Optimal Enrichment Temperature, Time and Materials for L-type Rotifer (Brachionus plicatilis) Cultured at a Low Temperature (저온 배양한 L-type 로티퍼(Brachionus plicatilis)의 적정 영양강화 수온, 시간 및 영양강화제 종류)

  • This study was undertaken to improve the survival and early life growth rates of cold-water fish by culturing rotifer (Brachionus plicatilis) with low-temperature tolerance. The enrichment experiment was carried out at different temperatures and over different time intervals. Cultivation of the rotifer at low temperatures was repeated, with the selected and cultured as the water temperature was gradually lowered from $20^{\circ}C$ to $10^{\circ}C$. Enrichment of the rotifer was completed using A, S, SCV and SCP. Enrichment was carried out after 6, 12 and 24 hours at three different temperatures (10, 15 and $20^{\circ}C$). In the growth experiments, the rotifer increased to approximately triple their original size, from $350{\pm}7.9ind./ml$ to $1,064{\pm}5.7ind./ml$ at $10^{\circ}C$ over 50 days. The fatty acid composition of the four enrichment materials was species-specific, with the highest ratios belonging to eicosapentaenoic acid (EPA, C20:5n-3) and docosahezaenoic acid (DHA, C22:6n-3) in SCP. The fatty acid composition of the rotifers was affected by the enrichment materials. The EPA (% of total fatty acid) was more than 2 % in SCP, which showed a higher ratio than the other enrichment materials. DHA was higher in S reaching 12.40 % at $15^{\circ}C$ for 24 hours. The highest levels of EPA (3.09 %) and DHA (11.65 %) were obtained after the rotifers were enriched with S at $20^{\circ}C$ for 12hours.

Development of A Network loading model for Dynamic traffic Assignment (동적 통행배정모형을 위한 교통류 부하모형의 개발)

  • For the purpose of preciously describing real time traffic pattern in urban road network, dynamic network loading(DNL) models able to simulate traffic behavior are required. A number of different methods are available, including macroscopic, microscopic dynamic network models, as well as analytical model. Equivalency minimization problem and Variation inequality problem are the analytical models, which include explicit mathematical travel cost function for describing traffic behaviors on the network. While microscopic simulation models move vehicles according to behavioral car-following and cell-transmission. However, DNL models embedding such travel time function have some limitations ; analytical model has lacking of describing traffic characteristics such as relations between flow and speed, between speed and density Microscopic simulation models are the most detailed and realistic, but they are difficult to calibrate and may not be the most practical tools for large-scale networks. To cope with such problems, this paper develops a new DNL model appropriate for dynamic traffic assignment(DTA), The model is combined with vertical queue model representing vehicles as vertical queues at the end of links. In order to compare and to assess the model, we use a contrived example network. From the numerical results, we found that the DNL model presented in the paper were able to describe traffic characteristics with reasonable amount of computing time. The model also showed good relationship between travel time and traffic flow and expressed the feature of backward turn at near capacity.

Changes in Feeding Habit and Body Composition of Black Sea Bream Acanthopagrus schlegeli Released in Eelgrass Zostera marina Bed (잘피밭 해역에 방류된 감성돔 Acanthopagrus schlegeli 치어의 식성 및 어체성분 변화)

  • Artificially-produced juvenile black sea bream Acanthopagrus schlegeli (total length $8.3{\pm}1.0\;cm$, body weight $11.2{\pm}4.2\;g$) were released in eelgrass, Zostera marina bed and their initial food organism and changes of body compositions were investigated for one month after release. Predation rates in fish sampled on 1st and 3rd days were 79%, increased up to 100% on 7th and 10th days, and then decreased on 30th days as 75%. Major prey organisms was composed of mainly Amphipoda and Gastropoda. Ratio of Amphipoda and Gastropoda in stomach were highest in fish sampled on 15th and 1st days after release, respectively. Crustacea and Algae were maintained about 20% during a sampling period. Visceral weight index (VWI) offish sampled 20th and 30th after release were significantly higher than that of initial. Carcass crude protein and lipid contents of released fish were showed significantly decreasing; however carcass n-3 HUFA composition was showed increasing tendency with the passage of time after release. Eelgrass bed was supposed to be helpful for the released fish to adjust their feeding habits and biochemical metabolism to the natural environment within a short period after release.

