Myo-Inositol Synthesis in the Milk of Lactating Rats (쥐 우유중의 Myo-Inositol 생성에 관한 연구)

  • A high concentration of myo-Inositol in rat's milk was observed (61-91mg. of myo-Inositol per 100g of milk) by gas-liquid chromatographic method, using a 3% SE-52 column. Feeding experiments showed that approximately 85% of myo-Inositol in milk was from dietary origin: the rest was considered to be synthesized by 1L-myo-Inositol-1-phosphate lyase. Results suggested that the biosynthesis was not sufficiently high to permit the maintenance of its myo-Inositol level in milk. However, study $using(^{14}C)-glucose$ injection into lactating female rats confirmed biosynthesis of myo-Inositol from glucose in mammary gland. This biosynthesis reached a maximum within an hour after $(^{14}C)-glucose$ injection intraperitoneally as lactose biosynthesis did. Study using $(^3H)-myo-Inositol$ confirmed that most of the myo-Inositol in milk was transported from blood plasma myo-Inositol against a concentration gradient. About four hours after the beginning of the injection of $(^{14}C)-glucose$, the specific radioactivity of myo-Inositol in milk was 8% of that of glucose in the blood. When $(^3H)-myo-Inositol$ was injected, the specific radioactivity of myo-Inositol in milk was about 26% of that of blood six hours after injection.

Effect of Myo-Inositol on In Vitro Maturation of Porcine Oocytes (Myo-inositol이 돼지 난모세포의 체외성숙에 미치는 영향)

  • This study was carried out to assess whether the addition of myo-inositol to maturation medium could improve porcine oocyte maturation in vitro. Oocytes were cultured for the first 22 h in Witten's medium containing 10IU/$m\ell$ PMSG, 10 IU/$m\ell$ HCG supplemented with or without myo-inositol. Subsequently, they were cultured for additional 22 h in Witten's medium without hormone supplemented with or without myo-inositol. When the porcine oocytes were cultured in maturation medium containing myo-inositol, the proportion of metaphase II oocytes 44h after culture was higher in the myo-inositol group(P<0.05). To study effects of cumulus cell on the maturation induced by myo-inositol, we examined the maturation status of cumulus-enclosed or cumulus-denuded porcine follicular oocytes. The rates of maturation were significantly higher in the cumulus-enclosed oocytes(P<0.05). However, the maturation rates of cumulus-denuded oocytes cultured in medium containing myo-inositol were higher than those of control group(P<0.05). Our results suggest that myo-inositol may affect meiotic progression of porcine follicular oocytes and supplementation of myo-inositol in maturation medium may be useful for the in vitro maturation of porcine follicular oocytes.

Increase of the D-chiro-inositol and D-Pinitol Contents by Abiotic Stress in the Buckwheat Seedlings (쓴메밀 유식물에서 비 생물학적 스트레스에 의한 D-chiro-inositol과 D-pinitol의 함량 증가)

  • D-chiro-Inositol, which is the isomer of myo-inositol, is a well known drug for the treatment of type II diabetes. The methylated form of D-chiro-inositol, D-pinitol and D-chiro-inositol are synthesized when the plants are exposed to the abiotic stresses such as drought, salinity and low temperature as osmoprotectants. In soybean, myo-inositol is converted to ononitol by O-methyltransferase, and ononitol is converted to D-pinitol by ononitol epimerase and finally converted to D-chiro-inositol by demethylase. However there have been some reports that in buckwheat, myo-inositol can be converted to D-chiro-inositol directly. This study was conducted to determine the changes of soluble cyclitols in buckwheat seedlings after exposure to salt and drought stresses by GC-FID. The results indicated that myo-inositol may be the precursor of D-chiro-inositol biosynthesis.

