Evaluation of Two Nematicides in the Initial Population Changes of the Soybean Cyst Nematode (콩시스트 선충의 초기밀도 변화에 있어서 두가지 살선충제의 효과검정)

  • The control efficiency of the nematicides, aldicarb and carbofuran, in the population dynamics of the soybean cyst nematode(Heterodera glycines)was evaluated. Nematode viability, penetration and reproduction were examined in greenhouse experiments. Nematode viability(movement) was greatly reduced by aldicard, but mot by carbofuran. Penetration was inhibited by both chemicals. Percentages of Pentration relative to the untreated check 3 days after treatment were 0.4% and 1.7% for aldicarb and carbofuran, respectively. Systemic effect ot the nematicides were observed with both nematicides but effect was greater with aldicarb than with carbofuran. The total effect of the nematicides, applied ay the time of planting soybean in infested soil, appeared to be less than the sum of the contact and systemic effects on the initial population dynamics of H. glycines.

Effect of pinewood nematode on the water content and early disease development of seedlings of susceptible Pinus densiflora and resistant Pinus × rigitaeda for breeding for resistance to pinewood nematode

  • Three-year-old seedlings of susceptible Pinus densiflora and resistant Pinus x rigitaeda were each inoculated with the pinewood nematode, Bursaphelenchus xylophilus, to compare disease development. Needle dehydration was evident on seedlings of P. densiflora by 20 days after inoculation, 10 days earlier than this symptom was observed on P. ${\times}$ rigitaeda. Xylem drying was more frequent in seedlings of P. densiflora than in that of P. ${\times}$ rigitaeda between 20 and 60 days after inoculation. No significant differences were found between P. densiflora and P. ${\times}$ rigitaeda for stem water content or for stem and leaf relative water content in current-year branches after nematode inoculation, but the average number of B. xylophilus recovered from stems differed significantly between the two groups. The number of B. xylophilus recovered from stems was negatively correlated with the stem water content and with stem and leaf relative water content. By the time the experiment was terminated at 60 days after inoculation, all 3 of the last group of P. densiflora seedlings had died, but 2 of the 3 remaining P. ${\times}$ rigitaeda hybrid seedlings were still alive. Additional studies are needed to further explore the specific mechanisms preventing nematode multiplication in the seedlings of resistant P. ${\times}$ rigitaeda.

Root-Knot Nematode (Meloidogyne incognita) Control Using a Combination of Lactiplantibacillus plantarum WiKim0090 and Copper Sulfate

  • Lactic acid bacteria (LAB) exert antagonistic activity against root-knot nematodes, mainly by producing organic acids via carbohydrate fermentation. However, they have not yet been used for root-knot nematode (Meloidogyne incognita) control owing to a lack of economic feasibility and effectiveness. In this study, we aimed to isolate organic acid-producing LAB from kimchi (Korean traditional fermented cabbage) and evaluated their nematicidal activity. Among the 234 strains isolated, those showing the highest nematicidal activity were selected and identified as Lactiplantibacillus plantarum WiKim0090. Nematicidal activity and egg hatch inhibitory activity of WiKim0090 culture filtrate were dose dependent. Nematode mortality 3 days after treatment with 2.5% of the culture filtrate was 100%, with a 50% lethal concentration of 1.41%. In pot tests, the inhibitory activity of an L. plantarum WiKim0090-copper sulfate mixture on gall formation increased. Compared to abamectin application, which is a commercial nematicide, a higher control value was observed using the WiKim0090-copper sulfate mixture, indicating that this combination can be effective in controlling the root-knot nematode.

Soil Chemical Characteristics and Comparison with Infested Status of Nematode(Meloidogyne spp.) in Plastic House Continuously Cultivated Oriental Melon in Songju (성주 지역 시설참외 연작지의 토양특성 및 토양선충 변화)

  • This study was conducted for ten years to evaluate the effective soil management for preventing the infection of root-knot nematode in the field of continuous cultivation with oriental melon under plastic house in Songju area of kyongbuk province. The content of available phosphate, total nitrogen, organic matter, CEC, and exchangeable base in the soil increased with the increase of continuous cultivation year. Especially salt content in the soil increased form 1.2 to 4.55 mS/cm and the yield of oriental melon dramatically decreased with the continuous cultivation year. The number of root-knot nematode was 91 per $300\;cm^3$ of soil in the field of continuous cultivation for 3 years and showed slight damage on the oriental melon, but it was 518 in the field of $4{\sim}6$ years continuous cultivation and showed that 50% of plants died in August, and the yield of late season was less than 50% compared to normal plant. For the seasonal changes in infection rate of root-knot nematode on oriental melon plant, 15% of the normal plant was infected by nematode in February and increased gradually by $10{\sim}20%$ per month, 60% of plants was infected in July. The density of root-knot nematode nymph was 167 in February and increased to 1,625 in August. The infection rate of nematode was 35%, and the number of nematode was about 54 in nursery soil originated from paddy soil, upland soil, and river sand. There were no relationship between the number of nematode and available phosphate or exchangeable base in the soil of plastichouse where oriental melon plants were grown.

