Effects of Saccharomyces cerevisiae Supplementation and Anhydrous Ammonia Treatment of Wheat Straw on In-situ Degradability and, Rumen Fermentation and Growth Performance of Yearling Lambs

  • The effects of Saccharomyces cerevisiae supplementation ($6.6{\times}10^8cfu$) and anhydrous ammonia treatment (3%) of wheat straw (WS) were investigated on in-situ dry matter (DM) degradability, and on rumen fermentation and growth performance of lambs. Rumen-fistulated Menemen sheep fed a diet with and without live yeast were used to assess the DM degradability characteristics of WS and ammonia-treated wheat straw ($WS_{NH3}$). Twenty-six yearling Menemen male lambs were fed in four groups. Lambs of control group (WS) received untreated WS without supplemental yeast, whereas other three groups were fed WS treated with anhydrous ammonia ($WS_{NH3}$ group), untreated WS and yeast (WS+YEAST group) or WS treated with anhydrous ammonia and yeast ($WS_{NH3}$+YEAST group). Supplemented live yeast (4 g/d) was added in the diet. Lambs were offered untreated or ammonia treated WS ad-libitum and concentrate was fed at 1% of live body weight. The degradability of the water-insoluble (fraction B) was significantly increased by all of the treatment groups. Potential degradability (A+B), effective DM degradability's (pe2, pe5, and pe8) and average daily weight gain increased only in $WS_{NH3}$+YEAST group (p<0.05). Voluntary DM intake was not increased by the treatments (p>0.05), but voluntary metabolizable energy and crude protein intake were increased by $WS_{NH3}$ and by $WS_{NH3}$+YEAST (p<0.05). Average daily rumen pH was not affected by any of the treatments, but average daily $NH_3$-N was significantly higher in the $WS_{NH3}$ and $WS_{NH3}$+YEAST groups, and total volatile fatty acids were significantly higher in the WS+YEAST and $WS_{NH3}$+YEAST groups. In conclusion, the improvement of feed value of WS was better by the combination of ammonia-treatment and yeast supplementation compared to either treatment alone.

  • 바이오디젤(BD)은 식물성 오일 또는 동물성 지방과 같이 재생산 가능한 원료로부터 유래된 긴 사슬 지방산의 단일 알킬에스테르로, 디젤연료에 비해 낮은 온도에서 연료특성이 열악한 것으로 알려져 있다. 디젤연료의 경우, 많은 저온 유동성 향상제가 개발되어 있지만 바이오 디젤은 디젤연료와 주요 구성성분이 다르기 때문에 디젤연료용 저온 유동성 향상제를 바이오디젤에 사용 시 저온 유동성 향상에 한계가 따른다. 이에 본 연구는 동절기에 바이오디젤의 저온 특성을 향상시키고자 에스테르 반응으로 합성한 알킬 메타크릴레이트 단량체(Stearyl methacrylate, Lauryl methacrylate)와 무수말레인산을 이용하여 저온 유동성 향상제를 합성하였고, 알킬아민을 이용한 개환 반응을 실시하였다. 이렇게 합성된 저온 유동성 향상제를 $^1H-NMR$ 및 FT-IR을 통해 분석하였으며, GPC로 분자량을 측정한 후 SoybeanBD와 PalmBD에 1000~10000 ppm의 농도로 첨가하여 저온특성을 조사하였다. 그 결과 SoybeanBD에 LMA2SMA6MA2-C8A 공중합체 첨가 시 유동점이 $12.5\;^{\circ}C$ 강하됨을 확인하였다.

