The importance of natural preservative has increased in recent years. The natural preservatives have been used in the field of foods, cosmetics and pharmacology. In the present work Artemisia sp., well recognized for their effect of antimicrobial activity, were extracted by methanol and water sequentially for selecting only water-soluble compounds that can be used as additives in food and cosmetics. Antimicrobial activities of water extracts from stem and leaf of Artemisia princeps var. orientalis were investigated by the disc diffusion method. Two gram positive bacteria(Staphylococcus aureus and Bacillus subtilis) and three gram negative bacteria(Escherichia coil, Agrobacterium tumefaciens and Pseudomonas putida) were used for antimicrobial activity studies. The water-soluble compounds from methanol extract showed higher antimicrobial activity than only water extract to these bacteria. Comparative evaluation of water-soluble metabolite profiles with caffeic acid that is known as an antimicrobial compound from Artemisia sp. was performed by high performance liquid chromatography with photo-diode array detection.
This study was preformed to make high quality fish cake made with silver pomfret, which is one of the savory, soft and delicious fishes by adding wasabi powder (WP). WP as natural additives was added to the ratio of 0.6%, 1.2%, 1.8%, 2.4%, 3%, a color values, texture, folding test, sensory evaluation, peroxide value, TBA value, and viable cell count were analyzed. L and a value were decreased significantly by the increase of the ratio of WP. However, b value was increased. In the folding test to show the flexibility of fish cake, it was measured AA in the all samples. As the result of measuring texture, hardness was increased significantly by the addition of WP, cohesiveness was the highest in 1.8 WP and 0.6 WP, Springiness showed no significant difference among samples. However gumminess and chewiness of fish cake adding wasabi powder were significantly higher comparing with control group. In sensory evaluation, 1.8 WP had the best score in overall acceptability. Fish cakes had lower peroxide value, TBA value and viable cell count after frying compared to the one without WP. These results indicate that fish cake could be prepared by adding the WP for high quality and functionality. Consequently, wasabi can be applied as a food preservative or additive in fish cake.

Citric acid is a natural preservative and is used to add a sour taste to foods and soft drinks. For the preparation of stable food additives of citric acid, a coating process using vegetable oil was developed. Coating materials used were purified soy bean oil and carnauba wax. The yield of coated citric acid was up to 95% in both cases. The contents of coating materials was 20~33% in the total composition and the coating efficiency was 95.2 ${\pm}$ 0.01%. The surface of coated citric acid was much smoother and more homogeneous than that of original citric acid according to SEM data. The coated citric acid can be used to as a stable food additive and also would be applied to nutraceuticals and cosmetic ingredients.
Sprassis crispa was cultivated using soybean curd whey, and its antimicrobial activities were examined against those of eight microorganisms that were foodborne pathogens or food-poisoning bacteria. The culture broth of soybean curd whey was superior in mycelium content (17.76 g/L) to that of the defined culture broth, and the ${\beta}$-glucan content was about 10.64 percent (w/w). The antimicrobial activities of the culture broth were confirmed against those of B. cereus, St. aureus, L. monocytogenes and S. typhimurium using the paper disk method. The antimicrobial activity was also maintained after the heat treatment and alcalase treatment. The filtrate with less than 3 kDa M.W. also showed the antimicrobial activity against four strains: B. cereus, St. aureus, L. monocytogenes and S. typhimurium. The minimum inhibitory concentration (MIC) was about 1.26 mg/mL in the B. cereus and 12.6 mg/mL in the St. aureus and L. monocytogenes. The S. typhimurium showed a MIC of 62.8 mg/mL. Thus, the culture of Sparassis crispa using soybean curd whey provides a thermally stable antimicrobial agent that can be used as a natural preservative in the biofood industry.
