포도 ASR (VvMSA) 단백질은 hexose transporter 유전자 VvHT1의 전사를 조절하는 조절 인자 중의 하나로서 sugar 및 abscisic acid (ABA) 신호에 의해 발현이 유도된다. 본 연구진은 ACP RT-PCR (annealing control primer reverse transcriptase-polymerase chain reaction) 방법을 이용하여 포도 과실발달 과정에서 조절되는 유전자 중 VvMSA와 동일한 cDNA (VlASR)를 클로닝하였다. 이 유전자는 착과 시기에 발현되기 시작하여 과실이 발달하면서 점점 증가하여 착과 후 10 주에 가장 많이 발현되며, 숙기 후반에는 도리어 발현양이 감소하였다. 포도 asr 유전자의 조절기작을 밝히기 위해, 이 유전자의 genomic clone을 분리하였다. 총 1375 bp로 이루어진 이 유전자 절편에는 open reading frame과 100 bp의 intron을 포함하고 있다. 약 600 bp 길이의 프로모터 내에는 sugar 신호전달과 연관이 있는 것으로 알려진 sugar box(sucrose box 3 +sucrose response box 1)가 있다. 프로모터 절편을 reporter 유전자와 연결하여 Arabidopsis에 도입하고 형질전환체를 분석한 결과, reporter 유전자는 sucrose 처리와 상관없이 항상 발현되었다. 이러한 결과는 포도에서 보고된 ASR/VvHT1를 매개로 하는 sugar/ABA 신호전달계가 asr 유전자가 없는 Arabidopsis에서는 작동되지 않음을 시사하고 있다.
Prostatic acid phosphatase (PAP)는 전립선 암의 진단에 널리 사용되는 표지자로서 1935년 처음으로 동정되었고 인체 전립선에 가장 많이 존재하는 탈 인산화효소이다. PAP는 prostate epithelial cells에서 합성되는 전립선 특이적인 효소로서, 산성 환경에서 효소활성을 띠는 acid phosphatase 그룹에 속한다. PAP는 전립선액에 풍부히 존재하여 수정, 정자부족증, 만성통증의 감소에 관여한다. 그러나 가장 눈에 띄는 기능은 ERK1/2와 MAPK 경로에 관계된 HER-2와 PI3P의 탈 인산화를 유도하여 세포 성장 신호를 억제하고 전립선 암의 억제자로 작용하는 것이다. 최근 PAP DNA 백신을 이용하는 임상시험이 현재 진행 중이고, PAP를 이용한 immunotherapy를 통해 전립선 암을 치료하는 방법이 FDA의 승인을 받아 시행되고 있다. 이러한 PAP의 임상적 중요성에도 불구하고 현재까지 PAP의 분자적 조절기작에 대한 이해는 제한적이라 PAP에 대한 많은 연구가 필요한 실정이다. PAP는 NF-${\kappa}B$, TNF-${\alpha}$, IL-1 및 androgen과 androgen receptor에 의하여 promoter region이 조절된다고 알려졌다. 본 총설에서는 현재까지 밝혀진 PAP 유전자 및 단백질의 특징들과 더불어 전립선 암에서의 PAP의 기능, 발현 조절, 역할들을 종합하였다.
C형 간염바이러스(hepatitis C virus; HCV)증식을 효과적이며 특이적으로 제어할 수 있는 유전산물을 개발하기 위하여 HCV 중식조절이자인 NS5B RNA replicase 존재에 의해 allosteric하게 그 활성 이 조절될 수 있는 HCV internal ribosome entry site (IRES) 표적 hammerhead 리보자임을 개발하였다. 우선 HCV IRES 염기서열 중+382 nucleotide(nt) 부위가 리보자임에 의해 가장 잘 인식되었음을 관찰하였다. 이러한 allosteric 리보자임은 NS5B RNA replicase와 특이적으로 결합하는 RNA aptamer 부위, aptamer와 NS5B와의 결합에 의해 리보자임 활성을 유도할 수 있도록 구조적 변이를 전달할 수 있는 communication module부위 및 HCV IRES의 +382 nt를 인지하는 hammerhead 리보자임 등으로 구성되도록 설계하였다. 특히 in vitro selection기법을 활용하여 NS5B 의존적으로 리보자임 활성을 증가시킬 수 있는 communication module 염기서열을 밝혀내었다. 이러한 리보자임은 단백질이 없거나 대조 단백질인 bovine serum albumin이 존재할 때에는 절단반응을 유도하지 못하였으나 HCV NS5B 단백질이 존재할 매에만 효과적으로 NS5B 농도 의존적으로 절단 반응을 유도할 수 있음을 관찰하였다. 이러한 allosteric 리보자임은 HCV중식의 효과적인 증식 억제 선도물질 뿐만 아니라 HCV 치료선도물질의 스크리닝용 도구 및 HCV 조절 인자를 탐색할 수 있는 HCV 진단용 리간드로서도 활용될 수 있을 것이다.