영양소 섭취 수준과 유방암 위험 - 대구.경북지역 환자-대조군 연구 - (Intakes of Energy and Nutrients and Risk of Breast Cancer - Case-Control Study in Daegu.Gyeongbuk Area, Korea -)

  • 본 연구는 대구 경북지역의 유방암 환자와 대조군을 대상으로 열량 및 영양소 섭취수준에 따른 유방암 발생의 상대 위험도를 분석하였다. 조사 대상자는 경북대학교 병원에서 유방암 진단을 처음 받고 입원한 103명의 유방암 환자와 유방암과 관련된 질환 및 전신 질환이 없으면서 연령(5세 내의 차이) 및 폐경 상태가 동일한 사람들을 대상으로 빈도 짝짓기를 통하여 선정한 159명의 지역사회 대조군이었다. 이들의 반정량적 식품섭취 빈도조사 결과를 분석하여 영양 섭취량을 산출하였고, 일반 특성에 대한 분석 결과에서 환자군과 대조군간 유의한 차이를 보인 요인들을 혼란변수로 통제한 후 열량 및 영양섭취 수준에 따른 상대 위험도를 분석하였다. 전체적으로 높은 BMI와 열량과 당질의 높은 섭취 수준은 유방암 발생의 상대적 위험을 유의하게 높이는 것으로 나타났고, 총 지방, 식이섬유, 포화지방산, 단일불포화지방산, 다불포화지방산, n-3 지방산, n-6 지방산, 비타민 E, 칼슘은 섭취수준이 높을수록 위험도가 유의하게 낮아지는 것으로 나타났다. 폐경 전 여성에서는 당질의 섭취 수준이 높을 때 유방암 위험이 유의하게 높고, 총지방, 식이섬유, 포화지방산, 단일불포화지방산, 다불포화지방산, n-6 지방산, 비타민 E, 엽산, 칼슘, 인, 칼륨의 섭취 수준이 높을 때 위험도가 유의하게 낮은 것으로 나타났다. 폐경 후 여성에서는 총지방, 콜레스테롤, 다불포화지방산, n-6 지방산의 섭취 수준이 높을 때 유방암 위험도가 유의하게 낮은 것으로 나타났다. 특히 지방의 경우 식물성 급원의 섭취 수준이 높을 때 유방암의 위험이 유의하게 감소되는 것으로 나타났다. 이상의 결과에서 대구 경북지역에서 유방암 발생 위험을 높일 가능성이 있는 영양섭취상태는 열량과 당질의 높은 섭취이며, 한편 유방암의 위험을 낮출 가능성이 있는 영양섭취 상태는 높은 지방의 섭취로 나타났다. 특히 지방의 경우 식물성 급원의 섭취가 높을 때 위험이 낮아지는 것으로 나타났다. 이는 지방 섭취량과 유방암 위험도 사이에 정의 상관성을 보고한 일부 국외의 연구 결과와는 상반되는 결과이며 그 원인으로는 한국인의 경우 식물성 지방 섭취의 비율이 서구인에 비해 상대적으로 높기 때문인 것으로 추정된다. 유방암의 위험을 낮출 가능성이 있는 미량 영양소로는 비타민 중에는 E와 엽산, 무기질로서는 칼슘, 인 그리고 칼륨으로 나타났다. 본 연구결과의 제한점으로서 조사 사례수가 적다는점을 들 수 있으므로 선행연구들의 결과와 상반된 결과를 보인 지방섭취와 유방암 위험과의 관련성에 대해서는 향후보다 많은 사례를 대상으로 한 조사 연구를 통해 재확인할 필요가 있다고 보며, 본 연구의 결과는 지역민의 유방암 예방을 위한 영양교육 자료의 일부로서 활용될 수 있다고 본다.

연화 및 연화주의 영양성분 및 항산화활성 (Nutritional components and antioxidant activities of Nelumbo nucifera Gaertner flower and its wine)

