Biocompatibility of Poly(MPC-co-EHMA)/Poly(L-1actide-co-glycolide) Blends

  • Poly(L-lactide-co-glycolide)(PLGA) was blended with poly[$\omega$-methacryloyloxyethyl phospho-rylcholine-co-ethylhexylmethacrylate (PMEH)] (PLGA/PMEH) to endow with new functionality i.e., to improve the cell-, tissue- and blood-compatibility. The characteristics of surface properties were investigated by measurement of contact angle goniometer, Fourier-transform infrared spectroscopy with attenuated total reflectance (FTIR-ATR) and electron spectroscopy for chemical analysis (ESCA). NIH/3T3 fibroblast and bovine aortic endothelial cell were cultured on control and PLGA/PMEH surfaces for the evaluation of ceil attachment and proliferation in terms of surface functionality such as the concentration of phosphoryl-choline. Also, the behavior of platelet adhesion on PLGA/PMEH was observed in terms of the surface functionality. The contact angles on control and PLGA/PMEH surfaces decreased with increasing PMEH content from 75$^{\circ}$ to about 43$^{\circ}$. It was observed from the FTIR-ATR spectra that phosphorylcholine groups are gradually increased with increasing blended amount of MPC. The experimental P percent values from ESCA analysis were more 3.28∼7.4 times than that of the theoretical P percent for each blend films. These results clearly indicated that the MPC units were concentrated on the surface of PLGA/PMEH blend. The control and PLGA/PMEH films with 0.5 to 10.0 wt% concentration of PMEH were used to evaluate cell adhesion and growth in terms of phosphorylcholine functionality and wettability. Cell adhesion and growth on PLGA/PMEH surfaces were less active than those of control and both cell number decreased with increasing PMEH contents without the effect of surface wettability. It can be explained that the fibronectin adsorption decreased with an increase in the surface density of phosphorylcholine functional group. One can conclude the amount of the protein adsorption and the adhesion number of cells can be controlled and nonspecifically reduced by the introduction with phosphorylcholine group. Morphology of the adhered platelets on the PLGA/PMEH surface showed lower activating than control and the number of adhered platelets on the PLGA/PMEH sample decreased with increasing the phosphorylcholine contents. The amount of fibrinogen adsorbed on the PLGA/PMEH surface demonstrated that the phospholipid polar group played an important role in reducing protein adsorption on the surface. In conclusion, this surface modification technique might be effectively used PLGA film and scaffolds for controlling the adhesion and growth of cell and tissue, furthermore, blood compatibility of the PLGA was improved by blending of the MPC polymer for the application of tissue engineering fields.

Early bone healing onto implant surface treated by fibronectin/oxysterol for cell adhesion/osteogenic differentiation: in vivo experimental study in dogs

  • Purpose: This study aimed to evaluate the effects of fibronectin and oxysterol immobilized on machined-surface dental implants for the enhancement of cell attachment and osteogenic differentiation, on peri-implant bone healing in the early healing phase using an experimental model in dogs. Methods: Five types of dental implants were installed at a healed alveolar ridge in five dogs: a machined-surface implant (MI), apatite-coated MI (AMI), fibronectin-loaded AMI (FAMI), oxysterol-loaded AMI (OAMI), and sand-blasted, large-grit, acid-etched surface implant (SLAI). A randomly selected unilateral ridge was observed for 2 weeks, and the contralateral ridge for a 4-week period. Histologic and histometric analyses were performed for the bone-to-implant contact proportion (BIC) and bone density around the dental implant surface. Results: Different bone healing patterns were observed according to the type of implant surface 2 weeks after installation; newly formed bone continuously lined the entire surfaces in specimens of the FAMI and SLAI groups, whereas bony trabecula from adjacent bone tissue appeared with minimal new bone lining onto the surface in the MI, AMI, and OAMI groups. Histometric results revealed a significant reduction in the BIC in MI, AMI, and OAMI compared to SLAI, but FAMI demonstrated a comparable BIC with SLAI. Although both the BIC and bone density increased from a 2- to 4-week healing period, bone density showed no significant difference among any of the experimental and control groups. Conclusions: A fibronectin-coated implant surface designed for cell adhesion could increase contact osteogenesis in the early bone healing phase, but an oxysterol-coated implant surface designed for osteoinductivity could not modify early bone healing around implants in normal bone physiology.

