Exploitation of the biologically active components in Youngia sonchifolia Max (고들빼기 생리활성물질의 검색)

  • Yongia sonchifolia Max. has been used as raw materials of traditional Kimchi and medicinal herb in Korea. This study was performed to investigate biologically active components in the plant. First, the writer carried out the experiment of antitumor screening test against Sarcoma-180A and the cytotoxic activity against Chinese hamster V-79 cells with methanol extract of the plant. And the aqueous solution of the extract from roots of Youngia sonchifolia Max. was partitioned into n-hexane. The concentrated extract of n-hexane layer was chromatographed on silica gel column and developed with n-hexane and ethylacetate. Two yellow elutes, on concentration, were recrystallized from ethylacetate, and the $R_f$ value of TLC of the crystal was 0.43. After analysis by $^{1}H-NMR$, $^{13}C-NMR$ and MS to confirm the structure, the author could identify the compound as bauerenyl acetate, a naturally occurring pentacyclic triterpene. The crystal was colorless plate and m.p. was $280{\sim}282^{\circ}C$.

  • 80% MeOH extract of black rices fractionated with n-hexane, EtOAc, and n-BuOH. Copper-induced LDL oxidation inhibition assay and ACAT inhibition assay examined with fractions. n-Hexane and EtOAc fractions showed high inhibition activity. We divided n-hexane fraction into three sub-layers(H1 - H3) and EtOAc into eight sub-layers(E1 - E8). H1, H2, E6, and E7 are showed higher inhibition activity than standard in Lp-PLA2 inhibition assay and H1 and E6 are showed higher ACAT inhibition activity than standard.

  • Antioxidant activity of a methanol extract of Symphyocladia latiuscula was evaluated by the thiocyanate method in the linoleic acid system. The methanol extract inhibited the peroxidation of linoleic acid in a dose-dependent manner. The MeOH extract was then sequentially partitioned with n-hexane, $CH_2Cl_2$, EtOAc, n-BuOH and H20. The antioxidant activity of the fractions increased in order of $CH_2Cl_2$, n-hexane, EtOAc, and n-BuOH. There was no activity found in $H_2O$ partitoned fraction by the thiocyanate method. Especially, the activities of the fractions of n-hexane and $CH_2Cl_2$ were comparable to that of 2,6-di-tert­butylhydroxytoluene (BHT). Column chromatography of the $CH_2Cl_2$ fraction over silica gel yielded cholesterol (1) and 2,3,6-tribromo 4,5-dihydroxybenzyl methyl ether (2) which were identified by instrumental analysis of MS and $^1H-$ and $^{13}C-NMR$. The latter (2) demonstrated significant antioxidant activity.

Anitimicrobial Effects of Zanthoxylum schinifolium Extracts against Multi-drug Resistant Acinetobacter baumannii (산초(Zanthoxylum schinifolium) 추출물의 다제내성 Acinetobacter baumannii 억제 효과)

  • The aim of this study was to investigate the possible utilization of Zanthoxylum schinifolium as a source of antimicrobial agents. The antimicrobial effects of Zanthoxylum schinifolium extracts were investigated against Acinetobacter baumannii, which is a multi-drug resistant pathogen, and 5 other pathogenic microorganisms. The hexane extract of Zanthoxylum schinifolium was more effective than the ethyl acetate, n-butanol and methanol extracts which were active against Acinetobacter baumannii 25, with minimum inhibitory concentrations(MIC) ranging from 0.8 mg/ml to 1.6 mg/ml. Tetracycline had no effect on Acinetobacter baumanniii. The hexane extract was highly active against Candida albicans IFO 6258, with an MIC of 1.5 mg/ml. In contrast, the ethyl acetate, n-butanol and methanol extracts showed no activity against the 5 pathogenic microorganisms. Furthermore, a combination of hexane extract and ethyl acetate extract was significantly more active against the 5 Acinetobacter baumannii strains than n-butanol and methanol. These results suggest that Zanthoxylum schinifolium extracts have great potential as antimicrobial compounds against multi-drug resistant pathogens, and further studies are warranted.

Constituents of Antioxidative Activity and Free Radical Scavenging Effect from Galla Rhois (Rhus javanica Linne) (오배자의 항산화 활성성분 및 자유라디칼 소거효과)

  • This study was carried out to investigate the structure of antioxidative constituents and the free radical scavenging effect of the main ingredients from Galla Rhois(Rhus javanica Linne). Antioxidative activities of n-hexane, EtOAc and BuOH extracts of Galla Rhois were similar or even higher than that of natural (tocopherol) or synthetic antioxidant (BHA). It is suggested that major fractions for the antioxidative activity of Galla Rhois were the n-hexane, EtOAc and BuOH extract compartments. In the subsequently experiment, one active compound from n-hexane extract, three active compounds from EtOAc extract and one active compound from BuOH extract were isolated. Their chemical structures were identified as syringic acid, protocatechuic acid, gallic acid methylester, gallic acid and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ on the basis of the speculation of spectral data and chemical reaction. Among the compounds, protocatechuic acid, gallic acid methylester and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ showed most potent radical scavenging effect using DPPH method.

