Synthesis and SAR of N-Chlorophenyl Substituted Piperrazinylethyl-aminomethylpyrazoles as 5-HT3A Inhibitors

  • 5-$HT_{3}$ receptor;5-$HT_{3A}$ receptor channel activity;Novel 5-$HT_{3}$ receptor channel current blockers;Chlorophenyl substituted piperazinylethylaminomethylpyrazoles; The 5-$HT_{3A}$ receptors are one of ligand-gated ion channels and are known to be involved in visceral pain, anxiety, or anticancer agent-induced nausea and vomiting. In present study, we designed novel skeletons based on the developed 5-$HT_{3}$ receptor antagonists and evaluated their effects on 5-$HT_{3A}$ receptor channel currents ($I_{5-HT}$) of a series of pyrazole derivatives having N-chlorophenylpiperazine functionality (6-9). We found that most of N-p-chlorophenyl substituted piperazinyl-pyrazole derivatives (7b, 7c, 7e and 7h) exhibited the high potency for the inhibition of $I_{5-HT}$, whereas the compound without chloride (6) or with m-chlorophenyl group (a serious of 8 and 9) showed the low potency. These result indicate that p-chlorophenyl group is might play an important role for increasing the inhibitory potency on $I_{5-HT}$.

Compensation of the Error Rate for the Non-invasive Sphygmomanometer System Using a Tactile Sensor

  • The Purpose Of This Paper Is To Use A Tactile Sensor To Compensate The Error Rate. Most Automated Sphygmomanometers Use The Oscillometric Method And Characteristic Ratio To Estimate Systolic And Diastolic Blood Pressure. However, Based On The Fact That Maximum Amplitude Of The Oscillometric Waveform And Characteristic Ratio Are Affected By Compliance Of The Aorta And Large Arteries, A Method To Measure The Artery Stiffness By Using A Tactile Sensor Was Chosen In Order To Integrate It With The Sphygmomanometer In The Future Instead Of Using Photoplethysmography. Since Tactile Sensors Have Very Weak Movements, Efforts Were Made To Maintain The Subject's Arm In A Fixed Position, And A 40hz Low Pass Filter Was Used To Eliminate Noise From The Power Source As Well As High Frequency Noise. An Analyzing Program Was Made To Get Time Delay Between The First And Second Peak Of The Averaged Digital Volume Pulse(${\Delta}t_{dvp}$), And The Subject's Height Was Divided By ${\Delta}t_{dvp}$ To Calculate The Stiffness Index Of The Arteries($Si_{dvp}$). Regression Equations Of Systolic And Diastolic Pressure Using $Si_{dvp}$ And Mean Arterial Pressure(Map) Were Computed From The Test Group (60 Subjects) Among A Total Of 121 Subjects(Age: $44.9{\pm}16.5$, Male: Female=40:81) And Were Tested In 61 Subjects To Compensate The Error Rate. Error Rates Considering All Subjects Were Systolic $4.62{\pm}9.39mmhg$, And Diastolic $14.40{\pm}9.62mmhg$, And Those In The Test Set Were $3.48{\pm}9.32mmhg,\;And\;14.34{\pm}9.67mmhg$ Each. Consequently, Error Rates Were Compensated Especially In Diastolic Pressure Using $Si_{dvp}$, Various Slopes From Digital Volume Pulse And Map To Systolic-$1.91{\pm}7.57mmhg$ And Diastolic $0.05{\pm}7.49mmhg$.

Electrical/Mechanical Diagnosis of Local Deterioration in 600V Shielded Twist Pair Cable in a Nuclear Power Plant (원전용 600V 차폐 꼬임쌍선 케이블의 국부열화에 대한 전기적/기계적 진단)

  • In this paper, we propose a electrical/mechanical method to effectively diagnose the local deterioration of a 10m long power shielded twist pair cable defined by the American Wire Gauge (AWG) 14 specification using electrical/mechanical methods. The rapid deterioration of the cable proceeded by using the heating furnace, which is based on the Arrhenius equations proceeds from 0 to 35 years with the deteriorated equivalent model. In this paper, we introduce a method to diagnose the characteristics of locally deteriorated cable by using $S_{21}$ phase and frequency change rate measured by vector network analyzer which is the electrical diagnostic method. The measured $S_{21}$ phase and rate of change of frequency show a constant correlation with the number of years of locally deteriorated cable, thus it can be useful for diagnosing deteriorated cables. The change of modulus due to deterioration was measured by a modulus measuring device, which is defined by the ratio of deformation from the force externally applied to the cable, and the rate of modulus change also shows a constant correlation with the number of years of locally deteriorated cable. Finally, By combining the advantages of electrical/mechanical diagnostic methods, we can efficiently diagnose the local deterioration in the power shielded cable.