Effects of molybdenum on myo-inositol uptake system in peripheral nerve isolated from lead-intoxicated rat. (Molybdenum이 납 중독 랫드의 말초신경내 myo-inositol uptake 시스템에 미치는 영향)

  • This study was carried out to elucidate the preventive mechanism of molybdenum on lead-induced neuropathy, An animal model of lead neuropathy was induced by feeding diet containing lead to Sprague-Dawley rat for three weeks. Four weeks aged Sprague-Dawley rats were divided into four groups : normal control group, 10ppm-lead treated group, 1mg/kg-molybdenum treated group, 10ppm-lead and 1mg/kg-molybdenum treated group. The parameters on neuropathy were examined by measuring concentration of myo-inositol and myo-inosito uptake in sciatic nerve. In the lead-treated rats, myo-inositol concentration and myo-inositol uptake rate were reduced by from 54% to 33% respectively. This deficit results from that myo-inositol uptake system which is carrier mediated and sodium-potassium dependent was inhibited by the lead treatment. However, the molybdenum administration significantly eliminated the impairment and maintained myo-inositol concentration to about 82% of normal level. These results suggest that lead-induced neurotoxicity was significantly reduced by administration of molybdenum and the mechanism might be partly normalization of myo-inositol uptake system in sciatic nerve.

  • In our previous studies, we reported that lead intoxicated nerve cell by inhibition of the Na$^{+}$-K$^{+}$ ATPase activity and reduction of myo-inositol in nerve cell. As the second series of experiments, in order to understand toxic mechanism of lead for nerve cell, the characteristics of myo-inositol transport system and the effect of lead on its system have been studied in the sciatic nerves of control and lead-treated rats. A lead intoxicated animal model was induced by feeding diet containing lead to Sprague-Dawley rat for two weeks. Four weeks aged Sprague-Dawley rats were divided into three group : normal control group, 10ppm-lead treated group, 100ppm-lead treated group. All rats were sacrified at the end of two weeks. The rate o myo-inositol transport by sciatic nerve isolated from lead-treated rat was significantly decreased compared with that of control rat. This deficit results from that myo-inositol transport system which is carrier mediated and sodium-potassium dependent was inhibited by the lead treatment (both 10ppm and 100ppm) due to increase of the Km value without affecting Vmax value for myo-inositol carrier. These observations suggest that the toxic mechanism of lead on nerve myo-inositol transport system might be a change of affinity without change of maximum transport velocity for carrier.

Myo-inositol Requirement in Diets for Juvenile Olive Flounder (Paralichthys olivaceus) (넙치 치어의 사료 내 myo-inositol 요구량)

  • A long-term (26 weeks) feeding experiment was conducted to examine the essentiality and requirement of inositol in diets for olive flounder because no information is available in the species. Five casein-gelatin based semi-purified diets were formulated to contain four different levels of myo-inositol (0, 0+antibiotic, 400, 800, and 1600 mg/kg, designated as M0, M0+, M400, M800, and M1600, respectively). One (M0+) of the control diets contained tetracycline hydrochloride (0.4%, wt/wt) as an antibiotic to inhibit biosynthesis of inositol by micro-organism in intestine of fish. Olive flounder at the early juvenile stage (initial body weight 1.22 g) were randomly distributed into fifteen 35 L tanks (48 fish/tank) and fed with one of the experimental diets (3 replicates per diet). At the end of the feeding trial, the weight gain, feed intake, specific growth rate, and protein efficiency ratio of fish fed diets containing higher levels of myo-inositol (M800 & M1600) were significantly higher than those of fish fed the other diets (P<0.05). Feed conversion ratio, survival, hematocrits, and hemoglobin of fish fed experimental diets were not significantly different among all the fish groups. Whole body compositions of fish were not different except for lipid content. The lipid content was significantly different between M0 and M400 diet groups. These results indicate that juvenile olive flounder requires dietary supplementation of myo-inositol in diets for normal growth and its optimum level seems to be approximately 800 mg myo-inositol/kg diet.

  • Inositols are cyclohexanehexol and they have been known to be nine stereomers. Scyllo-inositol, epi-inositol and muco-inositol could be synthesized from myo-inositol. Scyllo-inositol and epi-inositol were obtained by oxidation and reduction process from myo-inositol. Myo-inositol and epi-inositol were oxidized by treatment, in solution, with dilute hydrogen peroxide. In all cases, only axial hydroxyl groups were oxidized and monoketons were obtained. Reduction of myo-inosose-2 with sodium boron hydride was carried out in $pH2{\sim}3.$ The reduction products were equatorial alcohol but: reduction of DL-epi-inosose-2 by catalytic reduction produced axial alcohol obtained. Inositol could be esterified. Hexa-O-(p-hydroxy benzoyl)-esters of myo-inositol, scyllo-inositol, epi-inositol and muco-inositol were synthesized and their antimicrobial action on microbes were tested for application to food industry. As the results, it was found that the activities of muco-inositol ester was more vigorous than others.

