This experiment was undertaken to investigate whether methanol extract of fruit peel of Dimocarpus longan Lour. (MEFL) protects against kainic acid (KA)-induced seizure. Oral administration of MEFL (1, 2 and 4 mg/kg) increased KA (50 mg/kg)-induced survival rate and latency of convulsion onset, and deceased seizure scores and weight loss induced by intraperitoneal (i.p) injection of KA in mice. In addition, MEFL protected against cell death in the hippocampus of rat brain after KA-administration as analyzed by using TUNEL assay in rats. MEFL also significantly blocked seizure-form of electroencephalogram (EEG) power spectra induced by KA in rats. MEFL also inhibited elevation of [$Ca^{2+}$]i and increased [$Cl^-$]i induced by KA in cultured neuronal cells. Therefore, it is suggested that MEFL protects against seizure induced by KA, decreasing [$Ca^{2+}$]i.
배경: 흉부 및 흉복부 대동맥의 수술 중 대동맥 차단은 허혈성 척수 손상에 의한 하반신 마비와 같은 심각한 합병증을 유발할 수도 있어 수술 중 허혈성 척수손상을 예방하기 위한 여러 방법의 연구가 계속 되고 있다. 최근에 허혈성 대뇌 손상 모델에서 신경조직의 막전위 의존성 나트륨채널 길항제가 대뇌 보호 효과가 있다는 보고가 있다. 본 연구는 토끼의 허혈성 척수손상 모델에서 나트륨채널 길항제인 페니토인과 저체온의 척수보호효과를 비교해 보고자 시행되었다. 대상 및 방법: 뉴질랜드산 토끼의 신동맥직하부에서 복부대동맥을 25분간 차단하는 방식으로 척수허혈을 유도하였으며 각 군당 8마리씩 네 군으로 나누었다. 대조군과(S39) 저체온군은(S37) 대동맥 차단시간 동안 직장온도를 각기 $39^{\circ}C$와 $37^{\circ}C$로 일정하게 유지하면서 $22^{\circ}C$ 생리적 식염수만 2 mL/min의 속도로 연속 주입하였으며, 정상체온 및 저체온 페니토인 군은(P39, P37) 앞의 두 군과 동일한 방법으로 하되 생리적 식염수에 페니토인을 녹여 주입하였다(100 mg/50 mL). 수술 후 24시간 및 72시간이 경과한 다음 Tarlov scoring을 통해 신경학적 평가를 시행하였고 마지막 평가 후에는 객관적으로 신경손상의 정도를 정량화하기 위해 척수를 고정 처리하였다. 결과: 페니토인의 역행성 주입에 따른 심각한 문제는 없었으며 대조군에(S39) 속한 모든 동물은 완전 또는 심한 하반신 마비 소견을 보였다. 페니토인과(P39) 저체온(S37)군 모두 대조군에 비해 신경학적 평가는 유사한 정도로 우수한 결과를 보였다(p<0.05). 조직 병리학적 검사 결과, 대조군에 속한 모든 동물은 척수 회백질에서 심한 신경조직 괴사 때 보이는 전형적인 특징을 보여주었으며, TUNEL 염색에 양성인 신경세포도 높은 빈도로 관찰되었으나, 저체온 또는 페니토인 투여 군에서는 괴사현상이 유의한 정도로 감소하였으며, 상대적으로 매우 낮은 빈도의 TUNL 염색 양성세포가 관찰되었다(p<0.05). 그러나 저체온과 페니토인을 병용했을 때의 부가적인 척수보호효과를 조사해 본 결과 신경학적 평가와 조직병리학적 결과 모두 유의한 수준의 부가적인 효과는 없었다. 걸론: 결론적으로, 토끼의 허혈성 척수 손상 모델을 이용하여 페니토인과 저체온의 신경보호효과를 알아본 결과 신경학적 평가와 조직병리학적 검사 결과 모두 부가적인 효과는 보여주지 못했지만 각각의 경우 유사한 정도의 신경보호효과를 보여주었다.
