Variations in Mitochondrial DNA of Pleurotus sajor-caju (여름느타리 버섯류의 미토콘드리아 DNA 비교)

  • Five strains of Pleurotus sajor-caju were collected from some countries including India and Papua New Guinea. Four strains produced brown fruitbody but the other strain from Papua New Guinea produced white fruitbody. Monokaryons obtained from both strains producing brown fruitbody and white fruitbody were mated each other. They showed different mating types such as brown strain of $A_1A_2B_1B_2$ and white strain of $A_3A_4B_3B_4$. DNAs were isolated from mycelia of five strains of P. sajor-caju. Mitochondrial DNA was seperated from nuclear DNA by bisbenzimide-CsCl ultracentrifugation. Digestion of the fungal mitochondrial DNA with Eco RI restriction endonuclease yielded from ten to fourteen fragments. Two strains of the five strains showed different restriction pattern of mitochondrial DNA from the other three strains. Summation of the fragment sizes gave a mitochondrial DNA size of about 60 to 65kb.

Occurrence of a New Type of Pseudomonas syringae pv. actinidiae Strain of Bacterial Canker on Kiwifruit in Korea

  • Pseudomonas syringae pv. actinidiae strains, the causal agents of bacterial canker on kiwifruit, were isolated from Korea and Italy in 2011. Among 87 isolates, a total of six representative strains, three from Korea and three from Italy, were identified on the basis of biochemical and physiological tests. Identities were confirmed by PCR using P. syringae pv. actinidiae-specific primers PsaF1/R2, which amplified a 280-bp DNA fragment. The strains isolated from Korea in this study displayed BOX-PCR patterns similar to those isolated from Italy but different from those isolated previously in Korea or the pathotype P. syringae pv. actinidiae strain. The effector hopA1 and hopH1 genes, which are known to be present in strains isolated recently from France and Italy, were also present in P. syringae pv. actinidiae strains, SYS1, SYS2 and SYS4, isolated from Korea in this work. However, no amplicons of the expected size were obtained from strains previously isolated from Korea and Japan. In addition, the Korean strains isolated in this work belonged to haplotype I for the cts gene identical to those strains isolated from recent outbreaks in Italy. These results suggest that P. syringae pv. actinidiae strains isolated from Korea and examined in this work are a new type of strain similar to those found from recent outbreaks in Italy. This is the first report on the occurrence of cts haplotype I strains of P. syringae pv. actinidiae affecting kiwifruit plants in Korea.

The Antioxidant Effect of Cheonggukjang, Fermented Using the New Strain, Bacillus amyloliquefaciens NBF11-1 (신 균주 Bacillus amyloliquefaciens NBF11-1을 이용하여 발효한 청국장의 항산화효과)

  • This study aims to compare and analyze the antioxidant effect of Cheonggukjang's traditional fermentation strain, Bacillus subtilis NG24 which was a control of the study, in order to see the biological activity effect of the new strain, Bacillus amyloliquefaciens NBF11-1 that was first found in the surface of the bamboo stem, but hasn't been insufficiently researched. In the antioxidant activity experiment of the Cheonggukjang extract, the B.amyloliquefaciens NBF11-1 sample showed a significant increase in the total polyphenol extract content, compared to B.subtilis NG24(p=0.032). Also, compared to B.subtilis NG24, the sample containing B.amyloliquefaciens NBF11-1 showed a significant increase in SOD-like Activity, DPPH radical scavenging, and NO radical scavenging, as the concentration rose(p<.05). Additionally, $IC_{50}$ in each antioxidant activity experiment significantly decreased in the B.amyloliquefaciens NBF11-1 sample like SOD-like Activity(p=0.045), DPPH radical scavenging(p=0.041), and NO radical scavenging(p=0.019), compared to B.subtilis NG24.