Near-field limit in positioning the microphone for pressure measurements in using the near-field acoustical holography (근접 음향 홀로그래피에서 음압 측정용 마이크로폰의 근접 거리 한계)

  • The recently developed BEM-based NAH(nearfield acoustical holography) is a useful technique for identifying the sound source of vibrating objects. The acoustic parameters of a sound source can be reconstructed by using the vibro-acoustic transfer matrix, which is determined by means of BEM, and the sound pressure measured in the nearfield. Theoretically, one can come up with a very nice reconstructed result as the field plane gets near to the source surface. However, when a microphone is placed in the very close nearfield of the source surface, the scattering, reflection, or resonance in the gap between the source and the microphone can distort the acoustic field, and therefore, the measured field pressure would differ from the actual one in the absence of the microphone. In order to analyze this problem, the interference effect of the microphone is numerically calculated by using the nonsingular BEM that yields very small error in the nearfield. From this analysis, it is found that the prediction error of the field pressure decreases firstly and then increases as the microphone approaches the vibrating surface from the farfield to the close nearfield. It is noted that the microphone should be separated from the source surface by at least a diameter of the microphone for an error ratio less than 2% in the low frequency range less than about 2.7kHz. This means that if one wants to put a microphone in the very close nearfield. a microphone with small diameter should be used.

Photocatalytic Properties of WO3 Thin Films Prepared by Electrodeposition Method (전기증착법으로 제조된 WO3 박막의 광촉매 특성)

  • Tungsten trioxide ($WO_3$) is a promising candidate as a photocatalyst because of its outstanding electrical and optical properties. In this study, we prepare $WO_3$ thin films by electrodeposition and characterize the photocatalytic degradation of methylene blue using these films. Depending on the voltage conditions (static and pulse), compact and porous $WO_3$ films are fabricated on a transparent ITO/glass substrate. The morphology and crystal structure of electrodeposited $WO_3$ thin films are investigated by scanning electron microscopy, atomic force microscopy, and X-ray diffraction. An application of static voltage during electrodeposition yields a compact layer of $WO_3$, whereas a highly porous morphology with nanoflakes is produced by a pulse voltage process. Compared to the compact film, the porous $WO_3$ thin film shows better photocatalytic activities. Furthermore, a much higher reaction rate of degradation of methylene blue can be achieved after post-annealing of $WO_3$ thin films.

Preparation of a Hydrophobized Chitosan Oligosaccharide for Application as an Efficient Gene Carrier

  • To prepare chitosan-based polymeric amphiphiles that can form nanosized core-shell structures (nanopar­ticles) in aqueous milieu, chitosan oligosaccharides (COSs) were modified chemically with hydrophobic cholesterol groups. The physicochemical properties of the hydrophobized COSs (COSCs) were investigated by using dynamic light scattering and fluorescence spectroscopy. The feasibility of applying the COSCs to biomedical applications was investigated by introducing them into a gene delivery system. The COSCs formed nanosized self-aggregates in aqueous environments. Furthermore, the physicochemical properties of the COSC nanoparticles were closely related to the molecular weights of the COSs and the number of hydrophobic groups per COS chain. The critical aggregation concentration values decreased upon increasing the hydrophobicity of the COSCs. The COSCs effi­ciently condensed plasmid DNA into nanosized ion-complexes, in contrast to the effect of the unmodified COSs. An investigation of gene condensation, performed using a gel retardation assay, revealed that $COS6(M_n=6,040 Da)$ containing $5\%$ of cholesteryl chloroformate (COS6C5) formed a stable DNA complex at a COS6C5/DNA weight ratio of 2. In contrast, COS6, the unmodified COS, failed to form a stable COS/DNA complex even at an elevated weight ratio of 8. Furthermore, the COS6C5/DNA complex enhanced the in vitro transfection efficiency on Human embryonic kidney 293 cells by over 100 and 3 times those of COS6 and poly(L-lysine), respectively. Therefore, hydrophobized chitosan oligosaccharide can be considered as an efficient gene carrier for gene delivery systems.

