To elucidate the neuromodulation of neuropeptide Y and $\alpha,\;\beta$-methylene ATP on the neurogenic contraction of electrical perivascular nerve stimulation and the contractile response of noradrenaline from polygraph in the isolated renal artery of rabbit. 1. The neurogenic contraction induced by perivascular nerve stimulation was the voltage-dependent manner(10-100V) in the isolated renal artery of rabbit. 2. Neuropeptide Y(0.1uM) and $\alpha,\;\beta$-methylene ATP(1uM) increased the contractile responses of noradrenaline in the isolated renal artery of rabbit. 3. Neuropeptide Y(0.1uM) and $\alpha,\;\beta$-methylene ATP(1uM) increased the neurogenic contraction of electrical perivascular nerve stimulation in the isolated renal artery of rabbit. These results suggest that neuropeptide Y and $\alpha,\;\beta$-methylene ATP neuromodulated on the neurogenic contraction of electrical perivascular nerve stimulation on the isolated renal artery of rabbit.
The purpose of this study was to investigate the functional involvement of sympathetic nerve in the control of the microcirculation in the dental pulp with the aim of elucidation of the involvement of neuropeptides and sympathetic nerve in neurogenic inflammation. Experiments were done on the 7 cats anesthetised with sodium pentobarbital, and sympathetic nerve to the' dental pulp was stimulated electrically (10 Hz, 4 V, 1.5 ms, 3.5 mins). Ana-adrenoceptor antagonist phentolamine and a neuropeptide Y antagonist D-myo-inositol-1,2,6-trisphosphate (PP56) were injected close intra-arterially into the dental pulp without changing the systemic blood pressure. The probe of laser Doppler flowmeter was placed on the buccal surface of ipsilateral canine teeth to the stimulation, and pulpal blood flow was measured. Stimulation of the sympathetic nerve decreased pulpal blood flow by $55.24{\pm}7.74\;%$ (mean${\pm}$SEM, n = 13). Stimulation of the sympathetic nerve following the injection of the ${\alpha}$-adrenoceptor antagonist phentolamine ($0.1{\mu}g$/kg) caused decrease of pulpal blood flow by $14.35{\pm}3.43%$ (mean${\pm}$SEM, n=5). Phentolamine attenuated the sympathetic nerve-induced pulpal blood flow decrease by $74.02{\pm}9.32%$ (mean${\pm}$SEM) Stimulation of the sympathetic nerve following the injection of the neuropeptide Y antagonist PP56 (2.3 mg/kg) caused decrease of pulpal blood flow by $30.64{\pm}7.92%$ (mean${\pm}$SEM, n=6). PP56 attenuated the sympathetic nerve-induced pulpal blood flow decrease by $44.37{\pm}11.01%$ (mean${\pm}$SEM). These data provide evidences of the co-contribution of nerepinephrine and neuropeptide Y on the sympathetic nerve-induced vasoconstriction in the feline dental pulp. In addition, they show functional evidences that sympathetic nerve plays an active role in controlling the microcirculation of the dental pulp.
출산 전후의 황점볼락 및 조피볼락 어미의 뇌 조직에서 GtH 분비를 자극하는 신경호르몬으로 알려진 NPY를 검출하기 위해, 면역조직화학을 실시하고 뇌하수체내 GtH 분비세포의 활성을 서로 비교하였다. 두 어종 모두에서 뇌 조직중 NPY 양성반응을 나타내는 세포는 후각망울, 종뇌 및 중뇌에서, 신경섬유는 후각망울, 종뇌, 시각신경, 시상하부, 중뇌 및 시각엽에서 각각 관찰되었다. 뇌하수체내에서 NPY 양성반응을 나타내는 신경섬유는 성숙에 관계없이 두 어종 모두 앞원위부분의 AF 음성세포에 인접한 신경엽에 분포하였고, 성장 및 성숙기의 난모세포를 가진 어체에서는 앞원위부분의 신경엽과 중간원위부분의 GtH 분비세포에 인접한 신경엽에서 관찰되었다. 뇌하수체내의 GtH 분비세포는 출산전의 황점볼락 및 성숙기의 조피볼락 개체에서는 AF 염색성이 약했으나, 출산후 황점볼락 및 조피볼락의 GtH 분비세포는 출산전 및 성숙기에 비해 AF 염색성이 증가하였다. 두 어종에서 출산전 난소를 가진 개체들의 GtH 분비세포와 핵경의 크기는 출산후(황점볼락)이거나 출산 이후 휴지기의 난소를 가진 개체(조피볼락) 보다 유의하게 컸다(P<0.01).