Characterization of Pv92, a Novel Merozoite Surface Protein of Plasmodium vivax

  • The discovery and understanding of antigenic proteins are essential for development of a vaccine against malaria. In Plasmodium falciparum, Pf92 have been characterized as a merozoite surface protein, and this protein is expressed at the late schizont stage, but no study of Pv92, the orthologue of Pf92 in P. vivax, has been reported. Thus, the protein structure of Pv92 was analyzed, and the gene sequence was aligned with that of other Plasmodium spp. using bioinformatics tools. The recombinant Pv92 protein was expressed and purified using bacterial expression system and used for immunization of mice to gain the polyclonal antibody and for evaluation of antigenicity by protein array. Also, the antibody against Pv92 was used for subcellular analysis by immunofluorescence assay. The Pv92 protein has a signal peptide and a sexual stage s48/45 domain, and the cysteine residues at the N-terminal of Pv92 were completely conserved. The N-terminal of Pv92 was successfully expressed as soluble form using a bacterial expression system. The antibody raised against Pv92 recognized the parasites and completely merged with PvMSP1-19, indicating that Pv92 was localized on the merozoite surface. Evaluation of the human humoral immune response to Pv92 indicated moderate antigenicity, with 65% sensitivity and 95% specificity by protein array. Taken together, the merozoite surface localization and antigenicity of Pv92 implicate that it might be involved in attachment and invasion of a merozoite to a new host cell or immune evasion during invasion process.

급성 후방 십자 인대 파열의 관절경하 봉합술 및 자가 슬괵 이식건 보강술 - 수술 술기 - (Arthroscopic Repair of Acute Posterior Cruciate Ligament Rupture with Autogenous Hamstring Tendon Graft Augmentation - Technical Note)

  • 목적: 대퇴 근위 부착부가 파열된 급성 후방 십자 인대 파열에 있어서 후방 경격막 도달법을 이용한 관절경적 봉함술 및 보강술을 소개하고자 한다. 수술 술기: 슬관절의 관절경적 기본검사 후, 후내측 도달법과 후외측 도달법을 개설하여 후방 경격막 도달법을 확보한 후 파열되어 후방으로 이동된 후방 십자 인대 다발을 후내측 관절경적 시야 하에서 전내측에서 삽입한 Suture hook을 이용하여 봉합한다. 이 봉합사를 전내측으로 빼내어 견인하여 파열된 인대 다발을 과간 절흔으로 보낸 후 전방에서 추가 봉합을 시행한다. 남아 있는 후방 십자 인대 다발을 보존하면서 경골 및 대퇴골 터널을 만든 후에 자가 단일 다발 반건양 및 박 이식건을 터널을 통해 관절 내로 통과시키고 대퇴 터널을 통과 시킨 후방 십자 인대 봉합사과 함께 고정한다. 결론: 급성 후방 십자 인대 손상 시 특히, 대퇴 근위 부착부 파열의 경우 파열된 다발을 후방에서 관절경적 봉합술을 시행할 수 있으며 후방 경격막 도달법을 이용하여 남아있는 다발에 최소한의 손상을 주면서 경골 및 대퇴골 터널을 만들고 자가 슬괵 이식건을 보강함으로써 좋은 결과가 기대된다.

외측 반월상 연골 경골 후방 부착부 파열의 관절경적 All-Inside 봉합술 - 수술술기 - (Arthroscopic All Inside Repair of Lateral Meniscus Root Tear -Technical note-)

  • 목적: 전방 십자 인대 손상과 동반된 외측 반월상 연골 경골 후방 부착부 완전 방사상 파열 환자에서 전외측 및 전내측 도달법을 통한 all-inside 봉합 술기를 시술하여 온 바, 이를 소개하고자 한다. 수술 술기: 관절경을 전내측 도달법으로, 봉합용 갈고리(suture hook, $Linvatec^{TM}$, Largo, Florida, USA)를 전외측 도달법으로 관절에 삽입한 상태에서 봉합용 갈고리를 틀어서 외측으로 전위된 외측 반월상 연골 후각부 파열단의 대퇴골측 면에서 경골측 면으로 수직으로 통과시킨다. 갈고리 내로 PDS No. 1 ($Ethicon^{TM}$, Somerville, NJ, USA)를 통과시킨 후 봉합용 갈고리를 빼내고 전외측 도달법 입구로 봉합사의 양끝을 뽑아낸다. MAXON 2-0 ($Syneture^{TM}$, Norwalk, Connecticut, USA)를 봉합용 갈고리를 이용하여 경골 부착부 파열단 경골측 면에서 대퇴골측 면으로 봉합사를 수직으로 통과시켜 양끝을 전외측 도달법 입구로 빼내어 두 봉합사의 경골 측 끝 중 PDS를 MAXON에 결찰하여 MAXON의 대퇴골측 끝을 전외측 도달법으로 당겨 이를 PDS로 교체한 뒤 SMC(Samsung Medical Center) knot를 이용하여 결찰한다. 결론: 본 술기를 시행하여 외측 반월상 연골 후방 부착부 파열을 해부학적으로 봉합할 수 있으며 또한 반월상 연골의 연골 장력(hoop tension)을 효과적으로 회복시켜 줄 수 있을 것으로 생각된다.