Regulation of Wnt/β-catenin Signal Transduction in HT-29 Colon Cancer Cells by a Rhododendron brachycarpum Fraction (홍만병초 분획물에 의한 HT-29대장암 세포의 Wnt/β-catenin 신호전달 조절)

  • The aim of this study was to determine the anticancer activities of an 80% methanol extract and various fractions of Rhododendron brachycarpum (RB). The n-hexane fraction of RB showed the highest inhibitory activity (Inhibit concentration $50%=20.2{\pm}1.2{\mu}g/ml$) in HT-29 cells. Colony- and sphereforming abilities were significantly correlated with a decrease in the cell count and size. A TOP/FOP flash reporter assay revealed that the inhibitory activity of the n-hexane fraction of RB ($0.22{\pm}0.02$ fold change) was lower than that of the 80% methanol extract and that of other fractions. The n-hexane and ethyl acetate fractions of RB were predominantly dependent on the expression levels of intracellular ${\beta}-catenin$. Western blotting using $p-GSK3{\beta}$ with only the n-hexane fraction of RB was conducted to examine whether these secondary metabolites reduced ${\beta}-catenin$ degradation. Intracellular ${\beta}-catenin$ regulation resulted in quantitative changes in the nucleus. In summary, these results demonstrate the potential of the n-hexane fraction of RB as a natural anticancer agent.

Separation of Aromatics in Light Cycle Oil by Solvent Extraction - Re-extraction of Aromatics in Extract Phase - (용매 추출법에 의한 분해경유중의 방향족 분리 -추출상중의 방향족의 역수출-)

  • Recovery of aromatics in extract phase which was obtained by batch equilibrium extraction between light cycle oil(LCO) and dimethylsulfoxide(DMSO) solution as solvent was investigated by re-extraction. To select the most suitable re-extraction solvent for recovery of aromatics in extract phase, distribution equilibrium was measured between extract phase and solvents. The solvents used were benzene(B), toluene(T), m-Xylene(mX), n-hexane(Hx) and n-hexane(Hx) and n-Octane(Ot). From the distribution coefficients and yields of aromatics, Hx seemed to be the most suitable. Furthermore, effects of operation parameters for re-extraction of aromatics in the extract phase were studied by batch equilibrium re-extracion with Hx as solvent. Yields of aromatics were found to increase with increasing solvent/feed (extract phase) mass ratio(S/F), while distribution coefficients of aromatics were fixed irrespective of S/F used. Operating temperature did not affect distribution coefficients and yields of aromatics. Distibution coefficients and yields of naphthalene group(carbon numer : 10~12) increased with increasing cabon number. Mass transfer rates of aromatics were also measured with a batch stirred vessel.

Antioxidative and Free Radical Scavenging Effects of Rhus javanica Linne (오배자의 항산화 및 자유라디칼 소거효과)

  • This study was carried out to investigate the antioxidative activities of medicinal plants. Through the examination of methanol extracts from 30 species for radical scavenging effects using DPPH method, the extracts from Rhus javanica Linne, Smilax china Linne and Polygonum cuspidatum Siebold et Zucarinii showed strong antioxidative activity. Because of its highest antioxidative activity among 30 medicinal plants, radical scavenging effects of 4 different extract compartments (n- Hexane, EtOAc, BuOH and $H_2O$ extracts) from Galla Rhois MeOH extract of Rhus javanica Linne were examined by DPPH method and antioxidant effects on the 4 different extract compartments were tested by Ferric-Thiocyanate method. Antioxidative activities of n- Hexane, EtOAc and BuOH extracts were similar or even higher than that of natural (tocopherol) or synthetic antioxidants (BHA), suggesting that major fractions for the antioxidative activity of Rhus javanica Linne were the n- Hexane, EtOAc and BuOH extract compartments.

Anticoagulant and Fibrinolytic Activities of Hwanggeumchal Sorghum In Vitro (황금찰수수의 혈액응고저해 및 혈전용해 효과)

  • To examine whether miscellaneous cereal grains have an antithrombotic effect, we investigated the anticoagulant activity of 80% ethanol extracts from eleven selected miscellaneous cereal grains. The 80% ethanol extract of hwanggeumchal sorghum (Sorghum bicolor) showed the highest anticoagulant activity, followed by that of green foxtail millet grains, in terms of thrombin time (TT). When the ethanol extract of hwanggeumchal sorghum was sequentially fractionated with n-hexane, methylene chloride, ethyl acetate, and n-butanol, the majority of the TT-inhibitory activity was detected in the hexane and methylene chloride fractions. Whereas aspirin (final conc. 480 ${\mu}g/ml$) prolonged TT by 2-fold, the ethanol extract, hexane fraction, and methylene chloride fraction in the same dose prolonged TT by 2.2-fold, 2.9-fold, and 2.5-fold, respectively. The ethanol extract of hwanggeumchal sorghum could delay activated partial thromboplastin time (APTT) as well as prothrombin time (PT). Although the APTT-inhibitory activity of the ethanol extract was mainly partitioned into the hexane and methylene chloride fractions, the PT-inhibitory activity of the ethanol extract was solely partitioned into the hexane fraction. The APTT- and PT-inhibitory activities of these organic solvent fractions were more potent than those of the control warfarin (final conc. 3.13 mg/ml). The TT-inhibitory activity of the ethanol extract was heat-stable and acid-stable. The ethanol extract, hexane fraction, and methylene chloride fraction of hwanggeumchal sorghum appeared to possess a direct fibrinolytic activity toward fibrin clotting. These results show that hwanggeumchal sorghum can exert anticoagulant and fibrinolytic effects and, thus, have the potential to be applicable as antithrombotic dietary sources.

Studies on Antioxidative Substances of Ganoderma lucidum (영지의 항산화성 물질에 관한 연구)

  • To study antioxidative activities of Ganoderma lucidum, its extracts were fractionated by various organic solvents with different polarity the extracts were purified by thin chromatography, silicagel column chromatography and preparative liquid chromatography. In antioxidative activity tests using thiocyanate method, TBA method and weighing method, fraction 5 from the hexane extract and fraction II from the methanol extract showed antioxidative activity. When the antioxidative activities were expressed as TBA value using a homogeneous liver extracte of rats, the relative antioxidative activities of fraction 5 and fraction II were increased by 13.0% and 54.6%, respectively.
