The Effects of Crinum asiaticum on the Apoptosis Induction and the Reversal of Multidrug Resistance in HL-60/MX2

  • The present study investigated the anti-proliferative and chemosensitizing effects of Crinum asiaticum var. japonicum against multi-drug resistant (MDR) cancer cells. The 80% methanol extract, chloroform ($CHCl_3$) fraction and butanol (BuOH) fraction of C. asiaticum inhibited the growth of mitoxantrone (MX) resistant HL-60 (HL-60/MX2) cells. When HL-60/MX2 cells were treated with the $CHCl_3$ and BuOH fractions, DNA ladder and sub-G1 hypodiploid cells were observed. Furthermore, the fractions reduced BcI-2 mRNA levels, whereas Bax mRNA levels were increased. These results suggest that the inhibitory effect of C. asiaticum on the growth of the HL-60/MX2 cells might arise from the induction of apoptosis. Treatment of HL-60/MX2 cells with the fractions markedly decreased the mRNA levels of the multi-drug resistance protein-1 and breast cancer resistance protein. The $CHCl_3$ fraction and hexane fraction increased MX accumulation in HL-60/MX2 cells. These results imply that the $CHCl_3$ fraction of C. asiaticum plays a pivotal role as a chemosensitizer. We suggest that components of C. asiaticum might have a therapeutic potential for the treatment of MDR leukemia.

Formation of Dictyosome and Spherosome in Endosperm Cells of Panax ginseng during seed Formation (인삼(人蔘)(Panax ginseng)의 종자형성(種子形成)에 따른 배유세포(胚乳細胞)의 딕티오좀 및 Spherosome 형성)

  • This study has been carried out to investigate the development of dictyosome, and roles of dictyosome about the formation of spherosome in the endosperm cell during seed formation of Panax ginseng with electron microscope. The result is as follows; In the endosperm cells of early stage during seed formation of Panax ginseng, plastid, mitochondria, endoplasmic reticulum, dictyosome and ribosomes are evenly distributed in cytoplasm. Electron lucent vesicles derived from dictyosome are observed in endosperm cells. Vesicles that contain low electron density are derived from forming face of dictyosome and releases into the cytosol. This vesicles formed multi vesicular body or fused with the plasma membrane. The spherical spherosomes are formed from dictyosome containing the lipid materials of even electron density and are gradually increased in size and number. Dictyosome is located in between vacuole and spherosome and it's cisternae form a semicircle and a circle. Some membrane of the protein body that accumulate the storage protein are originate from the spherical vacuole which interfused between vesicles and vacuoles derived from dictyosome.

Organic Photovoltaic Devices on $HNO_3$-Treated Multilayer Graphene Electrodes

  • We reported on the characteristics of organic solar cells (OSCs) fabricated on $HNO_3$-treated multilayer graphene (MLG) transparent electrodes. MLG electrodes were prepared using a chemical vapor deposition and a multi-transfer process. Compared to organic solar cells (OSCs) on the ITO electrodes had a fill factor of 65.97%, and a power conversion efficiency (PCE) of 3.364%, OSCs on the MLG (three-layer graphene) electrodes with sheet resistance of $274{\pm}1{\Omega}$/square and transparency of 92.1% had a fill factor of 43.46%, and a power conversion efficiency (PCE) of 2.019%. However, OSCs on the HNO3-treated MLG electrodes with lower sheet resistance of $119{\pm}1{\Omega}$/square had a fill factor of 57.54%, and a PCE of 2.861%. The results would provide a promising method to improve the performance of large-area OSCs based on MLG electrodes.

Primary splenic hemangiopericytoma in a dachshund dog (닥스훈트의 원발성 비장 혈관주위세포종 증례)

  • A 7-year-old, castrated male dachshund dog with tumor of the spleen portion was referred to the Kangwon National University. The tumor removed surgically and tumor size was 6~7 cm. Histopathologically, this neoplasm was the presence of perivascular whorls of fusiform cells. The cells also arranged in interlacing bundles or storiform patterns. And this neoplasm consisted of spindle cells that often formed distinct whorls around a central capillary. Immunohistochemical analysis revealed multi-focally immunoreactivity for ${\alpha}$-smooth muscle actin, whereas not immunoreactive for desmin, S-100, von Willebrand factor. On the base of the histological and immunohistochemical results, this neoplasm was diagnosed as a canine hemangiopericytoma.

Morphological Variation Classification of Red Blood Cells using Neural Network Model in the Peripheral Blood Images (말초혈액영상에서 신경망 모델을 이용한 적혈구의 형태학적 변이 분류)

  • Recently, there have been researches to automate processing and analysing images in the medical field using image processing technique, a fast communication network, and high performance hardware. In this paper, we propose a system to be able to analyze morphological abnormality of red-blood cells for peripheral blood image using image processing techniques. To do this, we segment red-blood cells in the blood image acquired from microscope with CCD camera and then extract UNL fourier features to classify them into 15 classes. We reduce the number of multi-variate features using PCA to construct a more efficient classifier. Our system has the best performance in recognition rate, compared with two other algorithms, LVQ3 and k-NN. So, we show that it can be applied to a pathological guided system.

  • Photoelectrochemical solar cells such as dye-sensitized cells (DSSCs), which exhibit high performance and are cost-effective, provide an alternative to conventional p-n junction photovoltaic devices. However, the efficiency of such cells plateaus at 11-12%, in contrast to their theoretical value of 33%. Improvements in efficiency can only occur through a fundamental understanding of the underlying physics, materials, and device designs of DSSCs. A photoelectrode consisting of semiconducting oxide nanoparticles and a transparent conducting oxide electrode (TCO) is a key component of DSSC and design of photoelectrode materials is one of promising strategies to improving energy conversion efficiency. We introduce monodisperesed $TiO_2$ nanoparticles prepared by forced hydrolysis method and their superiority as photoelectrode materials was characterized with aids of optical and electrochemical analysis. Multi-layered TCO materials are also introduced and their feasibility for use as photoelectrodes is discussed in terms of optical absorption and charge collecting properties.

Improved Understanding of LeTID of Single-crystalline Silicon Solar Cell with PERC

  • Light elevated temperature induced degradation (LeTID) was noted as an issue in multi-crystalline silicon solar cells (MSSC) by Ram speck in 2012. In contrast to light induced degradation (LID), which has been researched in silicon solar cells for a long time, research about both LeTID and the mechanism of LeTID has been limited. In addition, research about LeTID in single-crystalline silicon solar cells (SSSC) is even more limited. In order to improve understanding of LeTID in SSSC with a passivated emitter rear contact (PERC) structure, we fabricated four group samples with boron and oxygen factors and evaluated the solar cell characteristics, such as the cell efficiency, $V_{oc}$, $I_{sc}$, fill factor (FF), LID, and LeTID. The trends of LID of the four group samples were similar to the trend of LeTID as a function of boron and oxygen.

  • One of the possible mechanisms of multi-drug resistance found in cancer cells is the over-expression of P-glycoprotein (P-gp). Studies have shown that compounds found in plants including vegetables and fruits not only have anticancer activities but may also modulate P-gp activity. The effect of flavonoids and organic isothiocyanates on P-gp activity was studied in human uterine sarcoma cell lines, MES-Sa (sensitive) and MES-SD/DX5 (resistant). The accumulation of daunomycin (DNM), a P-gp substrate, was approximately 10 times greater in the sensitive cell as compared to the resistant cells over the entire time course (up to 2 hrs).