Relative Effectiveness of Replacement Diets for Mass Production of the Brackish Water Flea Diaphanosoma celebensis (기수산 물벼룩(Diaphanosoma celebensis)의 대량생산을 위한 대체 먹이원의 효과)

  • This study examined the effect of formulated diets (composed of condensed freshwater $Chlorella$ or $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil sources derived from baker's yeast) on the population growth and fatty acid composition of the brackish water flea ($Diaphanosoma$ $celebensis$) to determine diet effectiveness for mass production. Results indicated that the highest maximum density, fecundity, and population specific growth rate (SGR) of the flea were observed using a diet of $Tetraselmis$ $suecica$ (control). However, results using condensed freshwater $Chlorella$ and the mixed treatment of $Schizochytrium$ sp.+$Crypthecodinium$ sp.+EPA oil did not significantly differ from the control treatment ($P$>0.05). Thus, $T.$ $suecica$ could be replaced with a combination diet of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil in condensed freshwater $Chlorella$ without growth interruption of $D.$ $celebensis$. As the percentage of combination diet (mix of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil) in condensed freshwater $Chlorella$ increased, the level of ARA, EPA, DHA, and n-3 HUFA in $D.$ $celebensis$ also increased. However, the population density of $D.$ $celebensis$ sharply decreased with the use of treated supplements C5M5 (CHL [50%] and $Crypthecodinium$ sp.+ $Schizochytrium$ sp. + EPA oil [MIX 50%]). The optimum proportions of CHL and MIX for $D.$ $celebensis$ within the mixture were 70% and 30%, respectively. Based on the results, the combination of $Schizochytrium$ sp., $Crypthecodinium$ sp., and EPA oil in condensed freshwater $Chlorella$ appears to be a stable and effective food source for mass production of $D.$ $celebensis$.

Nutrient Composition of Benthic Diatom Cultured on Plate at Four Areas in Spring (봄철 해역별 파판배양 부착규조류의 영양성분)

