The objective of this study was to find the way to prolong the storage time of sawdust-based oyster mushroom (Pleurotus osteratus) spent substrate (OMSS) by fermenting with potential probiotic microorganisms to recycle the otherwise waste of mushroom farms. To this purpose, lactic acid bacteria (LAB) were screened to select the best lactic acid-producing strains. Three strains of LAB (Lactobacillus plantarum Lp1', Pediococcus acidilacticii Pa193, L. plantarum Lp2M) were selected and in mixture they lowered the pH of the fermented OMSS to 3.81. fOMSS (fermented sawdust-based oyster mushroom spent substrate) could be stored at room temperature for at least 17 days without any deterioration of feed quality based on the pH, smell, and color. In dry matter disappearance rate in situ, commercial TMR (total mixed ration), OMSS and OMMM (oyster mushroom mycelium mass) showed no significant differences between the samples after 6, 12 and 24 h incubation except for 48 h. Two separate field studies were performed to test the effects of fOMSS supplement on the growth performance of postweaning Holstein calves. Field trials included groups of animals feeding calf starter supplemented with: Control (no supplement), AB (colistin 0.08% and oxyneo 110/110 0.1%), fOMSS (10% fOMSS) and fConc (10% fermented concentrate) and DFM (direct-fed microbials, average $10^9$ cfu for each of three LAB/d/head). Growth performance (average daily gain and feed efficiency) of the fOMSS supplement group was higher than that of AB followed by fConc and DFM even though there was no statistically significant difference. The Control group was lower than any other group. Various hematological values including IgG, IgA, RBC (red blood cell), hemoglobin, and hematocrit were measured every 10 days to check any unusual abnormality for all groups in trial I and II, and they were within a normal and safe range. Our results suggest that sawdust-based OMSS could be recycled after fermentation with three probiotic LAB strains as a feed supplement for post-weaning calves, and fOMSS has the beneficial effects of an alternative to antibiotics for a growth enhancer in dairy calves.

Food waste (FW) has been recognized as a critical problem in Korea and many research was conducted to efficiently utilize or treat FW. Main purpose of this research was to evaluate a feasibility for producing fermented organic fertilizer with vegetable waste (VW). Three different organic materials (saw dust, coco peat, and waste mushroom media) were mixed with VW at the rate of 30, 40, 50% respectively. Total days of composting experiment were 35 days and each sub samples were collected at every 5 days from starting of composting. Result showed that inner temperature of composting was increased to $60{\pm}4^{\circ}C$ within 5~10 days depending on varied organic materials and mixing ratio. Among different treatment, the highest increase of inner temperature was observed when 30% of saw dust was mixed with VW. After finishing composting experiment, maturity of each compost was evaluated with solvita and germination test. Maturity index (MI) of each treatment was ranged between 5~7 indicating that manufactured fertilizer was curing or finished stage. Calculated germination index (GI) was at the range of 57.83~101.16 depending on organic materials and mixing ratio. Both MI and GI showed that manufactured fertilizer was met for fertilizer criteria while control (VW only) was not adequate for composting. Overall, VW can be utilized for making organic fertilizer mixing with saw dust, coco peat and more research should be conducted to make high quality of organic fertilizer with vegetable waste.
Fungal diversity during composting was investigated by culture-independent rDNA sequence analysis. Composting was carried out with pig manure and mushroom cultural waste using a field-scale composter (Hazaka system), and samples were collected at various stages. Based on partial sequence analysis of large subunit (LSU) ribosomal RNA (rRNA) and sequence identity values, a total of 12 different fungal species were found at six sampling sites; Geotrichum sp., Debaryomyces hansenii, Monographella nivalis, Acremonium strictum, Acremonium alternatum, Cladosporium sphaerospermum, Myriangium durosai, Pleurotus eryngii, Malassezia globosa, Malassezia restricta, Rhodotorula glutinis, and Fusarium sporotrichioides. Geotrichum sp. of the class Saccharomycetes was the most predominant fungal species throughout the composting process (185 out of a total of 236 identified clones, or 78.4%), followed by Acremonium strictum (7.6%), Monographella nivalis (5.1%), and Pleurotus eryngii (3.8%). The prevalence of Geotrichum sp. was the lowest (61.1%) at the beginning of composting, and then gradually increased to 92.5% after 10 days of composting.
