무리우산버섯균의 배양적 특징과 목재부후 특성 파악 (The cultivate characteristics and the wood rotting ability and type of the Kuehneromyces mutabilis Sing. et A. H. Smith)

  • 무리우산버섯균의 기초적인 자료를 연구함으로써 앞으로 있을 인공재배 및 균사체 대량증식법 등의 연구에 있어서 발판이 되고자 연구한 결과는 다음과 같다. 1. 실험에 사용된 무리우산버섯균은 액체배지의 경우 Glucose pepton에서 202mg/18일로 균사생장이 가장 좋았으며, 고체배지는 YMG배지에서 81mm/8일로 가장 좋았다. 2. 균사생장 최적 온도는 $20{\sim}25^{\circ}C$이며, 최적 산도는 pH 5.5~6.0 이였으나 pH 6.0~7.0에서는 생장이 급속히 감소하였다. 3. 무리우산버섯균의 균사생장을 위한 최적영양원으로 탄소원에는 다당류인 Inulin이였으며, Dextrose를 제외하고는 균사생육에 있어서 큰 차이는 나지 않았다. 질소원으로는 무기태질소원인 $NaNO_3$였으며 다른 질소원들은 생장이 저조하였다. 4. 최적 톱밥배지 선정에 있어서 졸참나무가 가장 좋았으며 미송, 아카시나무, 상수리나무, 포플러 순으로 생장이 좋았고, Corn cob의 경우 생장이 가장 저조하였다. 5. 광학현미경과 주사형전자현미경으로 관찰한 바 목섬유의 세포벽의 일부분이 침식되고 세포 내강에서 중간층으로 점차 얇아지는 박벽화 현상이 관찰되었다. 6. 균사의 밀도는 도관과 방사유세포에 높게 나타났고, 일부 목섬유에서도 균사의 밀도가 높게 나타났다. 도관 내에 균사의 밀도가 높게 나타남에도 불구하고 목섬유와 유세포에 비해 분해가 거의 이루어지지 않았다. 7. PDA배지에 tannic acid를 첨가한 Bavendam 반응에서 배지의 재색이 암갈색으로 변하는 양성 반응을 보여 리그닌 분해 효소중의 하나인 laccase가 존재함이 확인되었다. 8. Dry weight에서 5.9%의 감소율을 보였으며, 이에 따른 pH의 변화는 6.07에서 4.31로 배지가 산성화 되었다. 또한 온수추출물에 있어서 톱밥과 미강(4:1 w/w)의 경우 12.98%의 추출량을 보여주었으며 무리우산버섯균이 배양된 배지의 경우 8.56%를 보여주었다.

Effects of Nutrient Composition on Yield and Quality of Mushroom in Lentinula edodes Cultivation Using Softwood Sawdust

  • This study was performed to evaluate the efficiency of using softwood as the sawdust medium for Lentinula edodes cultivation, effect of nutrient on the mycelial growth, spawning, the mushroom yield, and quality. The nitrogen nutrition significantly enhanced the mycelial growth of L. edodes. The glutamic acid in the L. leptolepis and P. koraiensis, and asparagine in the P. densiflora were appeared to slight increase in the mycelial growth. The vegetable oil showed very effective on the mycelial growth in the P. koraiensis sawdust medium. Carbon/nitrogen ratio of all the test was reduced after mycelial growth. The mycelial growth was exclusively dependent on reduction of carbon. The mushroom yield (32.7%) of the P. densiflora sawdust medium (carbon source: 3% active carbon, nitrogen source: 0.4% asparagines) was the best in mushroom production of L. edodes, followed by the Q. variabilis sawdust (35.4%) of the control medium. The diameter of mushroom cap was obtained from the P. densiflora sawdust (carbon source: 3% sucrose, nitrogen source: 0.4% potassium nitrate) and P. koraiensis sawdust (carbon source: 3% sucrose, nitrogen source: 0.4% potassium nitrate), and the P. koraiensis sawdust (carbon source: 3% xylose, nitrogen source: 0.4% glutamic acid, supplement: 0.05% amino acid), with values 71.5 mm, 71.5 mm and 72.1 mm, respectively. In the polypropylene bag cultivation, the weight losses of the block medium gradually increased for 80 days in the dark (13.8~16.8%) and then became stable in the range of 20.7~25.8%.

