A Study on the Effect of the Dendropanax Mobifera Extract on Anti-Hypertensive (황칠나무 추출물이 고혈압에 미치는 영향에 관한 연구)

  • This study examined the effects of an extract of Dendropanax morbifera on blood pressure, Angiotensin II, Angiotensin Converting Enzyme, Aldosterone, and lipid levels of spontaneously hypertensive rats. The groups were as follows: Control group, Hypertension control group, Water extract treated group, Ethanol extract treated group, n- hexane fraction treated group, Ethyl acetate fraction treated group, n- butanol fraction treated group, and Water fraction treated group. The blood pressure, and Angiotensin II, and Angiotensin Converting Enzyme, and Aldosterone levels were lower in the Ethyl acetate fraction treated group than in the hypertension control group. The change in blood pressure was lower in the Water extract treated group, Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group. The concentration of Angiotensin II was lower in the Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group(p<0.05). The level of Angiotensin Converting Enzyme was lower in the Ethyl acetate fraction treated group, Ethanol extract treated group, n- hexane fraction treated group, and n- butanol fraction treated group than the hypertension control group. The concentration of Aldosterone was lower in the Ethyl acetate fraction treated group, and n- butanol fraction treated group than the hypertension control group(p<0.05). In addition, the concentration lower in the Water extract treated group, Ethanol extract treated group, n- hexane fraction treated group than the hypertension control group. Overall, the effect of the Dendropanax morbifera extracts might be useful as anti-hypertensive, and functional food agents.

Inhibitory Activity of Asarum sieboldii against Wood Rot Fungi on Traditional Paper, Hanji (한지에서 셀룰로오스 분해 미생물에 대한 세신 추출물의 저해 활성)

  • Background: To investigate the possibility of using Asarum sieboldii as an environment-friendly fumigant for protecting organic cultural heritages, the inhibitory effect of A. sieboldii extract against wood rot fungi on Hanji was examined. Methods and Results: The physical, optical, and morphological properties of Hanji inoculated with Trametes versicolor and Tyromyces palustris, and exposed to the n-hexane fraction of A. sieboldii extract, were measured. The physical properties were expressed as weight loss, zero-span tensile strength and viscosity and the optical properties were depicted by luminance and chromaticity ($L^*$, $a^*$, and $b^*$). The results showed that, the n-hexane fraction of A. sieboldii extract inhibited the growth of fungi on Hanji, and preserved its condition. At a concentration of 25 mg, the n-hexane fraction of A. sieboldii extract maintained zero-span tensile strength, increased viscosity, and restricted discoloration of Hanji. It also was confirmed that the weight of fungi infested Hanji exposed to the extract did not decrease. Scanning electron microscopic images revealed that the spores and hyphae of T. versicolor and T. palustris were not present on Hanji during treatment with > 25 mg of the n-hexane fraction of A. sieboldii extract. Conclusions: These results indicate that the n-hexane fraction of A. sieboldii extract by virtue of its antifungal effectiveness may help in preserving Korean paper cultural heritages, including Hanji.

Inhibitory Effect of Methanolic Extract from Radix Trichosanthis on Melanin Synthesis (천화분 메탄올 추출물의 멜라닌 형성 억제 효과)

  • Objectives: In order to investigate the relationship of Radix Trichosanthis components and the melanin synthesis, the author has analyzed the cell viability and tyrosinase activity, melanin content and morphologic changes in n-hexane, EtOAc, n-BuOH, and H2O fraction. Methods: At first, in order to determine the concentration of the Radix Trichosanthis component, the author investigated the viability of B16 melanoma cell. To measure the effects of Trichosanthes kirilowii extracts (n-BuOH, n-Hexane, EtOAc, H2O fractions) on the viability of A549 cells, A549 cells were treated with various concentrations (from 0.5 to $25{\;}\mu\textrm{g}/ml$) of components of Trichosanthes kirilowii. After 24hrs, the cell viability was measured by MTT assay. The EtOAc components of Trichosanthes kirilowii decreased the viability of A549 cells in a dose-dependent manner. H2O and n-BuOH components had no cell toxicity till $25{\;}\mu\textrm{g}/ml$, the n-hexane component showed minor cell toxicity at $25{\;}\mu\textrm{g}/ml$ and the EtOAc component cell toxicity was revealed at $5{\;}\mu\textrm{g}/ml$ concentration. Results: 1. The results of tyrosinase activity and the Radix Trichosanthis component; n-hexane and EtOAc components controlled it effectively; the n-BuOH components were less effective. 2. The results of melanin content analysis showed that the n-hexane and EtOAc components effectively inhibited, the n-BuOH fraction inhibited less, and H2O component didn't inhibit the terminal melanin formation. 3. In the n-BuOH and H2O component there were no changes, but in the n-hexane component the melanin content was effectively inhibited. 4. In the EtOAc fraction, although the melanin content was inhibited, the cell count was evidently suppressed, Of all of the Radix Trichosanthis components, the n-Hexane and EtOAc fractions inhibited the melanin synthesis best, but owing to its toxicity, the EtOAc components inhibited the cell count. Conclusion: The above results demonstrated that Radix Trichosanthis n-hexane fraction efficiently inhibited the tyrosinase activity and melanin synthesis.

