The purpose of this study was to identify the relationship of symptom distress and natural killer cell cytotoxicity in breast cancer patients who had been radiation therapy and/or chemotherapy after surgery. Symptom distress measured by modified Lee's(1994) physical symptom questionnaire. For measuring the natural killer cell cytotoxic activity. 8ml to 10ml blood was collected from the subjects. Mononuclear cell was isolated by centrifuge of the blood and cultured by putting $Cr^{51}$, and reacted with target cell, K562 cell. Amount of $Cr^{51}$ was measured, and %lysis was calculated. The results were as follows. 1) Symptom distress score was 42.18, which is moderate symptom distress. 2) Natural killer cell cytotoxic activities were 42.18%lysis(effector : target cell ratio=100 : 1) and 28.05%lysis(effector : target cell ratio=50 : 1). 3) Correlation coefficients of symptom distress and natural killer cell cytotoxic activity were $-.134{\sim}-.461$. Though significant correlation was not found between total score of symptom distress and natural killer cell cytotoxic activity, 3('pain' 'feel hot on radiation site' and 'difficulty in breathing') of 19 symptom distress items and natural killer cell cytotoxic activity showed significant negative correlation(p<.05). These findings suggest that 1) breast cancer patients who had been radiation therapy and/or chemotherapy after surgery have moderate symptom distress and decreased natural killer cell cytotoxic activity. 2) The symptom distress was not related to natural killer cell cytotoxic activity.
The purpose of this study was to determine the effect of exercise program on natural killer cell cytotoxic activity(NKCA) in breast cancer patients who had been radiation therapy after surgery. The subjects in the experimental group consisted of 11 breast cancer patients, while the subjects in the control group consisted of 15. Subjects in the experimental group participated in exercise program for 8 weeks. Exercise program consisted of shoulder stretching, arm weight training and treadmill walking exercise. They started to exercise on treadmill for 20 minutes per day, 3 times a week at 40% of maximum heart rate, and increased intensity and duration of exercise so that they were running 30 minutes/day at 60% of maximum heart rate from the 3rd week to the 8th week. Natural killer cell cytotoxic activity were determined before and after the exercise program. For measuring the natural killer cell cytotoxic activity, 8ml to 10ml blood was collected from the subjects. Mononuclear cell was isolated by centrifuge of the blood and cultured by putting $Cr^{51}$, and reacted with target cell, K562 cell. Baseline demographic and medical data were compared between groups with the Fisher's exact test and Mann-Whitney U test. For effects of the exercise program, repeated measures ANOVA was used. The result was as follows; Natural killer cell cytotoxic activity(NKCA) in experimental group comparing with control group significantly increased after the exercise program in case of effector cell : target cell ratio is 100 : 1(p<0.05). The above result suggest that the exercise program for breast cancer patients undergoing radiation therapy after breast surgery may increase the natural killer cell cytotoxic activity.
Natural killer (NK) cells are a heteroguneous subpopulation of lymphocytes that spontaneously exhibit cytotoxic activity against various virus-Infected and neoplastic target cells without prior exposure to a specific antigen. It was thought that NK calls play an important role in immunosurvrillanre against viral agents and tumors, and in prevention of metastasis. Recently, several reports have indicated evidence that ginseng extracts show a significant stimulatory effect on the humoral and cellular immune responses. This evidence gives support to the suggestion that the anticarcinogenic effect of ginseng may be due to the effect of ginseng on the immunological system. Treatment with total, diol, and triol saponin resulted in an increase in NK cytotoxic activity, but no enhancement of the lytic activity due to the natural killer cytotoxic factor (NKCF). Therefore, these results suggest that the augmentation of NK activity by ginseng saponin fractions may not be due to the activation of NKCF lytic activity.
