the environmental factors on the variation of phytoplankton biomass were carried out at the 23 stations for four seasons in the Shiahae, southwestern coast of Korean Peninsula from February to October in 1995. I made an analysis on biological factor as chlorophyll a concentration as well as environmental factors such as water temperature, salinity and nutrients; ammonia, nitrite, nitrate, dissolved inorganic nitrogen, phosphate, N/P ratio, silicate and Si/P ratio. The waters in the Shiahae were not stratified due to the tidal mixing and high velocity of tidal current. And the high productivity in photic layer were supported by high nutrients concentration from freshwater on lands and bottom waters The low depth of transparency in the Shiahae had a bad influence upon primary production and marine biology. In Shiahae had a sufficient nutrients for primary production during a year. Especially dissolved inorganic nitrogen and silicate were high, the other side, phosphate was low. The source of nutrients in summer and silicate supply depend on input of freshwater from lands, the other side, dissolved inorganic nitrogen and phosphate were depend on rather supplied from bottom layer by the mixing and input of seawater from outside than input of freshwater from lands. Phosphate seemed to become a limiting nutrient for the primary production at all area of Shiahae in winter and at the northern parts in other seasons. However, dissolved inorganic nitrogen seemed to do it at the southern parts in other seasons except winter. Silicate didn't become a limiting nutrient for diatoms in Shiahae. Phytoplankton biomass as measured by chlorophyll a concentration was very high all the year round, it was controlled by the combination of the several environmental factors, especially of nitrogen, phosphorus and the physical factors such as light intensity. • To know the differences in ionic compositions in rain and snow as well as snow influence on the chemical characteristics of winter precipitation, precipitation samples were collected by the wet-only automatic precipitation sample, in winter(November-February) in the Iksan located in the northwest of Chonbuk from 1995 to 2000. The samples were analyzed for concentrations of water-soluble ion species, in addition to pH and electrical conductivity. The mean pH of winter precipitation was 4.72. According to the type of winter precipitation, the mean pH of rain was 4.67 and lower than 5.05 in snow. The frequencies of pH below 5.0 in rain were about 73%, while those in snow were about 30%. Snow contained 3 times higher concentrations of sea salt ion components originated from seawater than did rain in winter, mainly $Cl^-,\;Na^+$, and $Mg^{2+}$. Neglecting sea salt ion components, $nss-SO_4^{2-}$ and $NO_3^-$ were important anions and $NH_4^+$ and $nss-Ca^{2+}$ were important cations in both of rain and snow. Concentrations of $nss-SO_4^{2-}$ was 1.3 times higher in rain than in snow, while those of $nss-Ca^{2+}$ and $NO_3^-$ were 1.5 and 1.3 times higher in snow, respectively. The mean equivalent concentration ratio of $nss-SO_4^{2-}/NO_3^-$ in winter precipitation were 2.4, which implied that the relative contribution of sulfuric and nitric acids to the precipitation acidity was 71% and 29%, respectively. The ratio in rain was 2.7 and higher than 1.5 in snow. These results suggest that the difference of $NO_3^-$ in rain and snow could be due to the more effective scavenging of $HNO_3$ vapor than particulate sulfate or nitrate by snow. The lower ratio in snow than rain is consistent with the measurement results of foreign other investigators and with scavenging theory of atmospheric aerosols. Although substantial $nss-SO_4^{2-}$ and $NO_3^-$ were observed in both of rain and snow, the corresponding presence of $NH_4^+,\;nss-Ca^{2+},\;nss-K^+$ suggested the significant neutralization of rain and snow. Differences in chemical composition of non-sea salt ions and neutralizing rapacity of $NH_4^+,\;nss-Ca^{2+}$, and $nss-K^+$ between rain and snow could explain the acidity difference of rain and snow. Snow affected that winter precipitation could be less acidic due to its higher neutralizing rapacity.

