The experiments were carried out to investigate the biocontrol potential of Lysobacter capsici YS1215 on root-knot nematode and to characterize its lytic enzyme activities. L. capsici YS1215 showed chitinase and gelatinase activities on the medium containing 0.5% chitin or 0.5% gelatin as substrates. Cell growth of L. capsici YS1215 was highest at 6 days, and the highest activities of chitinase (4.0 unit $ml^{-1}$) and gelatinase (7.43 unit $ml^{-1}$) were observed on 3 and 5 days after incubation, respectively. To investigate the effect of L. capsici YS1215 on tomato growth and nematode infection, the plants in pot trial were treated with bacterial culture (BC), half of bacterial culture (HBC), only bacterial medium (BM), tap water (TW) and commercial nematicide (CN). HBC treatd plants showed the higher shoot fresh weight and dry weight on $5^{th}$week after incubation while BM, HBC and BC had consistently higher values than TW at $9^{th}$ week. HBC appeared to be the highest shoot fresh length at $9^{th}$ week. Both CN and BC showed lower number of egg mass, root gall, and population of juveniles in soil compared to BC, HBC, BM and TW. These results suggest that L. capsici YS1215 with its strong ability of lytic enzyme production can be one of the most significant candidates for biocontrol agents against root-knot nematodes.
Root-lesion nematode (Pratylenchus vulnus) and pin nematode (Paratylenchus sp.) were detected with high population density at the spray chrysanthemum greenhouse in Gumi, Gyeongbuk. The average density of P. vulnus and Paratylenchus sp. was 667 and 716 nematodes per 100 g soil and P. vulnus density were distributed 87% to the depth of $0{\sim}30$ cm in greenhouse. When spray chrysanthemum cv Chopin, was transplanted in September 2004 in greenhouse, P. vulnus were 854 nematodes/100 g soil in planting and increased 14,985 nematodes/100 g soils and 1g root after 10 weeks in harvest. Shoot weight and shoot height of spray chrysanthemum decreased 24.8% and 8.0% compared with non-infested P. vulnus after 10 weeks in greenhouse.
Severe damage on a greenhouse grown lettuce (Lactuca sativa L.) associated with Paratylenchus projectus is described for the first time. There were no marketable lettuce in entire greenhouse after three months of planting. Lettuce roots system appeared corky and most of feeder rootlets disappeared. Population densities of P. projectus were ranged from 320-11,600 nematodes per 100 ㎤ of soil. In an inoculation test in a greenhouse, P. projectus is proved as a causal agent of lettuce damage.
A bacterium having strong chitinolytic activity on $0.2\%$ colloidal chitin-containing agar medium was isolated from coastal soil in Korea. Based on the nucleotide sequence of conserved segment of a 165 rRNA gene, the bacterium was identified as Paenibacillus illinoisensis KJA-424. The population of P. illinoisensis KJA-424 and chitinase activity significantly increased for the first 2 days of incubation. On SDS-PACE analysis with $0.01\%$ glycol chitin, three protein bands (63, 54, and 38 kDa) with chitinolytic activity were detected tooted. The effect of P illinoisensis KJA-424 on the egg hatch of root-knot nematode (Meloidogyne incognita) was investigated. After 7 days of incubation with the chitinase-producing P. illinoisensis KJA-424, none of the eggs hatched, whereas a $39.8\%$ egg hatching rate was observed in the water control. Inverted and scanning electron microscopic observations demonstrated that P. illinoisensis KJA-424 deformed and destroyed the eggshell of M. incognita. In conclusion, chitinase-produced by p. illinoisensis KJA-424 caused the lysis of M. incognita eggshell and resulted in the inhibition of egg hatching in vitro.

Thirteen soybean cyst nematode (SCN) (Heterodera glycines) populations collected in Korea were examined in their HG type by their reproductivity on 7 Plant Introduction indicators for the identification of HG type. Six HG types were identified, HG type 0, 2, 5, 2.5, 1.2.7, and 2.5.7. HG type 2.5 was the most frequent (4 samples, 30.8%), followed by HG type 2.5.7 (3 samples, 23.0%). About 76.9% of SCN populations were reproduced on PI 88788, followed by PI 209332 (61.5%), PI 548316 ('Cloud') (30.8%), and PI 548402 ('Peking') (7.7%). No population could reproduce on PI 90763, PI 437654, thus, they could be used for resistant source for developing SCN resistant soybean in Korea.
To evaluate the mechanisms of pathogenicity and nonpathogenicity of Bursaphelenchus xylophilus and B. mucronatus isolated in South Korea, we used 4-year-old P. thunbergii seedlings and 20-cm long one-year-old stem cuttings of 5-year-old Pinus thunbergii, and studied distributions and multiplications of pine wood nematodes after inoculation. The distributions of B. xylophilus in the 20-cm pine stem cuttings were not significantly different from that of B. mucronatus. Conversely, the proliferation rate of B. xylophilus on mycelial mats of Botrytis cinerea was significantly different from that of B. mucronatus. The study using 4-year-old P. thunbergii seedlings also showed that B. mucronatus can migrate to distal portions of the pine seedlings the same as B. xylophilus, but the populations of B. xylophilus remaining in the pine seedlings were relatively larger than those of B. mucronatus. Therefore, we concluded that the pathogenicity of B. xylophilus could be strongly influenced by its ability to multiply.