Natural killer (NK) cells are innate cytotoxic lymphoid cells that actively prevent neoplastic development, growth, and metastatic dissemination in a process called cancer immunosurveillance. Regulation of the cytotoxic activity of NK cells relies on integrated interactions between inhibitory receptors and numerous activating receptors that act in tandem to eliminate tumor cells efficiently. Conventional chemotherapy is designed to produce an anti-proliferative or cytotoxic effect on early tumor cell division. Therapies designed to kill cancer cells and simultaneously maintain host anti-tumor immunity are attractive strategies for controlling tumor growth. Depending on the drug and dose used, several chemotherapeutic agents cause DNA damage and cancer cell death through apoptosis, immunogenic cell death, or other forms of non-killing (i.e., mitotic catastrophe, senescence, autophagy). Among stress-induced immunostimulatory proteins, changes in the expression levels of NK cell activating and inhibitory ligands and tumor cell death receptors play an important role in the detection and elimination by innate immune effectors including NK cells. Therefore, we will address how these cytotoxic lymphocytes sense and respond to high and low concentrations of drug-induced stress to the drug cisplatin, among the various types of drugs that contribute to their anticancer activity.
The number of splenic lymphocyte, serum prostaglandin $E_2$ level and natural killer cell activity were assayed after single whole body irradiation of a sublethal dose of $^{60}Co-{\gamma}$ ray to C57BL/6J mice. With a view to knowing the relationships between radiation induced prostaglandin $E_2$ level and the normal natural killer cell activity after natural killer cell-target cell conjugation, The change of normal natural killer cell activity were measured by administration of prostaglandin $E_2$ containing serum from irradiated mice. The results were summarized as follows; 1. The total number of splenic lymphocyte was significantly decreased by irradiation and the number was not affected by indometacin, prostaglandin synthesis inhibitor, treatment. 2. Serum prostaglandin $E_2$ level was increased in irradiated mice, but indometacin treated mice group showed low level of prostaglandin $E_2$. 3. In the case of irradiated mice, natural killer cell activity was not shown any difference between irradiated group and indometacin combined group. But when natural killer cell-target cell conjugations were exposed to the serum of each group during cytotoxic activity assay, whereas the normal natural killer cell activity was significantly decreased by treatment of serum from irradiated mice, the activity was not changed by treatment of indometacin pretreated mice serum. This result indicated that the prostaglandin $E_2$ induced by the radiation inhibited the post-target binding cytolytic process of natural killer activity.
Natural killer (NK) cells are innate lymphocytes that play an essential role in preventing cancer development by performing immune surveillance to eradicate abnormal cells. Since ex vivo expanded NK cells have cytotoxic activity against various cancers, including breast cancers, their clinical potential as immune-oncogenic therapeutics has been widely investigated. Here, we report that the pyrazole chemical XRP44X, an inhibitor of Ras/ERK activation of ELK3, stimulates NK-92MI cells to enhance cytotoxic activity against breast cancer cells. Under XRP44X stimulation, NK cells did not show notable apoptosis or impaired cell cycle progression. We demonstrated that XRP44X enhanced interferon gamma expression in NK-92MI cells. We also elucidated that potentiation of the cytotoxic activity of NK-92MI cells by XRP44X is induced by activation of the c-JUN N-terminal kinase (JNK) signaling pathway. Our data provide insight into the evaluation of XRP44X as an immune stimulant and that XRP44X is a potential candidate compound for the therapeutic development of NK cells.

Background: Chronic pain reportedly exerts complex effects on immune function. Natural killer (NK) cells are lymphocytes that play a critical role in cellular and innate immunity. This study examined changes in the subset populations and cytotoxic activity of peripheral blood NK cells in patients with chronic pain. Methods: Thirty patients with chronic moderate-to-severe pain (group P) and age-matched pain-free subjects (group NoP) were enrolled. Peripheral whole blood was analyzed for the percentage and expression of NK cell surface markers (CD56 and CD16) by flow cytometry. Cytotoxic activity was assayed by evaluating CD69 expression on $CD3^-/CD56^+NK$ cells. Results: The percentage of NK cells among total lymphocytes was not significantly different between groups P and NoP ($16.3{\pm}9.3$ vs. $20.2{\pm}10.5%$). Likewise, the percentages of two major NK cell subsets, $CD56^{bright}$ and $CD56^{dim}$, were also not significantly different between the two groups. However, the percentage of $CD56^{bright}/CD16^+$ subset, was slightly but significantly increased in group P ($1.0{\pm}0.9%$; P< 0.01) compared with group NoP ($0.5{\pm}0.6%$). The cytotoxicity of NK cells was not different between the two groups, showing similar CD69 expression (P vs. $NoP=29.2{\pm}15.2$ vs. $32.0{\pm}15.0%$). These findings were not influenced by pain intensity, opioid use, or disease causing pain in group P. Conclusions: NK cell cytotoxic activity and major subset populations, with the exception of an increased percentage of the $CD56^{bright}/CD16^+$ subset, are not significantly altered in patients with chronic severe pain.