It has been reported that alginic acid hydrolysate retains antimicrobial activity but the enzyme which hydrolyze alginic acid is not developed for industrial use. The authors developed chemical method for hydrolyzing alginic acid. For preparing alginic acid hydrolysate, equal quantity of alginic acid and ascorbic acid were added to water. Then the solution was heated at $121^{\circ}C$ for $20{\sim}30{\;}minutes$. The 4% solution of alginic acid hydrolysate was revealed relative viscosity 1.05, pH 3.2 and opaque whitish-yellow color. By addition of this hydrolysate to nutrient broth with the concentration of 0.1%, the growth of Bacillus sp. isolated from fish meat paste products was inhibited. The fish meat paste products containing 0.3% alginic acid hydrolysate prepared were prolonged their shelf life by 1 day stored at $30^{\circ}C$, 2 days at $20^{\circ}C$ and 4 days at $15^{\circ}C$.
Hen egg white lysozyme (HEWL) is very valuable as a natural preservative in food processing due to its selective bactericidal activity. HEWL which traditionally isolated by crystallization or freeze drying was simply separated from 13 different hen egg white (HEW) proteins by a single-step ultrafiltration. Freeze dried HEW (0.25%, w/v) dissolved in a citrate-phosphate buffer (pH 4.6) was ultrafiltered with a PM30 membrane under various operating conditions, by changing concentration, temperature, transmembrane pressure $({\triangle}P_T)$, and stirring speed. Optimum separation conditions were decided when maximal flux was obtained. Under the optimum separation conditions, the effect of membrane material and fouling on flux as time passed as well as lysozyme concentration, protein concentration, specific activity (SA) in the permeate were measured. Best separation conditions of HEWL with PM30 membrane were sample concentration 0.25%, temperature $35^{\circ}C$, ${\Delta}P_T\;30\;psi$, and stirring speed 300 rpm. During the first 12 min, the flux of YM30 was higher, but at the steady-state it was lower than that of PM30. The SA of the PM30 permeate was over 2 times higher in spite of the lysozyme and protein concentration being lower than that of YM30 permeate. The flux of 5 times used PM30 decreased 30% compared to a new PM30, but both had the same tendency in flux decrease when time passed. Both of them reached a steady-state after 35 min and remained at 70% of the initial flux. In the PM30 permeate, the lysozyme concentration and SA were 110 units/mL and 2,821 units/mg protein, respectively. Therefore, PM30 membrane separation was very effective for separation of antimicrobial lysozyme.
Yam has been recognized as healthy food due to its various biological activities, such as anti-obesity, antimicrobial, anticancer and immune-stimulation activities. In this study, antibacterial activities of 800 more natural plant extracts against yam-putrefactive psychrotrophic bacterium, Pseudomonas rhodesiae YAM-12, were evaluated to select a natural preservative, which is useful to long-term storage of yam and fresh-cut production. Finally, the clove oil was selected, and applied for the production of yam fresh-cut. The 1% of clove oil treated fresh-cut showed minor color changes during 31 days storage at $4^{\circ}C$ and the microbial growth was not detected. When the artificially contaminated fresh cut by dipping with P. rhodesiae YAM-12 suspension for 3 min was treated 1% clove oil, the microbial growth was identified to $10^4$ CFU/g from $10^3$ CFU/g with minor color changes. Whereas, treatment of sterilized water, or 100 ppm NaOCl into artificially contaminated fresh cut showed severe putrefaction $(10^8\;CFU/g)$ and color changes. Considering the previous reports that clove oil has antimicrobial, antioxidation, and antithrombosis activity, the use of clove oil into the yam fresh cut will provide market safety and consumer acceptability by prevention of microbial putrefaction and its biological activity.