대학생들의 메가 스포츠이벤트에 대한 의식변화 (University Students' Awareness of the Changes in Mega Sports Events)

  • 본 연구는 메가 스포츠이벤트가 대구지역 대학생에게 미치는 의식변화를 알아보는데 그 목적이 있다. 이러한 연구 목적을 달성하기 위해 2011 대구세계육상선수권대회에 직 간접적으로 참여한 대학생 남, 여 228명을 모집단으로 선정하여 설문조사를 하였다. 통계자료 처리는 t-test 검정, one-way Anova 분석을 실시하였으며, 이들 분석결과를 토대로 나타난 연구 결론은 다음과 같다. 첫째, 참가열정 정도에 따른 관여도, 지각된 편익, 지각된 비용, 지지 지속성은 그룹 간에 유의한 차이가 있는 것으로 나타났다. 둘째, 지역사회 애착심에 따른 관여도, 지각된 편익, 지각된 비용,지지 지속성은 대학생들의 애착심과 관여도는 높지 않았으며, 경제적인 부분은 매우 민감하게 인식하였고, 향후 대회의지지 지속성은 메가 스포츠이벤트의 개최를 원하는 것으로 나타났다. 셋째, 참가경험 유 무에 따른 관여도, 지각된 편익, 지각된 비용, 지지 지속성은 참여 경험자 그룹이 비경험자 그룹보다 더 높게 나타났다.

치주 큐렛의 절단 연 마모도 평가 (Evaluation of the wear of the periodontal curet's cutting edge)

  • The quality of periodontal instrument cutting edge is a basic element of effective root planing procedure. Using instruments, the sharp edge is changed into blunt or beveled edge. With the blunt instrument, the periodontal treatment can't be carried into accuracy and effective. The study on the wear of periodontal curet is insufficient, there are few publications about the change of sharpness of cutting egde after using instrument and a certen reports were published on the study of scanning electron microscope(SEM) examination. In this study, to declare the number of strokes for sharpening of instruments, the changes of cutting edge is measured by the clinical methods, tactile sensitivity examination and refraction light-white line test after scaling strokes and root planing strokes. SEM test was added for defined the changes of cutting edges. The 7/8 Gracey curets that have been never used was tested. Maxillary molars which were extracted from the School of Dental Medicine, Dankook University was used. Subjected teeth had attachment loss more than 6 mm in bucca-lingual surface and sufficient calculus of a band type in cervical area. The strokes of curet were executed 3, 5, 7, 9, 11, 13 times on scaling stroke and 10, 15, 20, 25, 30, 35 times on root planing stroke. A resident has periodontal experience over 3 years carried out the clinical examinations those tactile sensitivity examination and refraction light-white line test 5 times. The case there being tactile sensitivity certenly is 2, the case being felt tactile sensitivity is 1, and the case there not being tactile sensitivity is 0. The visual examination was recorded as following. The case that refracted white line is not recognised is 2, the case that uncerten is 1, and the case that acknowledged is 0. The results were obtained as follows. 1. After scaling strokes, the tactile sensitivity was reduced after 11 strokes and disappeared in 13 strokes. 2. In tactile sensitivity after root planing procedures, sensitivity was reduced after 25 strokes and disappeared in 35 strokes. 3. In case of visual examination, the detection of refracted white line was increased after 9 strokes of scaling procedures and the accuracy of wear wasn't showed after root planing procedures. 4. In SEM, metal projection was observed on new periodontal curet cutting edge and it was disappeared after scaling procedures. 5. In SEM, the cutting edge was showed changing linear into an aspect of the surface after 5 strokes of scaling procedures and 10 strokes of root planing procedures and showed beveled edge in 11 strokes of scaling procedures, 25 strokes of root planing procedures. The results of 3-type examination indicated that the sharpening of curet should be performed after 11 strokes of scaling procedures and 25 strokes of root planing procedures.

무치악 환자에서 기성 조립식 bar를 이용한 임플란트 피개의치 증례 (Implant-retained overdentures with pre-fabricated bar attachment system in edentulous patients)

