Expression Profiling of Genes involved in the Control of Pluripotency Using cDNA Microarray

  • To identify genes implicated in the control of pluripotency as well as characteristics of stem cells, we analyzed expression profiles of genes derived from mouse morulas, blastocysts, embryonic stem cells, mesenchymal stem cells, and uterus tissue using cDNA microarray. Comparative analyses of their expression profiles identified putative clones that expressed specifically in specific samples or not in a specific sample. The expression pattern of these candidate clones was analyzed using RT-PCR and non-radioactive in situ hybridization. Functional annotation of these clones on pluripotency and stem cell plasticity is in ongoing. These studies may further our understanding on the nature of the stem cells and molecular mechanisms underlying many facets of mammalian development and differentiation.

MOBILITY OF NATURAL TEETH AND OSSEOINTEGRATED IMPLANTS (자연 치아와 골유착성 임플랜트의 동요도에 관한 연구)

  • It is well known that implants showing no clinical mobility are successfully osseointegrated and have good prognosis. When implants are under load, their mobility begins to increase. It is of necessity to substantiate whether excessive load is on or premature occlusal force is acting prior to desirable osseointegration. Using Periotest unit, we could measure the pattern of mobility change. Consequently, osseointegrated treatment has come to success by intercepting progressive mobility and doing perceptive treatment according to the result of Periotest Value(PTV). In this study, we took records of intangible mobility of 70 osseointegrated implants. And we also measured the mobility of periodontally sound natural teeth as a standard from 30 dental personnel. Conclusions were summarized as followings ; 1. Lower lateral incisor has the highest PTV, whereas lower canine, upper canine, lower premolars and lower 1st molar have the lowest PTV in natural dentition. 2. There are little significant statistical difference of PTV between men and women in both(natural and implant) dentition. 3. In general, lower natural teeth show lower PTV than upper counterpart. 4. Mandibular implants have lower PTV than those of maxillary implants. 5. All of the successfully osseointegrated implants have lower PTV than those of periodontally healthy teeth.

Genetic Diversity and Speciation of Rana rugosa (Amphibia; Ranidae)

  • Horizontal starch gel electrophoresis for 29 populations (n=543) of the wrinkled frog, Rana rugosa, from Korea and Japan was peformed to assess the degree of genic variation and genetic diversity, and to understand the biogeographic pattern of distribution and speciation. A sum of 22 presumptive loci was screened from 17 enzymes and general proteins. Four loci, Aco, Est-3, Me-2, and Pgm, demonstrated high levels of polymorphism. The degree of average genetic variation of R. rugosa was P=22.7% (9.1-40.9%), Ho=0.086 (0.048-0.165) and He=0.090 (0.042-0.168). In the south-eastern region of the Korean peninsula (Chongsong, Yongchon, Ulsan, Kyongju, Pohang, yongdok and Ulchin), a few unique alleles in the Mpi locus were detected and their biogeographic implications were considered. The degree of genetic differentiation among the Korean populations was moderate (S=0.900), whereas the degree of genetic diversity between Korean and Japanese populations was notably high (S=0.687, D=0.293). This result corresponds with the data obtained by the mitochondrial cytochrome b gene sequence (Lee et al., 1999) suggesting that the Korean and Japanese R. rugosa might have evolved a specific level of genetic differentiation since their geographic isolation.

  • 본 논문은 지식경제부의 지원 하에 한국전자통신연구원 언어처리연구팀에서 2010년에 개발하고 있는 패턴기반 영한 메신저 대화체 문장 번역 시스템에 관한 것이다. 본 논문의 목표는 문어체 문장 위주의 패턴기반 영한 웹문서 자동번역 시스템을 대화체 문장 위주의 패턴기반 영한 메신저 자동번역 시스템으로 전환하고자 할 때, 특화하는 방법 및 모듈에 관해 기술하는 것이다. 영어권 Native speaker로부터 수집한 메신저 대화체 문장을 대상으로 번역률을 평가한 결과, 문어체 위주의 영한 웹 자동번역 시스템은 71.83%인 반면, 대화체 위주의 영한 메신저 자동번역 시스템은 76.88%였다. 대화체 문장을 대상으로 번역률을 5.05% 향상시킬 수 있었던 이유는 본 논문에서 제시한 특화 방법을 따른 결과라고 할 수 있다.

Experiment investigation on flow characteristics of open natural circulation system

  • Experimental research on flow characteristics of open natural circulation system was performed, to figure out the mechanism of the open natural circulation behaviors. The influence factors, such as the heating power, the inlet subcooled and the level of cooling tank on the flow characteristics of the system were examined. It was shown that within the scope of the experimental conditions, there are five flow types: single-phase stable flow, flash and geyser coexisting unstable flow, flash stable flow, flash unstable flow, and flash and boiling coexisting unstable flow. The geyser flow in flash and geyser coexisting unstable flow is different from classic geysers flow. The flow oscillation period and amplitude of the former are more regular, is a newly discovered flow pattern. By drawing the flow instability boundary diagram and sorting out the flow types, it is found that the two-phase unstable flow is mainly characterized by boiling and flash, which determine the behavior of open natural circulation respectively or jointly. Moreover, compared with full liquid level system, non-full liquid level system is more prone to boiling phenomenon, and the range of heat flux density and undercooling degree corresponding to unstable flow is larger.