  • 봄철 해역에 따른 부착미세조류의 영양성분 변화를 알아보기 위하여 본 실험을 수행하였다. 실험은 울진, 부안, 완도, 부산 지역에서 봄철인 2~4월 동안 실시되었으며 실험간의 오차를 줄이기 위해 실험 시작일의 차이는 최대 10일 이내로 하였다. 배양을 위한 실험해역의 기초 조사로 배양지역 연안해수의 수온, 염분, pH, 총질소, 총인, Chlorophyll-a의 측정을 하였고 각 지역의 부착미세조류를 수확하여 영양분석을 하였다. 실험에 사용된 파판은 전복 종묘생산에 이용되는 규조배양용 파판과 틀을 이용하였고 실험기간은 총 8주 동안 실시하였다. 2주마다 파판을 수거한 뒤 부착된 미세조류를 부드러운 솔로 분리하여 원심 분리한 후 지방산과 아미노산의 측정을 하였다. 또 각 시료의 일정량을 Sedwick-rafter counting chamber를 이용하여 검경ㆍ계수한 후 이를 단위면적당 개체수(cells/$\textrm{cm}^2$)로 환산하여 biomass를 구하였다. 봄철 지역간의 수온의 범위는 13.9~16.6$^{\circ}C$를 보였으며 울진이 가장 높고 부안지역이 가장 낮았다. 염분은 부안지역의 염분이 평균 2$\textperthousand$ 이상 낮았으며 그 외의 지역은 큰 차이가 없었다. pH는 7.77~8.21의 범위를 보이고 있었으며 질산염 농토는 부산이 0.265mg/$\ell$로 최대값을 보였고 울진이 0.032mg/$\ell$로 제일 낮았다. 인산염 농도는 완도지역이 0.018mg/$\ell$로 최대치를 보였고 부안은 검출되지 않았다. Chlorophyll-a의 농도는 울진지역이 0.66mg/$\ell$으로 제일 낮았으며 부산이 2.69mg/$\ell$, 부안지역이 2.27mg/$\ell$ 로 높은 값을 보였다. 봄철 지역에 따른 부착미세조류의 아미노산 분석결과는 울진이 실험지역 중 제일 높은 총 아미노산과 필수아미노산의 함량을 나타냈으며 각 지역별로 울진은 4주에 20.3%, 부안은 8주째 8.1%, 완도는 8주째 4.7%, 부산은 6주째 10.4%로 서로다른 시기에 높은 아미노산의 함량을 나타냈다. 실험지역 부착미세조류의 지방산 결과를 보면 전 실험기간동안 지방산 중 대부분은 $C_{18}$이하의 포화 또는 불포화 지방산이 주를 이루고 있었으며 palmitic acid( $C_{16:0}$)와 palmitoleic acid( $C_{16:0}$)의 양이 전체 지방산 함량중 37.47~75.94%로 대부분을 차지하고 있었다. 그리고 n-3HUFA(high unsaturated fat쇼 acids)인 EPA는 부안지역이 2주째 20.85%로 전체 지역중 함량이 제일 높았고, 울진은 실험기간중 6주가 14.76%로 제일 높은 함량을 보였다. 완도는 6주에만 10.97%의 함량을 보였으며 부산지역의 시료에서는 EPA의 검출이 없었다. 봄철 각지역에 설치된 파판에서의 부착미세조류 출현량을 보면 2주째의 부착미세조류량은 부안이 8.3$\times$$10^3$ cells/$\textrm{cm}^2$로 가장 높았고 부산이 0.6$\times$$10^3$ cells/$\textrm{cm}^2$로 가장 낮았다. 울진의 경우 0.7$\times$$10^3$ cells/$\textrm{cm}^2$로 부산과 비슷하게 낮은 출현량을 보였고 완도는 2.4$\times$$10^3$ cells/$\textrm{cm}^2$였다. 4주째의 출현량은 울진지역이 471.1$\times$$10^3$ cells/$\textrm{cm}^2$로 가장 높았으며 부안, 완도, 부산 순으로 출현량이 낮았고 특히 부산은 1.2$\times$$10^3$ cells/$\textrm{cm}^2$로 가장 낮았다. 6주째의 출현량은 울진 지역이 368.6$\times$$10^3$ cells/$\textrm{cm}^2$로 가장 높았으며 부안 89.8$\times$$10^3$ cells/$\textrm{cm}^2$, 완도 25.1$\times$$10^3$ cells/$\textrm{cm}^2$, 부산 5.9$\times$$10^3$ cells/$\textrm{cm}^2$ 순이었다. 8주째의 경우에도 울진, 부안, 완도, 부산이 각각 255.8$\times$$10^3$ cells/$\textrm{cm}^2$, 157.8$\times$$10^3$ cells/$\textrm{cm}^2$, 44.3$\times$$10^3$ cells/$\textrm{cm}^2$, 1.1$\times$$10^3$ cells/$\textrm{cm}^2$로 4주, 6주의 경우와 같은 경향을 보였다.보였다.