The Anti-complementary Activity of Exo-polymers Produced from Submerged Mycelial Cultures of Higher Fungi with Particular Reference to Cordyceps militaris

  • The anti-complementary activity (immuno-stimulating activity) was tested for the exo-polymers (extra-cellular polymer) produced from submerged mycelial cultures of 21 types of higher fungi. Anti-complementary activity of the exo-polymer from Cordyceps militaris showed the highest (69.0%) followed by Pleurotus ostreatus (63.7%) and Trametes suaveolens (61.4%). The mycelial growth rate and biomass doubling time of C. militaris in a 5 I air-lift fermenter were 0.0255 $h^{-1}$ and 27.2 h, respectively. The yield of the exo-polymer produced from the culture broth of C. militaris was 2.95 mg of dry weight/ml of culture broth. Sugar and amino acid compositions of this exo-polymer were analyzed.

Hypolipidemic Effect of Exo-Polymer Produced in Submerged Mycelial Culture of Five Different Mushrooms

  • The hypolipidemic effect of exe-polymer produced in submerged mycelial culture of Hericium erinaceus (HE), Auricularia auricula-judue (AA), Flammulina veluripes (FV), Phellinus pini (PP), and Grifola frondosa (GF) was investigated in dietary-induced hyperlipidemic rats. The animals were administered with exe-polymers at the level of 100 mg/kg body weight daily for four weeks. Hypolipidemic effect was achieved in all the experimental groups, however, HE exo-polymer proved to be the most potent one, which significantly reduced the plasma triglyceride ($28.9\%$), total cholesterol ($29.7\%$), low-density lipoprotein (LDL) cholesterol ($39.6\%$), phospholipid ($16.0\%$), and liver total cholesterol ($28.9\%$) level, when compared to the saline administered (control) group. The results of the present investigation strongly demonstrate the potential of HE exe-polymer in combating hyperlipidemia in the experimental animals.

Hypolipidemic Effect of Exo- and Endo-Biopolymers Pmduced from Submerged Mycelial Culture of Ganoderma lucidum in Rats

  • The hypolipidemic effect of the exe-biopolymer (EXBP) and endo-biopolymer (ENBP) produced from a submerged mycelial culture of Ganoderma lucidum was investigated in dietary-induced hyperlipidemic rats. Hypolipidemic effects were achieved in both the EXBP- and ENBP-treated groups, however, the former proved to be more potent than the latter. The administration of the EXBP (100 mg/kg body weight) substantially reduced the plasma total cholesterol, low-density lipoprotein (LDL) cholesterol, triglyceride, phospholipid levels, and atherogenic index by 31.0, 39.0, 35.4, 28.1, and 53.5%, respectively, when compared to the control group. The EXBP also lowered the liver total cholesterol, triglyceride, and phospholipid levels by 22.4, 23.1, and 12.9%, respectively. Furthermore, the high-density lipoprotein (HDL) cholesterol and ratio of HDL cholesterol to total cholesterol were significantly increased by as much as 24.2% and 47.6%, respectively.

인삼이 Aspergillus flayus의 생육 및 Aflatoxin 생성에 미치는 영향 (Effect of Panax ginseng on the Growth and Production of Aflatoxin by Aspergillus flayus)

  • This study was performed to investigate the effect of the Panax ginseng C. A. Meyer on the growth and production of aflatoxin by Aspergillus flayus ATCC 15517. Asp. fiavus with 10$^6$ conidia was incubated at 30$\circ$C for 7 days on YES broth containing 0.1%, 0.5%, and 1.0% of ginseng extract. After incubation, dry mycelial weight, pH, and production of aflatoxin were investigated. The results were as follows:There was no significant difference in dry mycelial weight by the addition of 0.1% and 0.5% ginseng extract. However, it was decreased to the rate of 13.7% by the addition of 1.0% ginseng extract in 7 days. pH changes in cultures were similar regardless of the concentration of ginseng extract. The pH values decreased to minimum in 5 days and again increased. Aflatoxin production was reduced as the concentration of ginseng extract increased. When compared to the control, the production of total aflatoxin significantly reduced to 56.7%, 54.0%, 53.3% in the media of 0.1%, 0.5% and 1.0% of ginseng extract, respectively. No significant difference was observed among ginseng extract groups.

