Association of SNP Haplotypes at the Myostatin Gene with Muscular Hypertrophy in Sheep

  • The myostatin gene of seven important meat (Beltex (Australia), Beltex$\times$Huyang (F1), Meat and Multi-Prolific Chinese Merino Fine Wool, Meat Chinese Merino Fine Wool and Dorper (South Africa)) and non-meat (Huyang and Kazak) sheep breeds was analyzed to study the genetic basis of muscular hypertrophy (double muscling) phenotype in sheep. SNPs, four in regulatory regions and several in the introns in the myostatin gene, were identified, and the former four SNPs were used for further studies. Twelve haplotypes were predicted by PHASE program, of which four main haplotypes (1, 3, 7, 9) were present in 90% of the 364 sheep in the study. Haplotypes 1-4 were mainly present in meat breeds while haplotypes 7 and 9 dominated the non-meat breeds. The association between haplotypes and average daily gain (ADG) was analyzed among 116 sheep with production data, Haplo2 (CGAA) and Haplo8 (TGAA) were identified to have significant (p<0.05) effect on ADG by the model (JMP5.1 software) taking into account the effects of breed, family background, haplotype, birth weight and sex. ADG of these haplotype groups also correlated well (r = 0.82) with hypertrophic phenotype scores. In conclusion, the mutations -956 (T$\rightarrow$C), -41 (C$\rightarrow$A) and 6223 (G$\rightarrow$A) involved in Haplo2 and 8 may be associated with the double-muscling trait by influencing myostatin function and be suitable markers in selecting meat sheep.

Effects of Castration on Androgen Receptor, IGF-I Ea, MGF and Myostatin Gene Expression in Skeletal Muscles of Male Pigs

  • Castration of male pig produces significant negative effects on skeletal muscle development. The androgen receptor (AR), two splice variants of insulin-like growth factor-I (IGF-I Ea and MGF) and the myostatin gene may play important roles in this process. In the present study, the expression of AR, IGF-I Ea, MGF and myostatin genes in three skeletal muscles, the brachialis, longissimus and semitendinosus, were studied using real-time quantitative RT-PCR. Our experimental design used 14 pairs of male Landrace sire${\times}$Yorkshire dam piglets. The two piglets in each pair were full sibs, one of which was castrated at 21 d of age; the other remained intact. The study group was divided into subgroups of equal size. Animals in the first subgroup were slaughtered at 147 d and those of the second at 210 d of age. Carcass weight and lean meat yield were similar between boars and barrows at 147 d of age (p>0.05), whereas barrows had lower carcass weight and less lean meat yield at 210 d of age (p<0.05). Castration caused down-regulation of AR gene expression at both 147 and 210 d of age (p<0.05). The two splice variants of the IGF-I gene from porcine skeletal muscle were cloned using RT-PCR, and it was found that MGF differs from IGF-I Ea in having a 52-base insert in the last coding exon of the mRNA. Both splice variants were down-regulated by castration only at 210 d of age (p<0.05). No differences in expression of the myostatin gene were observed between boars and barrows at either 147 or 210 d of age (p>0.05). These results suggest that the downregulation of AR, IGF-I Ea and MGF gene expression following castration helps to explain the negative effect of castration on skeletal muscle development.

Postnatal Expression of Growth/Differentiation Factor-8 (GDF-8) Gene in European and Asian Pigs

  • Myostatin (growth differentiation factor (GDF)-8), is one member of the transforming growth factor $\beta$ superfamily. Investigations of GDF-8 null mice and double-muscled cattle revealed that GDF-8 has a profound influence upon skeletal muscle growth. Therefore, the GDF-8 effect upon the productive performance of pigs is worth exploring. In the present study, the nucleotide sequences and expression levels of GDF-8 genes in European pigs (Landrace and Duroc) and Asian pigs (Taoyuan and Small-ear) were evaluated. Based upon their genetic background these breeds possess significantly distinct growth rate and muscle productionphenotypes. Our sequence data showed that the nucleotide sequences of European and Asian pigs were 100% similar. Postnatal expression of GDF-8 gene in skeletal muscles, from birth to 12 mo of age, among different breeds was measured. GDF-8 expression levels in the longissimus muscle of neonatal European breed littermates were the highest, however it declined significantly (p<0.05) at 1 and 3 mo, and then increased gradually at 6 to 12 mo. The Asian breeds, however, GDF-8 expression level increased markedly at 3 mo and maintained a constant level thereafter. The results indicate that rather than polymorphism within the GDF-8 functional sequence between European and Asia breeds, it was relative to the gene regulation in postnatal muscle growth.