Optimal Temperature for H2 Production and Population Growth of the N2-fixing Unicellular Cyanobacterial Strains from Korean Coasts (한국 연안산 질소고정 단세포 남세균 종주의 최적 성장 및 수소생산 온도)

  • Photobiological hydrogen production by nitrogen-fixing unicellular cyanobacteria has long been considered to be an environmentally sound and very promising method for the future supply of renewable clean energy. Using six Korean nitrogen-fixing unicellular cyanobacterial strains and the Synechococcus sp. strain Miami BG043511 we performed cultivation experiments to find out the strain-specific optimal temperature for population growth and $H_2$ production. Under $20^{\circ}C$ the population growth of all the tested strains was significantly retarded in contrasts to the faster and higher growth under 25, 30 or $35^{\circ}C$. The highest growth rates in all the 7 strains were measured under $30^{\circ}C$ while the maximal biomass yields were under $30^{\circ}C$ (strains CB-MAL 026, 054, and 055) or $35^{\circ}C$ (strains 002, 031, 058, and Miami BG043511). The difference between the maximal biomass yields at $30^{\circ}C$ and $35^{\circ}C$ was not greater than 10%. The quantity of photobiologically produced $H_2$ was only slight larger under $35^{\circ}C$ than that under $20^{\circ}C$. Our result may suggest a two-step process of $H_2$ production which includes rapid and sizable production of biomass at $30^{\circ}C$ and the following high $H_2$ production at $20^{\circ}C$ by the test strains of marine nitrogen-fixing unicellular cyanobacteria.

Potential Evaluation and Health Fostering Intrinsic Traits of Novel Probiotic Strain Enterococcus durans F3 Isolated from the Gut of Fresh Water Fish Catla catla

  • Over the last few years, marine environment was found to be a source of surplus natural products and microorganisms with new bioactive secondary metabolites of interest which can divulge nutritional and biological impact on the host. This study aims to assess the possible, inherent and functional probiotic properties of a novel probiotic strain Enterococcus durans F3 (E. durans F3) isolated from the gut of fresh water fish Catla catla. Parameters for evaluating and describing the probiotics described in FAD/WHO guidelines were followed. E. durans F3 demonstrated affirmative results including simulated bile, acid and gastric juice tolerance with exhibited significant bactericidal effect against pathogens Staphylococcus aureus, Salmonella Typhi, Escherichia coli and Pseudomonas aeruginosa. This can be due to the enterocin produced by E. durans F3 strain, which was resolute by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) gel with amplification of the anticipated fragment of a structural gene; enterocin A, followed by antibiotic susceptibility assessment. Effective antioxidant potentiality against ${\alpha}$-diphenyl-${\alpha}$-picrylhydrazyl free radicals including lipase, bile salt hydrolase activity with auto-aggregation and cell surface hydrophobicity was similarly observed. Results are proving the potentiality of E. durans F3, which can also be used as probiotic starter culture in dairy industries for manufacturing new products that imparts health benefits to the host. Finding the potent and novel probiotic strains will also satisfy the current developing market demand for probiotics.

Introduction of a speedy growing trait into Pleurotus eryngii by backcrossing (큰느타리(새송이)버섯에서 역교배를 통한 속성형질의 도입)

  • Two strains which have good traits in quality and speedy growing were selected based on previous report(Ryu, 2007) to breed a new strain carrying the traits in it. KNR2322 carrying high quality and KNR2503 carrying a speedy growing trait were cultivated to harvest monokaryons. The monokaryotic mycelia were characterized in growth rate, color, and stability. Selfing of monokaryons from KNR2322 and crossing between KNR2322- and KNR2503-derived monokaryons were performed. Two parental lines, $KNR2322-4{\times}37-6{\times}12(B)$ showing thremmatological values in weight and quality, 68.5g and 6.5 and $KNR2322-30{\times}KNR2503-60(A)$ was harvested in 13.5 days after scraping old medium were selected. Crossing between A and B resulted in two high quality and speedy growing strain, A8B8 and A8B10. Their days for harvest after scraping, weight, and quality were 14.0 days, 88.8g and 92.7g, 7.0 and 7.1. Originality tests were conducted with genomic DNAs of A8B8 and Keuneutari 3ho(commercial strain) by DNA fingerprinting a confrontation culture. The results showed polymorphism and a inhibition line between them. It suggested that the new strain have originality from the commercial strain. A8B8 have been registered in Korea Seed and Variety Service with as "Saesongi 1ho".