  • The growth and biochemical activities of Chlorella fusca were studied in the presence of different concentrations of either filtrates or mycelial mats of Saprolegnia ferax and Pythium graminicola. Low concentrations of both fungal filtrates exerted increase in total count, dry weight and in the biosynthesis of photosynthetic pigments, carbohydrates and nitrogen content. High concentrations showed inhibitory effect on both growth and biochemical activities of Chlorella fusca. Supplementation with different concentrations of dry mycelial mats of either fungi the culture of Chlorella showed elevation in biomass, dry weight, and biosynthesis of carbohydrates and nitrogen content especially at low concentrations. The contents of photosynthetic pigment were inhibited only at low concentrations. Neither the culture filtrate of Pythium nor Saprolegnia had cellulolytic activity, although polygalacturonase enzymes were detected, whereas chloroform-extract of both fungal filtrates showed blue spots under long wave light (366 nm).

  • H. erinaceum의 기능성 소재화 연구의 일환으로, 지금까지 체계적으로 연구된 바 없는 H. erinaceum의 액체배양을 시도하고. 이 버섯의 서로 다른 생물반응기하의 액체배양 특성을 규명하고 대량생산을 검토하였다. 5 L 용량의 jar fermenter와 air-lift fermenter에서의 액체배양의 비교실험을 통해 air-lift fermenter의 사용이 종균 배양에 적합한 것으로 나타났으며, 형태 분석을 통해 적정 크기($2{\sim}4mm$)의 pellet 형태의 유지가 대량 생산의 중요인자인 것을 규명하였다. Air-lift fermenter 하의 배양을 종균으로 하고, 적정 크기의 pellet을 유지하는 500 L까지의 jar fermenter를 사용한 회분 배양의 scale-up 실험결과, 30 및 500 L에서의 교반 및 통기속도는 각각 200 rpm, 1 vvm 및 150 rpm, 1 vvm이었다. 이 조건하에서 배양의 전 기간 중 적정 pellet을 유지하였으며, 최대 균사체량은 약 $13{\sim}14g/L$이었다.

Production of Exo-Polymers by Submerged Mycelial Culture of Cordyceps militaris and Its Hypolipidemic Effect

  • Hypolipidemic effect of exo-polymers(EPs) form submerged mycelial culture of Cordyceps militaris was investigated in male Sprague-Dawley rats. For a dose-dependent study, EPs were administered at the level of 50-100 mg/kg body weight (BW) and the data was compared with the saline administered control group. A significant reduction of both the plasma total cholesterol (TC) and triglyceride (TG) was registered under the influence of EPs. It reduced the lowe density lipoprotein (LDL) cholesterol level as much as 40.5%. Levels of high density lipoprotein (HDL) choloesterol did not vary significantly within the different experimental groups, but the HDL: TC ratio showed consistently a high value with the increasing dose. The effects of cultural conditions (pH and temperature) in mycelial growth and EPs production were studied. Both the biomass and EPs were produced in a wide range of pH and temperature.

  • Cellulose-degrading fungi were idenfied as Rhizoctonia solani AG2-2(IV), T. harzianum and C. cochliodes. Rhizoctonia solani AG2-2(IV) grows better in the acidified media of pH 4 and 5 than pH 6 and 7. Mycelial growth of T. harzianum and C. cochliodes was also higher in pH 4 and 5 than in pH 6 and 7. In order to relate the above findings to nutrient utilization, mycelial growth of R. solani AG2-2(IV) are evaluated with various carbon sources. R. solani AG2-2(IV) grows well in the order of mannose, cellobiose, glucose, xylose and arabinose. However, mycelial dry weights of T. harzianum were 98.7, 78.0, 72.3, 43.7 and 32.3mg in glucose, mannose, cellobiose, xylose, and arabinose, respectively. Mycelial dry weight of C. cochilodes was 118, 65, 57, 49, and 16mg in mannose, cellobiose, xylose, glucose, and arabinose, respectively. Result of cellulase assay of R. solani AG2-2(IV) and soil fungi was reffered as, R. solani AG2-2(IV) produced more cellulase on CMC substrate than on CEL and secretes more enzyme in floated condition than in water-immersed condition. T. harzianum secreted less amount of cellulase than R. solani AG2-2 and C. cochliodes. T. harzianum produced no enzyme on CEL under water-immersed condition. C. cochliodes produced similar amounts of cellulase on either CMC or CEL under both water-immersed and floated condition.
