Identification and Characterization of Alternaria iridiaustralis Causing Leaf Spot on Iris ensata in China

  • In 2016, a severe leaf spot disease was found on Iris ensata Thumb. in Nanjing, China. The symptom was elliptical, fusiform, or irregularly necrotic lesion surrounded by a yellow halo, from which a small-spored Alternaria species was isolated. The fungus was identified as Alternaria iridiaustralis based on morphological characteristics. The pathogenicity tests revealed that the fungus was the causal pathogen of the disease. Phylogenic analyses using sequences of ITS, gpd, endoPG, and RPB2 genes confirmed the morphological identification. This study is the first report of A. iridiaustralis causing leaf spots on I. ensata in China.

  • A seven-year-old female Jindo-dog was presented with a history of progressive abdominal distension. Except for severe bilateral abdominal swelling, other abnormal signs were not detected. The patient showed normal appetite and defecation. In the radiographic examination, the abdomen was filled with large masses. Suspected a certain neoplastic disease, laparotomy was taken through the cranial abdominal midline. Large pale-yellow masses were proliferated to fill the abdomen. In the masses, grey-brown or black portion presumed hemorrhagic or necrotic spots were found. Even though neoplastic tissues were not detected in the right kidney, they were infiltrated in the left kidney except for a part of the cortex. Obtaining the owner's consent, the patient was euthanized and samples were collecte for further study. In microscopic examination, the parenchyma of the medulla was substituted with tumor cells and the cortex was impressed by the expansive proliferation of the neoplastic tissues. This neoplasm was estimated as renal carcinoma originated from tubular epithelium, being based upon that tumor cells were largely cuboidal cells and they had obscure tubular forms.

Occurrence of Tan-brown Leaf Spot Caused by Pilidium concavum on Fragaria ananassa in Korea

  • In 2015, a new leaf spot disease was observed on strawberry seedlings in Wanju, Korea. Tanned brown spots appeared on the leaves of the infected plants, and often coalesced to form larger necrotic areas, resulting in the death of foliage. An isolate was obtained in pure culture. On the basis of morphological characteristics and molecular analysis of internal transcribed spacer rDNA sequence, the causal agent was identified as Pilidium concavum. Pathogenicity tests revealed the isolate was pathogenic to the leaves and fruits of strawberry. To the best of our knowledge, this is the first report of P. concavum causing tan-brown leaf spot on strawberry in Korea.

  • Turnip mosaic virus)TuMV-Ca) was isolated from a Chinese cabbage showing severe mosaic and black necrotic spots symptoms in Korea. The virus was identified as a strain of TuMV by its host range test, particle morphology, serology, double stranded RNA analysis. For detection of the virus, reverse transcription and polymerase chain reaction(RT-PCR) was performed with a set of 18-mer TuMV-specific primers to amplify a 876 bp DNA fragment The virus was rapidly detected from total nucleic acids of virus infected tissues as well as native viral RNA of purified virion particles by RT-PCR. Detection limit of the viral RNA by RT-PCR was 10 fg.

Identification and Characterization of Pseudocercospora cornicola Causing Leaf Spots on Cornus officinalis

  • Cornus officinalis plants that grow in several locations in Korea have been found to be infected with leaf spot disease. Symptoms include necrotic lesions, which are angular, irregularly shaped, vein-limited, and dark brown, on both sides of the leaves. The causal agent of the disease was identified to be Pseudocercospora cornicola based on the morphological characteristics of the fungus and molecular phylogenetic analysis of the obtained multi-locus DNA sequence data. This is the first report investigating P. cornicola found on C. officinalis in Korea.

  • 2014년 7월부터 9월까지 경상남도 합천 지역의 크리핑벤트그래스(Agrostis stolonifera) 퍼팅 그린에서 Curvularia trifolii에 의한 Curvularia 잎마름병이 관찰되었다. Curvularia 잎마름병은 크리핑벤트그래스 잎몸에서 수침상으로 시작되어 진한 괴사반점으로 변한 다음, 크기가 확대되어 회갈색 또는 밝은 갈색의 동그란 타원형 병반이 되었다. 반점의 둘레는 자주 분홍 또는 진한 갈색의 경계를 갖으며 둥그런 무리(halo)를 보였다. 괴사 반점들은 융합되어 불규칙한 모양이 되었고 잎끝 또는 잎 전체가 말랐다. 마른 잎몸은 회백색이나 황갈색을 나타냈다. 그 균은 균학적 특성과 16S rDNA 서열 분석에 의해 동정되었다. 병원균의 분생포자는 크기가 짧았고, 보통 3개의 격막을 가졌으며, 곧거나 자주 휘었는데 양끝 세포는 일반적으로 가운데 세포보다 투명하였다. 3개의 격막을 지닌 분생포자의 배지상 크기는 $26{\sim}28{\times}11{\sim}12{\mu}m$였다. 기주에 대한 균의 인공접종으로 병원성이 확인되었다. 본 논문은 C. trifolii가 크리핑벤트그래스에 Curvularia 잎마름병을 일으키는 우리나라 최초의 보고이다.