A New Methodology for the Assessment of Liquefaction Potential Based on the Dynamic Characteristics of Soils (I) : A Proposal of Methodology (지반의 동적특성에 기초한 액상화 평가법(I) : 이론제안)

  • In this study, a new methodology for the assessment of liquefaction potential is proposed. Since there is no data on the liquefaction damage in Korea, the dynamic behavior of fully saturated soils is characterized through laboratory dynamic tests. There are two experimental parameters related to the soil liquefaction resistance characteristics : the one is the index of disturbance determined by $G/G_{max}$ curve and the other is a plastic shear strain trajectory evaluated from stress-strain curve. The proposed methodology takes advantage of the site response analysis based on real earthquake records to determine the driving effect of earthquake. In the evaluation of liquefaction resistance characteristics, it is verified experimentally that the magnitude of cyclic shear stress has no influence on the critical value of plastic shear strain trajectory at which the initial liquefaction occurs. Cyclic triaxial tests under the conditions of various cyclic stress ratios and torsional shear tests are carried out far the purpose of verification. Through this study, the critical value at the initial liquefaction is found unique regardless of the cyclic stress ratio. It is also f3und that liquefaction resistance curve drawn with disturbance and plastic shear strain trajectory can simulate the behavior of fully saturated soils under dynamic loads.

Characterization of a new commercial strain 'Gumbit' in Pleurotus cornucopiae var. citrinopileatus (노랑느타리 품종 '금빛'의 특성)

  • A new commercial strain "Gumbit" of golden oyster mushroom was developed by hyphal anastomosis. It was improved with hybridization between monokaryotic strain derived from ASI 2295 and ASI 2703. The optimum temperature of mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $19{\sim}24^{\circ}C$, respectively. The pileus was golden to brilliant yellow color. Commercial strain "Gumbit" was not as prolific as the more commonly cultivated Pleurotus ostreatus in the conversion of substrate mass to mushrooms. However, cultivator can save money for mushroom growing on high-temperature season as like summer in Korea. Since picking individual mushrooms is tedious and often damages the fragile fruiting bodies compared with other species of oyster mushrooms.

  • A new approach has been proposed for the incremental analysis of the nonsteady state large deformation of planar anisotropic elastic-plastic sheet forming. A mathematical brief review of a constitutive law for the incremental deformation theory has been presented from flow theory using the minimum plastic work path for elastic-plastic material. Since the material embedded coordinate system(Lagrangian quantity) is used in the proposed theory the stress integration procedure is completely objective. A new return mapping algorithm has been also developed from the general midpoint rule so as to achieve numerically large strain increment by successive control of yield function residuals. Some numerical tests for the return mapping algorithm were performed using Barlat's six component anisotropic stress potential. Performance of the proposed algorithm was shown to be good and stable for a large strain increment, For planar anisotropic sheet forming updating algorithm of planar anisotropic axes has been newly proposed. In order to show the effectiveness and validity of the present formulation earing simulation for a cylindrical cup drawing and front fender stamping analysis are performed. From the results it has been shown that the present formulation can provide a good basis for analysis for analysis of elastic-plastic sheet metal forming processes.

A Study on the Coating Cracking on a Substrate in Bending II : Experiment (굽힘모드하에서의 코팅크랙킹의 분석II: 실험)

  • Fracture analysis of coating cracking on a substrate system described in a companion paper was applied and verified by four-point bending tests. The multiple cracking of coating was predicted using a fracture mechanics approach. The strain energy release rate (G) due to the formation of a new crack in a coating was obtained. A crack density vs. strain data of metallic and polymeric substrate was used to get the in-situ fracture toughness of coating with respect to various baking time and temperature. The $G_c$ was decreased as the baking temperature and time was increased. This paper gave insight about usefulness of four-point bending test for fracture toughness evaluation of coating and it gave a new method for in-situ coating toughness.
