담배 유식물의 생장과 Rubisco에 미치는 카드뮴의 효과에 대한 질산염의 영향 (Influence of Nitrate Against Effect of Cadmium on Growth and Rubisco in Seedling of Tobacco.)

  • 담배 유식물의 생장, 엽록소 및 광합성 효소에 대한 카드뮴과 질산염의 영향에 대해 연구하였다. 카드뮴에 의한 억제된 생장은 질산염에 의해 회복되지 않았다. 엽록소 a와 b의 함량은 카드뮴에 의해 감소하였으며, 카드뮴과 질산염 혼합처리구는 카드뮴 처리구보다 엽록소 함량이 감소하였다. 카드윰 처리구에서의 rubisco의 활성과 함량은 대조구에서의 함량보다 현저히 작았다. 이는 rubisco의 활성이 함량과 연관되어 있으며, rubisco의 활성화와 합성이 카드뮴에 의해 저해된다는 것을 의미한다. 카드뮴에 의해 감소된 rubisco의 활성과 함량은 질산염에 의해 더욱 더 감소되었다. 이와 비슷한 변화가 rubisco activase의 활성과 함량에서도 관찰되었다. 이 결과들은 카드뮴과 질산염에 의해 유도되는 rubisco의 변화가 rubisco activase와 서로 연관되어 있음을 의미하며, 질산염은 직접적으로 rubisco의 활성화와 합성에 관여 할 뿐만 아니라 rubisco activase를 통해서도 rubisco의 수준에 관여하는 것으로 해석된다.

Inhibition of methane formation and improvement of $H_2$ production by adding nitrate to anaerobic reactor

  • Anaerobic hydrogen production was executed using mixed anaerobic sludge. pH control (pH $5.5^{\sim}6.2)$ and heat treatment of anaerobic sluge at $92^{\circ}C$ was not effective for repressing the methane formation. The addition of $nitrate(500^{\sim}2,000\;mg/L\;KNO_3)$ made it possible to repress the methane formation, which resulted in the improvement of hydrogen production. By using SEM(Scanning Electron Microscope), less methane forming microbial with spagetti shape was observed when nitrate was supplied to anaerobic reactor.

Nitrogen Removal from Wastewaters by Microalgae Without Consuming Organic Carbon Sources

  • The possibility of microalgal nitrogen treatment was tested in wastewaters with a low carbon/nitrogen (C/N) ratio. Chlorella kessleri was cultured in the two different artificial wastewaters with nitrate as a nitrogen source: one contained glucose for an organic carbon source and the other without organic carbon sources. The growth rates of the two cultures were almost identical when the aeration rate was over 1 vvm. These results suggest that microalgae could successfully remove nitrogen from wastewater, as far as the mass transfer of $CO_2$, was not limited. Nitrate was successfully reduced to below 2 mg $NO_3^-$-N/ml from the initial nitrate concentration of 140 mg $NO_3^-$-N/ml in 10 days, even in the wastewater with no organic carbon source. Similar results were obtained when ammonium was used as the sole nitrogen source instead of nitrate. Higher concentrations of nitrogen of 140, 280, 560 and 1,400 mg/ml were also tested and similar amounts of nitrogen were removed by algal cultures without showing any substrate inhibition.

  • Tetrahydrofuran(THF)중에서 질산코발트(II)를 개시제로 사용하여 methyl methacrylate(MMA)의 중합반응을 연구하였다. 반응속도론적 연구 결과로 다음과 같은 중합속도식을 얻었다. $R_p=k\;[질산코발트(II)]^{0.5}\;[MMA]^{1.5}$ Chelate를 잘 형성하는 옥살산이 중합속도에 미치는 영향을 고려해 보면, 단위체인 MMA가 질산코발트(II)와 배위착물을 형성한다는 것을 추론할 수 있었다. 또 중합계의 겉보기 총 활성화에너지는 14.0kcal/mole이라는 것을 알았다.

  • Development of hematite-coated sand was evaluated for the application of the PRB (permeable reactive barrier) in the arsenic-contaminated subsurface of the metal mining areas. The removal efficiency of As(III) and As(V), the effect of anion competition and the capability of arsenic removal in the flow system were investigated through the experiments of adsorption isotherm, arsenic removal kinetics against anion competition and column removal. Hematite-coated sand followed a linear adsorption isotherm with high adsorption capacity at low level concentrations of arsenic (< 1.0 mg/l). When As(III) and As(V) underwent adsorption reactions in the presence of anions (sulfate, nitrate and bicarbonate), sulfate caused strong inhibition of arsenic removal, and bicarbonate and nitrate caused weak inhibition due to specific and nonspecific adsorption onto hematite, respectively. In the column experiments, high content of hematite-coated sand enhance the arsenic removal, but the amount of the arsenic removal decreased due to the higher affinity of As(V) than As(III) and reduced adsorption kinetics in the flow system, Therefore, the amount of hematite-coated sand, the adsorption affinity of arsenic species and removal kinetics determined the removal efficiency of arsenic in the flow system. arsenic, hematite-coated sand, permeable reactive barrier, anion competition, adsorption.