Essentiality of Dietary Eicosapentaenoic Acid and Docosahexaenoic Acid in Korean Rockfish, Sebastes schlegeli (조피볼락(Sebastes schlegeli) 사료의 EPA 및 DHA 필수성)

  • Feeding trials were conducted to determine the essentiality of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), and to compare the efficiency between EPA and DHA for juvenile Korean rockfish. Fish averaging 2.1 g were fed experimental diets containing different levels of EPA or DHA, and different combinations of EPA and DHA in two separate experiments. Graded levels ($0.0{\sim}l.75\%$) of dietary EPA or DHA as ethyl esters were substituted for a part of the $8\%$ beef tallow in the basal diet. After 5 weeks of the experimental period, weight gain, chemical composition of whole body, protein and lipid retention efficiency, hepatosomatic index, and fatty acid composition of liver were measured. Daily growth rate and feed efficiency were the lowest in fish fed the diets without EPA arid DHA. These responses were effectively improved by increasing EPA or DHA in the diets up to $1.0\%$ levels, and then reached a plateau between 1.0 and $1.75\%$ levels of either EPA or DHA. Protein and lipid retention efficiency were also improved with the high levels of dietary EPA or DHA. DHA was superior to EPA at the same level of each in weight gain, feed efficiency, and protein and lipid retention efficiency. Hepatosomatic index tended to decrease with increase of the dietary EPA or DHA levels. Lipid contents of whole body were increased with levels of EPA or DHA in the diets. Dietary EPA and/or DHA levels affected directly the fatty acid composition of liver polar lipid. EPA or DHA in the liver polar lipid were increased with levels of dietary EPA or DHA, respectively, whereas those in nonpolar lipid were not affected by the dietary levels of EPA and/or DHA. These finding indicate that either of the EPA or DHA is essential for normal growth of Korean rockfish, and the essential fatty acid requirement is $1.0\%$ of EPA and/or DHA in the diet. DHA is superior to EPA as essential fatty acid, and the dietary EPA/DHA ratio of less than 1.0 may be adequate for normal growth of Korean rockfish fed a diet enough n-3HUFA (EPA and DHA).

Effects of Dietary Probiotics Supplementation on Juvenile Korean Rockfish Sebastes schlegeli (치어기 조피볼락 Sebastes schlegeli 사료내 생균제 첨가효과)

  • A 12-week feeding trial was conducted to investigate the effects of dietary supplementation of probiotics as a feed additive for juvenile Korean rockfish Sebastes schlegeli. Four experimental diets supplemented with no probiotic(Control), Bacillus polyfermenticus(BP), Bacillus licheniformis(BL) or Bacillus polyfermenticus plus Saccharomyces cerevisiae(BP+SC) at $1.0{\times}10^7$ CFU/kg diet as a dry-mater(DM) basis were prepared by mixing with a basal diet. After 12 weeks of the feeding trial, fish fed BP+SC diet showed significantly higher weight gain(WG), feed efficiency(FE), specific growth rate(SGR) and protein efficiency ratio(PER) than those of fish fed control diet(P<0.05), however there were no significant differences in WG, FE, SGR and PER among fish fed the BP, BL and BP+SC diets. Fish fed BP and BP+SC diets showed significantly higher condition factor(CF) than that of fish fed control and BL diets. Fish fed BP, BL, BP+SC diets showed significantly higher hepatosomatic index(HSI) than that of fish fed control diet, however there was no significant difference in HSI among fish fed BP, BL and BP+SC diets. Fish fed BP+SC diet showed significantly lower serum glucose than that of fish fed control diet, however there was no significant difference in serum glucose among fish fed BP, BL and BP+SC diets. Fish fed BP+SC diet showed significantly higher respiratory burst activity(NBT assay) than that of the fish fed control and BL diets, however there was no significant difference in NBT assay between fish fed BP and BP+SC diets. Fish fed BP and BL diets showed significantly higher lysozyme activity than that of the fish fed control diet, however there was no significant difference in lysozyme activity among fish fed BP, BL and BP+SC diets. Fish fed BP and BP+SC diets showed significantly lower cumulative mortality than that of the fish fed control diet, however there was no significant difference in cumulative mortality among fish fed BP, BL and BP+SC diets after the challenge test. From these results, dietary B. polyfermenticus, B. licheniformis and B. polyfermenticus plus S. cerevisiae supplementation in juvenile Korean rockfish diet could enhance growth performances, non-speicific immunities and a higher resistance against the specific pathogen.
