Adsorption of Nicotinic Acid on the Porous Powders (니코틴 산의 다공성 분체 흡착)

  • Nicotinic acid was mixed with glass powders such as controlled pore glass (CPG), glyceryl controlled pore glass (GPG) and glass beads (GB) at room temperature. The physicochemical properties of nicotinic acid in the various mixtures were examined by differential thermal analysis, X-ray diffraction study. Infrared spectroscopy and BET gas adsorption measurements. The peak area at the melting point from the various mixtures of nicotinic acid and CPG was increased with an increase of nicotinic acid concentration while the broad peak area was remained unchanged in the DTA curve. As shown in the powder X-ray diffraction patterns, the crystalline peaks of nicotinic acid disappeared in mixture with CPG, suggesting the interaction of nicotinic acid and porous powders. It was found that the larger the content of CPG, the higher the ratio of an amorphous state to a crystalline state. BET isotherm showed that as the amount of nicotinic acid was increased, the specific surface area was reduced proportionally to nicotinic acid content of up to 40% and remained constant thereafter. Sublimation of nicotinic acid from the mixture of nicotinic acid and CPG was examined. A large quantity of nicotinic acid was retained in the mixture when stored on various temperatures in vacuo for 10 hours. The nicotinic acid mixtures with CPG or GPG showed a high dissolution rates of nicotinic acid in aqueous solution, especially in the initial dissolution stage. CPG is expected to be a good pharmaceutical excipient to reduce the crystallinity of drugs and to prevent sublimation of drugs.

Production of 6-hydroxynicotinic acid by nicotinic acid-assimilating Pseudomonas sp. (Pseudomonas sp.에 의한 nicotinic acid로부터 6-hydroxynicotinic acid의 생산)

  • For the production of 6-hydroxynicotinic acid from nicotinic acid, bacteria capable of assimilating nicotinic acid as a sole carbon, nitrogen and energy source were isolated from soils. Among them, SH-007, newly isolated strain having the best acitivity of nicotinic acid hydroxylase was selected and identified as Pseudomonas sp. The specific acitivity of nicotinic acid hydroxylase of Pseudomonas sp. was highest when the strain was cultured at $30^{\circ}C$ for 24 hrs in the medium (pH 7.5) containing 2 g nicotinic acid, 1 g $(NH_4)_2SO_4$, and 0.5 g peptone per liter. The addition of 1.5 g/l of nicotinic acid into the 24 hrs incubated culture medium resulted in the 12% higher specific acitivity of nicotinic acid hydroxylase than that of the non-added control after further 18 hrs incubation. In the 6-hydroxynicotinic acid production through the reaction with resting cells, 2.22 g/l of 6-hydroxynicotinic acid was produced from 2 g/l of nicotinic acid after incubation of 3 hrs under optimum conditions, which correpsonds to 98.2% of theoretical 6-hydroxynicotinic acid yield.

Determination of niacin profiles in some animal and plant based foods by high performance liquid chromatography: association with healthy nutrition

  • Vitamin $B_3$ (niacin) is essential for all living cells and plays a central role in energy metabolism and oxidative phosphorylation. Vitamin $B_3$, a water-soluble vitamin, is present in the form of nicotinic acid and nicotinamide, a monocarboxylic acid derivative of pyridine. While nicotinic acid is commonly effective in lowering cholesterol levels, unlike nicotinic acid, nicotinamide is ineffective on lipids. Presence rates of nicotinic acid and nicotinamide, which are the available forms of vitamin $B_3$, are different for each food. However, the studies in the literature are generally based on the analysis of total amount of vitamin $B_3$ in foods and the studies determining the profile of vitamin $B_3$ in foods are limited. The aim of the study was to determine the vitamin $B_3$ profiles of 10 kinds of animal based food and 10 different plant based food samples. In this study, 10 kinds of animal based food samples consisting of veal (veal steak fillet), chicken (breast), turkey meat (thigh), goat meat (leg, belly), lamb (leg, back, arm), mutton (belly), bovine meat (loin) and 10 different plant based food samples namely; barley, rye, wheat (bread), wheat (durum), oat, rice, dried pea, green lentil, red lentil and chickpea were studied by high performance liquid chromatography using post-column derivatization system. The presence rates of nicotinic acid and nicotinamide were determined in the meat samples as 30% and 70% and as 87% and 13% in the cereal and legume samples, respectively. Nicotinic acid levels were found in low amounts in the meat samples. The amounts of nicotinic acid in the cereal and legume samples were significantly higher than the meat samples. Consequently, the plant based foods such as cereals and legumes, with a ratio of 87% nicotinic acid presence, standout as the best source of nicotinic acid and encouraging regular intake of those cereals and legumes containing rich nicotinic acid would remove nicotinic acid deficiency in human.