  • Background: Nitric oxide is a short-lived effector molecule derived from L-arginine by the nitric oxide synthase(NOS). Nitric oxide plays a role in a number of physiologic and pathophysiologic functions including host defense, edema formation, and regulation of smooth muscle tone. Some kinds of cells including macrophage are known to produce large quantities of nitric oxide in response to inflammatory stimuli such as interleukin-$1\beta$(IL-$1\beta$), tumor necrosis factor-$\alpha$(TNF-$\alpha$), interferon-$\gamma$(IFN-$\gamma$) and lipopolysaccharide(LPS). Reactive oxygen species are also known to be important in the pathogenesis of acute cell and tissue injury such as acute lung injury model Methods: Using the RA W264.7 cells, we have examined the ability of oxidant hydrogen peroxide($H_2O_2$) to stimulate nitric oxide production and inducible NOS mRNA expression. Also, we have examined the effects of NOS inhibitors and antioxidants on $H_2O_2$ induced nitric oxide production. Results: Stimulation of RAW264.7 cells with combinations of 100 ng/ml IL-$1\beta$, 100 ng/ml TNF-$\alpha$, and 100 U/ml IFN-$\gamma$ or 100 U/ml IFN-$\gamma$ and $1{\mu}g/ml$ LPS induced the synthesis of nitric oxide as measured by the oxidation products nitrite($NO_2^-$) and nitrate($NO_3^-$). Addition of $250 {\mu}M-2$ mM $H_2O_2$ to the cytokines significantly augmented the synthesis of $NO_2^-$ and $NO_3^-$(p<0.05). When cells were incubated with increasing concentrations of $H_2O_2$ in the presence of IL-$1\beta$, TNF-$\alpha$ and IFN-$\gamma$ at constant level, the synthesis of $NO_2^-$ and $NO_3^-$ was dose-dependently increased(p<0.05). $N^G$-nitro-L-arginine methyl ester(L-NAME), dose dependently, significantly inhibited the formation of $NO_2^-$ and $NO_3^-$ in cells stimulated with LPS, IFN-$\gamma$ and $H_2O_2$ at constant level(p<0.05). Catalase significantly inhibited the $H_2O_2$-induced augmentation of cytokine-induced $NO_2^-$ and $NO_3^-$ formation(p<0.05). But, boiled catalase did not produce a significant inhibition in comparison with the native enzyme. Another antioxidant 2-mercaptoethanol and orthophenanthroline dose-dependently suppressed $NO_2^-$ and $NO_3^-$ synthesis(p<0.05). Northern blotting demonstrated that H:02 synergistically stimulated the cytokine-induced iNOS mRNA expression in RA W264.7. Conclusion: These results suggest that $H_2O_2$ contributes to inflammatory process by augmenting the iNOS expression and nitric oxide synthesis induced by cytokines.

  • In order to clarify the ecophysiological characteristics of Chenopodiaceae which widely distribute on saline and arid habitats, we collected 10 chenopodiaceous plant species, examined their inorganic and organic solute patterns, and confirmed several common physiological characteristics. In spite of high soil Ca/sup 2+/ contents, chenopodiaceous plants had a little water-soluble Ca within cells, but contained high contents of acid-soluble Ca particularly as a result of Ca-oxalate formation. These plant species also showed accumulation of inorganic ions such as K/sup +/, NO₃/sup -/ and Cl/sup -/, and Na/sup +/especially in saline habitats instead of K/sup +/ Meanwhile, with respect to nitrogen metabolism they retained high N contents in leaves, but showed very low amino acid contents. Additionally, they contained very little proline known to act as a cytoplasmic osmolyte. To ascertain whether this physiological characteristics in the field also can be found under controlled conditions, 7 chenopodiaceous plants (Atriplex gmelini, Corispermum stauntonii, Salicornia herbacea, Suaeda aspayagoides, Suaeda japonica, Chenopodium album var. centrorubrum, C. serotinum) were selected and cultivated under salt treatments. As well as field-grown plants, selected plant species showed similar solute pattern in growth experiment. In summary, the family of Chenopodiaceae represents the following physiological properties; high storage capacity for inorganic ions (especially alkali cations, nitrate and chloride), oxalate synthesis to maintain lower soluble Ca contents within cytoplasm, and low contents of amino acids. In addition to some characteristics mentioned above, the physiological plasticities of Chenopodiaceae which can properly regulate their ion and solute pattern according to soil conditions may enable its representative to grow in dry sand dune and salt marsh habitats.

  • A study on eutrophication in the downstream of the Nakdong River was conducted from October 1983 to September 1984. Nutrients concentration of the samples in this area was usually high and the nitrogen ratio to the phosphorus was as very high as $63.5{\sim}119.6$. The concentration of nutrients was in the range of $0.22{\sim}173{\mu}g-at/l$ for ammonia nitrogen, $26.7{\sim}187{\mu}g-at/l$ for nitrate nitrogen, $1.07{\sim}8.22{\mu}g-at/l$ for nitrite nitrogen, $58.7{\sim}231{\mu}g-at/l$ for total inorganic nitrogen and $0.44{\sim}4.43{\mu}g-at/l$ for phosphate phosphorus. The concentration of chlorophyll-a was in the range of $1.8{\sim}75.2\;mg/m^3$. Correlationship between concentration of nutrients and chlorophyll-a was not found. The concentrations of total suspended solid were in the range of $10.7{\sim}45.9\;ppm$, and $16.2{\sim}35.2\%$ of the total suspended solid was consisted of volatile suspended solid. According to the state of lacustrine nutrition, the state of the downstream of Nakdong River was equivalent to the eutrophication state in term of chlorophyll-a and inorganic nitrogen, and mid-nutritional state in term of phosphate.