First Report of an Unrecorded Nematode-Trapping Fungus Species Monacrosporium phymatopagum in Korea

  • A fungus that can capture nematodes by means of sessile adhesive knobs was isolated from rotten wood in Uiseong, Korea. It was found to produce single, spindle-shaped, 3-4 septate (commonly 4-septate) conidia, $44.8\;{\mu}m$ (range, $41.6-50.1\;{\mu}m$) long and $13.3\;{\mu}m$ (range, $10.7-15.4\;{\mu}m$) wide. Conidiophores were found to be hyaline, erect, straight, and $202.7-245.7\;{\mu}m$ high. On the basis of these morphological features, the fungus was identified as Monacrosporium phymatopagum. This is the first report of M. phymatopagum in Korea which can be a potential biological control resource of plant parasitic nematode.

A New Host of Citrus Nematode, Tylenchulus semipenetrans Cobb, in Korea (약용식물, 노루발(Pyrola japonica)에서의 감귤선충(Tylenchulus semipenetrans Cobb)의 기생)

  • During the course of study on plant parasitic nematodes associated with medicinal herbs in Korea, high population of different stages of citrus nematode, Tylenchulus semipentrans was found in association with Pyrola japonia Klenze in Imsil, Chunbuk province. The second stage larvae and males were isolated from rhizosphere soil ; immature and mature females were detached from the roots of infested plants of Pyrola japonica. Besides citrus it has been reported to parasitize on roots of Syringa vulgaris, Diospyros lotus, olive, grapevine, and pear in different parts of the world. This is the first record of T. semipenetrans parasitized on a medicinal plant, Pyrola japonica.

  • The frequent finding of soil contamination by dogs' feces are remarkable, and such soil contamination may be public health problem. Of 608 samples, 30(4.9%) were positive for nematode ova. Ten paint 3% of sail samples from parks and 7.9% from footpaths were found to contain nematode eggs. Of the helminth eggs detected, eggs of Toxocara spp. were the most commonly encountered, and mostly from the soil of footpaths. No eggs were recovered from the soil of school yards; the level of soil contamination was low in public play areas. Egg recovery rate of the soil to dogs' feces collected from footpaths was approximately 1:5. And the larvae demonstrated from the soil in the present study were nematode larvae, mainly recovered from parks.

Ophiostoma ips from Pinewood Nematode Vector, Japanese Pine Sawyer Beetle (Monochamus alternatus), in Korea

  • Japanese pine sawyer beetle (Monochamus alternatus) is an economically important pest in coniferous trees. Ophiostoma ips was isolated from the beetle and identified based on analysis of morphological properties and the ${\beta}$-tubulin gene sequence. The fungus easily produced perithecia with a long neck on malt extract agar and its ascospores were rectangular shaped. This is first report of Ophiostoma species associated with the pinewood nematode vector beetle in Korea.

  • We identified the five root-lesion nematode species, Pratylenchus crenatus, P. fallax, P. kumamotoensis, P. panamaensis and P. penetrans from intercepted in quarantine inspection over the past five years. Their diagnostic characters are including number of lip annuli, stylet length, shape of the labial region, presence or absence of males, structure of lateral fields, shape of spermatheca, length of the post-vulval uterine sac and shape of tail and so forth. We described the photos, measurements and morphological characters.

  • One hundred seventy five red pepper varieties were bioassayed for selecting resistant varieties to northern rootknot nematode, Meloidogyne hapla. Fifteen native varieties (IT 102794, 104806, 105516, etc) and two imported varieties were proved to be resistant while the currently cultivated varieties such as Hongtap, Kangsan, Hongsil, and Bookang were moderately resistant to the nematodes. Resistant varieties resulted in less nematode infection and development than did the susceptibles. Roots of the resistant strains had significantly higher esterase and peroxidase activities than did those of the susceptibles.
