A Study on Multiplication Response of "Tricholoma matsutake" (Pine Mushroom) Conidio to Cultural Media Environment (송이균(松茸菌) (Tricholoma matsutake)의 배양환경(培養環境)에 대한 증식반응(增殖反應)에 관한 연구(硏究))

  • This study was conducted to examine the physiology of pine mushroom mycelia cultured with various media for artificial culture of pine mushroom. The results obtained were as follows: 1) Among the various media, the medium composed of honey, boiled pine mushroom and soil extract fluid, fibrous root extract fluid, dry yeast, $KH_2PO_4$ inositol, folic acid, and biotin was the best for the growth of pine mushroom mycelium. 2) The optimum temperature for germinating pine mushroom spore and for culturing pine mushroom mycelium, was $24^{\circ}C$ and the optimum pH was 4.5. 3) There was no significant difference in growth between the mycelium separated from the tissue of pine mushroom sporophore and that separated from the spore. 4) No noticeable effect was found on the growth if such salts as $ZnSO_4$, $MnSO_4$, $MgSO_4$, $CaCl_2$ and ferric citrate were added to the Hamada's medium. 5) The addition of fibrous root extract promoted the growth of pine mushroom mycelium. 6) As a carbon source of artificial media, honey was more effective than glucose. 7) The culture infiltration of Mortierlla growing often in Fairy Ring was good for the growth of mycelium compared with the control. 8) The addition of fibrous root extract, inositol, biotin, and folic acid to artificial culture media was greatly effective in growth. When the temperature was lowered $19^{\circ}C$ after mycelium has appeared, the formation of primordium was observed.

  • Conditions for submerged culture of Agaricus campestris var. bisporus and the chemical composition of its mycelium were investigated. In shaking culture with TGY basal medium at $27{\sim}30^{\circ}C$, pH tended to increase upon culture period, mycelial growth was the highest on 12th day, with relatively high nitrogen content of 7% and sugar in the medium disappeared almost at the end of culture period. As a nitrogen source, ammonium phosphate (dibasic) gave relatively high mycelial yield and the addition of yeast extract gave rise to better results. As a carbon source, glucose was the best, fructose, maltose, lactose and sucrose gave the same results, and soluble starch was utilized slightly. Mushroom mycelium contained 48% of protein, 8 free amino acids including arginine, histidine, lysine, isoleucine, leucine, phenylalanine, proline, tyrosine and its protein consisted of most essential amino acids, with relatively high contents of lysine and threonine. Therefore, mushroom mycelium deserves to be a high quality protein food.

Proton Beam Sensitivity of Basidiospore and Mycelium in Pleurotus ostreatus (느타리버섯 포자와 균사체의 양성자 빔 조사에 대한 민감도)

  • To assess the effects of proton beam on radiation sensitivity of the basidiospore and mycelium of oyster mushroom (Pleurotus ostreatus), the $D_{10}$ values and $L_{50}$ (lethal 50%) values were analysed. By the proton beam radiation, the survival rate and germination rates increased at the dose of $10\;Gy{\sim}100\;Gy$ and then decreased significantly over 500 Gy. $L_{50}$ values of basidiospore and mycelium of Pleurotus ostreatus were over 500 Gy and 400 Gy, respectively. $D_{10}$ values were calculated from linear regression formulae ($D_{10}\;=\;-1/slope(b)$, y = a + bx) as 750 Gy and 1,250 Gy, respectively. Based on our experiment, the optimum dose of proton beam as a mutation source would be between from 500 Gy to 750 Gy for basidiospores and from 400 Gy to 1000 Gy for mycelium of oyster mushroom.

Enzyme activity of Basidiomycetes products in each cereals (곡물별 담자균 배양물의 효소활성)

  • In this study, growth rates and enzyme activities of mushroom mycelium were investigated in each cereals. Cultivation on hulled barley resulted in slightly faster mycelial growth as compared to other substrates. Enzyme activities were measured in different periods. In result, ${\alpha}$-amylase activity was higher in Pleurotus ostreatus and Phellinus linteus cultured cereals, whereas ${\beta}$-amylase activity was higher in Pleurotus ostreatus and Fomitella fraxinea cultured. Protease level did not affect kinds of cereal except Phellinus linteus. The color values lightness decrease by Basidiomycetes cultivation and redness and yellowness increased.

  • Flammulina velutipes is a popular edible basidiomycete mushroom found in East Asia and is commonly known as winter mushroom. Mushroom development showing dramatic morphological changes by different environmental factors is scientifically and commercially interesting. To create a genetic database and isolate genes regulated during mushroom development, cDNA libraries were constructed from three developmental stages of mycelium, primordium, and fruit body in F. velutipes. We generated a total of 5,431 expressed sequence tags (ESTs) from randomly selected clones from the three cDNA libraries. Of these, 3,332 different unique genes (unigenes) were consistent with 2,442 (73%) singlets and 890 (27%) contigs. This corresponds to a redundancy of 39%. Using a homology search in the gene ontology database, the EST unigenes were classified into the three categories of molecular function (28%), biological process (29%), and cellular component (6%). Comparative analysis found great variations in the unigene expression pattern among the three different unigene sets generated from the cDNA libraries of mycelium, primordium, and fruit body. The 19-34% of total unigenes were unique to each unigene set and only 3% were shared among all three unigene sets. The unique and common representation in F. velutipes unigenes from the three different cDNA libraries suggests great differential gene expression profiles during the different developmental stages of F. velutipes mushroom.

