  • The synthesis of 3a, 3b, 7a fro Benzpthiazepinone and 1,4-dihyropyridine derivatives is described. Benzothiazepinone nad 1,4-dihydropyridine derivatives were prepared according to literature procedure. The key reactions involve esterification and amidation of benzothize-pinone nad 1,4-dihydropyridine derivatives.

  • A mannitol dehydrogenase (MDH; EC 1.1.1.67) gene was cloned from the Sinorhizobium meliloti 1021 (KCTC 2353) genome and expressed in Escherichia coli. It was seen to have an open reading frame consisting of 1,485 bp encoding 494 amino acids (about 54 kDa), which shares approximately 35-55% of amino acid sequence identity with some known long-chain dehydrogenase/ reductase family enzymes. The recombinant S. meliloti MDH (SmMDH) showed the highest activity at $40^{\circ}C$, and pH 7.0 (D-fructose reduction) and pH 9.0 (D-mannitol oxidation), respectively. SmMDH could catalyze the oxidative/reductive reactions between D-mannitol and D-fructose in the presence of $NAD^+/NADH$ as a coenzyme, but not with NADP+/NADPH. These results indicate that SmMDH is a typical $NAD^+/NADH$-dependent mannitol dehydrogenase.

  • Here we report a highly sensitive and stable detection of $NAD^+$ and NADH by the electrode on which diaphorase (DI) is covalently immobilized in viologen polymer network. The network is prepared by the covalent formation of the structure by mixing propylamine viologen (PAV), poly(ethylene glycol)(400) diglycidyl ether (PEGDGE), an diaphorase (DI). The PAV/PEGDGE/DI modified electrode has the sensitivity of $0.02nA{\cdot}{\mu}M$ and the detection limit of $3{\mu}M$ with a response time of 2 s ($t_{90%}$) for NADH sensing.

  • This paper evaluates on the feasibility of MIL-STD-188-220 protocol as a medium access protocol over WNW(Wideband Network Waveform) by simulating and comparing with IEEE 802.11e-based protocol. WNW is newly designed waveform for next-generation broadband tactical communication system. This paper shows the feasibility of using the MIL-STD-188-220 protocol that shows better performance than IEEE 802.11e-based protocol on the particular environment.

Enhanced Expression of Cell Adhesion Molecules in the Aorta of Diabetic Mice is Mediated by gp91phox-derived Superoxide

  • Endothelial activation and subsequent recruitment of inflammatory cells are important steps in atherogenesis. The increased levels of cell adhesion molecules (CAM) have been identified in diabetic vasculatures, but the underlying mechanisms remain unclear. To determine the relationship among vascular production of superoxide, expression of CAM and diabetes, superoxide generation and expression of intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), E- and P-selectin in the aorta from control (C57BL/6J) and diabetic mice (ob/ob) were measured. In situ staining for superoxide using dihydroethidium showed an increased superoxide production in diabetic aorta, accompanied with an enhanced NAD(P)H oxidase activity. Immunohistochemical analysis revealed that the endothelial expression of ICAM-1 ($3.5{\pm}0.4$) and VCAM-1 ($3.8{\pm}0.3$) in diabetic aorta was significantly higher than those in control aorta ($0.9{\pm}0.5$ and $1.6{\pm}0.3$, respectively), accompanied with the enhanced expression of gp91phox, a membrane subunit of NAD(P)H oixdase. Furthermore, there was a strong positive correlation (r=0.89, P<0.01 in ICAM-1 and r=0.88, P<0.01 in VCAM-1) between ICAM-1/VCAM-1 expression and vascular production of superoxide. The present data indicate that the increased production of superoxide via NAD(P)H oxidase may explain the enhanced expression of CAM in diabetic vasculatures.

On the Root Rot of Ginseng(I) (인삼근부병에 관한 연구 1)

  • This study was conducted from April 1 to September 31, 1973. Ten strains of Fusarium spp. were isolated from the diseased ginseng in two local areas at Kangwha-Gun nad Kumsan-Gun in Korea. Among of them, 2 strains ($G_1$, $G_4$) did not have virulence to ginseng in reinoculation. Their cultural, morphological characteristic and hose virulence to pea seedling were examined. Taxonomical identification of 8 isolates followed by the method of Wollenweber, Snyder nad Toussoun, Booth, Matuo and Snyder. All of eight strains were identified as the Fusarium solani f.sp.pisi(Jones) Synd. et Hans.

Function of heat shock protein and Immune response (Heat shock protein의 기능과 면역 반응)

  • A study was made on enzymes of carbohydrate metabolism in T. concretivorus grown with and without glucose. The present results show that T. concretivorus possesses high activities of pentose shunt pathway and related enzymes, glucokinase, G-6-P dehydrogenase, 6-PG dehydrogenase, and phosphoglucoisomerase, but low activities of enzymes unique to EMP(fructose-1,6-diphosphate aldolase). Although the synthesis of the latter enzymes remains largely unaffected by the growth enviroment, that of the former is stimulated by glucose. And the failure to detect ED pathway enzymes in cells grown in thiosulate or thiosulfate-glucose medium eliminates the ED pathway as a significant route of glucose catabolism in T.concretivorus. These results suggest that pentose shunt pathway performs an energetic role in glucose metabolism by T.concretivorus with EMP as a subway. The absence of ED pathway and the presence of pentose shunt pathway which is the major route of catabolism in T.concretivorus are similar to those of other obligately chemolitho-trophic thiobacilli. The G-6-P and 6-PG dehydrogenase are both NAD and NADP specific, but MAD predominant. However, the 3-PGAL dehydrogenase is only NAD specific. Since the specific activity of 3-PGAL generated from glucose is converted mainly into pyruvate which is channeled into the TCA cycle. All enzymes of the TCA cycle tested and NADH oxidase are detected in the cells of T.concretivorus grown in thiosulfate. The specific activities of fumarase and isocitrate dehydrogenase are high and others are low. The presence of two isocitrate dehydrogenase (NAD-and NADP-linked) may have important regulatory function for this organism. The activity of NAD-oxidase, which is implicated in the energy generating metabolism, was very high in the crude cell-free extract of T.concretivorus, recording 55.11 m.mu. mole/min/mg protein. This well coincides with the fact that activities of NAD-linked G-6-P dehydrogenase, 6-PG dehydrogenase and 3-PGAL dehydrogenase were high.