Phellinus linteus was examined for its anticancer activity using an animal model. Water extract of Phellinus linteus was prepared from artificially cultivated mycelia. Neither toxicity nor abnormal changes of hematological parameters were observed in the rat given orally with high doses of drug extract for 15 days. ICR mice were transplanted with Sarcoma-180 tumor cells intraperitoneally and drug extract was daily given to the mice from 1 day after tumer transplantation for 3 weeks. Administration of drug extract significantly prolonged the survival duration of Sarcoma 180-transplanted mice. For the better understanding of the anticancer activity, we have examined the effect of the drug extract administration on various killer cell functions, such as natural killer(NK) cells, cytotoxic T-lymphocytes (CTL) and macrophages which have been known to be main effector cells in immune responses against tumors. The results from the 4 hr $^{51}Cr-release$ assay have shown that the drug extract augments mouse NK cell activity but neither CTL nor macrophages. It is possible, then, that the anticancer activity of the Phellinus linteus may be associated with augmentation of NK cell function in the cancerated hosts.
Aim : Natural killer (NK) cells are cytotoxic lymphocytes that lyse certain tumor- and virus-infected cells without any prior stimulation or immunization. This article aims to review the significance of evaluating peripheral blood NK cells to predict anti-tumor immune function and prognosis in cancer patients. Methods : PubMed was used to create a database for this review. Search words of cancer, natural killer cell, prognosis were used to retrieve related articles. References of the collected articles were also reviewed. Results : Current evidence indicates that decreased or absent NK cell count or activity is mostly associated with the development or progression of cancer. In patients with various types of cancer, NK cell activity was mostly associated with the cancer prognosis and survival despite some conflicting results. Conclusion : The data shows the evaluation of anti-tumor immunity in cancer patients through natural killer cell measurement still remains a controversial matter. However, it is clear that the NK cell activity plays an important role in cancer and is associated with prevention of both early stage and metastatic cancer.
Objectives : This study examined the effects of Bee venom on apoptosis in NCI-H157 human lung cancer cells and for promoting the apoptosis effects of Natural killer cell. Methods : Bee venom and Natural killer-92 cells were cultured either separately from or together with NCI-H157 cells for 24 hours. To figure out whether Bee venom enhances the cytotoxic effect of Natural Killer-92 cells, a cell viability assay was conducted. To observe the changes in Death receptors, apoptotic regulatory proteins and Nuclear $Factor-{\kappa}B$, western blot analysis was conducted. To observe the effect of Bee venom through an extrinsic mechanism, a transfection assay was conducted. Results : 1. Natural killer-92 cells and Bee venom significantly inhibited the growth of NCI-H157 cells and co-culture had more inhibitory effect than the separate culture. 2. Expressions of Fas, DR3, DR6, Bax, caspase-3, caspase-8, cleaved caspase-3, cleaved caspase-8 were increased, and expressions of Bcl-2 and cIAP were decreased. More efficacy was observed in co-culture than in separate culture. 3. Nuclear $Factor-{\kappa}B$ activation was clearly decreased. And co-culture showed much less activation than separate culture. 4. As a result of treatment for DR-siRNA, the reduced cell viability of NCI-H157 cells and the activity of Nuclear $Factor-{\kappa}B$ were increased. With this, it can be seen that Bee venom and Natural killer-92 cells have an effect on the cancer cells through the extrinsic mechanism. Conclusion : Bee venom is effective in inhibiting the growth of human lung cancer cells. Furthermore Bee venom effectively enhances the functions of Natural killer cells.
