The chemical characteristics, enzymatic saccharification, and ethanol fermentation of autohydrolyzed lignocellulosic material that was exposed to steam explosion were investigated using bagasse as the sample. The effects of the steam explosion on the change in pH, organic acids production, degrees of polymerization and crystallinity of the cellulose component, and the amount of extractive components in the autohydrolyzated bagasse were examined. The steam explosion decreased the degree of polymerzation up to about 700 but increased the degree of crystallinity and the micelle width of the cellulose component in the bagasse. The steam explosion, at a pressure of 2.55 MPa for 3 mins, was the most effective for the delignification of bagasse. 40 g/L of glucose and 20 g/L of xylose were produced from 100 g/L of the autohydrolyzed bagasse by the enzymatic saccharification using mixed cellulases, acucelase and meicelase. The maximum ethanol concentration, 20 g/L, was obtained from the enzymatic hydrolyzate of 100 g/L of the autohydrolyzed bagasse by the ethanol fermentation using Pichia stipitis CBS 5773; the ethanol yield from sugars was 0.33 g/g sugars.
On the purpose of exploiting cellulose resourse which is needed for artificial diet of silkworms, mulberry branch, eastern cotton wood branch, pitch pine pulp, poplar pulp, chaff, sawdust and rice straw were examined. Among the matters above mentioned. mulberry branch and pitch pine pulp were found to be replaceable as cellulose component in the artificial diet. By means of some chemical treatments, it is found that one of the extracted cellulose powder from mulberry branches is more useful in the larval growth.
Bacterial cellulose (BC) has played important role as new functional material for food industry and industrial products based on its unique properties. The interest in BC from static cultures has increased steadily in recent years because of its potential for use in medicine and cosmetics. In this study, we investigated culture condition for BC production by Acetobacter sp. F15 in static culture. The strain F15, which was isolated from decayed fruit, was selected on the basis of BC thickness. The optimal medium compositions for BC production were glucose 7%, soytone 12%, $K_2HPO_4$ 0.2%, $NaH_2PO_4{\cdot}_2H_2O$ 0.2%, lactic acid 0.05% and ethanol 0.3%, respectively. The strain F15 was able to produce BC at $26^{\circ}C-36^{\circ}C$ with a maximum at $32^{\circ}C$. BC production occurred at pH 4.5-8 with a maximum at pH 6.5. Under these conditions, a maximum BC thickness of 12.15 mm was achieved after 9 days of cultivation; this value was about 2.3-fold higher than the thickness in basic medium. Scanning electron micrographs showed that BC from the optimal medium was more compact than plant cellulose and was reticulated structure consisting of ultrafine cellulose fibrils. BC from the optimal medium was found to be of cellulose type I, the same as typical native cellulose.

In this study, 13 water soluble polymers(6 natural polymer, 7 synthetic polymer) were treated on Hanji. Mechanical properties, morphology and oxidation index with thermal aging were measured on the aged Hanji, dewaxed Hanji and polymer treated Hanji. 3 natural polymer(such as CMC, EC, MC) and 3 synthetic polymers(such as PVA 1500, 2000, PEG 1500) treated Hanji had higher strength than other polymer treated Hanji. The oxidation index of 3% methyl cellulose solution treated Hanji did a little increase with thermal aging. Finally, methyl cellulose was found to be the most efficient method for strengthening the dewaxed Hanji. The best aging safety and thermal stability were obtained at the methyl cellulose 3% water solution.
The binding of cellobiohydrolases to cullulose is a crucial initial step in cellulose hydrolysis. In the search for a detailed understanding of the function of cellobiohydrolases, much information concerning how the enzymes and their constituent catalytic and cellulose-binding changes during hydrolysis is still needed. The adsorption of purified two cellobiohydrolases (Ce17A and Ce16A) from Trichoderma reesei cellulase to microcrystalline cellulose has been studied. Cellobiohydrolase II (Ce16A) does not affect the adsorption of cellobiohydrolase I (Ce17A) significantly, and there are specific binding sites for both Ce17A and Ce16A. The adsorption affinity and tightness of the cullulase binding domain (CBD) for Ce17A are larger than those of the CBD for Ce16A. The CBD for Ce17A binds more rapidly and tightly to Avicel than the CBD for Ce16A. The decrease in adsorption observed when the two cellobihydrolases are studied together would appear to be the result of competition for binding sites on the cellulose. Ce17A competes more efficiently for binding sites than Ce16A. Competition for binding sites is the dominating factor when the two enzymes are acting together, furthermore adsorption to sites specific for Ce17A and Ce16A, also contributes to the total adsorption.

