It was observed that the hot-water extract of the bark of Kalopanax pictus N. had the highest anti-complementary activity among the 11 kinds of forest materials. Methanol-and ethanol-soluble portions had low anti-complementary activities, but crude polysaccharide. HKP-0 had a high activity of 80%. HKP-0 contained 54.8% of total sugar and 27.9% of protein. The neutral sugars of HKP-0 consisted of mainly arabinose, galactose and glucose. HKP-4 fraction obtained by cetavlon treatment of HKP-0 showed the highest anti-complementary activity of 90%. The activity was not changed by pronase digestion bu decreased greatly by periodate oxidation. HKP-4 consisted of mainly arabinose and glucose with molar ratio of 1.0 : 22.4, HKP-4-I, an unabsorbed fraction from HKP-4 on DEAE Sepharose CL-6B column showed higher yield and activity than those of absorbed fractions. HKP-4-I was homogeneous, and its molecular weight was about 25,000. HKP-4-I contained 84.0% of neutral sugar and consisted of arabinose and glucose with molar ratio of 1.0 : 11.2. The anti-complementary activity of HKP-4-I was not decreased by the treatment of polymyxin B, and the polysaccharide activated both classical and alternative pathway in complement system. Void volume fraction obtained from HKP-4-I hydrolyzed with ${\alpha}$-amylase on Sephadex G-25 column only had a high anti-complementary activity.
The heat decomposition rate of guanosine-5'-monophosphate was investigated in pH range from 5.52 to 7.00, and 0.1 of the ionic strength. The result showed that the rate was a first-order reaction and the rate of guanosine-5'-monophosphate loss was maximum near $pKa_2$. The loss of guanosine-5'-monophosphate was temperature dependent and followed to the Arrhenius equation in the temperature region from $93^{\circ}C$ to $108^{\circ}C$. The rate constant as function of temperature ($93^{\circ}C$ to $108^{\circ}C$) and neutral pH($pKa_2$, 6.0, to 7.0) was correlated by least-square fit of the experimental data; $$K=4.19{\times}10^{26}\;{\exp}\;[-1.3(pH+E/RT)]$$
The browning reaction of sugar derivatives, fructo-oligosaccharide, high maltose syrup(HMS), sorbitol and maltitol, and their effect on the appearance of jam and candy were investigated. The spectrophotometrie scanning of the absorbance between 230 nm and 700 nm could demonstrate the heat induced browning of the sugar derivatives. Fructo-oligosaccharide and HMS showed sharp increase in absorbance at 270-330 nm range by heating at $100{\sim}120^{\circ}C$ for 1 hr but sorbitol and maltitol did not show the increase in absorbance. When the pH was lowered red from neutral to 2.0, the absorbance of HMS and sucrose increased sharply, showing that these substances are relatively unstable in acidic heating compared to fructo-oligosaccharide. The addition of glycine enhanced the browing reaction of fructo-oligosaccharide and HMS, whereas little change was observed with sucrose, sorbitol and maltitol. These browning characterisitcs of sugar derivatives were reflected to the color development of apple jam and candy where they were used. Both fructo-oligosaccharide and HMS increased the yellowness of these products, while sugar alcohols reduced the yellowness compared to sugar.
A sensitive and quantiative method for the structural analysis of oligosaccharide was established for the glycoform analysis of glconproproteins. Inthis study, n-linked oligosaccharides of human IgG and bovine transferin were analyzed for the evaluation of the methydrate moiety ofthe method. Chrbohydrate moiety of glycoprotein was relased by hydrazinolysis and purified by paper chromatography. The oligosaccharides were labeled with a fluorescent bye, 2-aminobenzamide, for the enhancement of detection sensitivity. sialylated (acidic) oligosaccharides were separated from neutral oligosaccharide by employing a strong anion-exchange column(MonoQ) followed by the treatment with sialidase. Enzymatically desiayated fractions and neutral fractions of oligosaccharides were applied to normal-phase HPLC to resolve the peaks according to glucose unit (GU). The structure of separated molecules was further determined by sequential digestion with exoglycosidases. As a result, disialylated biantennary complextype oligosaccharide was found to be a major sugar chain in bovine transferrin (63%). In human IgG, core fucosylated asialobiantennary complex oligosaccharides were dominant. These results coincided well with reported results.
Fermented kale juices using four types of lactobacilli were produced in the present study. After 48 h of fermentation time, viable cell counts of all ferments reached an above $10^9CFU/mL$. The viability of the ferments after cold storage in the refrigerator for 4 weeks showed $10^8CFU/mL$ in all ferments. Among four types of fermented kale juices, the ferment of Lactobacillus acidophilus IFO 3025 indicated a good nutritional composition, including neutral sugar ($1,909.76{\mu}g/mL$), reducing sugar ($564.00{\mu}g/mL$, P<0.05), and protein contents ($160.06{\mu}g/mL$, P<0.05). The results of mineral composition analysis had the highest potassium value in all ferments ($854.16{\sim}895.07{\mu}g/mL$), particularly in the ferment of Lactobacillus brevis FSB-1 (P<0.001), which is necessary to sustain osmotic pressure, prevention of high blood pressure, and protein synthesis. Moreover, calcium, phosphorus, and magnesium contents related to bone health were generally sufficient in all ferments. Consequently, in this study, fermented kale juices may be suggested as a healthy fermented beverage with essential nutrients. However, the acceptability of the fermented kale juice to the Korean taste should be further investigated with a trained taste panel to determine whether inoculated fermentation could be an option for the consumers.
