Lipid Peroxidation of Hepatic Microsomal Drug-Metabolizing System in Hepatic Ischemia ands Reperfusion (간장내 허혈 및 재관류시 약물대사 효소계의 지질 과산화에 관한 연구)

  • This study was done to determine whether specific alterations exist in hepatic microsomal function after varying periods of ischemia (IS) and reperfusion (RP) during microsomal lipid peroxidation occurs. Rats were pretreated with $\alpha$-tocopherol to inhibit lipid peroxidation or with vehicle (soybean oil). Control animals were time-matched sham-ischemic animals. Four groups of animals were studied: Group 1 (sham), group 2 (30 mins IS), group 3 (60 mins IS) and group 4 (90 mins IS). After 1, 5 or 24 hr of reperfusion, liver microsomes were isolated and cytochrome P-450s were studied. In all vehicle-treated ischemic rats, serum ALT levels peaked at 5 hr and were significantly reduced by $\alpha$-tocopherol pretreatment. Similarly, microsomal lipid peroxidation was elevated in all vehicle-treated ischemic animal groups, but this elevation was prevented by $\alpha$-tocopherol pretreatment. Cytochrome P-450 content was significantly decreased in both group 3 and group 4. In all vehicle-treated ischemic animal groups, aminopyrine N-demethylase activity was significantly decreased for the entire reperfusion period. $\alpha$-Tocopherol inhibited reductions of cytochrome P-450 content and aminopyrine N-demethylase activity at both 1 hr and 5hr of reperfusion but did not affect the reduced levels of cytochrome P-450 content and aminopyrine N-demethylase activity at 24 hr of reperfusion. Aniline p-hydroxylase activity was significantly decreased in group 4, whereas it was increased in group 3. These decreases and increases were prevented by $\alpha$-tocopherol pretreatment. Our finding suggests that abnormalities in microsomal drug metabolizing function occur during hepatic ischemia and reperfusion in vivo and this is attributed to microsomal lipid peroxidation.

Total Phenolics of Dried Platycodon grandiflorum and Its Effect on Growth of Human Cancer Cell Lines (건조 도라지 추출물 및 분획물의 총 페놀계 화합물 함량 및 인체 암세포 증식 억제효과)

  • We investigated the effects of extracts from dried Platycodon grandiflorum on total phenolic content and growth of cancer cell lines (HT-29 and AGS). Total phenolic contents of acetone/methylene chloride (A+M) extract and methanol (MeOH) fraction were 4.53 and 27.22 TAE mg/100 g, respectively. Among the fractions, n-butanol (n-BuOH) fraction contained the highest phenolic content. Treatments of crude extracts and fractions significantly inhibited the growth of HT-29 and AGS cancer cell lines (p<0.05). Among the fractions, n-BuOH fraction exhibited the highest inhibitory effect and was then sub-fractionated by reverse phase flash column chromatography (rfc). The rfc 1-3 exhibited a higher inhibitory effect on proliferation of both cancer cells. The rfc 1 contained the highest phenol content. Our results showed that n-BuOH fraction possessed a potent inhibitory effect on proliferation of human cancer cells. We suggested that this anticancer activity was partially related to the content of phenolic compounds.

Quality Characteristics of Fermented Wine Using Nuruk by Aging Container and Period of Distilled Liquor (누룩 사용 발효주로 제조한 증류주의 숙성 용기 및 기간에 따른 특성)

  • This study aimed to determine the quality characteristics of fermented wine using Nuruk according to the type of aging container (jar or stainless container) and aging time. Alcohol analysis was initiated at 40.4~39.3%, and alcohol contents in the jar and stainless container decreased as aging time increased. Volatile acid content slightly increased in the reduced pressure (RP) jar and stainless container and showed a different result in the atmospheric pressure (AP) container. Amount of acetic acid did not change either in the jar or stainless container as aging time increased. In the case of 2-thiobarbituric acid (TBA), RP was lower than AP after distillation, and TBA value did not change much either in the jar or stainless container. In the case of fusel alcohol by aging time, n-propanol content did not increase either in the jar or stainless container as aging time increased. Iso-butanol content was reduced in the jar but did not change in the stainless container. In the AP container, more aromatic components were measured on the 180th day of aging time than after distillation. High boiling point aromatic components increased in particular. In the stainless container, more aromatic components were generated on the 180th day of aging, but their amount was relatively lower than in the jar. In the jar aging sensory test, the sensory score at RP 50 cmHg was highest on the 180th day of aging. In the case of AP $80^{\circ}C$ liquor, sensory score on the 180th day was the most improved.

  • This study was conducted to investigate the characteristics of humic substances and the fractional distribution of organic nitrogen in Korean paddy soils. The results are obtained as follows: 1. The content of humus in soils used was 665-2680 mg/100g and the average contents of humic acid in normal paddy soils and in sandy paddy soils were 1436 mg/100g, 970 mg/100g respectively. 2. The humic acid diagrams classed by Kumada method belong to B type, P type and Rp type. 3. The content of Mineral - N to Total - N was 1.04-2.86% and the average contents of that in sandy paddy soils and in normal paddy soils were 1.72% and 1.75% respectively. 4. The fractionations of acid hydrolysable organic - N in sandy paddy soils were Amino acid - N (31.52%), Humin - N (20.63%), Amino sugar - N (18.70%), and Unknown - N (25.73%).