A Case Report of Olivopontocerebellar Atrophy (Multiple Systemic Atrophy-Cerebellum) Patient Complicating Sleep Disorders and Gait Disturbance (수면장애와 보행장애를 호소하는 올리브교소뇌위축증 환자 증례보고 1례)

  • This case report describes a patient with olivopontocerebellar atrophy accompanied by sleep disorder and gait disturbance whose condition was improved by treatment with Korean medicine. The 61-year-old woman, who was diagnosed with olivopontocerebellar atrophy (Multiple Systemic Atrophy-Cerebellum), was admitted to hospital twice and treated with Korean medicine (acupuncture and herbal medicine) and rehabilitation. The Korean medicine was Gwibiondam-tang-gami and Jaeumgeonbi-tang-gami. Clinical symptoms were assessed by the Modified Bathel index, functional independent measurement, Berg balance scale, and Unified Multiple System Atrophy rating scale. A brain MRI at the one-year follow up after onset showed similar progress but clinical symptoms were improved after treatment, and the evaluation index score increased. Multiple system atrophy, a type of degenerative neurological disease, has no targeted treatment. In this situation, although this report describes a single case, Korean medicine treatment could provide a meaningful improvement in the sleep disturbance and gait disorder symptoms of patients with olivopontocerebellar atrophy.

  • The application of software engineering is not common in the development of astronomical observation system. While there were component-wise developments in the past, large-scale comprehensive system developments are more common in these days. In this study, current methodologies of development are reviewed to select a proper one for the development of astronomical observation system and the result of the application is presented. As the subject of this study, a project of operation software development for an astronomical observation system which runs on the ground is selected. And the output management technique based on Component Based Development which is one of the relatively recent methodologies has been applied. Since the nature of the system requires lots of arithmetic algorithms and it has great impact on the overall performance of the entire system, a prototype model is developed to verify major functions and performance. Consequently, it was possible to verify the compliance with the product requirements through the requirement tracing table and also it was possible to keep to the schedule. Besides, it was suggested that a few improvements could be possible based on the experience of the application of conventional output management technique. This study is the first application of the software development methodology in the domestic astronomical observation system area. The process and results of this study would contribute to the investigation for a more appropriate methodology in the area of similar system development.

  • The Immersion Grating Infrared Spectrometer (IGRINS) is a near-infrared wide-band high-resolution spectrograph jointly developed by the Korea Astronomy and Space Science Institute and the University of Texas at Austin. IGRINS employs three HAWAII-2RG Focal Plane Array (H2RG FPA) detectors. We present the design and fabrication of the detector mount for the H2RG detector. The detector mount consists of a detector housing, an ASIC housing, a Field Flattener Lens (FFL) mount, and a support base frame. The detector and the ASIC housing should be kept at 65 K and the support base frame at 130 K. Therefore they are thermally isolated by the support made of GFRP material. The detector mount is designed so that it has features of fine adjusting the position of the detector surface in the optical axis and of fine adjusting yaw and pitch angles in order to utilize as an optical system alignment compensator. We optimized the structural stability and thermal characteristics of the mount design using computer-aided 3D modeling and finite element analysis. Based on the structural and thermal analysis, the designed detector mount meets an optical stability tolerance and system thermal requirements. Actual detector mount fabricated based on the design has been installed into the IGRINS cryostat and successfully passed a vacuum test and a cold test.

A Study on Design Development Direction for Designing Aging Friendly City through The Case Analysis (사례분석을 통한 고령친화도시 디자인 개발방향연구)

  • The purpose of this study is to suggest a foundational design direction for seniors, according to paradigm shift in aging society. This paper researched on current state of the aging society through literature study and defined the theoretical concept that set the foundation of the study. Secondly, for building an age-friendly city, relevant studies on seven cities were selected and their regional design guidelines were analyzed. As a result, essential design elements for building the aging-friendly city were defined as safety, recognition, and accessibility. A detailed design guideline that applies the three elements with understanding on seniors is required to construct the environment for a senior-friendly city. Ultimately, the result of this study is expected to be in practical use as a fundamental resource for age-friendly city design.