목 적 : 규칙적인 운동이 신경보호 효과와 도파민성 신경원의 재구축, 운동기능 향상에 영향을 미친다는 실험실적 연구결과에도 불구하고, 아직까지 파킨슨병 질환자의 트레드밀 운동이 뇌신경 변화에 영향을 미치는지에 대해서는 논란이 되고 있는 상황이다. 더군다나, 증상의 진전이 흑질선조체의 뇌신경 변화에 의한 것인지, 운동에 의한 전반적인 효과인지, 의욕에 영향을 받은 것이지 또한 확실치 않은 상황이다. 이에 본 연구자는 트레드밀 운동이 파킨슨 유발 실험쥐의 뇌신경 변화를 유발하는 것을 밝히고자 본 실험을 수행하였다. 방 법 : 본 실험에서는 파킨슨 모델을 만들기 위해 수컷 C57BL/6 쥐에 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine(MPTP) 30 mg/kg과 프로베네시드 20 mg/kg을 매 12시간마다 10회 투여(총 5일)하여 파킨슨병을 유발하였다. 이후 운동군을 경사도 $0^{\circ}$, 18 m/min의 속도로, 하루 40분의 트레드밀 운동을 수행하였다. 운동수행의 마지막에는 모든(염류 비교군, 비운동 비교군) 동물의 뇌를 적출하여 신경원성, 신경화학적 변화가 어떤지 비교군, 비운동군과 비교분석하였다. 본 실험에서 Synphilin 단백질은 알파시누크린의 발현 징후로 사용되었다. 흑질과 선조체의 뇌세포를 western blotting에 의해 염색하여 분석하였다. 결 과 : 염류 비교군의 경우 synphilin 단백질의 발현이 발견되지 않았다. 파킨슨 유발을 위한 MPTP(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) 투여는 알파시누크린의 응집을 의미하는 synphilin 단백질의 발현이 급증하였다. 하지만, 트레드밀 운동군에서는 synphilin 단백질의 발현이 비운동군에 비해 유의하게 낮았다. 이는 트레드밀 운동이 알파시누크린의 응집도를 낮추는데 영향을 미친다는 것으로 사료된다. 결 론 : 본 연구에는 트레드밀 운동이 파킨슨 모델의 뇌에서 알파시누크린 응집체의 제거를 촉진하고, 병의 진행, 세포사멸을 억제하는 것으로 밝혀졌다.
In the present study, we synthesized a series of novel 7-methoxy-N-(substituted phenyl)benzofuran-2-carboxamide derivatives in moderate to good yields and evaluated their neuroprotective and antioxidant activities using primary cultured rat cortical neuronal cells and in vitro cell-free bioassays. Based on our primary screening data with eighteen synthesized derivatives, nine compounds (1a, 1c, 1f, 1i, 1j, 1l, 1p, 1q and 1r) exhibiting considerable protection against the NMDA-induced excitotoxic neuronal cell damage at the concentration of $100{\mu}M$ were selected for further evaluation. Among the selected derivatives, compound 1f (with $-CH_3$ substitution at R2 position) exhibited the most potent and efficacious neuroprotective action against the NMDA-induced excitotoxicity. Its neuroprotective effect was almost comparable to that of memantine, a well-known NMDA antagonist, at $30{\mu}M$ concentration. In addition to 1f, compound 1j (with -OH substitution at R3 position) also showed marked anti-excitotoxic effects at both 100 and $300{\mu}M$ concentrations. These findings suggest that $-CH_3$ substitution at R2 position and, to a lesser degree, -OH substitution at R3 position may be important for exhibiting neuroprotective action against excitotoxic damage. Compound 1j was also found to scavenge 1,1-diphenyl-2-picrylhydrazyl radicals and inhibit in vitro lipid peroxidation in rat brain homogenate in moderate and appreciable degrees. Taken together, our structure-activity relationship studies suggest that the compound with $-CH_3$ substitution at R2 and -OH substitution at R3 positions of the benzofuran moiety might serve as the lead exhibiting potent anti-excitotoxic, ROS scavenging, and antioxidant activities. Further synthesis and evaluation will be necessary to confirm this possibility.

Oxidative stress is closely associated with various diseases and is considered to be a major factor in ischemia. NAD(P)H: quinone oxidoreductase 1 (NQO1) protein is a known antioxidant protein that plays a protective role in various cells against oxidative stress. We therefore investigated the effects of cell permeable Tat-NQO1 protein on hippocampal HT-22 cells, and in an animal ischemia model. The Tat-NQO1 protein transduced into HT-22 cells, and significantly inhibited against hydrogen peroxide ($H_2O_2$)-induced cell death and cellular toxicities. Tat-NQO1 protein inhibited the Akt and mitogen activated protein kinases (MAPK) activation as well as caspase-3 expression levels, in $H_2O_2$ exposed HT-22 cells. Moreover, Tat-NQO1 protein transduced into the CA1 region of the hippocampus of the animal brain and drastically protected against ischemic injury. Our results indicate that Tat-NQO1 protein exerts protection against neuronal cell death induced by oxidative stress, suggesting that Tat-NQO1 protein may potentially provide a therapeutic agent for neuronal diseases.