Water and 75% ethanol extracts were prepared from defatted green tea seeds and evaluated for their anti-yeast activities. The antimicrobial activities of defatted green tea seed extracts (DGTSEs) were tested against food-spoilage bacteria, yeasts, and molds. DGTSEs exhibited antimicrobial activities with minimum inhibitory concentrations of $39{\sim}1,250{\mu}g/mL$ against three bacteria, two molds, and all tested yeast strains. Ethanol extract showed higher antimicrobial activity than water extract. The stability of anti-yeast activities of DGTSEs was examined under different conditions of temperature, pH, and NaCl concentrations. The anti-yeast activities of DGTSEs were stable at pH 3~9, 0~20% NaCl, and $100^{\circ}C$ for 30 min. However, anti-yeast activities of DGTSEs decreased upon heating at $70^{\circ}C$ for 24 h or $121^{\circ}C$ for 15 min. DGTSEs were applied to food models to determine their inhibitory effects on yeast film formation. Water and 75% ethanol extracts were effective in preventing yeast film formation at concentrations more than 156 and $39{\mu}g/mL$ in soy sauce, 156 and $78{\mu}g/mL$ in pickle sauce, and 78 and $39{\mu}g/mL$ in kimchi, respectively.

An annual plant, Eclipta prostrata (Linn) is a member of the Asteraceae plant family and inhabited in tropical or subtropical regions of the world. Through many previous researches, E. prostrata has been extensively studied for its hepatoprotective effect, antivenom potential against viper venom, antioxidant, hair-growth, wound-healing efficacy and so on. In this study, for better understanding of the potential of E. prostrata as skin protectant, we conducted the experiments evaluating the antioxidant and antiaging efficacy. To this end, 50% ethanolic extract of E. prostrata and its ethyl acetate fraction were prepared and investigated. For the evaluation of antioxidant capacity of the samples, $FSC_{50}$ and $OSC_{50}$ were estimated. As a result, $OSC_{50}$ of ethyl acetate fraction was 2.7 times superior to $OSC_{50}$ of L-ascorbic acid, a well known antioxidant agent. Futhermore E. prostrata showed notable reactive oxygen species (ROS) scavenging effect and protective effect against $H_2O_2$ in the celluar level as well. Especially, in the $^1O_2$ induced hemolysis test, $64{\mu}g/mL$ of ethyl acetate fraction showed greater than 6 times increased retardation effect compare to control which means E. prostrata has remarkable antioxidant capacity. To validate the antiaging effect of the samples, we conducted elastase inhibition assay using elastase solution extracted from human skin fibroblasts, Hs68. As a result, $16{\mu}g/mL$ of each sample showed 6.8% and 14.0% of elastase inhibition respectively. Finally, antimicrobial activity of E. prostrata was assessed to validate the possibility as alternative preservative. From the result, ethyl acetate fraction showed oustanding antimicrobial activity as of methyl paraben, a well known chemical preservative. In conclusion, these results suggest that E. prostrata can be used as natural skin protectant or preservative as natural ingredient in food or cosmetics industry.

A study was carried out to produce antimicrobial peptides from zein treated with pretenses of six kinds. Among the pretenses of six kinds, zein hydrolysate treated with pepsin showed the highest antimicrobial activity. The zein hydrolysate with pepsin was fractionated with membrane filter (30,000 10,000 and 3,000 molecular weight cut-off) and antimicrobial activity was measured for each fractions. Antimicrobial activity appeared greatly in the fraction below 3,000 (molecular weight cut-off) . The fraction was re-fractionated by HPLC and substances of two peaks collected as a sample to measure antimicrobial activity. All of both peaks showed the antimicrobial activity but 1st peak exhibited a consistently higher antimicrobial activity than 2nd peak. Minimum inhibitory concentrations (MIC) were between 2.5 and 3.0 mg/mL. The peptide was heat-stable since antimicrobial activity was maintained after treated with heat for 20 min at $121^{\circ}C$. N-terminal amino acid sequence of peptide fractionated by HPLC was leucine, glutamic acid, proline, phenylalanine, aspartic acid and argenine. These results indicated that peptide isolated from zein hydrosate with pepsin can use as a natural preservative ingredient in food industry.