This study aimed to investigate whether nociceptin is implicated in the, trigeminovascular responses to electrical stimulation of trigeminal ganglion in rats. An open cranial window was prepared on the right parietal bone of male Sprague-Dawley rats. Trigeminovascular system was stimulated by electrical stimulation of trigeminal ganglion (ETS; 5ms, 5Hz, 3V). Neonatal capsaicin treatment was performed with subcutaneous administration of capsaicin (50mg/kg) within the first 24 hours after birth. Changes in regional cerebral blood flow were continuously measured through the cranial window by laser-Doppler flowmetry, and the expression of nociceptin-like immunoreactivity was determined by immunohistochemistry. ETS caused increases in regional blood flow of pial arteriole in a voltage-dependent manner. ETS markedly and voltage-dependently increased the expression of nociceptin-like immunoreactivity in dura mater ipsilateral rather than contralateral to ETS. The nociceptin-like immunoreactivity was markedly reduced by pretreatments with calcitonin gene-related peptide(8-37) ($CGRP_{8-37},\;a\;CGRP_1$ receptor antagonist), L-733060 (a $NK_1$ receptor antagonist), and $[Nphe^1]$ nociceptin(1-13)$NH_2$ (a selective and competitive nociceptin receptor antagonist) as well as by neonatal capsaicin treatment. These results suggest that the electrical stimulation of trigeminal ganglion causes prominent expression of nociceptin within dura mater, in which not only neuropeptides inducing substance P and CGRP but also nociceptin are implicated in the trigeminovascular responses to electrical trigeminal ganglion stimulation.
Oxytocin is a neuropeptide produced primarily in the hypothalamus and plays an important role in the regulation of mammalian birth and lactation. It has been shown that oxytocin has important cardiovascular protective effects. Here we investigated the effects of oxytocin on vascular reactivity and underlying the mechanisms in human umbilical vein endothelial cells (HUVECs) in vitro and in rat aorta ex vivo. Oxytocin increased phospho-eNOS (Ser 1177) and phospho-Akt (Ser 473) expression in HUVECs in vitro and the aorta of rat ex vivo. Wortmannin, a specific inhibitor of phosphatidylinositol 3-kinase (PI3K), inhibited oxytocin-induced Akt and eNOS phosphorylation. In the rat aortic rings, oxytocin induced a biphasic vascular reactivity: oxytocin at low dose (10-9-10-8 M) initiated a vasorelaxation followed by a vasoconstriction at high dose (10-7 M). L-NAME (a nitric oxide synthase inhibitor), endothelium removal or wortmannin abolished oxytocin-induced vasorelaxation, and slightly enhanced oxytocin-induced vasoconstriction. Atosiban, an oxytocin/vasopressin 1a receptor inhibitor, totally blocked oxytocin-induced relaxation and vasoconstriction. PD98059 (ERK1/2 inhibitor) partially inhibited oxytocin-induced vasoconstriction. Oxytocin also increased aortic phospho-ERK1/2 expression, which was reduced by either atosiban or PD98059, suggesting that oxytocin-induced vasoconstriction was partially mediated by oxytocin/V1aR activation of ERK1/2. The present study demonstrates that oxytocin can activate different signaling pathways to cause vasorelaxation or vasoconstriction. Oxytocin stimulation of PI3K/eNOS-derived nitric oxide may participate in maintenance of cardiovascular homeostasis, and different vascular reactivities to low or high dose of oxytocin suggest that oxytocin may have different regulatory effects on vascular tone under physiological or pathophysiological conditions.