  • 본 연구는 연화 및 연화주에 대한 식품학적 기초자료를 제공하고자 연화 및 연화주의 영양성분, 항산화성분 및 다양한 모델에서의 항산화활성을 평가하였다. 연화 및 연화주의 수분함량은 각각 85.90 및 92.87%이었고, 조회분 함량은 1.04 및 0.15%, 조단백질 함량은 1.91 및 1.70%, 조지방 함량은 0.30 및 0.11%, 탄수화물 함량은 10.85 및 5.17%이었다. 연화 및 연화주의 총 아미노산 함량은 각각 2,168 및 6,341 mg/kg 이었고 필수 아미노산은 각각 485 및 2,246 mg/kg으로 전체 아미노산 함량의 약 26.0 및 35.4%이었다. 연화에는 fructose가 0.14 g/100 g 함유되어 있었고 연화주에는 glucose 1.85 g/100 g이 함유되어 있었으며 지방산 조성은 연화의 경우, palmitic acid(C16:0)가 약 38.63%로 가장 높았고, 연화주의 경우 oleic acid(C18:1n9c)가 약 76.24%의 비율로 가장 높았다. 무기질 함량은 연화(390 mg/100 g) 및 연화주(27.40 g/100 g)에서 모두 K이 가장 높았다. 유기산 함량은 연화에서 citric acid(2,222 mg/kg)가 가장 많았고, 연화주는 lactic acid(5,427.90 mg/kg)가 가장 많았다. 한편, 연화 에탄올 추출물 및 열수 추출물의 항산화활성은 연화주 동결건조물에 비해 높았으며 이는 총 페놀 함량의 결과와 유사한 경향을 나타냈다. ORAC value는 연화 에탄올 추출 및 열수 추출물에서 각각 $272.9{\pm}7.6$$275.3{\pm}2.6mM\;TE/g$을 나타낸 반면, 연화주 동결건조물에서는 $11.5{\pm}0.4mM\;TE/g$로 나타났다. 이상의 연구 결과로부터 연화 추출물은 다양한 항산화 평가모델에서 항산화 활성을 나타내었으며 전통방식에 의해 제조된 연화주의 항산화 활성은 연화에 함유된 일부 항산화 성분의 침출에 기인된 것으로 사료되며, 연화추출물 및 연화주는 체내 산화적 스트레스 저감에 일부 도움을 줄 것으로 판단되었다.

Influence of Wheat Straw Pelletizing and Inclusion Rate in Dry Rolled or Steam-flaked Corn-based Finishing Diets on Characteristics of Digestion for Feedlot Cattle

  • Eight Holstein steers ($216{\pm}48kg$ body weight) fitted with ruminal and duodenal cannulas were used to evaluate effects of wheat straw processing (ground vs pelleted) at two straw inclusion rates (7% and 14%; dry matter basis) in dry rolled or steam-flaked corn-based finishing diets on characteristics of digestion. The experimental design was a split plot consisting of two simultaneous $4{\times}4$ Latin squares. Increasing straw level reduced ruminal (p<0.01) and total tract (p = 0.03) organic matter (OM) digestion. As expected, increasing wheat straw level from 7% to 14% decreased (p<0.05) ruminal and total tract digestion of OM. Digestion of neutral detergent fiber (NDF) and starch, per se, were not affected (p>0.10) by wheat straw level. Likewise, straw level did not influence ruminal acetate and propionate molar proportions or estimated methane production (p>0.10). Pelleting straw did not affect ($p{\geq}0.48$) ruminal digestion of OM, NDF, and starch, or microbial efficiency. Ruminal feed N digestion was greater (7.4%; p = 0.02) for ground than for pelleted wheat straw diets. Although ruminal starch digestion was not affected by straw processing, post-ruminal (p<0.01), and total-tract starch (p = 0.05) digestion were greater for ground than for pelleted wheat straw diets, resulting in a tendency for increased post-ruminal (p = 0.06) and total tract (p = 0.07) OM digestion. Pelleting wheat straw decreased (p<0.01) ruminal pH, although ruminal volatile fatty acids (VFA) concentration and estimated methane were not affected ($p{\geq}0.27$). Ruminal digestion of OM and starch, and post-ruminal and total tract digestion of OM, starch and N were greater (p<0.01) for steam-flaked than for dry rolled corn-based diets. Ruminal NDF digestion was greater (p = 0.02) for dry rolled than for steam-flaked corn, although total tract NDF digestion was unaffected (p = 0.94). Ruminal microbial efficiency and ruminal degradation of feed N were not affected (p>0.14) by corn processing. However, microbial N flow to the small intestine and ruminal N efficiency (non-ammonia N flow to the small intestine/N intake) were greater (p<0.01) for steam-flaked than for dry rolled corn-based diets. Ruminal pH and total VFA concentration were not affected ($p{\geq}0.16$) by corn processing method. Compared with dry rolled corn, steam-flaked corn-based diets resulted in decreased acetate:propionate molar ratio (p = 0.02). It is concluded that at 7% or 14% straw inclusion rate, changes in physical characteristics of wheat straw brought about by pelleting negatively impact OM digestion of both steam-flaked and dry-rolled corn-based finishing diets. This effect is due to decreased post-ruminal starch digestion. Replacement of ground straw with pelleted straw also may decrease ruminal pH.

