Influence of Alcohol Consumption on the Serum hs-CRP Level and Prevalence of Metabolic Syndrome -Based on the 2015 Korean National Health and Nutrition Examination Survey - (알코올 섭취량이 혈청 hs-CRP 수준 및 대사증후군 유병률에 미치는 영향 - 2015 국민건강영양조사 자료 이용 -)

  • Elevated high-sensitivity C reactive protein (hs-CRP) levels and metabolic syndrome are considered important predictors of cardiovascular mortality. This study examined the influence of the alcohol consumption level on the hs-CRP level and the prevalence of metabolic syndrome in South Koreans. The study subjects were 3,884 participants (${\geq}19$ years) from the KNHANES in 2015. The subjects were divided into four groups according to their alcohol consumption; None (none-alcohol consumption), Low (less than 15 g/day), Medium (15~29.9 g/day), and High (over than 30 g/day). The odds ratio for high-risk hs-CRP of men was significantly lower in the Low or Medium groups compared to the None group. In women, the odds ratio for high-risk hs-CRP was not decreased in the Low or Medium groups but was significantly higher in the High group than in the None group. Compared to nondrinkers, the prevalence of metabolic syndrome was lower in the Low group and Medium group in both men and women, whereas it was significantly higher in the High group in men only. In particular, the prevalence of low HDL-cholesterol was lower in all drinking groups. When the confounding factors were adjusted, the odds ratio for low HDL-cholesterol appeared to decline in the Low groups in both men and women. On the other hand, in the Medium group, women showed an elevated odds ratios of high blood pressure (HBP) and high fasting plasma glucose but men showed an elevated odds ratios of HBP and high triglyceride levels. The overall results suggest that low alcohol consumption is more appropriate than medium alcohol consumption. Nevertheless, more study will be needed to evaluate the appropriate alcohol consumption level.

  • In order to assign NMR signals, conditions for the specific labeling of cytochrome $c_3$ of D. vulgaris Miyazaki F through the culture in a minimal medium were established. Phenylalanine residue was specifically deuterated at more than 85% efficiency. Cytochrome $c_3$ has two phenylalanine residues. The signals of one phenylalane were missing and this was tentatively assigned to Phe20.

  • The gravimetric determination of scandium by di-(2-ethylhexyl)phosphate(DEHPA) as a precipitant and the mole ratio of Sc-DEHPA arecipitation obtained in ethanol medium have been investigated. Scandium can be determined by gravimetric method of precipitation of Sc-DEHPA and the mole ratio of Sc-DEHPA is found as 1 :2 in ethanol medium.

  • Conditions for the production and regeneration in Lactobacillus bulgaricus protoplasts were investigated. Protoplasts of L bulgaricus strains were obtained by treatment with mutanolysin and lysozyme together in a protoplast forming buffer containing 0.02 M N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid (HEPES) (pH 7.0) and 0.5 M sucrose. High protoplast yield was obtained from cells cultured in the de Man, Rogosa and Sharpe(MRS) medium at the middle to late logarithmic growth phase. Regeneration was efficiently accomplished with a complex medium containing 1% sucrose, 20 mM $MgCl_2$, 5% gelatin, and 0.5% bovine serum albumin. The frequency of regeneration of protoplasts was 10~20% after 5 days of incubation at $30^{\circ}C$.

  • Enterobacter sp. BL-2 excretively produced a unique cationic polyglucosamine biopolymer PGB-1 comprised of more than 95% D-glucosamine in an acetate-mediated culture condition. The excretion of the biopolymer PGB-1 was closely associated with the cellular morphology of Enterobacter sp. BL-2, a feature highly dependable on the pH of the medium. The initially formed uneven and irregular surface cells were aggregated into the cell-biopolymer network structure connected by the adhesion modules of the cell-bound biopolymer. The excretive production of the biopolymer PGB-1 coincided with the disruption of the cell-biopolymer network, most actively at the medium pH of 8.0.

  • In order to use the symbiotic bacteria from ethomophatogenic nematodes as a biological control agent for agriculture, the cultural condition for maintaining phase I and antifungal activity was investigated. Symbiotic bacteria (SB) 1 stain from nematodes were selected from the three strains isolated from entomopathogenic nematodes. The growth of the SB 1 strain in NB, TSB, TY and YS medium was higher than that of the SB 2 and SB 3 strain. The packed cell volume of the SB 1 strain was reduced in NB medium which showed radical pH change. Phase I of the SB 1 strain was maintained in TSB medium after being stored for 2 weeks at $4^{\circ}C$. Culture broth with the SB 1 strain in TSB medium for 6 days and 7 days showed antifungal activities against Rhizoctonia solani KACC 40142, Botrytis cinerea Pers. KACC 40854, and Botrytis cinerea Pers. KACC 41008. Culture broth with the SB 1 strain in TSB medium containing 100 mM L-proline for 5 days showed antifungal activities against Rhizoctonia solani KACC 40142, and Botrytis cinerea Pers. KACC 40854.