표고버섯 골목의 바이오에탄올 생산 자원으로서의 활용 가능성을 모색하고, 새로운 당화 공정으로 주목받고 있는 초임계수 가수분해 공정에 대한 기초 기술을 제공하기 위하여 버섯 골목으로 사용되는 상수리나무 정상재를 다양한 초염계수 가수분해 공정에 적용하고 분해산물의 분석을 실시하였다. 초임계수 가수분해 반응 시간 및 온도의 증가에 의하여 수용성 분해산물 및 비수용성 잔사의 색 진해짐, 분해율의 증가 그리고 비수용성 잔사의 결정화도가 증가하였다. 반응 압력의 증가는 낮은 반응 온도와 짧은 반응 시간 조건에서만 분해 산물의 색변화 및 분해율의 증가에 영향을 주었다 초염계수 가수분해는 반응 초기에 hemicellulose의 분해가, 반응 후기에는 cellulose 분해가 진행되었다. 반응 시간의 증가는 당화 수율을 향상시켰고 반응 온도의 증가는 $415^{\circ}C$까지는 당화 수율을 증가시켰지만 $415^{\circ}C$ 이상에서는 급격히 감소시켰다. 낮은 반응 온도조건에서 반응 압력의 증가는 당화 수율을 증가시컸지만, 높은 반응 온도 조건에서는 오히려 당화 수율을 감소시켰다. 확인된 당화 수율의 결과를 토대로 상수리나무 정상재의 최적 초엄계수 가수분해 조건은 $415^{\circ}C$, 23 MPa, 60초로 결정하였으며, 당화 수율은 2.68%로 확인되었다. 결정된 최적 조건에서 골목의 초임계수 가수분해를 실시한 결과 당화 수율이 3.58%로 정상재의 당화 수율보다 높았다. GC-MS를 이용한 수용성 분해산물의 분석 결과, 주요 분해 산물은 1,1'-oxybis-benzene과 1,2-benzendicarboxylic acid로 확인되었고, 낮은 분해율을 보인 반응 조건에서는 pentadecanoic acid, 14-methyl-heptadecanoic acid 등과 같은 지방산류가 공통적으로 검출되었다. 반응 온도 및 시간이 증가함에 따라 phenol, benzene류의 증가가 확연되었지만 비수용성 잔사에서는 반응 압력의 차이에 의한 분해 산물의 변화는 없었다. 비수용성 잔사의 성분 분석 결과 60.6~79.2%의 holocellulose를 함유하고 있었고, 산 촉매 가수분해에 의하여 49.2~67.5%의 당화 수율을 보여주었다. 희산을 이용한 수용성 분해산물의 2차 가수분해에 의하여 수용성 분해 산불 내 당화 수율이 큰 폭으로 향상되었다.
BW를 이용하여 느타리를 재배함에 있어 면실박을 대체할 수 있는 재료를 찾기 위해 시험이 진행되었다. 일반성분 분석에서 BW가 면실박에 비해 단백질 함량은 다소 떨어졌으나 자실체의 생육특성에서 BW 혼합 시 대조구와 수량 등의 생육특성이 유사함을 확인할 수 있었고, 자실체의 구성아미노산 조성 확인결과 처리구 간 아미노산의 함량은 큰 차이를 나타나지 않았다. 따라서 BW가 느타리 배지제작에 있어 면실박을 대체하여 사용될 수 있을 것으로 판단된다.
우수한 약리기능성을 보이는 검은비늘버섯의 병재배에 적합한 새로운 품종 진황을 교잡육종법으로 육성하였다. 진황은 포플라톱밥을 이용한 병제배와 폐면을 이용한 상자재배가 모두 가능하여 재배농가에서 생산이 용이하나, 약용 또는 기능성버섯으로서의 인지도가 낮으므로 소비 확대를 위한 약리효과의 홍보가 필요하다. 진황 검은비늘버섯의 주요특성은 다음과 같다. 가. 균사배양 최적용도는 $25^{\circ}C$ 자실체발생 및 생육온도는 $15 {\sim}18^{\circ}C$이다. 나. 자실체의 색깔은 황갈색이며 갓표면에 흰색~갈색의 인피가 덮혀 있으며 점성이 있다. 다. 병당 유효개채수가 평균 26개로 많고 대도 굵어 품질이 양호하다.