Objectives : This study was undertaken to investigate the effect of noncompetitive NMDA receptor blocker, MK801 on motor recovery, SSEP and pathology in spinal cord injured rat. Methods : The effects of MK801 on neuronal function protection, SSEP, and pathology were measured on spinal cord injury rats which were divided into 6 groups according to dose, time of drug delivery and magnitude of injury. Spinal cord injury was made with the magnitude of 25gm-cm and 50gm-cm on 42 rats. BBB locomotor function test was performed to evaluate the motor power recovery in hindlimb for 2 weeks after injury. After motor function test was completed, SSEP was measured. Amplitude and latency of the P1, N1 peak was measured and compared between groups. Finally rats were sacrificed, and pathologic findings including measurement of area of necrotic cord were studied and compared between groups. Results : Motor recovery at 2 weeks was better in MK801 group comparing to saline control group. SSEP at 2 weeks showed no difference in N1, P1 latencies, but significantly greater amplitude in MK801 group, compared to saline control group. On light microscope, there was no specific histologic differences between experimental groups. The cystic necrotic area in coronal plane was measured and compared in each group. The necrotic area was significantly smaller in MK801 1mg/kg group(delivered after injury) than vehicle group. The necrotic area in MK801 5mg/kg group and MK801 1mg/kg group(delivered before injury) was smaller than vehicle group even though it was not statistically significant. Conclusion : From the above result, it is speculated that NMDA blocker, MK801 can improve impaired neuronal function in spinal cord injury.
Familial Amyotrophic lateral sclerosis (FALS) is a progressive neurodegenetative disorder induced by mutations of the SOD1 gene. Heat shock protein 27 (HSP27) is well-defined as a stress-inducible protein, however the its role in ALS protection has not yet been established. To investigate the role HSP27 may have in SOD1 mutant-mediated apoptosis, human SOD1 or HSP27 genes were fused with a PEP-1 peptide in a bacterial expression vector to produce a genetic in-frame fusion protein, which was then transduced into cells. We found the purified PEP-1-HSP27 fusion proteins can be transduced efficiently into neuronal cells and protect against cell death by enhancing mutant SOD1 activity. Moreover, transduced PEP-1-HSP27 efficiently prevents protein aggregation produced by oxidative stress. These results suggest that transduced HSP27 fusion protein may be explored as a potential therapeutic agent for FALS patients.
루이보스(Aspalathus linearis; RB)는 콩과식물로서 남아프리카 Cape Province의 북서부 산악지역에 자생한다. 루이보스차 침제는 가는 가지와 잎의 발효산물로서 플리보노이드류와 페놀산이 있어 강한 항산화활성을 갖는 것으로 알려져 있다. 본 연구에서는 저산소증모델에서 루이보스차 침제가 배양한 흰쥐 대뇌세포의 스트레스를 완화하는지에 대한 연구하였다. 배지로 누출된 LDH의 정량실험에 의하면 루이보스는 정상산소환경 및 저산소증에서 함량 의존적으로(10-100 $\mu\textrm{g}$/ml) 각각 6-18% 및 2-24%의 세포생존율을 증가시켰다(16 DIV 세포, 처리 후 3일째). CFP-Hsc70 단백질을 표현시킨 신경세포의 모양을 관찰하였을때 루이보스(50 $\mu\textrm{g}$/ml)는 저산소처리 후 5일에 세포체에 수포가 있는 세포의 수를 대조군(55.4$\pm$4.59%)에 비하여 유의하게 감소시켰다(40.9$\pm$6.3%). 이러한 결과들은 루이보스차가 저산소증에서 신경세포를 보호함을 의미하며, 신경세포 손상을 예방 또는 치료하는데 응용될 수 있을 것으로 보인다.
백년초를 80% (v/v) 에탄올-물 혼합용액으로 추출하여 총페놀 함량, 총플라보노이드 함량, 산화방지능, 신경세포 보호효과, AChE 및 BChE 활성저해능을 평가하였다. 백년초 100 g FW당 총페놀 함량은 409.9 mg GAE, 총플라보노이드 함량은 72.2 mg CE이었다. 백년초(100 g FW)의 산화방지능은 ABTS, DPPH, ORAC법에서 각각 381.2, 298.2, 3,219.9 mg VCE였다. 백년초 추출물은 $H_2O_2$의 산화 스트레스로부터 세포 내 산화 스트레스를 감소시켜 신경세포 PC-12의 생존율을 농도의존적으로 증가시켰다. 또한, 백년초 추출물은 AChE 및 BChE 활성을 저해하였다. 본 연구 결과로부터 백년초의 세포 내 산화 스트레스 감소에 의한 신경 세포 보호능과 AChE 및 BChE 저해능이 확인 되었으며, 이러한 결과를 통해 향후 백년초를 이용한 기능성 제품 및 여러 가공품 개발에 활용이 가능할 것으로 기대한다.