  • In the previous work, as we have reported on the reduction of penta-O-acetyl-myo-inosose-2 and penta-O-acetyl-DL-epi-inosose-2, axial and equatorial alcohol have been produced. The synthesis of various inositol 5-nitro-2-furoate are described. The esters have been obtained by reaction of 5-nitro-2-furoylchloride with inositol stereomers in chloroform at low temperature. The ester have been characterized by paper chromatography and other determination. For the applicable purpose of food industry, their antimicrobial activities on microbes were tested, and it was found that the antimicrobial activity of muco-inositol ester was superior to the others, and considered that result would be related to the stereochemical structure.

Study on the Effect of Cysteine and Myo-inositol on In Vitro Maturation of Porcine Oocytes

  • This study was carried out to investigate the effect of morphology of oocytes, kinds of media, cysteine and myo-inositol supplementation on IVM rate of porcine oocytes. Cumulus- enclosed oocytes were incubated in maturation NCSU-23 and TCM-199 medium with supplementation with 3, 5, 10, 20 mM myo-inositol and 0.05, 0.1, 0.5, 1.0 mM cysteine. 1. When classified by morphology, excellent, good and fair of cumulus-enclosed oocytes were incubated for 48 hrs and the IVM rate were $14.2{\pm}3.7%{\sim}58.7{\pm}4.0%$, respectively. The rate were greater in oocytes with excellent cumulus cells than those without cumulus cells. 2. The IVM rate of oocytes cultured in TCM-199 and NCSU- 23 medium supplementation or non-supplementation with 1.0 mM myo-inositol were $7.5{\pm}4.5%,\;45.0{\pm}4.8%\;and\;4.4%,\;42.5{\pm}4.2%,\;18.0{\pm}5.2%$, respectively. Supplementation with myo-inositol significantly increased the IVM rate of oocytes. 3. The IVM rate of oocytes cultured in NCSU-23 medium supplementation of 3, 5, 10, 20 mM myo-inositol for 48 hrs were $47.5{\pm}4.5%,\;57.5{\pm}4.2%,\;62.5{\pm}4.9%,\;50.0{\pm}5.2%$, respectively. The IVM rate of oocytes in NCSU-23 medium supplemented with 10 mM myo-inositol were significantly increased compared to control ($42.5{\pm}4.0%$). 4. The IVM rate of oocytes cultured for 48 hrs in NCSU-23 media supplement with 0.3, 0.5, 1.0, 2.0 mM myo-inositol were $50.0{\pm}4.5%,\;62.5{\pm}4.2%,\;52.5{\pm}4.9%,\;45.0{\pm}4.2%$, respectively. The IVM rate of oocytes in NCSU-23 medium supplemented with 10 mM cysteine were significantly increased compared to control ($42.5{\pm}4.0%$).

  • myo-Inositol, a growth factor for Saccharomyces cerevisiae (S. cerevisiae), has been known to be incorporated into phosphatidylinositol (PI), which is a kind of phospholipid in the cell membrane, by a membrane-associated PI-synthesizing enzyme. The deficiency of myo-inositol in S. cerevisiae adversely affected the membrane structure and function. On the basis of biochemical functions of myo-inositol, the effect of deficiency of myo-inositol on the aerobic glucose metabolism was investigated by measuring specific oxygen uptake rate (Q$_{O2}$) used as an indicator representing the respiratory capacity of S. cerevisiae in batch and continuous cultures. The respiratory capacity of aerobic glucose metabolism in S. cerevisiae was also monitored after glucose pulse-addition in a continuous culture (D=0.2, 1/hr), in which glucose was utilized through respiratory metabolism. The deficiency of myo-inositol was found to lead to both the decrease of the maximum specific oxygen uptake rate (Q$_{O2max}$) observed from the batch as well as in the continuous culture experiment and the decrease of the respiratory capacity of aerobic glucose metabolism of S. cerevisiae determined from the glucose pulse-addition experiment, in which the glucose flux into respiratory and fermen- tative metabolism was quantitatively analyzed.