Button mushroom compost (BMC) was prepared by fermenting the mixture of waste button mushroom bed collected from Boryeong area in South Korea (4): sawdust (8) : pig and fowl manure (1) for 40 days at $30^{\circ}C$. The BMC compromised diverse microorganisms including aerobic bacteria $8.1{\times}10^6cfu\;g^{-1}$, Gram negative bacteria $1.7{\times}10^7cfu\;g^{-1}$, genus Bacillus $6.4{\times}10^6cfu\;g^{-1}$, genus Pseudomonas $1.5{\times}10^4cfu\;g^{-1}$, actinomycetes $1.0{\times}10^4cfu\;g^{-1}$, and fungi $3.5{\times}10^3cfu\;g^{-1}$. BMC was used as a microbial inoculant for estimating the mobilization of heavy metals in soil or plant. When metal solubilization potential of BMC was assessed in a batch experiment, the inoculation of BMC was shown to increase the concentrations of water soluble Co, Pb, Cd, and Zn by 29, 26, 27, and 43% respectively, than those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 17, 15, 18, and 21% respectively in Co, Pb, Cd, and Zn contaminated soils. Moreover, enhanced accumulation of Co, Pb, Cd, and Zn in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the BMC. The apparent results suggested that the BMC could effectively be employed in enhancing phytoextraction from the soils contaminated with heavy metals such as Co, Pb, Cd, and Zn.
본 연구는 균상재배 느타리버섯부산물(SMS)의 반추동물 조사료원으로서의 가치를 평가하기 위하여 in situ 반추위 소실율 및 면양을 이용한 in vivo 영양소 소화율, 체내 질소이용성 및 섭취행동 패턴을 평가하였다. SMS에 당밀 5% (w/w, 건물기준), 유산균(Lactobacillus plantarum) 0.5% (v/w) 그리고 효모 (Saccharomyces cerevisiae) 0.5% (v/w)를 첨가하여 30일간 혐기발효 시킨 후 반추위캐뉼라가 장착된 평균체중 620 kg Holstein 육우 2두를 이용하여 in situ 소실율을 평가하였다. 면양대사실험은 평균체중 48kg의 숫 면양 6두 이용하여 $3{\times}3$ 라틴방각법으로 하였다. 대조구는 볏짚 100%를, 처리구는 볏짚의 25%와 50%를 생균발효 SMS로 각각 대체 급여하였다. 균상재배 버섯부산물의 반추위 in situ DM 및 NDF 이용성은 생균발효처리에 의해 개선되었다(P<0.001). 생균발효 SMS를 볏짚의 25% 및 50% 대체하여 면양에게 급여하였을 때, 단백질과 섬유소(NDF, ADF, CF) 소화율은 감소하였으나 (P<0.05), 조회분 소화율은 증가하였으며(P<0.001), 면양체내 단백질 축척량은 차이가 없었다(P>0.05). 볏짚을 생균발효 SMS로 대체 급여함에 따라 면양의 반추시간은 평균 28% 감소하였다(P<0.05). 생균발효 SMS는 볏짚의 76% 수준의 에너지가를 보였다. 결과적으로, 균상재배 느타리버섯부산물은 유지목적의 반추동물 조사료원으로서 보다 적합한 것으로 사료되었다.
한국버섯원균영농조합에서 품종보호등록된 화성1호와 다포자임의교배법으로 육성 선발되었던 PSC109를 교잡하여 봄-가을 균상재배에 적합한 화성5호를 육성하였다. 버섯의 대가 백색이고 곧으며 과습이나 갈변에 강한 특성을 가진다. 대조품종 수한1호와 비교하여 대는 길면서 약간 가늘고 갓색은 덜 진한 편이다. 버섯의 대조직은 단단하면서 탄력성이 있고, 씹음성과 깨짐성이 높다. URP-primer를 이용한 RAPD실험으로 두 모균주와 동일하지 않으면서 일부 밴드를 갖는 것을 알 수 있었다. 화성5호는 자체 시험재배에서 대조품종 수한1호보다 발이는 3-4일 늦으나 16.6% 높은 수량성을 보였고, 농가실증재배에서 서로 다른 재배조건에서 다소 수량의 차이는 있었으나 안정적인 재배가 이루어졌다. '화성5호'의 주요 재배특성으로 첫째, 수한1호와 비교하여 대가 길고, 갓은 얕은 깔때기형이다. 둘째, 균사배양 온도는 배지온도를 $20-26^{\circ}C$정도로 수한1호보다는 낮게 관리하여 주는 것이 좋다. 셋째, 버섯발생 온도는 $13-18^{\circ}C$, 자실체의 적정 생육온도는 $14-18^{\circ}C$으로 봄부터 가을철까지의 기간에 재배하기에 좋은 품종이다. 넷째, 균사배양중 $27^{\circ}C$ 이상으로 배지온도가 높아지면 배지 표층이 단단해지면서 발이가 늦어지거나 버섯 발생이 적게 될 수도 있다. 다섯째, 대가 길고 곧으며 대의 표면이 백색이며 단단하면서 탄력이 있다. 여섯째, 버섯 생육시기에 관수를 하며 관리해 주는 것이 좋으며, 수한1호보다 환기요구량이 조금 높다.