The objective of this study was to investigate the enolization reaction and the antioxidant activity of caramelization products (CPs) obtained by caramelization browning of glucose and fructose solutions prepared at a pH ranging from 7.0 to 12.0 at varying temperatures ($80-180^{\circ}C$). The degradation of sugars rapidly increased at a high alkaline pH (10.0-12.0), and fructose degraded more rapidly than glucose (p<0.05). As the pH increased, the degree of sugar enolization was higher in fructose than in glucose. Browning and the formation of intermediate degradation products increased with the increase in heating temperatures. The browning development was dependent upon the type of sugar, and it was generally higher at alkaline pH than at neutral pH. The reducing power and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of the CPs increased with the increase in browning and formation of large amounts of intermediates. Therefore, the CPs with pronounced antioxidant activity can be prepared by heating fructose or glucose solutions that have a very alkaline pH to high temperatures.

Fiber dimensions, their derived values and chemical properties of cotton stalks (Gosypium hirsutum L), jute stick (Corchorus capsularis) and dhancha (Sesbania aculeate) have been examined to assess their suitability for paper production. Cotton stalks have a good derived values especially slender ratio, which is comparable to hardwood. The flexibility coefficient of these three non wood plants is better than hardwood. Anatomical analysis shows higher percentage of fibers and vessels than in general non wood plants. Lignin, $\alpha$-cellulose and pentosan contents in these three nonwood plants are within the range of hardwood. Neutral sugar analysis of cotton stalks, jute stick and dhancha shows that the glucose in the major sugar followed by xylose and mannose. The arabinose and galactose are present in minor amount. Alkaline nitrobenzene oxidation of cotton stalks, jute stick and dhancha wood meal exhibits that these nonwood plant lignins mainly consist of syringyl (S) and guaiacyl (V) units. The S/V ratios are 1.6, 1.2 and 2.1 for cotton stalks, jute stick and dhancha, respectively.
A bacterial strain capable of producing a novel bioflocculant was isolated from a biofilm sample and identified as Bacillus megaterium G31. The highest biopolymer yield was achieved when the organism was cultivated in a medium containing acetate as the sole carbon source and ($NH_4)_2HPO_4$as the nitrogen source. In kaolin suspension, the flocculating activity was highest at 170 mg I$^{-1}$ and decreased at the higher bioflocculant concentrations. The crude bioflocculant produced by the organism was purified by ethanol precipitation and gel permeation chromatography. The FTIR spectrum of the purified bioflocculant revealed that the bioflocculant might be a heterogeneous polysaccharide composed of hexosamines and neutral sugars. The analysis of sugar components of the bioflocculant using high performance anion-exchange chromatography showed that the sugar constituents of the bioflocculant were glucosamine, fucose, galactosamine, galactose, glucose, mannose in approximate molar ratio of 4 : 1 : 6 : 3 : 8 : 19. Its flocculating activity was stimulated by various cations. The bioflocculant was thermo-stable and retained 64% of its original activity after heating at $100^{\circ}C$ for 50 min.
This study was conducted to examine the physiochemical characteristics (PC) of a sausage with added dietary fiber and stevioside mixture, as well as the blood properties (BP) of rats with feeding of the sausage. There were no significant differences between the control and the treatments in the PC; in case of treatments, as additive contents of dietary fiber (1-3%) and stevioside (0.05-1.5%), redness ($a^*$) gradually increased, but yellowness ($b^*$) rapidly decreased (p<0.05). The pH level and water holding capacity (WHC) also tended to decrease. Thiobarbituric acid reactive substances (TBARS) were of little change. However, the texture of sensory evaluation and the adhesiveness of texture analysis had significantly low values (p<0.05). BP, such as blood sugar, high density lipoprotein (HDL) cholesterol, low density lipoprotein (LDL) cholesterol and triglyceride (TG), did not increase and total cholesterol (TC) actually decreased (p>0.05). However, body weights of rats increased with treatments. Overall, it is suggested that adding dietary fiber and stevioside mixture to sausages can moderately improve the redness, and also reduce the levels of sugar, cholesterol and neutral lipids in the blood without noticeably affecting the WHC, oxidant stability, appearance, and taste.
The physicochemical and in vitro physiological properties of soluble dietary fiber (SDF) from wax gourd (Benincasa hispida) pulp and peel were investigated. The pulp was composed of 11.4% SDF and 24.3% insoluble dietary fiber (IDF), while the peel contained 3.2% SDF and 43.3% IDE The predominant sugar in the SDF of the wax gourd pulp and peel was uronic acid, followed by galactose and rhamnose. The SDFs from the wax gourd pulp and peel gave similar elution patterns, with 4 main neutral sugar and uronic acid peaks eluted by 0.4, 0.5, 1, and 2 M ammonium acetate buffer. The pulp SDF had a much higher glucose retardation index (GRI) than the peel SDF for all measurement times. The pulp SDF showed strong growth-inhibiting activity against Escherichia coli and Clostridium perfringens, whereas the peel SDF produced strong growth-promoting activity against Bifidobacterium longum, Bifidobacterium infantis, and Lactobacillus brevis when compared to glucose.