가정용 소형 퇴비화용기에 의한 부엌쓰레기의 분산식 퇴비화 I. 실험실 조건에서 퇴비화 연구 (Decentralized Composting of Garbage in a Small Composter for Dwelling House I. Laboratory Composting of the Household Garbage in a Small Bin)

  • 주택용 소형 퇴비화용기에 의한 부엌쓰레기의 퇴비화 가능성을 검토하기 위하여 두가지 용기(Type 1과 Type 2)를 제작하여 실험실 실험을 통하여 조사하였다. 두 용기의 구조는 같으나 차이점은 Type 1은 보온을 하였고 Type 2는 보온을 하지 않았다. 겨울철 실험을 통하여 Type 2는 우리나라의 기후 여건에 사용이 불가능한 것으로 판단되어 봄철 및 여름철 실험에서 제외하였다. 따라서 Type 1에 대한 계절 실험결과를 요약하면 다음과 같다. 1) 퇴비화기간 중 상승한 최고온도는 봄철 $58^{\circ}C$, 여름철 $57^{\circ}C$, 겨울철 $41^{\circ}C$였다. 따라서 우리나라의 기후조건에서는 반드시 보온이 필요하다는 것이 확인되었다. 2) 퇴비화원료물질의 무게는 8주 후 평균 62.5% 그리고 부피는 평균 74% 감소하였다. 3) 퇴비충의 밀도는 봄철 0.7kg/l, 여름철 0.8kg/l, 겨울철 1.1kg/l였다. 4) 수분함량은 전 퇴비화기간동안 큰 변동이 없었으며 8주 후 봄철 75.6%, 여름철 76.6%, 겨울철 76.6%였다. 5) pH는 계절에 따라 큰 차이를 보였으며 여름철에 가장 높았고, 겨울철에 가장 낮았다. 8주 후 봄철 6.13, 여름철 8.62, 겨울철 4.75였다. 6) 퇴비화시간의 경과에 따른 회분 및 유기물 함량은 유기물 분해속도가 빠를수록 증감현상이 뚜렷하였고 cellulose 및 lignin은 증가, hemicellulose 함량은 감소하였다. 7) 질소함량은 3.1-5.6%로 높았으며, 특히 여름철에 가장 높았다. 암모늄태 질소는 전반기에 증가했다가 감소하는 경향이었으며 겨울철 2주째 3,243mg/kg, 봄철 3주째 6,052mg/kg, 여름철 6주째 30,828mg/kg으로 가장 높았다. C/N율은 퇴비화시간이 경과함에 따라 감소하였으나 차이는 크지 않았다. 그러나 질산화는 봄 및 여름철에만 일어나고 겨울철에는 거의 일어나지 않았다. 8) 휘발성 및 고급유기산함량은 초기에 증가했다가 계절적으로 시기적 차이는 있으나 감소하였다. 총 유기산의 최고농도는 겨울철 2주째 10.1%, 봄철 2주째 5.8%, 여름철 4주째 15.7%였다. 9) 퇴비의 부숙도와 무관하게 각 무기성분함량은 $P_2O_5$ 0.9-4.4%, $K_2O$ 1.6-2.9%, CaO 2.4-4.6%, MgO 0.30-0.80%였다. 10) CN 및 각종 중금속함량도 퇴비화시간에 따라 큰 변화가 없었으며 각 함량범위는 계절 구분없이 CN 0.11-28.89mg/kg, Zn 24-l30mg/kg, Cu 5-219mg/kg, Cd 0.8-14.3mg/kg, Pb 7-42mg/kg, Cr ND-30mg/kg, Hg $ND-132.16\;{\mu}g/kg$이었다.