본 연구에서는 감각성 neuropeptide인 CCRP의 치수혈류 조절에 관해 교감신경과의 유기적 관계를 연구함으로써 CGRP의 치수혈류 조절기전을 밝히고자 하였다. 열두 마리의 전신마취된 고양이에서 실험하였으며 CGRP를 혈관을 통해 전신적 또는 국소적으로 투여하였다. 견치에서 치수혈류의 변화를 측정하고 paired t-test로 통계분석 하였으며 $95\%$ 수준에서 유의성을 검증하였다. CGRP $(0.31{\mu}g/kg)$를 전신정맥으로 주사시, 전신혈압에 현저한 영향을 나타내면서 치수혈류는 평균 $68.85\%$의 일차적인 증가와 감소를 보였고 이차적으로 다시 평균 $161.8\%$ 증가하였다가 감소하였다. CCRP를 저용량 $(0.03{\mu}g/kg)$으로 국소적으로 투여시, 치수혈류는 평균 $2.92\%$의 미약한 증가를 나타내었다. 교감신경을 전기자극시 (10Hz, 4V, 1.5ms), 전신혈압은 영향을 받지 않으면서 치수혈류가 유의하게 평균 $57.88\%$ 감소하였다. 교감신경 자극으로 치수혈류가 저하되어 있는 동안 주입한 CCRP는 저하된 치수혈류를 유의하게 회복시켰다. CGRP의 이 치수혈류 증가 효과는 $CGRP_{8-37}$에 의해 효과적으로 차단되었다.
Pancreatic polypeptide family펩타이드들의 뇌혈관 평활근 수축성에 미치는 효과를 관찰하고, cyclic nucleotide 활성제 및 칼륨통로개방제와의 상호작용을 관찰하기 위하여 다음과 같은 실험을 하였다. 체중 $20{\sim}30\;g$의 개를 사혈 희생시켜 두개골을 절개한 후 뇌저동맥과 중뇌동맥을 적출하였다. 적출된 동맥편은 $4^{\circ}C$의 생리적식염수 내에서 나선형 절편으로 만들어 0.3%의 CHAPS 용액에 침잠시킴으로써 내피세포층을 제거한 후 95% $O_2$와 5% $CO_2$의 혼합기체로 포화된 $37^{\circ}C$의 Krebs-Henseleit 용액을 포함한 적출근편실험조에서 등척성 장력을 측정하였다. 1. PP, PYY 및 NPY는 뇌동맥 나선형절편을 농도의존적으로 수축시켰으며, 그 효력과 효능은 PYY가 가장 강하였고, 그 다음이 NPY, 그리고 PP의 순이었다. 이들의 효력은 노르아드레날린보다 20내지 200배 강하였으며, 그 중 PYY는 5-HT 보다도 강한 효력을 보였다. 2. Cyclic AMP 활성제인 forskolin과 cyclic GMP 활성제인 sodium nitroprusside는 뇌동맥절편의 기본장력을 감소시켰으며, PP, PYY 및 NPY 유발수축 역시 농도의존적으로 억제하였다. 이 때 forskolin의 기본장력억제작용이 sodium nitroprusside보다 강한 효력을 나타내었다. 3. 칼륨통로 개방제인 RP 49356, P 1060 및 BRL 38227은 기본장력에 대해서는 공히 농도의존적으로 억제하였으나, PP, PYY 및 NPY 유발수축에 대해서는 P 1060만이 농도의존적으로 억제하였고, RP 49356 및 BRL 38227은 약간 억제하는 경향만을 보였는데, 특기할 것은 저농도의$(0.1\;{\mu}M)$ BRL 38227이 이들 펩타이드 유발수축을 오히려 증가 시켰다는 것이다. 4. 기본장력에 대해서, 칼륨통로개방제들은 forskolin의 이완작용에 유의한 영향을 미치지 못하였으나, 그중 P 1060과 BRL 38227은 sodium nitroprusside의 이완작용을 상승적으로 강화하였다. PYY$(0.1\;{\mu}M)$유발 수축작용에 대해서, 칼륨통로 개방제들은 forskolin의 수축억제작용에 대해서는 약간 길항하는 경향만을 보였고, sodium nitroprusside의 수축억제작용은 유의하게 길항하였다. 이상의 결과들을 종합하면, 개의 뇌혈관에서는 NPY 뿐만 아니라 PYY도 혈관수축기전에 중요한 역할을 한다고 볼 수 있으며, 그 수축작용의 기전에는 세포내 cAMP 및 cGMP 활성도의 변화가 포함된다고 사료된다. 또 칼륨통로개방제들은 pancreatic polypeptide family의 뇌혈관수축작용에 대하여 제제 및 농도에 따라 다양한 영향을 미치므로 이에 대한 향후의 더욱 세밀한 연구가 요구된다.