Gene Expression in the Muscles of young and Mature Channel Catfish (Ictalurus punctatus) as Analyzed by Expressed Sequence Tags and Gene Filters

  • To generate expressed sequence tags for genomics research involving genetic linkage analysis, to examine gene expression profiles in muscles of channel catfish in a non-normalized muscle cDNA library, to compare gene expression in young and mature channel catfish muscles using the EST reagents and gene filters to demonstrate the feasibility of functional genomics research in small laboratories. 102 randomly picked cDNA clones were analyzed from the catfish muscle cDNA library. Of the sequences generated, 90.2% of ESTs was identified as known genes by identity comparisons. These 92 clones of known gene products represent transcriptional products of 24 genes. The 10 clones of unknown gene products represent 8 genes. The major transcripts (70.1% of the analyzed ESTs) in the catfish muscle are from many major genes involved in muscle contraction, relaxation, energy metabolism and calcium binding such as alpha actin, creatine kinase, parvalbumin, myosin, troponins, and tropomyosins. Gene expression of the unique ESTs was comparatively studied in the young and adult catfish muscles. Significant differences were observed for aldolase, myostatin, myosin light chain, parvalbumin, and an unknown gene. While myosin light chain and an unknown gene (CM 192) are down-regulated in the mature fish muscle, the aldolase, myostatin, and parvalbumin are significantly up-regulated in the mature fish muscle. Although the physiological significance of the changes in expression levels needs to be further addressed, this research demonstrates the feasibility and power of functional genomics in channel catfish. Channel catfish muscle gene expression profiles provide a valuable molecular muscle physiology blueprint for functional comparative genomics.

Association between expression levels and growth trait-related SNPs located in promoters of the MC4R and MSTN genes in Spinibarbus hollandi

  • Melanocortin 4 receptor: (MC4R) and Myostatin (MSTN) are two important growth trait-related genes in animals. In this study, we showed that two SNPs, MC4R-719A>G and MSTN-519C>T, found in the promoters of the MC4R and MSTN genes, respectively, are both associated with growth traits in Spinibarbus hollandi. Furthermore, we observed that there were significant associations between the expression levels of the MC4R and MSTN genes and these two growth trait-related SNPs. The expression level of MC4R gene in brain was lower in GG genotype fish with extremely high growth performance than that in AA genotype fish with extremely low growth performance. Expression level of the MSTN gene in muscle was lower in TT genotype fish with extremely high growth performance than that in CC and CT genotype fish with lower growth performance. The results indicated that these SNPs located in the promoters of MC4R and MSTN are associated with growth-related traits through modification of gene expression levels. The MSTN and MC4R SNPs may have useful application in effective marker-assisted selection aimed to increase output in S. hollandi.

고리매(Scytosiphon lomentaria) 에탄올 추출물이 마이오스타틴 활성과 고 급식으로 유도된 비만 제브라피쉬에 미치는 영향 (Effect of Scytosiphon lomentaria Ethanol Extracts on Myostatin Activity and Zebrafish Obesity Induced by High Feeding)

  • 생활습관 개선을 통한 근육량향상은 대사 증후군의 위험을 줄이는 것으로 보고되었다. 본 연구에서는 갈조류인 고리매(Scytosiphon lomentaria) 에탄올 추출물이 골격근 성장억제조절 단백질인 마이오스타틴 신호전달을 억제하는지와 고 급식으로 유도된 비만 제브라피시의 대사 항상성에 대한 효과를 확인하였다. 고리매 에탄올 추출물(10 ㎍/ml)은 pGL3- (CAGA) 12-루시퍼라제 분석에서 마이오스타틴(1 nM/ml) 신호를 완전히 차단하였다. 또한 웨스턴 블롯 분석에서 마이오스타틴 신호를 차단하여 Smad2 인산화가 억제되는 것을 확인하였다. 제브라피쉬의 치어에 대한 연구는 고 급식 대조군 그룹의 체내 포도당 농도는 정상 급식 대조군 그룹보다 유의하게 높았지만, 12.5 ug의 고리매 에탄올을 처리한 고 급식 그룹과 18.75 ug의 고리매 에탄올로 처리한 고 급식 그룹의 체내 포도당 수준은 정상 급식 대조군 그룹과 유사하였다. 고 급식 그룹의 GLUT2 유전자의 mRNA 발현 수준은 정상 급식 대조군 그룹에 비해 현저히 낮았다. 하지만, 고리매 에탄올 추출물을 처리한 실험군 그룹의 GLUT2 유전자의 발현은 정상 급식 대조군 그룹의 GLUT2 유전자의 발현과 거의 유사하게 나타났다. 그러므로 고리매 에탄올 추출물은 GLUT2유전자의 발현 조절을 통하여 체내 포도당 조절이 가능함을 보여준다. 본 연구의 결과는 고리매 에탄올 추출물이 대사 증후군 치료에 도움을 주는 소재 및 마이오스타틴 억제제로서의 가능성을 시사합니다.