Transfer of Isolated Nuclei from Pleurotus florida into Protoplasts of Pleurotus ostreatus (느타리버섯 원형질체내(原形質體內)에 사철느타리버섯 핵(核)의 전이(轉移))

  • The transfer of the isolated nuclei from P. florida into protoplasts of P. ostreatus was induced with polyethylene glycol and $CaCl_2$. Three types of transfer products of nuclei were obtained when transferred to MMM. Type 1 colonies were more vigrously growing mycelium and stable on MCM. One of the type 1 colonies, appeared segregation on MCM plus benomyl. The mycelium did not form clamp connection. These results suggest that type 1 colonies were nuclear hybrids or allodiploids. Type 2 was main products of nuclear transfer. The mycelium formed clamp connection and fertile on sawdust media. Type 3 was very slow growing or non-viable colonies after debris of nuclei or chromosomes transfer into recipient protoplasts. Isozyme pattern of esterase in type 1 produced a new band. Type 2 and type 3 could be characterized by parental bands.

Radiation Sensitivity of Basidiospore and Mycelium in Pleurotus ostreatus

  • To assess the effects of gamma-ray (Co-60) on radiation sensitivity and genetic similarity of the basidiospore and mycelium in oyster mushroom, Pleurotus ostreatus, the D$_{10}$ values and RAPD patterns were analysed. Three strains were isolated from basidiospores (PO-Bl, -B2, and - B3 from 2 kGy irradiation group) and five strains from mycelia (PO-Ml, -M2 from 1 key, PO-M3 from 2 kGy, and PO-M4 and -M5 from 2+1 key irradiation group). The D$_{10}$ values of basidiospore and mycelium of P. Preurotus ostreatus were 1,250 Gy and 500 Gy, respectively. The growth rates of the eight strains on the five media were various and the activities of extracellular chitinases of them were generally higher than those of the control. By the gamma-ray radiation, 22-25% of genetic similarities were changed in the basidiospore strains and 23-36% of them in the mycelium strains. From these results, it seems that the basidiospore could be more radio-resistant than the mycelium of P. ostreatus and that the genetic similarity of the mycelium of P ostrentus could be changed easier than that of the basidiospore by the gamma-ray radiation.ion.

  • The study has been carried out to invetigate the possibility of oak mushroom (shiitake) cultivation on Ban Oak (Quercus incana) growing naturally in Simlaregion, India. The survival and growth of oak mushroom mycelium and fruit body formation on Q. incana as well as the composition of the log were compared with that of Bristle-tooth oak (Q. serrata) which are being used for mushroom cultivation in Korea. The results are summarized as follows. 1. The content of alcohol-benzen extract, NaOH extract, NaOH extract and ash were higher in Q. incana than in Q. Serrata. While cellulose and pentosan were less in the formers. 2. The mycelial growth of oak mushroom were more rapid on sawdust medium of Q. Serrata than on Q. incana. However the mycelial growth on the later were more compact. 3. The mycelial growth of oak mushroom were more rapid on the logs of Q. serrata than Q. incana. The mycelium survived well on both two species, and no difference in the survival rate of mycelium were observed. 4. The first fruit bodies on logs of Q. Serrata and Q. incana were appeared 16 months inoculation of spawn. 5. In view of the above results. it seems that the cultivation of oak mushroom (shiitake) on Ban oak (Q. incana), growing in India, is possible.

Cultivation technique using plastic container and selection the superior strain of nameko mushroom (Pholiota nameko) (맛버섯(Pholiota nameko) 우량균주 선발 및 병 재배법)

  • These experiment was conducted to find the superior strain selection, cultivation technique and optimum environmental condition of nameko mushroom culture using plastic container. The results was following as Mycelium of Pholiota nameko grown well at MCM and Hamada media, and its media acidity was pH 6~7. The optimum temperature condition for growing mycelium was $25^{\circ}C$. Under $15^{\circ}C$ and above $30^{\circ}C$ of temperature condition, mycelium growing speed was delayed remarkably. Among the 29 strains of nameko mushroom, the most productive strains was JNM19007, JNM19026, JNM19027 and JNM19028. The optimum media composition rate for produce fruitbody was pine sawdust 80% + wheat bran 20%. In this condition, the average fruitbody amount was 188g per 1,100cc container. The optimum post-culturing period was 50 days and mushroom sprout appeared 7 days after old mycelia removed. The suitable temperature was $12^{\circ}C$ for induce sprout, growing period was $16^{\circ}C$ and the optimum relative humidity was 95% in all culturing periods.

Protoplast Isolation and reversion from Pleurotus spodoleucus (참느타리버섯의 원형질체(原形質體) 분리(分離) 및 환원(還元))

  • This experiment was undertaken to investigate proper conditions for protoplast formation from Pleurotus spodoleucus. Novozym 234 was the most effective enzyme and a high yield of protoplasts was obtained. Combination of enzymes did not improve this result. Sucrose gave the best result to support the release and maintain the stability of protoplasts. The optimal reaction time of mycelium with the lytic mixture was 3 hrs. in shaking condition at 120 strokes $min-^1$. When mycelium of P. spodoleucus was cultured for 3 days on mushroom complete agar medium at $27^{\circ}C$, the formation of protoplasts was effective. The mushroom complete agar medium stabilized with 0.6 M sucrose was the most effective for reversion of protoplasts.
