A Fibrinolytic Enzyme from Bacillus amyloliquefaciens D4-7 Isolated from Chungkook-Jang; It′s Characterization and Influence of Additives on Thermostability. (청국장으로부터 분리한 Bacillus amyloliquefaciens D4-7이 분비하는 혈전용해효소의 특성 및 열안정성에 미치는 첨가물의 효과)

  • Bacillus amyloliquefaciens D4, which produces a strongly fibrinolytic enzyme, was isolated from Chungkook-Jang, a traditional Korean soybean-fermented food. B. amyloliquefaciens D4 was mutated with N-methyl-N-nitro-N-nitrosoguanidine (MNNG) to yield a series of mutants with increasing levels of fibrinolytic enzyme production. After mutation, a mutant D4-7 was obtained with fibrinolytic activity about eight times stronger than the parent strain. The fibrinolytic activity of B. amyloliquefaciens D4-7, reached a maximum, when the producer was cultivated in 2% Isolated Soy Protein (ISP) broth for 48 h at $37^{\circ}C$. Compared to commercial fibrinolytic enzymes, the cell-free culture supernatant of B. amyloliquefaciens D4-7 showed stronger activity than plasmin and streptokinase. The optimum temperature and pH were $50^{\circ}C$ and 10.0 and thermostability was increased by the addition of glycerol, glucose, and NaCl.

Construction of Killer Yeasts by Spheroplast Fusion (포도주용 Killer Yeast의 개발)

  • This study was performed to construct killer wine yeasts which might suppress the growth of wild yeasts, reduce the consumption of starter and condense the fermentation period. Saccharomyces cerevisiae M524, a commercial wine yeast, was treated with N-methyl-N'-nitro-N-nitrosoguanidine to induce auxotrophic mutants, i.e., CHM $2(thr^-)$, CHM 3 $(asp^-)$ and CHM 6 $(tyr^-)$. These auxotrophs were fused successfully with a killer yeast, S. cerevisiae $1368R({\alpha}\;his\;4\;kar\;1-1(kil-k)\;(k_0)$, respiratory deficient) using sphoroplast techniques and the fusants were designated as CHF 21$(th^-\;kil^+)$, CHF 22$(thr^-\;kil^+)$, CHF 31$(asp^-\;kil^+)$ and GHF 61$(tyr^-\;kil^+)$. Combined cultivation of CHF 31 with 1368R or S. cerevisiae $5{\times}47$ (killer sensitive) proved out that CHF 31 had the characteristic of killing and produced the same amount of ethanol as the prototroph, M524.

  • This study was carried out to investigate the antimutagenic affect of crude and heated comfrey extract on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), benzo${(\alpha)}$pyrene (B${(\alpha)}$P) and 3-amino-1, 4-dimethyl-5H-pyri-do [4,3-b] indole (Trp-P-1). In spore rec-assay using Bacillus subtilis $H17(rec^+)$ and $M45(rec^-),$ crude comfrey extract showed strong antimutagenic effects on MNNG in the concentration of $40{\mu}l/disc$(p<0.01). In the Ames test using Salmonella typhimurium TA98 and TA100, the crude comfrey extract suppressed about 43% and 52% in the mutagenesis induced by $B{(\alpha)}P.$ However, the heated comfrey extract strongly suppressed about 75% and 76% in the mutagenesis in Salmonella typhimurium TA98 and TA100 induced by Trp-P-1(p<0.01).

  • Prophage cured strain derivatives from Luctobacillirs araei YIT 9018 were isolated from thermoinducible mutant of the parent lysogenic strain. Two thermoinducible mutants were isolated from L. casei YIT 9018 strain treated with N-methyl-N'-nitro-N-nitrosoguanidine. Prophage cured strains were selected after heat induction of thermoinducible strains at $42^{\circ}C$ for 30 min in MRT medium containing anti- 4 FSV serum. The prophage cured strains, L. casei HYM 1213 and L. casei HYM 4024, could be used an indicator strain for temperate phage $\phi$ FSW. The growth, lactic acid producing ability and carbohydrates fermentation of L. casei HYM 1213 were similar to the parent L. cmei YIT 9018 strain, but A. casei HYM 4024 was not. One of the prophage cured strain, L. cmei HYM 1213, could be used industrially .to produce lactic acid beverages because this strah could not induce the virulent phage$\phi$FSV. The physiological characterization of L. casei HYM 1213 strain was similar to the parent L. casei YIT 9018 strain.