數種 韓藥材의 齒牙美白 效果에 관한 實驗的 硏究 (Study on the effects of herbal extracts on tooth whitening, antioxidant, nitric oxide synthesis and Streptoccocus mutans.)

  • Fifteen herbs used in oral medicine were extracted in ethanol and screened for tooth whitening effect, radical scavenging activity, inhibition of inducible nitric oxide synthase activity and anti-bacterial activity on Streptoccocus mutans. The results are as followed: 1. The tooth whitening effect of toothpaste containing 1 $\%$ ethanol extracts of 15 herbs was tested in brushing method using artificially stained HAT tablet and bovine tooth specimens. Toothpaste containing 1$\%$ of Vucia unijuga, Illicium verum, Angelica dahurica and Piper longum extracts showed tooth whitening effects on both HAT tablet and bovine tooth specimens. But the others did not show a considerable activity. 2. The antioxidant activity of ethanol extracts of 15 herbs was tested using the method of 1,1-diphenyl-2-picryl hydrazyl (DPPH) reactivity. Two ethanol extracts of Cimicifuga heracleifolia and Phyllostachys nigra var. henonis were found to be the most effective on radical scavenging activity. C. heracleifolia and P. nigra var. henonis extracts removed 86$\%$ and 81 $\%$ of DPPH radical at 0.01$\%$, compared with butylated hydroxy toluene as positive control. 3. The inhibition activity of ethanol extracts on nitrate production In RAW264.7 cell stimulated by lipopolysacchride was tested using the Griess reagent. But all extracts did not inhibit nitrate production. 4. The antibacterial activity on Streptoccocus mutans was tested by paper disk method. But no one extract showed any anti - bacterial activity.

Regulation of LPS-induced Nitric Oxide Synthase Activity by Cigarette Smoke in Mouse Brain

  • Nitric oxide(nitrogen monoxide, NO) plays important physiological roles, but excessive generation can be toxic. NO is present in cigarette smoke at up to 1,000 ppm, and probably represents one of the greatest exogenous sources of NO to which humans are exposed. We investigated whether cigarette smoking reduces the production of endogenous NO and whether it influences the action of lipopolysaccharide(LPS) to induce nitric oxide synthase activity in mouse brain. Mice(C57BL6/J) were exposed to cigarette smoke for 8 weeks. LPS was injected intraperitoneally in single or combination with the exposure to cigarette smoke. Six hours after the injection of LPS, mice were sacrificed and sera and brains were collected. Serum concentrations of nitrate and nitrite were not charged by 4-week smoke exposure, but were significantly increased by 6 and 8 weeks of smoke exposure. Interestingly, cigarette smoke reduced the elevation in serum nitrate and nitrite concentrations produced by LPS after 4-week smoking exposure. NO synthase(NOS) activity in brain was upregulated by LPS-administration. However, cigarette smoke exposure remarkably and consistently decreased the LPS-induced activity in mouse brain. This result suggests that cigarette smoking may affect against overproduction of the endogenous NO by LPS through the inhibition of NOS activity induced by LPS in brain or by modulation of the LPS action for the induction of NOS activity. We also suggest the possibility that the exogenous NO evolved in cigarette smoke enables feedback inhibition of NOS activity or other possibility that it attenuates the toxicity of endotoxin LPS in vivo by unknown mechanisms, which should be further studied.

질산염과 산소에 의한 생물학적 퍼클로레이트 환원의 저해 (Inhibition of Biological Perchlorate Reduction by Nitrate and Oxygen)

  • 본 연구에서는 질산염(${NO_3}^-$)과 용존산소(DO)에 의한 생물학적 퍼클로레이트 환원의 저해 그리고 전자공여체를 차지하기 위한 이러한 전자수용체들의 경쟁적 관계를 조사하고자 염소이온프로브(chloride ion probe)를 직접적인 측정방법으로 사용하였다. 퍼클로레이트 환원미생물을 포함하는 플라스크에서 염소이온프로브를 이용하여 염소생성 (=퍼클로레이트 환원)을 모니터링하였다. 2 mM 퍼클로레이트의 생물학적 환원은 2 mM의 질산염에 의해 저해를 받았으며, 염소이온 생성율이 퍼클로레이트가 단독으로 존재하는 경우에 비해 30% 정도 감소하였고, 아세테이트가 제한된 상태이며 질산염과 퍼클로레이트가 공존하는 경우는 염소이온의 생성율이 약 70% 정도 감소하였다. 7-8 mg/L의 용존산소와 2 mM의 퍼클로레이트가 공존하는 조건에서 아세테이트의 공급정도와 상관없이 퍼클로레이트의 생물학적 환원작용이 완벽하게 저해를 받았다.