Cellulose based electro-active paper(EAPap) is considered as a new smart material which has a potential to be used for biomimetic actuators and sensors. Beside of the natural abundance, cellulose EAPap is fascinating with its biodegradability, lightweight, high mechanical strength and low actuation voltage. When the external stress is applied to EAPap, it can generate the electrical output due to its piezoelectric property. Using piezoelectric behavior of EAPap, we studied the feasibility of EAPap as mechanical strain sensor applications and compared to commercial strain sensor. By measuring the induced output voltage from the thin piezoelectric cellulose EAPap under static and dynamic force, we propose cellulose EAPap film as a potential strain sensor material.
The sequence coding for carboxymethylcellulase (CMCase, CelC) was isolated from the DNA of Salmonella typhimurium URl. Comparison between the deduced amino acid sequence of CelC (368 amino acid residues, Molecular mass 41 kDa) and that of the previously published CMCase revealed that this enzyme belongs to the cellulase family 8 and D. The protein was overproduced in Escherichia coli using T7 expression system, and its activity was confirmed by CMC-SDS-PAGE. When the overexpressed CelC protein was tested on cellulose-type substrates, the recombinant protein is able to degrade cellulose-type substrates, such as CM-cellulose, xylan, avicel, lichenan, and laminarin. Optimal temperature and pH for enzyme activity were found to be 50$^{\circ}C$ and pH 6.5, respectively.
Chitosan(CS) and cellulose acetate(CA) composite films were prepared using formic acid as a cosolvent by casting, solvent evaporating and neutralization method. This study examines if the blending method, which uses formic acid as a cosolvent is efficient in improving the mechanical properties of CS film, especially wet strength and elongation. Formic acid is an effective cosolvent for the blend of CS and CA. Under wet condition, tensile strength and elongation of the composite films were obviously higher than those of the films made from pure CS. FTIR, DSC, and X-ray diffraction showed that the composite films exhibit a high level of compatibility and that strong interaction between the CS and CA was caused by intermolecular hydrogen bonding. The affinity series of composite film to transition metal ions are Cu(II) > Cd(II) > Cr(III). The adsorption of Cu((II) ion was shown to be highly pH sensitive.
In order to develop bacterial cellulose (BC) with antimicrobial activity against pathogenic microorganisms, silver and chitosan were incorporated into BC, respectively. Experiment results showed that antimicrobial activity against pathogenic microorganisms was improved with increasing silver concentration. Chitosan also showed a direct proportion between its concentration and antimicrobial activity. These results suggest that antimicrobial effects of BC using silver and chitosan are well proven to be effective. We also tested the stainability of BC with natural colorant for the application of food industry. Stainability of BC was enhanced with increasing natural colorant concentration. Decolorization of BC stained was observed by dipping it into distilled water with one hour-intervals. As a result, there was no significant difference. Combination of natural colorant-stainability and antibiosis of BC is expected to be useful in making colored antibiotic BC in various industrial application areas, considering its antimicrobial activity, high stainability and low decolorization tendency.
Cellulose-digesting bacteria were isolated from hindgut of Allomyrina dichotoma (Coleoptera: Dynastidae). The bacterial isolates were identified as Yersinia sp. and Bacillus sp. The addition of the identified bacteria to diet increased growth rate of the cetoniid beetle, Protaetia brevitarsis senlensis (Coleoptera: Cetoniidae), probably by digesting cellulose nutrient contained in the oak tree sawdust diet. An additive of wheat flour at more than 10% to the sawdust diet significantly enhanced growth of P. brevitarsis senlensis. Trimmed branches of apple trees have been disposed in the apple farms and could be used for a diet component of the cetoniid beetle when the cellulose-digesting bacteria were mixed with the derived-sawdust. Resulting manure from mass rearing of P. brevitarsis senlensis contained high organic matters and trace amounts of toxic metals. When the manure were splayed on soil, it was effective as a natural compost and significantly stimulated lettuce growth. This research suggests a model technology to use cellulose-digesting bacteria to use for culturing grub, which results in natural recycles of trimmed branches in apple farms as grub diet, and to use grub manure as a natural compost.