본 논문에서는 ZnS:Mn/ZnS 코어-쉘 양자점을 유기금속전구체의 열분해 방법으로 합성하였다. 쉘의 형성온도를 135$^{\circ}C$로 고정한 반면 코어 나노입자의 합성 온도 조건을 다양화 하여 각 조건하에서 형성된 양자점들의 특성을 조사하였다. 실험을 통해 얻은 양자점들은 UV-Vis, 액체 photoluminescence (PL) spectroscopy 방법으로 광 특성을 조사하였으며, 또한 XRD, HR-TEM, 및 EDXS 분석으로 입자크기와 조성 등을 측정하였다. 실험 결과 가장 좋은 광 특성을 보인 나노입자의 합성조건은 코어와 쉘 모두 135$^{\circ}C$인 것으로 밝혀 졌으며, 이 조건에서 얻은 양자점은 583 nm 의 PL 발광 피이크와 42.15%의 높은 양자효율을 나타내었다. HR-TEM 으로 측정한 ZnS:Mn/ZnS 양자점의 평균 입자크기는 지름이 약 4.0 - 5.4 nm 정도였으며, 특히 150$^{\circ}C$의 온도 조건에서는 타원형의 입자가 형성되는 것이 관찰되었다.
혈관이 좁아지거나 막혀서 생기는 허혈은 심장, 뇌, 다리와 같은 인체의 여러 장기에 영향을 미친다. 최근 혈관신생 분야에서 많은 진전이 있어 기존 치료법으로 치료가 되지 않는 허혈성 환자들의 치료 가능성에 대한 기대가 많아졌다. 혈관형성은 여러 개의 인자들과 세포들이 관여하는 복잡한 과정이기 때문에, 한 개의 인자보다는 여러 인자들을 병합하는 요법이 점점 많이 시도되고 있는데, 이런 병합요법의 한 예로 전사인자를 전달하는 전략을 생각할 수 있다. 이에 본 연구에서는 cardiac ankyrin repeat protein (CARP)의 유전자를 대상으로 그 혈관신생 능력을 혈관내피세포에서 조사하였다. 아데노바이러스 벡터 내에 human CARP의 cDNA를 클로닝하여 재조합 아데노바이러스를 제조하였으며, 이를 이용한 유전자 전달실험 결과, CARP 유전자 전달 군에서 유의하게 혈관내피세포의 중식과 모세관 구조 형성, 그리고 vascular endothelial growth factor의 발현 등을 증가시킴을 확인하였다. 본 연구 결과는 CARP가 혈관신생 연구의 새로운 목표 유전자로서 그 기능에 대한 많은 연구가 필요함을 뒷받침해준다.
본 연구에서는 잘 정의된 덴드리틱 폴리스티렌-블록-선형 폴리(3차 부틸 아크릴레이트) 공중합체를 성공적으로 합성하였다. 음이온 중합법을 통해 합성된 폴리스티렌($M_n$=1000 g/mol)을 외곽부분으로 함유하는 덴드론의 수산기 그룹을 토실화(tosylation), 아지드화(azidation), 환원(reduction) 반응을 통해 아민기로 전환하였다. 한편, 선형 poly(t-butyl acrylate)는 DMF 용매에서 benzyl 2-bromopropanoate/Cu(I)Br, PMDETA/t-butylacrylate를 각각 개시제/촉매 시스템/단량체로 사용하여, 원자이동라디칼 중합법(atom transfer radical polymerization, ATRP)을 통해 합성할 수 있었고, 말단을 카르복시산 그룹으로 전환하기 위해 수소기체 환경하에서 Pd/C 을 사용하여 탈벤질화(debenzylation) 반응을 수행하였다. 마지막으로, 합성된 덴드리틱 및 선형 블록들을 다이메틸아미노피리딘(4-(dimethylamino)pyridine, DMAP)과 다이아이소프로필카보다이이미드(N,N'-diisopropylcarbodiimide, DIPC)를 이용한 아미드 커플링 방법을 통해 최종 덴드리틱-선형의 블록공중합체를 합성하였다. 합성된 블록공중합체를 수소원자핵공명법 및 겔침투크로마토그래피 방법을 통해 조사한 결과, 잘 정의된 분자량 및 낮은 분자량 분포를 확인할 수 있었다.
