Curcumin modulates the apolipoprotein B mRNA editing by coordinating the expression of cytidine deamination to uridine editosome components in primary mouse hepatocytes

  • Curcumin, an active ingredient of Curcuma longa L., can reduce the concentration of low-density lipoproteins in plasma, in different ways. We had first reported that curcumin exhibits hypocholesterolemic properties by improving the apolipoprotein B (apoB) mRNA editing in primary rat hepatocytes. However, the role of curcumin in the regulation of apoB mRNA editing is not clear. Thus, we investigated the effect of curcumin on the expression of multiple editing components of apoB mRNA cytidine deamination to uridine (C-to-U) editosome. Our results demonstrated that treatment with $50{\mu}M$ curcumin markedly increased the amount of edited apoB mRNA in primary mouse hepatocytes from 5.13%-8.05% to 27.63%-35.61%, and significantly elevated the levels of the core components apoB editing catalytic polypeptide-1 (APOBEC-1), apobec-1 complementation factor (ACF), and RNA-binding-motif-protein-47 (RBM47), as well as suppressed the level of the inhibitory component glycine-arginine-tyrosine-rich RNA binding protein. Moreover, the increased apoB RNA editing by $50{\mu}M$ curcumin was significantly reduced by siRNA-mediated APOBEC-1, ACF, and RBM47 knockdown. These findings suggest that curcumin modulates apoB mRNA editing by coordinating the multiple editing components of the edito-some in primary hepatocytes. Our data provided evidence for curcumin to be used therapeutically to prevent atherosclerosis.

Nonlinear vibration and primary resonance of multilayer functionally graded shallow shells with porous core

  • This research studies the primary resonance and nonlinear vibratory responses of multilayer functionally graded shallow (MFGS) shells under external excitations. The shells considered with functionally graded porous (FGP) core and resting on two types of nonlinear viscoelastic foundations (NVEF) governed by either a linear model with two parameters of Winkler and Pasternak foundations or a nonlinear model of hardening/softening cubic stiffness augmented by a Kelvin-Voigt viscoelastic model. The shells considered have three layers, sandwiched by functionally graded (FG), FGP, and FG materials. To investigate the influence of various porosity distributions, two types of FGP middle layer cores are considered. With the first-order shear deformation theory (FSDT), Hooke's law, and von-Kármán equation, the stress-strain relations for the MFGS shells with FGP core are developed. The governing equations of the shells are consequently derived. For the sake of higher accuracy and reliability, the P-T method is implemented in numerically analyzing the vibration, and the method of multiple scales (MMS) as one of the perturbation methods is used to investigate the primary resonance. The results of the present research are verified with the results available in the literature. The analytical results are compared with the P-T method. The influences of material, geometry, and nonlinear viscoelastic foundation parameters on the responses of the shells are illustrated.

A Rare Case of Primary Duodenal Follicular Lymphoma

  • Primary duodenal follicular lymphoma is rare and presents as multiple, small polyp-like lesions on endoscopy. If this lesion is suspected, an endoscopic biopsy is crucial for diagnosis. A watchful wait would be appropriate management as it has a fairly indolent clinical course. Herein, we present a rare case of primary duodenal follicular lymphoma.

  • The increasing incidence of multiple primary carcinomas occuring in the upper aerodigestive tract is well documented, with the accepted incidence being as high as 20-30%. The fiberoptic endoscopy has also enabled visualization of areas previously inaccessible without general anesthesia. A prospective panendoscopic examination of the upper aerodigestive tract was peformed on 104 patients with squamous cell carcinoma of head & neck in our hospital between 1989 and 1994. Five second primary cancers (4.8% :2 stomach, 2 esophagus, 1 lung cancers) were detected endoscopically. These finding should reinforce the belief that head & neck cancer is a panmucosal disease of the aerodigestive tract that silent second primary cancers are not uncommon. So every effort should be done to detect second primary cancers in head & neck squamous cell carcinomas. Panendoscopy has proved valuable in achieving that.

  • Primary hryperparathyroidism is a relatively rare disease entity in Korea. It's characterized by severe skeletal and renal changes due to hypersecretion of parathyroid hormone, and rarely shows peptic ulceration, hypertension, pancreatitis and impaired mentality. Recently the determination of the serum calcium level has become a routine laboratory test and the awareness of primary hyperparathyroidism has been incerased, the disease is being diagnosed with increasing frequency. Primary hyperparathyroidism is most commonly caused by parathyroid adenoma and rarely hyperplasia, cancer of parathyroid glands. The authors operated sucessfully a case of primary parathyroid adenoma by diagnosed by nuclear medical diagnostic work-up. The patient had anterior neck mass(soft, non tender nodule) on physical examination, multiple goiter on thyroid ultrasonogram and scan. The parathyroid lesion was difficult to find preoperatively.

