  • Fructose 1,6-diphosphate(FDP), a glycolytic metabolite, is reported to ameliorate inflammation and inhibit the nitric oxide production in murine macrophages stimulated with endotoxin. It is also reported that FDP has cytoprotective effects against hypoxia or ischemia/reperfusion injury in brain and heart, and may play a protective role in ultraviolet B (UVB, 280~320 nm)-injured keratinocyte by attenuating prostaglandin (PG)-E$_2$production and cyclooxygenase (COX)-2 expression, which are possibly through blocking the intracellular reactive oxygen species (ROS) accumulation. Therefore FDP is considered to act as a potent antioxidant especially in the skin. We conducted the several safety tests (single-dose toxicity, primary skin irritation test, eye irritation test, skin sensitization test, phototoxicity test, photosenitization test and human patch test) to see if FDP is safe in case used for the skin application. Our data obtained hitherto suggest that FDP is very safe if applied to the skin.

  • The antiinflammatory mechanism of NSAIDs is attributed to the reduction of prostaglandin synthesis by the direct inhibition of cyclooxygenase. Inhibition of prostaglandin production in organs such as stomach and kidney can result in gastric lesions, nephrotoxicity and increased bleeding. In this study, newly designed COX-2 inhibitors, synthesized 1,2-benzothiazine derivatives, were screened in vitro for selectivity of COX-1 and COX-2 inhibition properties. Lead compounds in the structure-activity relationship were studied to synthesize new highly selective COX-2 inhibitors.13 determine inhibitory effect of COX-2, synthesized 1,2-benzothiazine derivatives were screened with accumulation of prostaglandin by lipopolysaccharide (LPS) in aspirin-treated macrophages and murine macropharge cell. Some of synthesized 1,2-benzothiazine derivatives were shown to be effective as selective COX-2 inhibitory activity. Others exhibited a preferential inhibition of COX-2, although some COX-1 inhibitory activity was still present. As a conclusion, simple monomer derivatives were more active than dimer derivatives. Substitution of halogen (Br, C1) on the benzothiazine nucleus slightly enhanced inhibition activity.

Anthraquinones with Immunostimulating Activity from Cassia tora L.

  • Many of plants had been reported having immunostimulating activity. This study reports the immunostimulating activity of Cassia tora L. (Leguminosae) seed, by means of solvent extraction method. Ethanol extract and solvent fractions, n-hexane, chloroform, ethylacetate, n-butanol and aqueous layer of Cassia tora L. seed were tested for immunostimulating activity in vitro. The ethylacetate-soluble fraction caused significant inhibition on the production of nitric oxide by murine macrophages (RAW 264.7), and mouse splenocytes were also stimulated at the concentration of 10 pgfmL. Three anthraquinones, chrysophanol (1), isochrysophanol (2) and aloe-emodin (3) with immunostimulating activity were isolated from the ethylacetate-soluble fraction of Cassia tora L. seed through activity-monitored fractionation and isolation method. These results permit Cassia tora L. to be useful as one the of natural immunostimulating crops.

  • Polygonum cuspidatum which is widely distributed in Korea has been used as treatments of dermatitis, gonorrhea and inflammation in traditional medicine. We examined anti-inflammatory activities by the inhibition of NO production in RAW264.7 murine macrophages cells. Phytochemical examination of Polygonum cuspidatum led to the isolation and characterization of emodin (1), emodin 8-O-${\beta}$-D-glucopyranoside (2), emodin 1-O-${\beta}$-D-glucopyranoside (3). Antioxidative activities of these compounds were determined by measuring the radical scavenging effects on DPPH radicals. Compounds 1 and 3 showed potent activities compared with L-NMMA. These results suggested that the antraquinone compounds isolated from Polygonum cuspidatum might be used as antiinflammatory agents.

  • The aim of this study was to evaluate capability of pertussis toxin (PT) to active mouse macrophages. The investigations were undertaken to determine whether the role played by this toxin required the A-protomer of the toxin to ADP-ribosylate a guanine nucleotide binding protein (a class I activity) or was dependent on the binding of B-oligomer of the toxin to the surface of target cells (a Class II activity). The results of these experiments have established that the mechanism of macrophage activation with PT seems to be dependent upon a Class II activity of the toxin.

  • MeOH extract obtained from the Kochia scoparia (KS) was observed to inhibit tumor necrosis factor-alpha (TNF-alpha), prostaglandins (PGs) and nitric oxide(NO) production in a lipopolysaccharide (LPS)-stimulated murine macrophage cell line, RAW 264.7. These effects of MeOH-KS were based on modulation of iNOS and COX-2 level. Western blot analysis showed that MeOH-KS reduced the iNOS and COX-2 level in LPS activated macrophages, in a dose dependent manner without cNOS and COX-1 protein level. (omitted)

  • Synthetic polymers, biological polymer such as collagen and gelatin are employed extensively as backbones in the construction of hydrogels or cell/tissue scaffoldings for various biomedical applications. In the present study, we investigated the effect of collagen and gelatin on the inducible nitric oxide synthase (iNOS) gene expression in the mouse macrophage cell line RAW 264.7. The production of nitric oxide and expression level of iNOS mRNA were induced by both of collagen and gelatin in dose-dependent manner. (omitted)

  • Nitric oxide (NO) is known to work as an important signaling molecule involved in regulating a wide range of biological activities in the neuronal, vascular, and immune system. NO and its metabolites mediate a number of host defence functions and are also implicated in the pathogenesis of tissue damage associated with inflammation. Cyclohexylimminobenzoxathiol LYR-64 compound inhibited LPS-induced NO production in murine macrophages Raw264.7 with an IC50 value of 0.7 uM with 95.9% inhibition at 3 uM, 63.5% at 1 uM and 30.2% at 1 uM.   • Aromatic diamine JSH-21 showed an IC50 value of 9.2 uM with 74.5% inhibition at 30 uM, 53.5% at 10 uM and 24.5% at 3 uM on LPS-induced NO production in murine macrophages Raw 264.7. To examine whether inhibitory effect on NO production by JSH-21 was attributed to influence on iNOS expression, iNOS transcript and protein were analyzed by sequantitative RT-PCR and immunoblot analysis. Consistent with previous result on NO production, treatment of the Raw 264.7 cells with JSH-21 decreased the LPS-induced expression of iNOS transcript and protein in a dose-dependent manner with IC50 values of about 10 uM.