  • 치아 상실에 이어 심각한 치조골 소실을 보이는 무치악 환자들은 전통적인 방식의 총의치를 사용하는데 어려움이 많다. 이러한 문제를 보완할 수 있도록 임플란트를 이용한 피개의치가 사용되고 있다. SFI-bar$^{(R)}$는 진료실에서 직접 조정이 가능한 기성품으로 복잡한 기공과정을 필요로 하는 기존의 바 시스템보다 간결하여 시간과 비용을 절감할 수 있다. 첫 번째 증례는 55세 남자 환자로 기존 하악 임플란트-지지 피개의치가 파절되었다는 주소로 내원하였고 내구성이 있으며 통증이 없는 의치를 원하였다. 파절된 Locator$^{(R)}$ 어태치먼트를 제거 후 바를 이용한 피개의치로 치료하였다. 두 번째 증례는 파킨슨병을 앓고 있는 77세 여자 환자로 하악 치조골이 심각하게 흡수되어 의치가 자주 탈락한다는 주소로 내원하였고 통증이 없고 유지력이 있는 의치를 원하였다. 하악 전치부에 두개의 임플란트를 식립한 후 임플란트에 연결된 어댑터에 바를 연결하였다. 그 후 바에 맞는 여성부를 새로운 의치에 장착하여 치료하였다. 2년의 추적 관찰 결과 상기 환자 모두 심미적, 기능적으로 만족할만한 결과를 보였으므로 증례를 보고하고자 한다.

양식 전복(Haliotis discus hannai)으로부터 분리된 Vibrio harveyi의 생화학적 특성 및 병원성 (The Pathogenicity and Biochemical Characteristics of Vibrio harveyi Isolated from the Pacific Abalone, Haliotis discus hannai)

  • 최근 어린 전복의 대량 폐사가 국내의 전복 종묘 양식장에서 발생하였다. 죽은 어린 전복의 마리 수는 전체 사육 마리 수의 약 50% 이상이었으며, 그 크기는 각장 3 cm 전후였다. 폐사의 현상은 어린 전복이 부착 기질로부터 탈락되어 힘이 없이 바닥에 깔려 있거나, 뒤집어진 상태에서 스스로 일어나지 못하였다. 이러한 개체들은 대부분 근육에 수포를 형성하고 있는 병리학적 특징을 나타내었다. 폐사 직전의 전복으로부터 3개의 균이 분리되었으며, 이들은 16S rDNA 염기서열 분석에 의해 모두 V. harveyi로 동정되었다. 또한 이들을 어류에서 분리된 V. harveyi 36개의 균들과 염기서열을 비교 분석한 결과, 어류에서 분리된 V. harveyi는 genogroup a와 b로 구별되며, 전복에서 분리된 3균주는 genogroup a와 새로운 genogroup c에 속하였다. 그중 WA AG-1과 WA CS-5 균을 건강한 전복에 인위 감염시킨 결과, 자연 감염된 개체와 같은 외부 및 병리조직학적 증상을 나타내면서 폐사하였으며, 그 반수 치사농도는 각각 $1.0{\times}10^3cfu\;animal^{-1}$$1.7{\times}10^4cfu\;animal^{-1}$이었다.

북한이탈여성들의 심리사회적자원에 관한 질적사례연구 (Qualitative Case Study on Psychosocial Resources of North Korean Female Defectors Living in South Korea)

  • According to the survey by the Ministry of Unification, 28,133 live in South Korea as of september 2015 and the number has been steadily increased. It is interesting to note that the majority of defectors were women. Statistics say that the total of women defectors was 1,138 in 2002 and consisted of 55.5%. But the number increased to 2,706 in 2011 and consisted of 70.5%(Wikipedia, 2015). Most of them have trauma, because they experienced terrible events like prostitution, sexual abuse, domestic violence, etc. while they were living in South Korea. They had to get over a lot of challenges they never expected. They had to do whatever they need to do for their survival in South Korea as well as in North Korea. Objectives: This study focuses on identifying the psychosocial resources available for them in order to help them out living here. Method: For this purpose, this study conducted unstructured in-depth interviews with four women defectors from North Korea. Their age bracket is between the ages of 50 and 58 and they have stayed over five years in South Korea. They were recommended by social welfare workers, who had dealt with them over 20 years. For the reliability of this study, the three methods were performed such as three-month close relationship, three-person triangular verification, and diversification of resources-the field notes and observation notes. Results: As a result of this study, the resources may be labeled into three main clusters of themes such as 1) Self-Preservation - 'Self-Love', 'Family-Attachment', 'Share the Goods', 'Live in Harmony', 'Avoidance of conflict'; 2) Self-Conquest - 'Endurance', 'Earnestness and Effort', 'My own Activities', 'Chat', 'Put Down', 'Appreciation and Optimism'; 3) Self-Presence Awareness - 'I was a Precious Daughter', 'I am Mother'. Also there are thirteen sub categories to be considered. Conclusions: This study focuses on identifying how women defectors have used or can use the psychosocial resources available for them, rather than what they have struggled with. It is meaningful to identify in a positive light that this study provides how they have adjusted to a totally different context for their survival and what psychosocial resources they have used. This study can give an idea about how they may receive proper and practical help from the government programs in order to become better used to new living circumstances in South Korea. As well, this study can provide meaningful criteria and guidance with family life educators for women defectors and social workers working in counseling and social welfare areas. This study contributes to better understanding about how they are using these psychosocial resources.