Effect of Lactobacillus mucosae on In vitro Rumen Fermentation Characteristics of Dried Brewers Grain, Methane Production and Bacterial Diversity

  • The effects of Lactobacillus mucosae (L. mucosae), a potential direct fed microbial previously isolated from the rumen of Korean native goat, on the rumen fermentation profile of brewers grain were evaluated. Fermentation was conducted in serum bottles each containing 1% dry matter (DM) of the test substrate and either no L. mucosae (control), 1% 24 h broth culture of L. mucosae (T1), or 1% inoculation with the cell-free culture supernatant (T2). Each serum bottle was filled anaerobically with 100 mL of buffered rumen fluid and sealed prior to incubation for 0, 6, 12, 24, and 48 h from which fermentation parameters were monitored and the microbial diversity was evaluated. The results revealed that T1 had higher total gas production (65.00 mL) than the control (61.33 mL) and T2 (62.00 mL) (p<0.05) at 48 h. Consequently, T1 had significantly lower pH values (p<0.05) than the other groups at 48 h. Ammonia nitrogen ($NH_3$-N), individual and total volatile fatty acids (VFA) concentration and acetate:propionate ratio were higher in T1 and T2 than the control, but T1 and T2 were comparable for these parameters. Total methane ($CH_4$) production and carbon dioxide ($CO_2$) were highest in T1. The percent DM and organic matter digestibilities were comparable between all groups at all times of incubation. The total bacterial population was significantly higher in T1 (p<0.05) at 24 h, but then decreased to levels comparable to the control and T2 at 48 h. The denaturing gradient gel electrophoresis profile of the total bacterial 16s rRNA showed higher similarity between T1 and T2 at 24 h and between the control and T1 at 48 h. Overall, these results suggest that addition of L. mucosae and cell-free supernatant during the in vitro fermentation of dried brewers grain increases the VFA production, but has no effect on digestibility. The addition of L. mucosae can also increase the total bacterial population, but has no significant effect on the total microbial diversity. However, inoculation of the bacterium may increase $CH_4$ and $CO_2$ in vitro.

Effect of Carbohydrate Sources and Levels of Cotton Seed Meal in Concentrate on Feed Intake, Nutrient Digestibility, Rumen Fermentation and Microbial Protein Synthesis in Young Dairy Bulls

  • The objective of this study was to investigate the effect of levels of cottonseed meal with various carbohydrate sources in concentrate on feed intake, nutrient digestibility, rumen fermentation and microbial protein synthesis in dairy bulls. Four, 6 months old dairy bulls were randomly assigned to receive four dietary treatments according to a $2{\times}2$ factorial arrangement in a $4{\times}4$ Latin square design. Factor A was carbohydrate source; cassava chip (CC) and cassava chip+rice bran in the ratio of 3:1 (CR3:1), and factor B was cotton seed meal levels in the concentrate; 109 g CP/kg (LCM) and 328 g CP/kg (HCM) at similar overall CP levels (490 g CP/kg). Bulls received urea-lime treated rice straw ad libitum and were supplemented with 10 g of concentrate/kg BW. It was found that carbohydrate source and level of cotton seed meal did not have significant effects on ruminal pH, ammonia nitrogen concentration, microbial protein synthesis or feed intake. Animals which received CC showed significantly higher BUN concentration, ruminal propionic acid and butyric acid proportions, while dry matter, organic matter digestibility, populations of total viable bacteria and proteolytic bacteria were lower than those in the CR3:1 treatment. The concentration of total volatile fatty acids was higher in HCM than LCM treatments, while the concentration of butyric acid was higher in LCM than HCM treatments. The population of proteolytic bacteria with the LCM treatments was higher than the HCM treatments; however other bacteria groups were similar among the different levels of cotton seed meal. Bulls which received LCM had higher protein digestibility than those receiving HCM. Therefore, using high levels of cassava chip and cotton seed meal might positively impact on energy and nitrogen balance for the microbial population in the rumen of the young dairy bull.