Intron retention decreases METTL3 expression by inhibiting mRNA export to the cytoplasm

  • Methyltransferase-like 3 (METTL3), a key component of the m6A methyltransferase complex, regulates the splicing, nuclear transport, stability, and translation of its target genes. However, the mechanism underlying the regulation of METTL3 expression by alternative splicing (AS) remains unknown. We analyzed the expression pattern of METTL3 after AS in human tissues and confirmed the expression of an isoform retaining introns 8 and 9 (METTL3-IR). We confirmed the different intracellular localizations of METTL3-IR and METTL3 proteins using immunofluorescence microscopy. Furthermore, the endogenous expression of METTL3-IR at the protein level was different from that at the mRNA level. We found that 3'-UTR generation by intron retention (IR) inhibited the export of METTL3-IR mRNA to the cytoplasm, which in turn suppressed protein expression. To the best of our knowledge, this is the first study to confirm the regulation of METTL3 gene expression by AS, providing evidence that the suppression of METTL3 protein expression by IR is an integral part of the mechanism by which 3'-UTR generation regulates protein expression via inhibition of RNA export to the cytoplasm.

Does Natural Disasters Have an Impact on Poverty in East Java, Indonesia?

  • There is a strong association between poverty levels and the probability of natural disasters. East Java, however, exhibits a distinct pattern. While the rate of poverty is declining, natural disasters are becoming more severe. Considering that East Java is an area with a high risk of natural disasters and a high poverty rate, this study aims to estimate the effect of environmental preservation and the magnitude of the impact of disasters as measured by the Disaster Risk Index (IRBI) on poverty. The 3SLS model is used on secondary data from 38 districts/cities from 2015 to 2021 as an analytical database. Based on the estimation results, there are 3 findings in this study: (i) the role of government, population development, and economic activity have a strong influence on nature conservation; (ii) nature conservation has a strong influence on disaster risk; and (iii) the disaster risk index has a strong effect on poverty. As a result, areas with a high level of disaster risk have a slower rate of poverty reduction. The role of this research is to show the need for the government's role in improving the quality of natural disaster mitigation anticipation, economic activity, and the role of the population in a sustainable manner.

Occurrence of a Natural Intergeneric Hybrid between Rhodeus pseudosericeus and Acheilognathus signifer (Pisces: Cyprinidae) from the Namhangang (river), Korea (한강납줄개 Rhodeus pseudosericeus와 묵납자루 Acheilognathus signifer (Pisces: Cyprinidae)의 속간 자연 잡종 출현)

  • A natural intergeneric hybrid between Rhodeus pseudosericeus and Acheilognathus signifer was collected in their sympatric area, the Jucheongang (stream), the Namhangang (river), Korea. Morphological characters as well as mitochondrial cytochrome b (mt-cyb) gene were investigated to confirm the natural hybrid origin. In the comparison of morphological characters, the natural hybrid appeared to have intermediate characters between two parental species in several key characters such as the band of dorsal fins, the color pattern of body, and the body spot and stripe. Also, the collected specimen had an incomplete lateral line with a large number of pored scales. In analysis of mt-cyb gene, it revealed that the maternal species of the natural hybrid appeared to be R. pseudosericeus due to their 100% sequence identity. Thus, such results gave convincing evidence for the occurrence of a natural intergeneric hybrid between Rhodeus pseudosericeus and Acheilognathus signifer.

A Study on the Expression Method and Characteristics of Ecology Design in Commercial Space (상업공간에서의 자연요소 표현방법 및 특성에 관한 연구)

  • In the commercial space, the modern consumers want to consume not only product but also culture. Thus, the modern commercial space tries to induce the customer's concern and purchase to the differentiated design. The introduction of this 'the natural element' delivers the pleasure and stability to the consumer buying process and availability is enlarged. Therefore, the purpose of study is gain that expression and characteristic method of the natural element in the commercial space. The detailed study method are as follows. First, the study looks into the natural element expression tendency in the modern space. Second, the expression type of the natural element was classified as 'Reappearance', 'Transformation', and 'Fuse' based on the preceding research. And according to the content of the expression type, subdivided method of 'Inclusion' and 'Replication' of 'Reappearance' and method of 'Imitation' and 'Association' of 'Transformation' and method of 'Juxtaposition' and 'Combine' of 'Fuse'. Third, the result of analyze the characteristic of expression of the natural element of the besides the commercial space case 20 place is as follows. First, 'Inclusion' of 'Reappearance' introduced the external scenery or planned garden as the inside through the opening. 'Replication' is used for decorative purposes or functional purposes as the natural element. 'Imitation' of 'Transformation' imitated the form, pattern, and color of the natural element and was mainly expressed in the wall and objet. The method of 'Association' expressed the concept of the natural element for the whole of the space and a portion of the wall and objet repetitively. As for 'Juxtaposition', mixture of 'Reappearance' had a good visual effect, because it can be obtained the external-internal nature. As for 'Combine', mixture of 'Replication' and 'Imitation' was the most common method. Thus, the study results are expected to be utilized as base date in designing the commercial with development of the natural element application method.

Purification and Partial Characterization of a Peroxidase from Perilla Callus

  • Cotyledons of perilla6 were cultured on MS medium containing 0.5 mg/l NAA and 0.5 mg/l BA for 7 weeks. The activity of perilla peroxidase was observed to increase following culture stages as assessed by peroxidase assay. A peroxidase (POD) was purified from perilla tissue cultured on MS medium for 7 weeks. The peroxidase was purified using ion exchange and gel nitration chromatography. The perilla peroxidase had a molecular mass of 30 kDa by SDS-PAGE. We showed that the N-terminal amino acid sequence of this protein shared 67% identity with the tea peroxidase. As indicated by SDS-PAGE, the banding pattern of the 30 kDa polypeptide present in total soluble protein from perilla tissue was increased following culture stages. Immunoblot analysis indicated that perilla peroxidase protein appeared after 3 weeks of perilla tissue culture, and continued to increase with extended duration of tissue culture for at least 7 weeks.