  • The thermotolerant methylotrophic yeast Hansenula polymorpha is an attractive model organism for various fundamental studies, such as the genetic control of enzymes involved in methanol metabolism, peroxisome biogenesis, nitrate assimilation, and resistance to heavy metals and oxidative stresses. In addition, H. polymorpha has been highlighted as a promising recombinant protein expression host, especially due to the availability of strong and tightly regulatable promoters. In this study, we investigated the possibility of employing human serum albumin (HSA) as the fusion tag for the secretory expression of heterologous proteins in H. polymorpha. A set of four expression cassettes, which contained the methanol oxidase (MOX) promoter, translational HSA fusion tag, and the terminator of MOX, were constructed. The expression cassettes were also designed to contain sequences for accessory elements including His8-tag, $2{\times}(Gly_4Ser_1)$ linkers, tobacco etch virus protease recognition sites (Tev), multi-cloning sites, and strep-tags. To determine the effects of the size of the HSA fusion tag on the secretory expression of the target protein, each cassette contained the HSA gene fragment truncated at a specific position based on its domain structure. By using the Green fluorescence protein gene as the reporter, the properties of each expression cassette were compared in various conditions. Our results suggest that the translational HSA fusion tag is an efficient tool for the secretory expression of recombinant proteins in H. polymorpha.

  • This study was carried out to investigate the effect of sawdust bedding barn on manure handing, management labour and physiological aspect of dairy cows, and then to establish the criteria on the optimum utilization method of sawdust bedding barn. 46 tie stall barns and 49 sawdust bedding barns were surveyed to cmopare the milk productivity between two different barns, and also 5 tie stall barns, 15 sawdust bedding barns and 1 rice hull bedding barn were selected to study the utilization situation of sawdust barn in Kyung-Ki province area. The major results obtained were as follows; 1. The roof material of sawdust barn were consisted of 66% vinyl house, 23% PVC light and 11% slate and galvanum coated tin. Most of the floor structure was earth ground with the rate of the approximately 82%. 2. The average occupied area of sawdust bed per cow was 15.2 $m^2$, depth that 30 cm and the utilization period was 12 months. 3. Milk Yield was significantly higher at sawdust barn than at tie stall barn(P<0.01). Bacterial and somatic cell count in raw milk were less at the sawdust barn than in tie stall barn. However, there was not significance difference between two barns. 4. The labour hour needed to cow management in the sawdust barn was approximately 48% of that of tie stall barn. 5. The temperature and moisture content measured in sawdust bed were closely affected by seasonal ambient temperature. The skin and hair of cow were much cleaner at the PVC light roofed sawdust barn than any other roof materials. 6. The additives used for improving of fermentation did not show any effect on temperature and moisture content in sawdust bed. When the ambient temperature was $30.4^{\circ}C$, the surface temperature of measured 1 cm above the sawdust bed was $12.2^{\circ}C$ lower and the temperature of 100 cm above the sawdust bed was $2.4^{\circ}C lower under shading net facility than that of vinyl roofed one.7. The hoof length of miking cow was 7.95 cm in tie stall and 9.19 cm in sawdust barn with high significance (P<0.01). And disease occurence such as mastitis and foot-rot tended to decrease in the sawdust barn. 8. The number of total bacteria and coliform bacteria were less in the sawdust bed compared with earth ground resting area. And a parasite strongyloides papillosus was detected but without any infected cow. 9. The nitrate($NO_3N$) content in non-roofed earth ground resting area and earth ground under the sawdust bed was likely to pollute the ground water. 10. In economic point of view, rice hull bedding barn was the cheapest among different systems. And in the sawdust bedding barn PVC light + slate roofed barn was most desirable, and vinyl roofed one the least.

  • Salt accumulation in the plastic film house soils under continuous cultivation condition causes problems such as salt damages to plants, nitrate accumulation in vegetables, groundwater contamination, etc. due to excess application of fertilizers. Objective of this research was to find an optimum adsorbent to reduce salt concentration in the soil solution of plastic film house soils, where crop injuries have been observed due to the salt accumulation. The soils were significantly high in available P $(1,431{\sim}6,516mg\;kg^{-1}),\;NO_3-N\;(117.60{\sim}395.73mg\;kg^{-1})$, exchangeable Ca $(4.06{\sim}11.07\;cmol_c\;kg^{-1})$ and Mg $(2.59{\sim}18.76\;cmol_c\;kg^{-1})$, as compared to those of the average upland soils in Korea. Soils were treated with each of adsorbent such as ion-exchange resin, zeolite, rice bran, etc. at 2% level and prepared into saturated-paste samples. After equilibrium, soil solution was vacuum-extracted from the soil and measured for changes of the pH, EC, and concentrations of $Ca^{2+},\;Mg^{2+},\;K^+,\;Na^+,\;{NH_4}^+,\;{PO_4}^{3-}\;and\;{NO_3}^-$. Rice bran effectively removed ${PO_4}^{3-}\;and\;{NO_3}^-$ in the soil solution up to 100%. Efficiency was decreased in the orders of rice bran > ion-exchange resin > zeolite. Removal efficiencies of zeolite and ion-exchange resin for $Ca^{2+}$ were ranged from 1 to 65% and from 7 to 61%, respectively. Ion-exchange resin was also effective for removing $Mg^{2+},\;K^+,\;Na^+,\;and\;{NH_4}^+$. Overall results demonstrated that rice bran and ion-exchange resin could be applicable for salt accumulated soil to remove the respective anion and cation.