  • The purpose of this investigation is to observe tha variation of vitamin $B_{12}$, nicotinic acid and folic acid contents in relation to the growing years of Panax Ginseng roots. The contents of the vitamins were estiamted microbiologically with Lactobacillus leichmannii, Lactobacillus arabinosus and Streptococcus faecalis, respectively. It is found that the content of vitamin $B_{12}$ in Panax Ginseng roots somewhat increases according to their growing years except 3-year-old roots. It is probable that the lower content in these roots should be due to the cultivating soil. The content of nicotinic acid in all the roots is not significantly different. The result of paper chromatography using the concentrated extract of the roots suggests that there exists some other substance besides nicotinic acid and nicotinamide, of which Rf value is 0.5 in comparison with the Rf 0.75 of nicotinic acid and nicotinamide. It is thought that this substance stimulate the growth of L. arabinosus. The content of folic acid is significantly different. The content level is the highest in the 4-year-old roots and the lowest in the 6-year-old roots.

Inhibiting Effect of Nicotinic Acid Hydrazide on Corrosion of Aluminum and Mild Steel in Acidic Medium

  • The corrosion behavior of aluminum and mild steel in hydrochloric acid medium was studied using a nicotinic acid hydrazide as inhibitor by potentiodynamic polarization, electrochemical impedance spectroscopy technique and gravimetric methods. The effects of inhibitor concentration and temperature were investigated. The experimental results suggested, nicotinic acid hydrazide is a good corrosion inhibitor for both aluminum and mild steel in hydrochloric acid medium and the inhibition efficiency increased with increase in the inhibitor concentration. The polarization studies revealed that nicotinic acid hydrazide exhibits mixed type of inhibition. The inhibition was assumed to occur via adsorption of the inhibitor molecules on the aluminum and mild steel surface and inhibits corrosion by blocking the reaction sites on the surface of aluminum.

Effects of Nicotinic Acid Deficiency on the Levels of Various Metabolites in the Serum of Quail (Nicotinic acid 결핍이 메추리 혈청의 여러 대사물질 수준에 미치는 효과)

  • Body weight gain in the niadn deficient group of quail was markedly lowered as compared to that of the control group, but heart, kidney and liver weight were slighdy reduced reladve to the body weight. Nicotinic acid deficiency resulted in the significant increase of serum glucose level but the serum cholesterol, albumin and total protein levels were not affected to any extent. Glutamic oxaloacetate iransaminase and glutamic pyruvate transaminase activities were significandy enhanced but alkaline phosphatase and lactic dehydrogenase activities were not influenced. Tryptophan and tyrosine levels were remarkably reduced and a similar observation was also made with aspartic acid, glutamic acid and alanine plus serine. However, the levels of basic amino acids such as arginine, histidine and lysine plus branched chain amino acids such as isoleucine, leucine and valine were not affected.

Synthesis of Sesamol Derivatives and Biological Activities (Sesamol 유도체의 합성 및 생물활성)

  • Sesamol, one of constituents isolated from sesame seed (Sesamum indicum L.) has been known to have natural antioxidant property. Nicotinic acid and clofibric acid have been used for treatment of hyperlipidemia. Sesamol derivatives prepared by conjugation with nicotinic acid (SJ-301) and clofibric acid (SJ-302) were expected to have various biological activity caused by synergistic effect of sesamol. Thus, this study were carried out to investigate the synthesis of sesamol derivatives from seasamol and antihyperlipidemia drugs and their biological activities. As a result, SJ-301 showed a stronger antihyperlipidemia activity in vivo and inhibitory effect on $TNF-{\alpha}$ release in vitro compared to the original agents such as sesamol, nicotinic acid and clofibric acid.

  • The kinetics of oxidation of nicotinic acid by peroxomonosulfate (PMS) has been studied in acetate buffers. Stoichiometry of the reaction corresponds to the reaction of one mole of the oxidant with a mole of nicotinic acid. N${\rightarrow}$O product has been confirmed both by UV visible and IR spectroscopy. The reaction is second order viz. first order with respect to each reactant. Activation parameters have also been evaluated. A plausible reaction mechanism is mentioned and the derived kinetic rate law accounts for experimental observations.

Transdermal Permeation Characteristics of Anti Cholesterol Ointment using Nicotinic acid N-oxide (Nicotinic acid N-oxide를 이용한 항콜레스테롤 연고의 경피 투과 특성)

  • Transdermal therapeutic system(TTS) is often used as the method of drug dosage into the epidermic skin. Natural polymer were selected as ointment material of TTS. We investigated the permeation of natural polymer ointment containing drug in rat skin using horizontal membrane cell model. Permeation properties of materials were investigated for water-soluble drug such as Nicotinic acid N-oxide in vitro. These results showed that skin permeation rate of drug across the composite was mainly dependent on the property of ointment base and drug. Proper selection of the polymeric materials which resemble and enhance properties of the delivering drug was found to be important in controlling the skin permeation rate. This result suggests a possible use of natural polymer ointment base as TTS of antihyperlipoproteinemic agent.