Layered mixed metal hydroxy double salts (HDS) with the formulas $(Cu_{0.75}Ni_{0.25})_2(OH)_3NO_3$ ((Cu, Ni)-HDS) and $Cu_2(OH)_3NO_3$ ((Cu, Cu)-HDS) were prepared via slow hydrolysis reactions of CuO with $Ni(NO_3)_2$ and $Cu(NO_3)_2$, respectively. The crystal structures, morphologies, bonding natures, and magnetic properties of (Cu, Ni)-HDS and (Cu, Cu)-HDS were characterized with X-ray diffraction (XRD), scanning electron microscopy (SEM), Fourier transformation infrared spectroscopy (FT-IR), thermogravimetric analysis (TGA), and a superconducting quantum interference device (SQUID). Even though (Cu, Ni)-HDS has a similar layered structure to that of (Cu, Cu)-HDS, the bonding nature of (Cu, Ni)-HDS is slightly different from that of (Cu, Cu)-HDS. Therefore, the magnetic properties of (Cu, Ni)-HDS are significantly different from those of (Cu, Cu)-HDS. The origin of the abnormal magnetic properties of (Cu, Ni)-HDS can be explained in terms of the bonding natures of the interlayer and intralayer structures.
Hemodynamic shear stress, the dragging force generated by blood flow, is known as an anti-atherogenic factor. We tested whether lysyl-tRNA synthetase (KRS) will be utilized as an agent controlling shear-sensing systems. KRS was previously known to be secreted as a pro-inflammatory agent. Here we found that KRS inhibited various shear-stimulated signaling pathways. We further found that KRS binds to detergent-resistant membrane (DRM), indicating that KRS binding molecules exist in DRM, specialized regions of the plasma membrane. DRM plays important roles in a variety of cellular processes and consists of gangliosides, signaling molecules and cytoskeletons. We then determined that KRS was colocalized with integrins ${\alpha}4$, ${\alpha}5$ and $av{\beta}3$. In addition, KRS was shown to be associated with sialic acid, existing at the end of gangliosides. Interestingly, the adherent effect of KRS was inhibited by pretreatment with sialic acid. Moreover, treatment of endothelial cells with neuraminidase appeared to inhibit both the KRS adhesion to endothelial cells and shear-stimulated signaling. In conclusion, KRS is likely to be utilized as a vascular regulator.
Many researchers studied cell culturing on surfaces with chemical functional groups. Previously, we reported surface properties of plasma polymerized ethylenediamine (PPEDA) films deposited by plasma enhanced chemical vapor deposition with various plasma conditions. Surface properties of PPEDA films can be controlled by plasma power during deposition. In this work, to analyze correlation of cell adherence/proliferation with surface property, we cultured rat intestinal epithelial cells-18 on the PPEDA films deposited with various plasma powers. It was shown that as plasma power was decreased, density of cells cultured on the PPEDA film surface was increased. Our findings indicate that plasma power changed the amine density of the PPEDA film surface, resulting in density change of cells cultured on the PPEDA film surface.
Intracellular expression of RNA decoys, such as TAR or RRE decoy, has been previously shown to protect immune cells from human immunodeficiency virus type 1 (HIV-1) replication by inhibiting the binding of the HIV-1 regulatory protein to the authentic HIV RNA sequence. However, HIV-1 challenge experiments of primary human T cells, which express the RNA decoy, demonstrated that the cells were only transiently protected, and hence, more improved protocols for HIV-1 inhibition with the RNA decoys need to be developed. In this report, in order to develop a more effective RNA decoy, we analyzed and compared the ability of a series of RNA decoy derivatives in inhibiting HIV-1 replication in CEM cells. Using an improved tRNA cassette to express high levels of RNA decoy transcripts in cells, we found that co-expression of both TAR and RRE decoys, in the form of an aligned sequence in a single transcription cassette, much more potently blocked cells from HIV-1 than the expression of only one kind of RNA decoy. This observation will have an important implication for experiments involving optimization of clinical applications in RNA decoy-based gene therapy against HIV-1.