  • Talaromyces luteus 2004, a thermophilic mutant of T. luteus 6112 was obtained by mutagenesis with N-methyl-N'-nitro-N-nitrosoguanidine. T. luteus 2004 produced thermophilic carboxymethylcellulase (CMCase), and other polysaccharide enzymes: avicellase, xylanase, and ${\beta}-glucosidase$. Induction of CMCase production was shown at the highest level in 3% carboxymethylcellulose (CMC) minimal broth, indicating that CMC could work as an inducer. However, glucose and D-cellobiose showed catabolite repression for CMCase production which was under the control of CMC utilization. Optimal conditions for CMCase activity were at $70^{\circ}C$ and pH 4.0, suggesting that CMCase of T. luteus 2004 was a thermophilic enzyme.

  • Two regulatory mutants of Trichoderma reesei QM 9414 were isolated by treatment with N-methyl-N'-nitro-N-nitrosoguanidine, and the effects of various inducers on the carboxymethyIcellulose (CMC) and filter paper (FP) production were investigated. Induction of CMCase and FPase production of mutants was shown higher level than wild type strain in 1% lactose minimal broth. When induced by glucose, wild type showed glucose-repression for CMCase and FPase production and mutants showed glucose-derepression. Mutant 1 showed 8.38 fold higher CMCase activity and 5.68 fold higher FPase activity than wild type stain. Mutant 2 showed about 8.42 fold higher CMCase activity and 5.41 fold higher FPase activity than wild type strain. Enzyme activities from the mutants and wild type had the same optimum pH of 4.8 and optimum temperature of $60^{\circ}C$.

  • Antimutagenic effects of boiled water extract and tannin from persimmon leaves were studied by using Ames test, spore rec assay and SOS chromotest. Strong antimutagenic activities toward aflatoxin B1(AFB1), dimethyl-aminobiphenyl(DMAB), N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 4-nitroquinoline-1-oxide(4-NQO) were observed when boiled water extract and tannin from the persimmon leaves were added in the Salmonella typhimurium TA 100. In spore rec assay using Bacillus subtilis H17($rec^{+}$) and M45($rec^{-}$), boiled water extract and tannin from the persimmon leaves considerably inhibited the mutagenesis induce by MNNG. In SOS chromotest using Escherichia coli PQ37, these samples also exhibited strong antimutagenic activity toward 4-NQO. The tannin was more effective than boiled water extract of persimmon leaves in the antimutagenicity tests.

  • The desmutagenic activity of the water extract of Cassia tara L on the mutagenicity induced by 4-nitroquinoline 1-oxide (4-NQO) and N-methyl-N-nitro-N'-nitrosoguani-dine (MNNG) was studied using the SOS Chromotest with Escherichia coli PQ37. The inhibition rates of water extract of Cassia tara L. at concentration of $100{\mu}g$/assay were 27.5% and 40% against 4-NQO and MNNG. The water extract of Cassia tara L. was separated into methanol soluble and methanol insoluble parts. The methanol soluble part exhibited higher inhibition effects than the methanol insoluble part against the mutagenic activities of 4-NQO and MNNG. Step-wise fractionation of methanol soluble part was done to obtain methanol, ethyl acetate and water fractions. Among these fractions, water fraction had the strongest inhibitory effects of 23.0 and 19.0% against mutagenicities of MNNG and 4NQO, respectively. The results clearly indicated that the water fraction showed much stronger antimutagenicity against MNNG than 4NQO. The inhibition rates of aqueous fraction of methanol-soluble from water extracted Cassia tara L. at concentrations of 1.0, 10, 100 and $250{\mu}g$/assay were 8.0%, 12.0%, 25.5% and 43.0%, respectively. The water fraction showed the inhibitory effects with dose response against the mutagenic activities induced by MNNG.

The mutant strain produced gibberellic acid (70 mg/l) more than that of wilde type. A fermentation medium containing glucose, $NH_4N0_3$, $MgS0_4$, $KH_2P0_4$ and trace elements was deve]oped for the maximal production of a gibberellic acid by the mutanL The Guctuating cultural temperature that was vaded from 300e to 20DC resulted in higher GA yield than that of fixed cu1tura] temperature at $28^{\circ}C$.