  • Multiple color selection techniques are successful in identifying quasars from wide-field broadband imaging survey data. Among the quasars that have been discovered so far, however, there is a redshift gap at 5 ≲ z ≲ 5.7 due to the limitations of filter sets in previous studies. In this work, we present a new selection technique of high redshift quasars using a sequence of medium-band filters: nine filters with central wavelengths from 625 to 1025 nm and bandwidths of 50 nm. Photometry with these medium-bands traces the spectral energy distribution (SED) of a source, similar to spectroscopy with resolution R ~ 15. By conducting medium-band observations of high redshift quasars at 4.7 ≤ z ≤ 6.0 and brown dwarfs (the main contaminants in high redshift quasar selection) using the SED camera for QUasars in EArly uNiverse (SQUEAN) on the 2.1-m telescope at the McDonald Observatory, we show that these medium-band filters are superior to multi-color broad-band color section in separating high redshift quasars from brown dwarfs. In addition, we show that redshifts of high redshift quasars can be determined to an accuracy of Δz/(1 + z) = 0.002 - 0.026. The selection technique can be extended to z ~ 7, suggesting that the medium-band observation can be powerful in identifying quasars even at the re-ionization epoch.

Mechanism for the Action of Co-culture (공배양의 작용기전에 관한 연구)

  • Objective: A number of studies to improve in vitro culture conditions have been tried over past ten years by using co-culture system with helper somatic cells. However, the mechanism of coculture is poorly understood. This study was designed to understand the mechanism for the mode of actual action of co-culture using co-culture system of ICR strain's 1-cell embryos with human oviduct epithelial cells by examining the effect of conditioned medium and contactless coculture using a cell culture insert on the embryo development and by measuring the level of superoxide anion from conditioned medium after co-culture. Methods: ICR strain's zygote embryos were cultured in medium alone (control), coculture, conditioned medium, or contactless coculture system for 6 days. Conditioned media (CM) were prepared as following 5 groups. All CM were collected after culturing oviduct cells for 2 days. CM-1 was stored at $-20^{\circ}C$ until use, and CM-2 was prepared just before use as a culture medium. CM-3 was cocultured with embryos and retrieved just before use. CM-4 and CM-5 were derives from the microfilteration of CM-2 and CM-3, respectively, using Microcon-10 (10 kDa molecular weight cut-off). The percentage of the embryos developed to hatched blastocyst stage and the level of superoxide anion in supernatant from medium alone culture (control), coculture, and contactless coculture were measured. Results: The rates of embryo development to the hatched blastocyst stage were significantly higher in coculture (43%) than in control (0%) (p<0.05). The CM-1 group had no embryo development since 2-cell embryonic stage, whereas the CM-2, CM-3, CM-4 and CM-5 groups had the improved development to 4 or 8 cell embryo stage, but the similar rate of development to hatched blastocyst compared to control. The effect of coculture on embryo development was disappeared in the contactless coculture group. The level of superoxide anion was significantly reduced in coculture group compared to control. Conclusion: It is concluded that the present coculture system overcomes the 2-cell block in vitro and improves the embryo development. This beneficial effect may be due to the direct cell-cell contact between embryo and helper cells or the removal of deleterious components from medium rather than the embryotrophic factors.

Optimization of Mass cultivation Media for the Production of Biomass and Natural Colourants from Two Marine Cyanobacteria by a Mathematical Design of Experiments

  • Optimization of chemicals in the large scale sea water medium and inoculum for biomass and natural colourants production in the marine cyanobacteria, Phomidium tenue BDU 46241 (phycoerythrin producer) and P.valderianum BDU 30501 (phycocyanin producer) was carried out by experiments in L8 orthogonal array. Mathematical analysis revealed the significance of these factors. The factor(s) that critically control the yield varied with the organism and the end-product further, the desirable level of these factors between the normal and a higher level tested was identified and improved media were evolved. In both cyanobacteria, higher level of $K_2$$HPO_4$, $NaNO_3$ and inoculum with normal level of ferric ammonium citrate was found to be desirable for biomass production and additionally, higher level of $MgSO_4$ for pigment production. The level of other factors varied with the organism and the end-product. Confirmation experiments showed that the clues obtained based on mathematical experimentation are valid. In P.tenue, the medium optimized for biomass production increased the yield of biomass by 495% and the medium optimized for phycoerythrin production increased the yield of biomass by 408% with 30% increase in phycoerythrin content of the biomass. Similarly in P.valderianum, the medium optimized for biomass production increased the yield of biomass by 224% and the medium optimized for phycocyanin production increased the yield of biomass by 143% with 44% increase in phycocyanin content of the biomass.

Secretion of Human Angiogenin into Periplasm and Culture Medium with Its Eukaryotic Signal Sequence by Escherichia coli

  • The synthesis and secretion of human angiogenin in E. coli by the natural leader sequence has been studied. We constructed a recombinant plasmid containing human angiogenin cDNA which encompassed all the coding region including leader sequence required for secretion. The recombinant plasmid was introduced into a suitable E. coli host. The angiogenin was detected in the culture medium and periplasm upon the induction of gene expression. The molecular weight of the secreted angiogenin was identical to that of authentic angiogenin purfied from human plasma when estimated by SDS-PAGE and immunoblotting. showing that the natural leader sequence was recognized and processed by the secretion machinery of E. coli. The angiogenin concentration in the culture medium reached a maximum within 2 h when expressed at $37^{\circ}C$ with 0.02~2 mM IPTG. In contrast, the expression level increased gradually over time up to 11 h at $23^{\circ}C$ with 0.002~2 mM IPTG and at $37^{\circ}C$ with 0.002 mM IPTG.