Antioxidative Effect of Chaenomelis Fructus Ethanol Extract (모과 에탄올 추출물의 항산화 효과)

  • This study was performed to investigate the antioxidant effect of 80% (v/v) ethanol extract from Chaenomelis Fructus (CF). Total flavonoids and total polyphenols in the extract were also measured spectrophotometrically. The extraction yield was 9.23g/100g CF. The extract was further fractionated by partition with n-hexane, chloroform, ethylacetate, butanol, and water. The water fraction showed the highest extraction yield of all fractions. The n-hexane method and compared with the properties of the commerical antioxidant BHT. The activities of the n-hexane fraction were the highest of all fractions. In addition, there was strong positive correlation between antioxidant activities and levels of antioxidative compounds, such as flavonoid and polyphenols, in CF fractions, suggesting that these antioxidative compounds may contribute to the antioxidative effect of CF.

Studies on the Organic Matters in River Sediments (하천저질중의 유기물질에 관한 연구)

  • The writer measured methemoglobin formation with solvent (n-hexane, ethylacetate and methanol) organic extracts from the sediments of the Wonju stream run through Wonju city on Octover 2nd, 1985. Hemoglobin was converted to methemoglobin with the n-hexane extract obtained from the sediment of the St. C and St. D was polluted industrial wastewater, but not with that from the St. B was polluted with the urban wastewater. The formation of methemoglobin was remarkable with the all solvent extracts from the sediment of the uper stream of the Wonju stream. Doses of the solvent extracts for 20% methemoglobin formation in the researched Wonju streams were as follows; 0.095~0.28 mg/m.$\ell$ in n-hexane extract. 0.85-1.3 mg/ m$\ell$ in methanol extract and 1.95~2.80 mg/m$\ell$ in ethylacetate extract.

Cytotoxicity and Antimicrobial Effect of the Extract of Salvia plebeia (배암차즈기 추출물의 세포독성과 항균효과)

  • The comparison of $IC_{50}$ values of Salvia plebeia R. Br. extracts on L1210, $P388D_1$ cancer and Vero normal cell lines showed that the n-hexane soluble extract of S. plebeia R. Br. retains the most growth-inhibitory activity against tumor cell lines. The minimum inhibitory concentration (MIC) of the extract against microorganisms were also examined. Antimicrobial activity of amocla and ketoconazole as references was compared to those of other solvent extracts such as $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The antimicrobial activity of extract had growth inhibition activity against gramnegative bacteria, gram-positive bacteria and fungi $(MIC\;>\;200\;{\mu}g/ml)$ except for n-hexane extract. Seven bacterial strains were tested for in vitro susceptibility to the extract of S. plebeia R. Br. However the n-hexane extract of S. plebeia R. Br. inhibited the growth of several bacterial strains (MIC values between 100 and $200\;{\mu}g/ml$ for gram positive bacteria, $25\;{\mu}g/ml$ for P. putida).

Anti-termite Activity of Tamanu Bark Extract (Calophyllum inophyllum L.)

  • This study aimed to analyze the anti-termite properties of tamanu (Calophyllum inophyllum L.) stem bark extracts against subterranean termites, specifically, Coptotermes curvignathus. The bark powder of C. inophyllum was extracted using different solvents, such as n-hexane, ethyl acetate, and methanol, using the maceration method. Anti-termite testing was performed using two paper disc methods: no- and two-choice tests. Whatman test paper was dripped with the extract solutions at concentrations of 4%, 6%, 8%, and 10% (w/v). Subsequently, the treated paper disc was placed into an acrylic tube, and the subterranean termite was added. The parameters utilized in the test included termite mortality and the weight loss of the test paper. The results revealed that the total extract yield of C. inophyllum stem bark was 30.24%. Furthermore, the extractive substances from C. inophyllum bark exhibited anti-termite activity. The most favorable outcomes were obtained with the n-hexane and ethyl acetate extracts at a concentration of 10%. The termite mortality and weight loss of the test paper were respectively 66% and 5.67% for the n-hexane extract and 66.67% and 6.19% for the ethyl acetate extract. In addition, the n-hexane extract contained friedelan-3-one, while the ethyl acetate extract contained 1,2-benzene dicarboxylic acid, dinonyl ester, and friedelan-3-one. The results suggested that these compounds are responsible for the observed anti-termite activity.