A neem based product, Suneem-G (granules) alone as well as in combination with urea was tested against root-knot nematode, Meloidogyne incognita, on okra (Abelmoschus esculentum) in clay pots of 20-cm-diameter, containing 2 kg sterilized soil, inoculated with 2,000 freshly hatched second stage juveniles (J$_2$) of M. incognita. Application of different dosages of Suneem-G with or without urea significantly reduced nematode population and incidence of galling on okra roots and increased vegetative plant growth compared with the untreated control. Suneem-G with urea was the most effective and its effect increased with the increase of dosage. Suneem-G 6 g+urea 25 g per pot gave maximum plant growth and root gall reduction, as plants shoot and root lengths were increased by 84 and 58% and root galling was reduced by 58%, respectively. The lowest dosage of Suneem-G alone, 2 g per pot increased shoot and root lengths of plants by 25.7 and 17%, respectively, and reduced root galling by 24%. The lowest effect was recorded in the pots treated with 25 g urea alone; shoot and root lengths were increased and root galling was decreased by 14% each.
This study was carried out to provide basic information on physiological changes of 4-year-old seedlings of Pinus thunbergii inoculated with pine wood nematodes by analyzing changes of symptom development, stem and needle water content and nematode populations in stem. Twenty days after inoculation, needles were discolored as an external symptom, and xylem drying and pith browning occurred at the above and below of the inoculation sites as an internal symptom. However, xylem drying began to occur 10 days after inoculation, which was determined by the difference in drying status of xylem and cortex between control and inoculated seedlings. Although population of pine wood nematode increased from 5 to 10 days after inoculation, it has increased dramatically from 10 to 20 days after inoculation when both internal and external symptoms appeared. As the time passed by after inoculation with pine wood nematodes, water content of stem and relative water content in current needles and branch gradually decreased. As the number of nematodes increases, water content of stem and relative water content of current needles and branches decreased significantly. There was a positive relationship between the number of nematodes and xylem drying and/or disease development, but the number of nematodes rapidly decreased as seedlings become severely diseased.
Distribution and population of plant parasitc nematodes were investigated using 70 soil and root samples in total collected from six locations in Gyeongbug and Gyeongnam provinces. Among nematodes identified, Meloidogyne spp. was the most prevalent with $91.4\%$ in frequency cf detection. Frequency of detection was $87.1\%$ for Tylenchus sp., $24.3\%$ for Hirschmanniella, $23.3\%$ for Aphelenchus sp., and $20\%$ for Aphelenchoides sp.. Meloidogyne spp. was most abundant from samples collected in Namji and the number of nematodes dectected was 2,104 per $300cm^3$ soil. The number of Meloidogyne spp. detected was 671 from Milyang, 401 from Samrangjin. The number of nematodes was 57 from Samrangjin for Tylenchus sp., 127 from Gupo for Pratylenchus, 26 for Aphelenchoides sp. and 34 for Criconemoides from Samrangjin. Population of Meloidogyne spp. was the highest in Namji and Milyang where pepper and cucumber or pepper and chinese cabbage was cultivated in multiple croppings, however, population of Pratylenchus spp., and Meloidogyne spp. was the highest in Samrangjin where strawberry was cultivated as a single crop. Population in Pratylenchus and Meloidogyne was the highest in Gupo where tomato and chinese cabbage were cultivated alternatively, however, population of nematodes was generally low in Seongju and Yucheon where watermelon-rice or pepper-rice were cultivated in sequence. Meloidogyne spp. and Pratylenchus spp. appear to be economically important nematode species in cropping by controlled horticulture. In this study, 15 species of plant parasitic nematodes were identified.
A survey was conducted during $April{\sim}May$ 2004 to determine the occurrence and population density of plant-parasitic nematodes in ginseng (Panax ginseng C.A. Meyer) growing fields, in major ginseng growing regions of Chungbuk, Chungnam, Gyeongbuk and Kyongki provinces. The survey revealed presence of eleven species of plant-parasitic nematodes namely, Criconemoides morgensis, Ditylenchus destructor, Helicotylenchus dihystera, Meloidogyne incognita, M. hapla, Paratylencgus lepidus, Pratylenchus penetrans, Psilenchus hilarulus, Trichodorus similis, Tylenchorhynchus claytoni and Xiphinema americanum. Frequency and density of each species were highly variable. M. incognita and M. hapla were the predominant species, their infestation observed in 46.3 and 39.4% fields with an average density of $78{\sim}254\;and\;76{\sim}211$ nematodes per $300\;cm^3$ soil, respectively. Whereas, T. similis and X. americanum were rarely observed; only in 2.3 and 1.8% of surveyed fields and their density was $10{\sim}17\;and\;7{\sim}10$ individuals per $300\;cm^3$ soil, respectively. They are recorded herewith for the first time from ginseng fields of Korea. In nematode-infestated fields, stunted plant growth with chlorotic leaves, and wilted plants were observed in patches.