Structural Identification of a Non-Glycosylated Variant at Ser126 for O-Glycosylation Site from EPO BRP, Human Recombinant Erythropoietin by LC/MS Analysis

  • A variant peak was detected in the analysis of RP-HPLC of rHu-EPO, which has about 7% relative content. Fractions of the main and the variant peaks were pooled separately and further analyzed to identify the molecular structure of the variant peak. Total mass analysis for each peak fraction using ESI-TOF MS shows differences in molecular mass. The fraction of the main peak tends to result in higher molecular masses than the fraction of the variant. The detected masses for the variant are about 600-1000 Da smaller than those for the main peak. Peptide mapping analysis for each peak fraction using Asp-N and Glu-C shows differences in O-glycopeptide profiles at Ser126. The O-glycopeptides were not detected in the fraction of the variant. It is concluded that the variant peak is non-O-glycosylated rHu-EPO and the main peak is fully O-glycosylated rHu-EPO at Ser126.

  • Dendrolimus punctatus tetravirus (DpTV) has been identified as a new member of the genus Omegatetravirus of the family Tetraviridae that may be related serologically to Nudaurelia capensis virus ($N{\omega}V$). To establish the function of DpTV RNA genome and to better understand the mechanism of viral replication, the putative RNA-dependent RNA polymerase (RdRp) domain has been cloned and expressed in Escherichia coli. The recombinant protein was purified on a Ni-chelating HisTrap affinity column and demonstrated to initiate viral RNA synthesis in a primer-independent manner but not by terminal nucleotidyle transferase activity in the presence of $Mg^{2+}$ and RNA template. Mutation of the GDD to GAA interferes with the residues at the polymerase active site and metal ions, and thus renders the polymerase inactive.

Cattle Production on Small Holder Farms in East Java, Indonesia: I. Household and Farming Characteristics

  • A general household survey was carried out in the village of Sonoageng in East Java, Indonesia with the aim to assess their socio-economic status, and the crop and livestock production system prevailing in the area. Of the households interviewed (164), 52% are landless, 35% own land or have a combination of own and shared land, and 12% do not own land but have access to land by sharing. Nearly two thirds (65%) of the households raise cattle; most of them own animals, 8% reared shared animals only. The dominant crop was rice; other crops grown were soya bean, groundnut, maize and sugar cane. Nearly half (47%) the household income was derived from off-farm work (non-agricultural activities), 33% from crops, 13% from livestock (mainly cattle), and 7% from agricultural labour. Most households kept 1 to 2 animals and only 21 out of 164 households earned more than Rp. 500,000 (~250 US$) per annum from livestock. The most prevalent type of livestock production in the area could be characterized as small-scale cow/calf operation, either by landless households or those with <0.4 ha of land.

High-Performance Liquid Chromatographic Analysis of Chrysin Derivatives on A $Nova-Pak^{\circledR}C_{18}$ Column

  • A high-performance liquid chromatographic method has been developed for the separation and quantification of chrysin and synthetic chrysin derivatives (12 chrysin alkyl and 7 chrysin acyl derivatives). The chromatography was performed using a $Nova-Pak^{\circledR}C_{18}$ column. A RP-HPLC was performed by using a binary mixture (MeOH-10 mM H$_3$PO$_4$) as a mobile phase, and the column temperature was maintained at room temperature. A flow rate was 1.0 ml/min, and the effluent was monitored at a wavelenth of 280 nm. The retention times for chrysin acyl and alkyl derivatives were within 10 minutes and 20 minutes, respectively. The absolute recovery of samples were all over 96%. The detection limits were 0.1~18 ng at S/N = 3 ratio.

Determination of Co(II) Ion as a 4-(2-Thiazolylazo)resorcinol or 5-Methyl-4-(2-thiazolylazo)resorcinol Chelate by Reversed-Phase Capillary High-Performance Liquid Chromatography

  • Determination of Co(II) ion as a 4-(2-thiazolylazo)resorcinol(TAR) or 5-methyl-4-(2-thiazolylazo)resorcinol(5MTAR) chelate was accomplished by reversed-phase capillary high-performance liquid chromatography (RP-Capillary-HPLC) using a Vydac $C_4$ column and MeCN-water mixture as mobile phase. The effect of change in pH and MeCN percentage of the mobile phase on the retention factor, k and peak intensity were evaluated. It was found that 30% MeCN (v/v) of pH 5.60 or 7.20 was adequate as mobile phase when TAR or 5MTAR is used. Detection limit (D.L., S/N=3) in each case was $2.0\;{\times}\;10^{-7}$M (11.8 ppb) and $3.0\;{\times}\;10^{-7}$ M (17.7 ppb). The Co(II) ion in mineral and waste water was determined with the optimum column and mobile phase.

Procyanidins from Acer komarovii Bark

  • The bark of Acer komarovii was collected, ground, and extracted with 70% aqueous acetone to obtain concentrates. The concentrates were suspended in $H_2O$, and then successively partitioned with n-hexane, dichloromethane and ethylacetate to be freeze dried. A portion of ethylacetate fraction was chromatographed on a Sephadex LH-20 and a RP C-18 column with various aqueous MeOH-$H_2O$ (1:0, 1:1, 1:2, 1:5, 1:7, 1:9, 1:10, 3:1, and 4:1, v/v) eluents. Four compounds were isolated; (-)-epicatechin (9.6 g), procyanidin A2 (epicatechin-($4{\beta}{\rightarrow}8$, $2{\beta}{\rightarrow}O{\rightarrow}7$)-epicatechin) (1.3 g), procyanidin B2 (epicatechin-($4{\beta}{\rightarrow}8$)-epicatechin) (40.0 mg), and cinnamtannin B1 (epicatechin-($4{\beta}{\rightarrow}8$, $2{\beta}{\rightarrow}O{\rightarrow}7$)-epicatechin-($4{\beta}{\rightarrow}8$)-epicatechin) (690 mg). The analysis of the bark procyanidins showed that the basic unit constituting condensed tannins was only (-)-epicatechin. This study, for the first time, report the procyanidins of Acer komarovii bark.