Antimicrobial Activity and Antimutagenesis of Cinnamon (Cinnamomum cassia Blume) Bark Extract (계피추출물의 항균 작용과 항돌연변이원성)

  • In order to develop antimicrobial substances, many kinds of medicinal herbs were extracted with absolute ethanol and then antimicrobial activities against various microorganisms were investigated. Ethanol extract from cinnamon bark showed the strongest antimicrobial activity on the growth of almost all submitted microorganisms. Specially, molds such as Aspergillus sp. and Pencillium sp. were inhibited strongly. Therefore, the crude antimicrobial substance from the ethanol extract was fractionated with various solvents such as n-hexane, chloroform, ethyl acetate, and n-butyl alcohol and then their antimicrobial activities were tested. Among the various solvent fractions from the ethanol extract, n-hexane fraction was the best in antimicrobial activity especially against molds. There were no significant changes in antimicrobial activity of the n-hexane fraction by heat treatment at $100^{\circ}C$ for 60 min or $121^{\circ}C$ for 15 min and by the change of pH 4.0~10.0. We could get the results that the n-hexane fraction of cinnamon bark extract showed not only antimutagenicity but also no mutagenicity by Ames test with Salmonella typhimurium TA 98 and TA 100.

The Antioxidant and Antitumor Effects of the Extract of Bulnesia sarmientia (Bulnesia sarmienti 추출물의 항산화 및 항암효과)

  • Present study have been performed to develop Bulnesia sarmienti as a functional food. Methanol, n-hexane, chloroform, ethyl acetate and butanol extracts of Bulnesia sarmienti contained total phenol by 5.81 to 7.47%. It is high content than fruits which were known as high contests of total phenol. The electron donating ability of the extract of Bulnesia sarmienti were increased along with increasing concentrations of extracts. At $500{\mu}g/mL\;and\;1000{\mu}g/mL$, the all extracts showde more than 80% of scavenging abilities, which means the equal effect of the antioxidant, BHT. Nitrite scavenging abilities were measured as follows: methanol, butanol, 5.53, 5.77% at $100{\mu}g/mL$, respectively. The ethyl acetate extract was 73.29% at $1000{\mu}g/mL$ which showed the highest activity and methanol, butanol, n-hexane, chloroform and water extract were 65.65, 65.02, 47.49, 52.51, 45.54% which also showed relatively high activities. The growth inhibitory effects of each solvent extract on tumor cell were as follows: test against SUN-1, the gastric carcinoma cell, exhibited the highest inhibitory effects at $100{\mu}g/mL$ where the n-hexane extract was 61.6%. The ethyl acetate and water extracts did not revealed any inhibitory effects. Hela, the uterine carcinoma cell, exhibited the highest inhibitory effects at $100{\mu}g/mL$ where the n-hexane extract was 75.1%. The water extracts did not revealed any inhibitory effects. HT-29, the colon carcinoma cell, also exhibited the highest inhibitory effects at $100{\mu}g/mL$ where n-hexane extract was 57.4%. In conclusion, Bulnesia sarmienti have been shown the antioxidant and antitumor effects, and that it is expected to be developed as functional foods.

Antioxidative Effect of Angelica acutiloba Kitagawa Ethanol Extract (일당귀 에탄올 추출물의 항산화 효과)

  • This study was conducted to investigate the antioxidant effect of 80% ethanol extracts from Angelica acutiloba Kitagawa (A. acutiloba Kitagawa) in vitro. The extract was further fractionated subsequently by n-hexane, chloroform, ethylacetate, n-butanol and water. Antioxidative activities of different fractions were examined by 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical generation, Rancimat test, thiobarbituric acid (TBA) value, nitrite scavenging activity, inhibition of lipid peroxidation and peroxide value (POV) in linoleic acid in comparison with the commercial antioxidant butylated hydroxy toluene (BHT). Antioxidant activities of n-hexane fraction of Angelico acutiloba Kitagawa ethanol extract were the highest among fractions and were a little less than that of BHT. Nitrite scavenging activity showed the most remarkable effect at pH 12. These results suggest that ethanol extracts of A. acutiloba Kitagawa can be used in natural antioxidant source.