  • Pear black necrotic leaf spot (PBNLS) on pear trees (Pyrus pyrifolia) is caused by a Korean isolate of Apple stem grooving virus (ASGV-K). Yellow spots were detected in Phaseolus vulgaris (kidney bean) and Chenopodium quinoa which were grown near the diseased pears in year 2000 through 2003. The ASGV-K, the causative agent of PBNLS, was detected from the symptoms of the diseased kidney bean plant and C. quinoa. ASGV-harboring fungi were also isolated from symptomatic plants and from soils surrounding the infected plants. The ASGV-harboring fungus was identified and characterized as Talaromyces flavus. Ecopathological studies showed that the number of ASGV-harboring fungi on the pear leaves was not correlated with differences in temperature or severity of symptoms. Additionally, there was no difference in fungus frequency among the orchard locations or different host plants. Although the frequency of fungi isolated from the soil was not affected by changes in temperature or location, the fungi occurred at higher densities in the rhizosphere than in the plants themselves.

First Report of Freesia sneak virus in Freesia spp. in Korea

  • In March, 2013, twenty symptomatic freesia plants (10 plants of cultivar Shiny Lemon and 10 plants of cultivar Shiny Gold), with striking virus-like symptoms were collected in Cheongju, Korea. The plants showed chlorotic, coalescing, interveinal, whitish, necrotic, mosaic, mottling or dark brown-to-purple necrotic spots on leaves. Freesia crude sap was directly analyzed by transmission electron microscopy, which potyvirus particles as well as long virus-like particles were detected. Total RNA extracts were analyzed for the infection of Freesia sneak virus (FreSV) by reverse transcription (RT)-PCR with primers specific to FreSV coat protein (CP) gene based on the sequences of FreSV isolates (GenBank No. GU071089, FJ807730 and DQ885455), showing 9 of 20 plants were infected. All 1305bp RT-PCR products were cloned and sequenced. Comparisons of nucleotide and deduced amino acid sequences using BLAST and bioinformatics tools resulted in 99 to 100% sequence identity with FreSV isolates FOV, Virginia, and Italy, confirming FreSV in 9 symptomatic freesia plants. Of 9 determined cDNAs of FreSV isolates, sequences of 5 cDNA clones were identical (GenBank No. AB811437) and sequences of 4 cDNA clones were identical (GenBank No. AB811792). To our knowledge, this is the first report of FreSV from Freesia spp. in Korea.

  • 경북 청도지역의 단 고추 과일에 나타난 큰 괴저 반점 및 과일 괴저와 잎에 엽맥 녹대 및 기형 병징을 일으키는 바이러스는 오이모자이크바이러스의 새로운 계통인 CMV-NP로 분류동정되었다. CMV-NP는 직경이 26 nm의 구형이었으며, VC/RT-PCR 유전자 진단결과 CMV로 확인되었다. CMV-NP는 오이(Cucumis sativus) 등 9개 지표식물에 전신감염을 일으켰으며, 명아주(Chenopodim amaranticolor; C. quinoa)에 국부감염을 일으켰다. CMV-NP는 담배(N. rustica)와 번행초(Tetragonia expansa)의 접종잎과 상엽에 특이한 괴저 원형 반점을 일으켰다. 특히 오이에서는 상엽에 큰 퇴록 원형반점과 엽맥퇴록 병징을 일으켰으며 독말풀(Datura stramonium)에서는 병원성이 없었다.

Ribgrass Mosaic Tobamovirus Occurred on Chinese Cabbage in Korea

  • A tobamovirus, Ribgrass mosaic virus (RMV), was identified newly from chinese cabbage (Brassica campestris L. pekinensis) in Korea. Virus disease incidence of RMV on chinese cabbage was 37.9% in alpine area on August in 1993. RMV induced the symptoms of necrotic ring spots, necrotic streak on midrib and malformation. RMV, Ca1 and Ca3 isolate, could infect 35 species out of 45 plants including Chenopodium amaranticolor. Physical properties of RMV Ca1 isolate were very stable as 10.8 over for dilution end point, $95^{\circ}C$ for temperature inactivation point and 18 weeks for longevity in vitro. RMV had the soil transmission rate of 75.0% for the chinese cabbages, 'Chunhawang' and 'Seoul' cultivars. The purified virions of RMV had the typical ultraviolet absorption spectrum of maximum at 260 nm and minimum at 247 nm. RMV of Ca1 isolate was related serologically with antisera of Tobacco mosaic virus (TMV)-Cym, TMV-O and Pepper mottle virus, but not related with antiserum of Odontoglossum ring spot virus. coat protein gene of RMV-Ca1, sized 473 nucleotides, encoded 158 amino acid residues. Nucleotide identity of RMV-Ca1 CP gene was 96.4% with RMV-Shanghai (GenBank accession No. of AF185272) from China and 96.0% with RMV-Impatiens (GenBank accession No. of AM040974) from Germany. Identity of amino acids between RMV-Ca1 and the two RMV isolates was 96.8%. Specific three primers were selected for rapid and easy genetic detection of RMV using Virion Captured (VC)/RT-PCR method.