Characterization of the Outer Membrane-Associated 2-Furaldehyde Dehydrogenase from Klebsiella pneumoniae (Klebsiella pneumoniae 균주의 세포외막에서 분리한 2-furaldehyde dehydrogenase의 특성에 관한 연구)

  • An outer membrane-associated 2-furaldehyde dehydrogenase, catalyzing the oxidation of 2-furaldehyde to 2-furoic acid from Klebsiella pneumoniae was purified to homogeneity and characterized. The enzyme showed its highly specific dependency on $\beta$-$NAD^{+}$. Enzyme activity was monitored during purification by using substrate 2-furaldehyde and coenzyme $\beta$-$NAD^{+}$ by means of high performance liquid chromatography. The outer membrane was successfully collected by the methods of Percoll density gradient ultracentrifugation and ultracentrifugation after preferential solubilization of the membrane with $Mg^{2+}$ and Triton X-100. The enzyme was purified by the series of procedures including extraction of outer membrane protein with EDTA and lysozume, and fractionation by column chromatography on QAE-Sephades Q-50, and subsequently Sephadex G-100. The enzume showed its optimal activity at $85^{\circ}C$, pH 9.5, and in the presence of 1.5% (vol/vol) Triton X-100. The enzyme exhibited a native molecular size of 88,000 by nondenaturing polyacrylamide gel electrophoresis and had an apparent Km of 4.72mM for 2-furaldehyde.

Resveratrol attenuates lipopolysaccharide-induced dysfunction of blood-brain barrier in endothelial cells via AMPK activation

  • Resveratrol, a phytoalexin, is reported to activate AMP-activated protein kinase (AMPK) in vascular cells. The blood-brain barrier (BBB), formed by specialized brain endothelial cells that are interconnected by tight junctions, strictly regulates paracellular permeability to maintain an optimal extracellular environment for brain homeostasis. The aim of this study was to elucidate the effects of resveratrol and the role of AMPK in BBB dysfunction induced by lipopolysaccharide (LPS). Exposure of human brain microvascular endothelial cells (HBMECs) to LPS ($1{\mu}g/ml$) for 4 to 24 hours week dramatically increased the permeability of the BBB in parallel with lowered expression levels of occluding and claudin-5, which are essential to maintain tight junctions in HBMECs. In addition, LPS significantly increased the reactive oxygen species (ROS) productions. All effects induced by LPS in HBVMCs were reversed by adenoviral overexpression of superoxide dismutase, inhibition of NAD(P) H oxidase by apocynin or gain-function of AMPK by adenoviral overexpression of constitutively active mutant (AMPK-CA) or by resveratrol. Finally, upregulation of AMPK by either AMPK-CA or resveratrol abolished the levels of LPS-enhanced NAD(P)H oxidase subunits protein expressions. We conclude that AMPK activation by resveratrol improves the integrity of the BBB disrupted by LPS through suppressing the induction of NAD(P)H oxidase-derived ROS in HBMECs.

Rooting and Survival Rate as Affected by Various Types and Concentrations of Auxin on 'Maehyang' Strawberry in Cutting Propagation ('매향' 딸기의 삽목 번식 시 옥신의 종류 및 처리 농도에 따른 발근율과 생존율)

  • This study was conducted to examine the effect by different types and concentrations of auxin on the rooting and growth of strawberry (Fragaria × ananassa Duch. cv. Maehyang) cuttings in the greenhouse. The NAD (1-naphthylacetamide), IBA (indole-3-butyric acid), and IAA (3-indoleacetic acid) were applied with a 1 hour soaking as 50, 100, 150, and 200 mg·L-1, respectively. The non-treatment was set as the control. The cuttings of strawberry were transplanted in the strawberry seedling tray filled with coir medium on June 4, 2020. The humidification was carried out for 2 weeks. The average relative humidity, daytime temperature, and nighttime temperature inside the humidification tunnel was 63.4 ± 15%, 29.3 ± 5℃, and 16.2 ± 5℃, respectively. There was no significant difference in rooting rate on the control, IBA, and IAA treatments. However, significantly low rooting rates were observed in NAD treatments. The survival rates were significantly higher in the control and IBA with 50 mg·L-1 than in other treatments. The number of leaves was the highest in IBA with 100 mg·L-1. The root length was the longest in the control. More number of roots were counted in IAA with 100 and 150 mg·L-1. The dry weight of root was the heaviest in the control. The total root length, root surface, number of root tips, and number of root forks were significantly higher in the control. As a result, control, IAA, and IBA showed similar shoot and root growth. However, NAD showed the worst root and shoot growth. Consequently, compared with IAA and IBA, NAD was not appropriate plant growth regulator of rooting for cutting propagated strawberries.