Effect of Saline Concentrations on Biological Nitrification in Batch Reactor

  • This study was carried out on 4 batch reactors to determine the specific ammonium oxidizing rate (SAOR), specific nitrate forming rate (SNFR) and inhibitory degree of nitrifying activities with saline concentrations. Under salt free condition ammonia was consumed during the reaction period within 200 min. When the salt level increased to 10, 20 and 30 g $NaClL^{-1}$ in reactor, ammonia depletion took 250, 300 and above 350 min, respectively. During concentration above 10 g $NaClL^{-1}$, there was nitrite accumulation. Also, at 30 g $NaClL^{-1}$ ammonia did not depleted and $NO_2{^-}$-N accumulated until the final reaction. Nitrate formation rates decreased with increasing salt concentration. SAOR and SNFR showed a decreasing trend as salinity concentrations were increased. The SAOR was reduced from 0.2 to 0.08 mg $NH_4{^+}$-N $g^{-1}VSS\;day^{-1}$ as the salt concentration increased from 0 to 30 g $NaClL^{-1}$. Similarly, the SNFR decreased from 0.26 kg $NO_3{^-}$-N $kg^{-1}VSS\;day^{-1}$ at saline free to 0.1 kg $NO_3{^-}$-N $kg^{-1}VSS\;day^{-1}$ at saline 30 g L-1. A severe inhibition of nitrifiers activity was observed at increased salt concentrations. The inhibition ratio of specific ammonium oxidation rates were 17, 47 and 60% on the reactor of 10, 20 and 30 g $NaClL^{-1}$ added, respectively. The inhibition ratio of specific nitrate forming rates also were inhibited 30, 53 and 62% on the reactor of 10, 20 and 30 g $NaClL^{-1}$ added, respectively. As the salinity concentrations increased from 0 to 30 mg $NaClL^{-1}$, the average MLSS concentration increased from 1,245 to 1,735 $mgL^{-1}$. The SS concentration of supernatant in reactor which settled about 30 minutes was not severely difference between concentration of salt free reactor and one of those high salt contained reactors.

고정상 담체를 충진한 BER에서 HRT, 전류밀도 및 담체 충진율 변화가 질산성 질소 제거효율과 전류이용효율에 미치는 영향 (The effect of HRT, current density, and packing ratio on nitrate nitrogen removal efficiency and current efficiency in BRM-BER)

  • BER at different packing ratios of bio-ring media(BRM) was tested to investigate the effect of varying hydraulic retention time (HRT) and current density on the nitrate removal and current efficiency. In the preliminary batch mode experiment of BERs, current density was applied at 2.0 A/$m^2$, 4.0 A/$m^2$, 4.8 A/$m^2$, which correspond to the designation of reactor #1, #2, #3, respectively. The reactor #2 showed a highest nitrate removal rate of 162.0 mg $NO_3{^-}$-N/L/d, and the kinetics of nitrate removal rate was defined as the Zero order reaction. In the primary experiment of BERs, four BERs packed with BRM were operated in varying HRT and current, and the packing ratios of reactor #1, #2, #3 and #4 were 0%, 8%, 16%, 24%. respectively. This results of the experiments indicated that the nitrate removal rate and current efficiency were increased significantly cause of growing of autotrophic denitrification microorganisms on the surface of cathode and media by increasing of the current density and decreasing of HRT. However, The decreasing of nitrate removal rate and current efficiencies were observed in the condition of HRT of 5.25 hr and 4.8 A/$m^2$ of current density. With more increasing current density and decreasing of HRT, the hydrogen inhibition occurred at the surface of cathode. Moreover, nitrate removal rate by autotrophic denitrification microorganisms attached on the media surface was observed to be limited by no longer increasing dissolved hydrogen concentration of each reactor. In conclusion, the highest nitrate nitrogen removal and current efficiency could be achieved when the BER was operated at the conditions of 7 hr HRT, current density of 4.0 A/$m^2$, and 16% packing ratio. And it was found that the amount of nitrate removal by microorganisms attached on the surface of cathode and media (BRM) was 178.2 mg/L and 52.2 mg/L respectively. and the amount of nitrate removal per MLVSS was 0.435 g $NO_3{^-}$-N/g $MLVSS{\cdot}d$ and 0.336 $NO_3{^-}$-N/g $MLVSS{\cdot}d$.