Effects of the slaughter weight of non-lean finishing pigs on their carcass characteristics and meat quality

  • The present study aimed to assess the feasibility of increasing the slaughter weight (SW) of non-lean finishing pigs to improve their meat quality. A total of 36 (Landrace × Yorkshire) × Duroc gilts and barrows were slaughtered at 115 (Av), 125 (Hi), or 135 (XHi) kg, followed by physicochemical analyses and sensory evaluation on their longissimus dorsi (LD) and Semimembranosus (SM) muscles. Backfat thickness was greater (p < 0.05) for the XHi (31.2 mm) and Hi (29.3 mm) groups than for Av (25.0 mm). Dressing percentage and yield of the belly per whole carcass were also slightly greater for XHi and Hi vs. Av. The intramuscular fat (IMF) content of SM was greater for XHi (2.64%) than for Av (1.83%) and Hi (2.04%) and also was correlated with SW (r = 0.55). The pH value, lightness, redness, drip loss, shear force, and moisture and protein contents of LD and SM, as well as IMF content of LD, were unaffected by SW. Percentages of 14:0, 16:0, and total saturated fatty acids (FA) were less for Hi and XHi vs. Av in SM, those of total unsaturated FA, 18:2, 20:4, and n-6 being opposite; FA composition of LM was not influenced by SW except for a reduced 18:0 percentage for XHi vs. Av. The sensory score was less for XHi vs. Av for odor in fresh LD and SM, and less for Hi and XHi vs. Av for aroma in fresh LM; scores for color, drip loss, marbling, and acceptability were unaffected by SW. As for cooked muscles, none of the scores for color, aroma, flavor, juiciness, tenderness, and acceptability was affected by SW, except for a greater LD color score for Hi and XHi vs. Av. Collectively, the results suggested that the increased yield of the carcass and belly due to increased SW is outbalanced negatively by excessive backfat deposition in production efficiency, whereas the SW increase exerts little influence on overall sensory quality of fresh or cooked meat. Production of non-lean market pigs overweighing 115 kg therefore will be uneconomical unless consumers pay a substantial premium for the over-fattened pork.

  • While searching for the bacteria which are responsible for degradation of pesticide in soybean field soil, a novel bacterial strain, designated 5-5T, was isolated. The cells of the strain were Gram-staining-positive, aerobic and non-motile rods. Growth occurred at 10-42℃ (optimum, 30℃), pH 5.5-9.0 (optimum, pH 7.0-7.5), and 0-2% (w/v) NaCl (optimum, 1%). The predominant fatty acids were C15:0 anteiso, C17:0 anteiso, and summed feature 8 (C18:1 ω7c and/or C18:1 ω6c). The predominant menaquinone was MK-9 (H2). Diphosphatidylglycerol, glycolipids, phosphatidylinositol, and phosphatidylglycerol were the major polar lipids. Phylogenetic analysis of 16S rRNA gene sequences indicated that strain 5-5T is a member of the genus Sinomonas and its closest relative is Sinomonas humi MUSC 117T, sharing a genetic similarity of 98.4%. The draft genome of strain 5-5T was 4,727,205 bp long with an N50 contig of 4,464,284 bp. Genomic DNA G+C content of strain 5-5T was68.0 mol%. The average nucleotide identity (ANI) values between strain 5-5T and its closest strains S. humi MUSC 117T and S. susongensis A31T were 87.0, and 84.3 % respectively. In silico DNA-DNA hybridization values between strain 5-5T and its closest strains S. humi MUSC 117T and S. susongensis A31T were 32.5% and 27.9% respectively. Based on the ANI and in silico DNA-DNA hybridization analyses, the 5-5T strain was considered as novel species belonging to the genus Sinomonas. On the basis of the results from phenotypic, genotypic and chemotaxonomic analyses, strain 5-5T represents a novel speciesof the genus Sinomonas, for which the name Sinomonas terrae sp. nov. is proposed. The type strain is 5-5T (=KCTC 49650T =NBRC 115790T).