Clinical Features of Multiple Primary Malignancies: a Retrospective Analysis of 72 Chinese Patients

  • There is a scarcity of reports addressing patients with multiple primary malignancies (MPM), especially for Chinese cses. The aim of this study was to present a detailed analysis of Chinese patients presenting with at least two primary malignancies. Particularly, the clinical characteristics and survival between synchronous and metachronous MPM were compared. Out of 6,545 cases, 72 patients (1.1%) including 39 males (54.2%) and 33 females (45.8%) were diagnosed as MPM, giving a male: female sex ratio of 1.2:1. Of these, there were 16 (22.2%) cases of synchronous MPM (7 males, 9 females), 55(76.4%) metachronous (31 males, 24 females), and 1 "mixed form". For first tumor diagnosis time, synchronous MPM patients generally presented later than the metachronous cases. The top three sites for malignancies with metachronous MPM were colorectum, head and neck, and lung, while for synchronous they were lung, colorectum and breast. Among MPM patients, the median survival time was 15.7 years and the 5-year survival was 56%, and there was statistical difference in MPM categories (P < 0.05). The median survival time was 17.3 years and 3.8 years for metachronous and synchronous MPM patients, respectively. In comparison with synchronous MPM patients, those metachronous had a longer survival. This studies increase understanding of the clinical features of Chinese MPM patients and suggest that those presenting with metachronous cancers have a higher incidence and a better prognosis.

Release Profile of Peptide from Biodegradable Microspheres: Comparison of Blending and Multiple Emulsion Method (고분자 혼합법과 다중 에멀젼법에 의해 제조된 생분해성 미립구로부터 펩타이드의 용출에 관한 연구)

  • The novel microsphere blending and multiple emulsion method by single process was tried to prepare sustained release microspheres which release a physiologically active substance for long periods of time. A drug was separately dissolved in each of two or more oils containing biodegradable polymers to give the primary oil phases. The primary oil phases were dispersed in single aqueous phase in succession. From the drug-dispersed solution, the organic solvent was removed to produce microspheres. The accelerated drug release from the microsphere formulation prepared by single process through the multiple emulsion method was very similar to a physical blending of separately prepared microspheres using the same polymers. But long term release was not same. In this study, leuprorelin acetate loaded poly(lactide-co-glycolide) microsphere formulation for one-month delivery was developed by the multi-emulsion method followed by solvent extraction/evaporation method.

Association of Higher Continuity of Primary Care with Lower Risk of Hospitalization among Children and Adolescent Patients (어린이와 청소년 환자에서 일차의료의 지속성과 입원 위험도의 관련성)

  • This study aims to describe levels and distribution of the continuity of primary care among children and adolescent patients who are 2-19 years old, and analyze the effects of it on the risk of hospitalization. Study population was 2-19 year old child and adolescent patients as of 2002, who had more than three ambulatory care visits in the years of 2002-3 and whose most frequent provider was the primary care practices (189,660 persons). Association of levels of primary care with the risk of hospitalization was evaluated using multiple event survival analysis. Outcome variables were whether the patient had hospitalized or not, and whether the patient had hospitalized due to ambulatory care-sensitive conditions or not. Multiple event survival analysis revealed statistically significant association of the levels of primary care with the risk of hospitalization. Hazard ratio was 1.34 [1.27-1.41] at the medium level of continuity and 1.47 [1.39-1.55] at the lower level where outcome variable was whether the patient had been hospitalized or not. Hazard ratios were 1.35 [1.21-1.50] at the medium level of continuity and 1.60 [1.44-1.78] at the lower level, where outcome variable was whether the patient been had hospitalized due to ambulatory care-sensitive conditions or not. This study produced some evidences on the benefits of continuity of care, which will in turn support the introduction of personal doctor registration program in the future.

  • Many disorders and abnormalities are accompanied by cavitary lesoin of the lung and one of the most common causes of cavitary lung disease are primary and metastatic lung neoplasms. but cavitary formation of primary lung cancer is not frequent and cavitary or cystic pulmonary metastases of this is also rare. We report a case of cavitary pulmonry metastases of primary lung cancer proven by bronchoscopic biopsy and chest CT. The patients was 60 year-old heavy smoker who had no known underlying lung diseases 7 years ago and complained chronic cough. The chest CT showed primary lung cancer in right low lobe with multiple cavitary or cystic metastases in both lungs and multiple lymphatic metastases.

A Case of Primary Cutaneous Aspergillosis on the Scalp and the Neck (두피 및 경부에서 발생한 원발성 피부 아스페르길루스증의 1례)

  • The cutaneous aspergillosis is one of the most common dermatologic manifestations of disseminated infections associated with the Aspergillus organisms, but the isolated primary cutaneous disease itself can rarely occur in an immunocompetent host. We report a case of the primary cutaneous aspergillosis on the scalp and the neck in a 39-year-old immunocompetent male patient. There was a single purulent ulcer surrounded by the erythematous indurated plaque on the scalp with multiple satellite papules and a multi-lobulated granulomatous plaque with a crust on the neck. Skin biopsy demonstrated a fungus, the Aspergillus, in the deep dermis as the etiologic agent. No evidence of involvement in other organs was found. The patient exhibited no other apparent systemic diseases nor immunologic defects. An elliptical excision and a primary closure was performed, and the adjuvant antifungal treatment, oral itraconazole, was applied to prevent the recurrence by the satellite lesions.