Effects of maternal dietary energy restriction on breast muscle fibre development in the offspring of broiler breeders

  • Objective: The effects of maternal dietary energy levels on breast muscle fibre development in offspring of broiler breeders were investigated. Methods: A total of 480 20-week-old Arbor Acres (AA) healthy female broiler breeders, with an average body weight of 2.33±0.01 kg, were randomly divided into 4 treatment groups with 6 replicates and 20 broiler breeders for each replicate and fed a corn and soybean meal diet with 100%, 80%, 70%, and 50% energy levels, respectively. Approximately 300 eggs per treatment were collected for incubation for 6 days. Then, 120 0-day-old female broilers at each energy level were randomly selected and divided into 6 replicates with 20 broilers for each replicate, with this experimental phase with the offspring lasting for 49 days. Results: Compared with the 100% energy group, the breast muscle fibre diameter at embryonic day 21 in the 80% energy group was significantly reduced (p<0.05). In the 80% energy group, the muscle fibre density of the breast increased significantly (p<0.05) at embryonic days 15 and 21. The breast muscle fibre diameter of the offspring in each group was significantly decreased (p<0.05) on the 1st day. The breast muscle sarcomere length of the embryos in the 80% energy group was significantly higher (p<0.05) than those in the 70% and 50% energy groups. Compared with the 100% energy group, the expression of the myostatin gene in the offspring was significantly decreased (p<0.05). Conclusion: In conclusion, the effects of a maternal dietary energy level of 80% in this study were found to be optimal for breast muscle fibre development in offspring, which indicated that the metabolic energy level of AA broilers of 9.36 MJ/kg for the mid-term diet for laying eggs has a more practical significance.

제주마에서 Myostatin 유전자 변이 특성 구명 (Characterization of Myostatin Gene Variants in Jeju Horses)

  • 제주마는 한국의 말 품종으로 제주도에서 오랜 기간 자생하여 왔다. 제주마의 기원은 몽골마에서 유래한 것으로 추정되었다. 그러나 최근 연구에서 제주마와 몽골 토종마를 비교 한 결과, 유전적으로 가까우나 제주마가 몽고마와 독립적으로 진화해 온 것으로 보고되었다. 제주마는 경주마로 이용되어 경주 능력이 주요한 경제 형질로 활용되고 있다. Myostatin (MSTN) 유전자는 다양한 포유류에서 골격근량에 영향을 미치는 것으로 연구되었다. Thoroughbred에서 MSTN 유전자 내 존재하는 변이가 경주 능력 및 스태미너에 영향을 미치는 것으로 알려져 있다. 우리는 제주마 1,433두를 포함하여 여러 말 품종의 MSTN 유전자내 변이 빈도를 비교하였다. MSTN 유전자의 g.66493737 변이 중 장거리 적성 유전자형(TT)의 경우 제주마에서 빈도가 0.826으로 한라마(0.285) 및 Thoroughbred (0.252)보다 높은 것으로 확인되었다. 또한 다른 연구를 참고하여 8종 말에서 g.66493737 빈도를 비교하였다. 그 결과, 경주마로 활용되는 품종은 CC형의 빈도가 높았으나, TT형의 빈도는 낮은 것으로 확인되었다. 반대로 승용마 및 역용마 등의 품종은 TT형의 빈도가 높은 것으로 확인되었다. 제주마의 경주 거리(400 m, 800 m, 900 m, 1,000 m, 1,110 m, 1,200 m)별로 유전자형과 도착 기록을 비교 분석했다. 그 결과 1,000 m 이하의 경주에서는 CT형이 TT형보다 더 빠른 도달 기록을 보였다. 그러나 1,110 m 이상의 경주에서는 거의 동일한 결과가 확인되었다. 본 연구에서는 제주마의 MSTN 유전자 변이가 경주 거리 적성과 관련이 있을 수 있음을 시사하였다. 이러한 결과는 추후 연구를 통해 제주마에서 경주 거리 적성 및 경주 능력과 연관된 마커 개발을 위한 자료로 활용이 가능할 것으로 사료된다.

Biotechnologies for Improving Animal Metabolism and Growth - A Review

  • Biotechnology will play critical role in improving animal productivity. Animal growth rate and muscle deposition potential can be greatly improved by the application of biotechnology and biotechnological products. Administration of recombinant somatotropin (ST) or other compounds such as IGF-1 and growth hormone-releasing peptides (GHRPs) can enhance growth rate and carcass lean percentage. Gene transfer offers a powerful approach to manipulate endocrine system and metabolic pathways toward faster growth and better feed efficiency. Biotechnology is also extensively used for improving metabolism and activity of gut microorganisms for better nutrient digestibility. Knockout of growth-inhibiting genes such as myostatin results in considerable acceleration of body weight and muscle growth. Animal growth can also be improved by the use of gene therapy. Immunomodulation is another approach for efficient growth through controlling the activity of endogenous anabolic hormones. All the above aspects will be discussed in this review.

Gene-editing techniques and their applications in livestock and beyond

  • Genetic modification enables modification of target genes or genome structure in livestock and experimental animals. These technologies have not only advanced bioscience but also improved agricultural productivity. To introduce a foreign transgene, the piggyBac transposon element/transposase system could be used for production of transgenic animals and specific target protein-expressing animal cells. In addition, the clustered regularly interspaced short palindromic repeat-CRISPR associated protein 9 (CRISPR-Cas9) system have been utilized to generate chickens with knockout of G0/G1 switch gene 2 (G0S2) and myostatin, which are related to lipid deposition and muscle growth, respectively. These experimental chickens could be the invaluable genetic resources to investigate the regulatory pathways and mechanisms of improvement of economic traits such as fat quantity and growth. The gene-edited animals could also be applicable to the livestock industry.
